Zines are so cool, man! â Thatâs not what Throwrug, or most zines, are about. Jesse Hamiltonâs story points out that Karl & co. donât print Throwrug for us, they print it for themselves. We hope Karl will be happy with the story, but we doubt that will happen. After all, we\u27re spotlighting a figure in the community who would rather remain in shadow. But donât fret, Karl. Persist in place or shift in space? Evaluating the adaptive capacity of species to climate change
Assessing the vulnerability of species to climate change serves as the basis for climate-adaptation planning and climate-smart conservation, and typically involves an evaluation of exposure, sensitivity, and adaptive capacity (AC). AC is a speciesâ ability to cope with or adjust to changing climatic conditions, and is the least understood and most inconsistently applied of these three factors. We propose an attribute-based framework for evaluating the AC of species, identifying two general classes of adaptive responses: âpersist in placeâ and âshift in spaceâ. Persist-in-place attributes enable species to survive in situ, whereas the shift-in-space response emphasizes attributes that facilitate tracking of suitable bioclimatic conditions. We provide guidance for assessing AC attributes and demonstrate the framework's application for species with disparate life histories. Results illustrate the broad utility of this generalized framework for informing adaptation planning and guiding species conservation in a rapidly changing climate.publishedVersio
SGLT2 inhibition promotes glomerular repopulation by cells of renin lineage in experimental kidney disease
Aim: Sodium glucose co-transporter-2 (SGLT2) inhibitors stimulate renal excretion of sodium and glucose and exert renal protective effects in patients with (non-)diabetic chronic kidney disease (CKD) and may as well protect against acute kidney injury (AKI). The mechanism behind this kidney protective effect remains unclear. Juxtaglomerular cells of renin lineage (CoRL) have been demonstrated to function as progenitors for multiple adult glomerular cell types in kidney disease. This study assesses the impact of SGLT2 inhibition on the repopulation of glomerular cells by CoRL and examines their phenotypic commitment. Methods: Experiments were performed in Ren1cre-tdTomato lineage-trace mice. Either 5/6 nephrectomy (5/6NX) modeling CKD or bilateral ischaemia reperfusion injury (bIRI) mimicking AKI was applied, while the SGLT2 inhibitor empagliflozin (10 mg/kg) was administered daily via oral gavage for 14 days. Results: Both 5/6NX and bIRI-induced kidney injury increased the number of glomerular CoRL-derived cells. SGLT2 inhibition improved kidney function after 5/6NX, indicated by decreased blood creatinine and urea levels, but not after bIRI. In line with this, empagliflozin in 5/6NX animals resulted in less glomerulosclerosis, while it did not affect histopathological features in bIRI. Treatment with empagliflozin resulted in an increase in the number of CoRL-derived glomerular cells in both 5/6NX and bIRI conditions. Interestingly, SGLT2 inhibition led to more CoRL-derived podocytes in 5/6NX animals, whereas empagliflozin-treated bIRI mice presented with increased levels of parietal epithelial and mesangial cells derived from CoRL. Conclusion: We conclude that SGLT2 inhibition by empagliflozin promotes CoRL-mediated glomerular repopulation with selective CoRL-derived cell types depending on the type of experimental kidney injury. These findings suggest a previously unidentified mechanism that could contribute to the renoprotective effect of SGLT2 inhibitors.</p

Randomized Trial of Time-Limited Interruptions of Protease Inhibitor-Based Antiretroviral Therapy (ART) vs. Continuous Therapy for HIV-1 Infection
Background
The clinical outcomes of short interruptions of PI-based ART regimens remains undefined. Methods
A 2-arm non-inferiority trial was conducted on 53 HIV-1 infected South African participants with viral load/ml and CD4 T cell count \u3e450 cells/”l on stavudine (or zidovudine), lamivudine and lopinavir/ritonavir. Subjects were randomized to a) sequential 2, 4 and 8-week ART interruptions or b) continuous ART (cART). Primary analysis was based on the proportion of CD4 count \u3e350 cells(c)/ml over 72 weeks. Adherence, HIV-1 drug resistance, and CD4 count rise over time were analyzed as secondary endpoints. Results
The proportions of CD4 counts \u3e350 cells/”l were 82.12% for the intermittent arm and 93.73 for the cART arm; the difference of 11.95% was above the defined 10% threshold for non-inferiority (upper limit of 97.5% CI, 24.1%; 2-sided CI: â0.16, 23.1). No clinically significant differences in opportunistic infections, adverse events, adherence or viral resistance were noted; after randomization, long-term CD4 rise was observed only in the cART arm. Conclusion
We are unable to conclude that short PI-based ART interruptions are non-inferior to cART in retention of immune reconstitution; however, short interruptions did not lead to a greater rate of resistance mutations or adverse events than cART suggesting that this regimen may be more forgiving than NNRTIs if interruptions in therapy occur
Association between HIV replication and serum leptin levels: an observational study of a cohort of HIV-1-infected South African women
<p>Abstract</p> <p>Background</p> <p>Advanced HIV infection can result in lipoatrophy and wasting, even in the absence of ongoing opportunistic infections, suggesting that HIV may directly affect adipose tissue amount and distribution.</p> <p>Methods</p> <p>We assessed the relationship of fat (measured using anthropometry, DEXA, MRI scans) or markers related to glucose and lipid metabolism with viral load in a cross-sectional sample of 83 antiretroviral-naĂŻve HIV-1-infected South African women. A multivariable linear model was fitted to log<sub>10</sub>VL to assess the combined effect of these variables.</p> <p>Results</p> <p>In addition to higher T cell activation, women with viral load greater than the population median had lower waist circumference, body mass index and subcutaneous abdominal fat, as well as lower serum leptin. We demonstrate that leptin serum levels are inversely associated with viral replication, independent of the amount of adipose tissue. This association is maintained after adjusting for multiple variables associated with disease progression (i.e., cellular activation and innate immunity effector levels).</p> <p>Conclusions</p> <p>Our results demonstrate that serum leptin levels are inversely associated with viral replication, independent of disease progression: we postulate that leptin may affect viral replication.</p
Americansâ Attitudes toward the Affordable Care Act: What Role Do Beliefs Play?
How do people form their attitudes toward complex policy issues? Although there has long been an assumption that people consider the various components of those issues and come to an overall assessment, a growing body of recent work has instead suggested that people may reach summary judgments as a function of heuristic cues and goal-oriented rationalizations. This study examines how well a component-based model fits Americansâ evaluations of the Patient Protection and Affordable Care Act of 2010, an important and highly contentious piece of legislation that contained several constituent parts. Despite strong partisan disagreement about the law, we find that Democrats and Republicans both appear to evaluate the law as a function of their beliefs and what the law would do as well as their confidence in those beliefs. This finding implies that correcting misperceptions and increasing awareness of the components of legislation have the potential to change attitudes
