Group B streptococcal infection and activation of human astrocytes.
BACKGROUND:Streptococcus agalactiae (Group B Streptococcus, GBS) is the leading cause of life-threatening meningitis in human newborns in industrialized countries. Meningitis results from neonatal infection that occurs when GBS leaves the bloodstream (bacteremia), crosses the blood-brain barrier (BBB), and enters the central nervous system (CNS), where the bacteria contact the meninges. Although GBS is known to invade the BBB, subsequent interaction with astrocytes that physically associate with brain endothelium has not been well studied. METHODOLOGY/PRINCIPAL FINDINGS:We hypothesize that human astrocytes play a unique role in GBS infection and contribute to the development of meningitis. To address this, we used a well- characterized human fetal astrocyte cell line, SVG-A, and examined GBS infection in vitro. We observed that all GBS strains of representative clinically dominant serotypes (Ia, Ib, III, and V) were able to adhere to and invade astrocytes. Cellular invasion was dependent on host actin cytoskeleton rearrangements, and was specific to GBS as Streptococcus gordonii failed to enter astrocytes. Analysis of isogenic mutant GBS strains deficient in various cell surface organelles showed that anchored LTA, serine-rich repeat protein (Srr1) and fibronectin binding (SfbA) proteins all contribute to host cell internalization. Wild-type GBS also displayed an ability to persist and survive within an intracellular compartment for at least 12 h following invasion. Moreover, GBS infection resulted in increased astrocyte transcription of interleukin (IL)-1β, IL-6 and VEGF. CONCLUSIONS/SIGNIFICANCE:This study has further characterized the interaction of GBS with human astrocytes, and has identified the importance of specific virulence factors in these interactions. Understanding the role of astrocytes during GBS infection will provide important information regarding BBB disruption and the development of neonatal meningitis
Increased human pathogenic potential of Escherichia coli from polymicrobial urinary tract infections in comparison to isolates from monomicrobial culture samples
The current diagnostic standard procedure outlined by the Health Protection Agency for urinary tract infections (UTIs) in clinical laboratories does not report bacteria isolated from samples containing three or more different bacterial species. As a result many UTIs go unreported and untreated, particularly in elderly patients, where polymicrobial UTI samples are especially prevalent. This study reports the presence of the major uropathogenic species in mixed culture urine samples from elderly patients, and of resistance to front-line antibiotics, with potentially increased levels of resistance to ciprofloxacin and trimethoprim. Most importantly, the study highlights that Escherichia coli present in polymicrobial UTI samples are statistically more invasive (P<0.001) in in vitro epithelial cell infection assays than those isolated from monomicrobial culture samples. In summary, the results of this study suggest that the current diagnostic standard procedure for polymicrobial UTI samples needs to be reassessed, and that E. coli present in polymicrobial UTI samples may pose an increased risk to human health
Computed tomography measures of nutrition in patients with end-stage liver disease provide a novel approach to characterize deficits
Aim
Patients with cirrhosis and end-stage liver disease (ESLD) develop severe nutrition deficits that impact on morbidity and mortality. Laboratory measures of nutrition fail to fully assess clinical deficits in muscle mass and fat stores. This study employs computed tomography imaging to assess muscle mass and subcutaneous and visceral fat stores in patients with ESLD.
Methods
This 1:1 case-control study design compares ESLD patients with healthy controls. Study patients were selected from a database of ESLD patients using a stratified method to assure a representative sample based on age, body mass index (BMI), gender, and model for end-stage liver disease score (MELD). Control patients were trauma patients with a low injury severity score (<10) who had a CT scan during evaluation. Cases and controls were matched for age +/- 5 years, gender, and BMI +/- 2.
Results
There were 90 subjects and 90 controls. ESLD patients had lower albumin levels (p<0.001), but similar total protein levels (p=0.72). ESLD patients had a deficit in muscle mass (-19%, p<0.001) and visceral fat (-13%, p<0.001), but similar subcutaneous fat (-1%, p=0.35). ESLD patients at highest risk for sarcopenia included those over age 60, BMI< 25.0, and female gender. We found degree of sarcopenia to be independent of MELD score.
