    Three Composite Cross Populations (CCPs) of winter wheat were created in 2001, one from crossing high baking quality parents (called QCCP), one based on high yielding parents (YCCP), and one created from both parent sets combined (YQ). All three CCPs were grown for several years in the UK and in Germany under conventional and organic cropping management. Based on properties of the different parent sets, yields were predicted to be highest in the YCCP and lowest in the QCCP, with the YQCCP in between. However, data showed that the YCCP was not significantly higher-yielding than the more generalist YQCCP in five out of six sites; in fact, the YQCCP even tended to outyield the YCCP on average over all sites, and this effect was significant at the German conventional site. Our results suggest that high diversity present in generalist populations of wheat may convey yield advantages

    Structure and play: rethinking regulation in the higher education sector

    This paper explores possible tactics for academics working within a context of increasing regulation and constraint. One suggested tactic is to move outside of a creativity-conformity binary. Rather than understanding creativity and conformity as separable, where one is seen as excluding the other, the authors consider the potential of examining the relationships between them. The theme of 'structure and play' illustrates the argument. In the first part of the paper, using various examples from art and design - fields generally associated with creativity - the authors explore the interrelatedness of creativity and conformity. For example, how might design styles, which are generally understood as creative outcomes, constrain creativity and lead to conformity within the design field? Is fashion producing creativity or conformity? Conversely, the ways in which conformity provides the conditions for creativity are also examined. For example, the conformity imposed by the state on artists in the former communist bloc contributed to a thriving underground arts movement which challenged conformity and state regulation. Continuing the theme of 'structure and play', the authors recount a story from an Australian university which foregrounds the ongoing renegotiation of power relations in the academy. This account illustrates how programmatic government in a university, with its aim of regulating conduct, can contribute to unanticipated outcomes. The authors propose that a Foucauldian view of distributed power is useful for academics operating in a context of increasing regulation, as it brings into view sites where power might begin to be renegotiated

    Multilingual gendered identities: female undergraduate students in London talk about heritage languages

    In this paper I explore how a group of female university students, mostly British Asian and in their late teens and early twenties, perform femininities in talk about heritage languages. I argue that analysis of this talk reveals ways in which the participants enact ‘culturally intelligible’ gendered subject positions. This frequently involves negotiating the norms of ‘heteronormativity’, constituting femininity in terms of marriage, motherhood and maintenance of heritage culture and language, and ‘girl power’, constituting femininity in terms of youth, sassiness, glamour and individualism. For these young women, I ask whether higher education can become a site in which they have the opportunities to explore these identifications and examine other ways of imagining the self and what their stories suggest about ‘doing being’ a young British Asian woman in London

    Filtration artefacts in bacterial community composition can affect the outcome of dissolved organic matter biolability assays

    Inland waters are large contributors to global carbon dioxide (CO2) emissions, in part due to the vulnerability of dissolved organic matter (DOM) to microbial decomposition and respiration to CO2 during transport through aquatic systems. To assess the degree of this vulnerability, aquatic DOM is often incubated in standardized biolability assays. These assays isolate the dissolved fraction of aquatic OM by size filtration prior to incubation. We test whether this size selection has an impact on the bacterial community composition and the consequent dynamics of DOM degradation using three different filtration strategies: 0.2&thinsp;µm (filtered and inoculated), 0.7&thinsp;µm (generally the most common DOM filter size) and 106&thinsp;µm (unfiltered). We found that bacterial community composition, based on 16S rRNA amplicon sequencing, was significantly affected by the different filter sizes. At the same time, the filtration strategy also affected the DOM degradation dynamics, including the δ13C signature. However, the dynamics of these two responses were decoupled, suggesting that filtration primarily influences biolability assays through bacterial abundance and the presence of their associated predators. By the end of the 41-day incubations all treatments tended to converge on a common total DOM biolability level, with the 0.7&thinsp;µm filtered incubations reaching this point the quickest. These results suggest that assays used to assess the total biolability of aquatic DOM should last long enough to remove filtration artefacts in the microbial population. Filtration strategy should also be taken into account when comparing results across biolability assays.</p

    The fate of the homoctenids (Tentaculitoidea) during the Frasnian-Famennian mass extinction (Late Devonian)

    The homoctenids (Tentaculitoidea) are small, conical-shelled marine animals which are amongst the most abundant and widespread of all Late Devonian fossils. They were a principal casualty of the Frasnian-Famennian (F-F, Late Devonian) mass extinction, and thus provide an insight into the extinction dynamics. Despite their abundance during the Late Devonian, they have been largely neglected by extinction studies. A number of Frasnian-Famennian boundary sections have been studied, in Poland, Germany, France, and the United States. These sections have yielded homoctenids, which allow precise recognition of the timing of the mass extinction. It is clear that the homoctenids almost disappear from the fossil record during the latest Frasnian “Upper Kellwasser Event”. The coincident extinction of this pelagic group, and the widespread development of intense marine anoxia within the water column, provides a causal link between anoxia and the F-F extinction. Most notable is the sudden demise of a group, which had been present in rock-forming densities, during this anoxic event. One new species, belonging to Homoctenus is described, but is not formally named here

