Rapid Evolution of BRCA1 and BRCA2 in Humans and Other Primates

The maintenance of chromosomal integrity is an essential task of every living organism and cellular repair mechanisms exist to guard against insults to DNA. Given the importance of this process, it is expected that DNA repair proteins would be evolutionarily conserved, exhibiting very minimal sequence change over time. However, BRCA1, an essential gene involved in DNA repair, has been reported to be evolving rapidly despite the fact that many protein-altering mutations within this gene convey a significantly elevated risk for breast and ovarian cancers. Results: To obtain a deeper understanding of the evolutionary trajectory of BRCA1, we analyzed complete BRCA1 gene sequences from 23 primate species. We show that specific amino acid sites have experienced repeated selection for amino acid replacement over primate evolution. This selection has been focused specifically on humans and our closest living relatives, chimpanzees (Pan troglodytes) and bonobos (Pan paniscus). After examining BRCA1 polymorphisms in 7 bonobo, 44 chimpanzee, and 44 rhesus macaque (Macaca mulatta) individuals, we find considerable variation within each of these species and evidence for recent selection in chimpanzee populations. Finally, we also sequenced and analyzed BRCA2 from 24 primate species and find that this gene has also evolved under positive selection. Conclusions: While mutations leading to truncated forms of BRCA1 are clearly linked to cancer phenotypes in humans, there is also an underlying selective pressure in favor of amino acid-altering substitutions in this gene. A hypothesis where viruses are the drivers of this natural selection is discussed.National Institutes of Health R01-GM-093086, 8U42OD011197-13National Science Foundation BCS-07115972Burroughs Wellcome FundMolecular Bioscience

The proceedings are presented of a conference, jointly sponsored by the National Aeronautics and Space Administration, the U.S. Environmental Protection Agency, and the University of Maryland. The purpose of the Conference was to assemble representatives of federal and state government agencies engaged in research on the condition and evolution of the Chesapeake Bay to compose a status report, to present current activities and future plans, and to recommend a long-range future course of policies and programs

Impact of Dietary Fat Source on Beef Display Life

    Get PDF
This study was conducted to evaluate the effects of dietary fat source with modified distillers grains plus solubles (MDGS) on beef display life. Steers were fed either a corn control, full-fat MDGS, de-oiled MDGS, or de-oiled MDGS plus corn oil diet. Strip loins were aged for 2, 9, 16 and 23 days and placed under retail conditions for 7 days. Results suggest that feeding MDGS to cattle increases polyunsaturated fatty acid content of beef and has the potential to reduce beef color and lipid stability in comparison to corn diets. These data indicate that feeding MDGS to cattle may decrease beef display life. Addition of corn oil to de-oiled MDGS decreased redness and increased discoloration and lipid oxidation in comparison to corn control diets

CS as a Graduation Requirement: Catalyst for Systemic Change

    Get PDF
Since President Obama\u27s announcement of the Computer Science for All Initiative in 2016, there has been a surge in the number of districts that are planning for or newly implementing computer science (CS) offerings at their schools. Chicago Public Schools (CPS) is the first large school district to have adopted Computer Science as a high school graduation requirement, taking this significant step along the path towards systemic change. The foundation was laid eight years ago when an informal alliance was formed between a CPS high school CS teacher, a CPS administrator, and three university computer scientists

The Relationship between Marbling, Superoxide Dismutase, and Beef Tenderness

    Get PDF
This study was conducted to evaluate the relationships between animal oxidative status (as indicated by superoxide dismutase [SOD] activity) to marbling and beef tenderness. Prime and Select-grade strip loins were selected and aged for 2, 7, 14, 21, and 28 days for Warner Bratzler shear force, Troponin-T, and SOD activity. Results showed that meat exhibiting higher levels of marbling had lower shear force values and thus were more tender. Low-marbled samples tended to have a greater tenderness response to aging. The effect of oxidative stress, however, was not evident in this study as SOD values were similar. Although the effects of oxidative stress on beef tenderness are still unclear, results from this study provide a conceptual foundation for a new research perspective on meat tenderness

Searches for neutrinoless double beta decay

Neutrinoless double beta decay is a lepton number violating process whose observation would also establish that neutrinos are their own anti-particles. There are many experimental efforts with a variety of techniques. Some (EXO, Kamland-Zen, GERDA phase I and CANDLES) started take data in 2011 and EXO has reported the first measurement of the half life for the double beta decay with two neutrinos of 136^{136}Xe. The sensitivities of the different proposals are reviewed.Comment: 8 pages, prepared for TAUP 201

Experimental constraints on a dark matter origin for the DAMA annual modulation effect

A claim for evidence of dark matter interactions in the DAMA experiment has been recently reinforced. We employ a new type of germanium detector to conclusively rule out a standard isothermal galactic halo of Weakly Interacting Massive Particles (WIMPs) as the explanation for the annual modulation effect leading to the claim. Bounds are similarly imposed on a suggestion that dark pseudoscalars mightlead to the effect. We describe the sensitivity to light dark matter particles achievable with our device, in particular to Next-to-Minimal Supersymmetric Model candidates.Comment: v4: introduces recent results from arXiv:0807.3279 and arXiv:0807.2926.