Conclusions
These results support previous research demonstrating substantial nutrition deficits in ESLD patients that are not adequately measured by laboratory testing. Patients with ESLD have significant deficits of muscle and visceral fat stores, but a similar amount of subcutaneous fat

Laser Guide Star for Large Segmented-Aperture Space Telescopes, Part I: Implications for Terrestrial Exoplanet Detection and Observatory Stability
Precision wavefront control on future segmented-aperture space telescopes
presents significant challenges, particularly in the context of high-contrast
exoplanet direct imaging. We present a new wavefront control architecture that
translates the ground-based artificial guide star concept to space with a laser
source aboard a second spacecraft, formation flying within the telescope
field-of-view. We describe the motivating problem of mirror segment motion and
develop wavefront sensing requirements as a function of guide star magnitude
and segment motion power spectrum. Several sample cases with different values
for transmitter power, pointing jitter, and wavelength are presented to
illustrate the advantages and challenges of having a non-stellar-magnitude
noise limited wavefront sensor for space telescopes. These notional designs
allow increased control authority, potentially relaxing spacecraft stability
requirements by two orders of magnitude, and increasing terrestrial exoplanet
discovery space by allowing high-contrast observations of stars of arbitrary

Differential negative reinforcement of other behavior to increase compliance with wearing an anti-strip suit
Using a changing-criterion design, we replicated and extended a study (Cook, Rapp, & Schulze,
2015) on differential negative reinforcement of other behavior (DNRO). More specifically,
educational assistants implemented DNRO to teach a 12-year-old boy with autism spectrum
disorder to comply with wearing an anti-strip suit to prevent inappropriate fecal behavior in a
school setting. The duration for which the participant wore the suit systematically increased from
2 s at the start of treatment to the entire duration of the school day at the termination of the study.
Moreover, these effects were generalized to a new school with novel staff and persisted for more
than a year. These findings replicate prior research on DNRO and further support the use of the
intervention to increase compliance with wearing protective items, or medical devices, in
practical settings
Electrostatic potential on human leukocyte antigen: implications for putative mechanism of chronic beryllium disease.
The pathobiology of chronic beryllium disease (CBD) involves the major histocompatibility complex class II human leukocyte antigen (HLA). Although occupational exposure to beryllium is the cause of CBD, molecular epidemiologic studies suggest that specific (Italic)HLA-DPB1(/Italic) alleles may be genetic susceptibility factors. We have studied three-dimensional structural models of HLA-DP proteins encoded by these genes. The extracellular domains of HLA-DPA1*0103/B1*1701, *1901, *0201, and *0401, and HLA-DPA1*0201/B1*1701, *1901, *0201, and *0401 were modeled from the X-ray coordinates of an HLA-DR template. Using these models, the electrostatic potential at the molecular surface of each HLA-DP was calculated and compared. These comparisons identify specific characteristics in the vicinity of the antigen-binding pocket that distinguish the different HLA-DP allotypes. Differences in electrostatics originate from the shape, specific disposition, and variation in the negatively charged groups around the pocket. The more negative the pocket potential, the greater the odds of developing CBD estimated from reported epidemiologic studies. Adverse impact is caused by charged substitutions in positions 55, 56, 69, 84, and 85, namely, the exact same loci identified as genetic markers of CBD susceptibility as well as cobalt-lung hard metal disease. These findings suggest that certain substitutions may promote an involuntary cation-binding site within a putatively metal-free peptide-binding pocket and therefore change the innate specificity of antigen recognition
The effect of match standard and referee experience on the objective and subjective match workload of English Premier League referees
The aim of the present study was to examine the effect of match standard and referee experience upon the objective and subjective workload of referees during Premier and Football league soccer matches. We also examined the relationship between heart rate (HR) and ratings of perceived exertion (RPE) for assessing match intensity in soccer referees. Heart rate responses were recorded using short-range telemetry and RPE scores were collected using a 10-point scale. Analysis revealed a significant relationship between mean match HR and match RPE scores (r = 0.485, p < 0.05, n =18). There were significant differences in match HR (Premier league 83.6 2.6 %HRmax vs. Football league 81.5 2.2 %HRmax, p < 0.05) and match RPE scores (Premier league 7.8 0.8 vs. Football league 6.9 0.8, p < 0.05) between standards of competition. Referee experience had no effect upon match heart rate and RPE responses to Premier and Football league matches. The results of the present study demonstrate the validity of using HR and RPE as a measure of global match intensity in soccer referees. Referee experience had no effect upon the referees’ objective and subjective match workload assessments, whereas match intensity was correlated to competition standard. These findings have implications for fitness preparation and evaluation in soccer referees. When progressing to a higher level of competition, referees should ensure that appropriate levels of fitness are developed in order to enable them to cope with an increase in physical match demands