    Daily evaluation of 26 precipitation datasets using Stage-IV gauge-radar data for the CONUS

    New precipitation (P) datasets are released regularly, following innovations in weather forecasting models, satellite retrieval methods, and multi-source merging techniques. Using the conterminous US as a case study, we evaluated the performance of 26 gridded (sub-)daily P datasets to obtain insight into the merit of these innovations. The evaluation was performed at a daily timescale for the period 2008–2017 using the Kling–Gupta efficiency (KGE), a performance metric combining correlation, bias, and variability. As a reference, we used the high-resolution (4&thinsp;km) Stage-IV gauge-radar P dataset. Among the three KGE components, the P datasets performed worst overall in terms of correlation (related to event identification). In terms of improving KGE scores for these datasets, improved P totals (affecting the bias score) and improved distribution of P intensity (affecting the variability score) are of secondary importance. Among the 11 gauge-corrected P datasets, the best overall performance was obtained by MSWEP V2.2, underscoring the importance of applying daily gauge corrections and accounting for gauge reporting times. Several uncorrected P datasets outperformed gauge-corrected ones. Among the 15 uncorrected P datasets, the best performance was obtained by the ERA5-HRES fourth-generation reanalysis, reflecting the significant advances in earth system modeling during the last decade. The (re)analyses generally performed better in winter than in summer, while the opposite was the case for the satellite-based datasets. IMERGHH V05 performed substantially better than TMPA-3B42RT V7, attributable to the many improvements implemented in the IMERG satellite P retrieval algorithm. IMERGHH V05 outperformed ERA5-HRES in regions dominated by convective storms, while the opposite was observed in regions of complex terrain. The ERA5-EDA ensemble average exhibited higher correlations than the ERA5-HRES deterministic run, highlighting the value of ensemble modeling. The WRF regional convection-permitting climate model showed considerably more accurate P totals over the mountainous west and performed best among the uncorrected datasets in terms of variability, suggesting there is merit in using high-resolution models to obtain climatological P statistics. Our findings provide some guidance to choose the most suitable P dataset for a particular application.</p

    CD74-downregulation of placental macrophage-trophoblastic interactions in preeclampsia

    Rationale: MWe hypothesized that Cluster of differentiation 74 (CD74) downregulation on placental macrophages, leading to altered macrophage-trophoblast interaction, is involved in preeclampsia. Objective: Preeclamptic pregnancies feature hypertension, proteinuria and placental anomalies. Feto-placental macrophages regulate villous trophoblast differentiation during placental development. Disturbance of this well-balanced regulation can lead to pathological pregnancies. Methods and Results: We performed whole genome expression analysis of placental tissue. CD74 was one of the most downregulated genes in placentas from preeclamptic women. By RT-PCR, we confirmed this finding in early onset (<34 gestational week, n=26) and late onset (≥34 gestational week, n=24) samples from preeclamptic women, compared to healthy pregnant controls (n=28). CD74 protein levels were analyzed by Western blot and flow cytometry. We identified placental macrophages to express CD74 by immunofluorescence, flow cytometry and RT-PCR. CD74-positive macrophages were significantly reduced in preeclamptic placentas compared to controls. CD74-silenced macrophages showed that the adhesion molecules ALCAM, ICAM4, and Syndecan-2, as well as macrophage adhesion to trophoblasts were diminished. Naïve and activated macrophages lacking CD74 showed a shift towards a pro-inflammatory signature with an increased secretion of TNF , CCL5, and MCP-1, when co-cultured with trophoblasts compared to control macrophages. Trophoblasts stimulated by these factors express more CYP2J2, sFlt1, TNF and IL-8. CD74-knockout mice showed disturbed placental morphology, reduced junctional zone, smaller placentas and impaired spiral artery remodeling with fetal growth restriction. Conclusions: CD74 downregulation in placental macrophages is present in preeclampsia. CD74 downregulation leads to altered macrophage activation towards a pro-inflammatory signature and a disturbed crosstalk with trophoblasts

    TEAD and YAP regulate the enhancer network of human embryonic pancreatic progenitors.

    The genomic regulatory programmes that underlie human organogenesis are poorly understood. Pancreas development, in particular, has pivotal implications for pancreatic regeneration, cancer and diabetes. We have now characterized the regulatory landscape of embryonic multipotent progenitor cells that give rise to all pancreatic epithelial lineages. Using human embryonic pancreas and embryonic-stem-cell-derived progenitors we identify stage-specific transcripts and associated enhancers, many of which are co-occupied by transcription factors that are essential for pancreas development. We further show that TEAD1, a Hippo signalling effector, is an integral component of the transcription factor combinatorial code of pancreatic progenitor enhancers. TEAD and its coactivator YAP activate key pancreatic signalling mediators and transcription factors, and regulate the expansion of pancreatic progenitors. This work therefore uncovers a central role for TEAD and YAP as signal-responsive regulators of multipotent pancreatic progenitors, and provides a resource for the study of embryonic development of the human pancreas
