Pennsylvanian Stratigraphy of Douglas County, Kansas: Glacioeustatically Modulated Cyclic Deposition on a Remarkably Smooth Shelf
The Middle and Late Pennsylvanian succession of Kansas consists of two types of cyclothems, lithologic successions that consist of several members that repeat in a particular order. Cyclothems are a few meters to a few tens of meters thick. One type of cyclothem, the Kansas cyclothem, or here the limestone-rich cyclothem, consists of interbeds of limestone and shale, where the amounts of limestone are approximately equal to the amount of shale or dominate the succession. This trip will study one such cyclothem in the Oread Limestone at Stop 3 and possibly another cyclothem at the optional Stop 4. Siliciclastic cyclothems are dominated by shale and may contain an incised valley filled partly by fluvial or estuarine sandstone. The Lawrence Shale, which we will see at Stops 2 and 3, contains such features.
While many cyclothems are incomplete, both types of cyclothem may contain dark gray to black, fissile, phosphatic core shale beds, commonly a meter or less thick, and an underlying transgressive deposit beneath. Overlying deposits represent regression or progradation of the shoreline: either lime mudstone grading to grainstone or clay-rich marine shale grading to coarse siltstone or fine sandstone. Paleosols and fossils of marine invertebrates are common and indicate that deposition took place in both marine and non-marine conditions. Some marine deposits show a paleosol overprint.
The deposits indicate that the area was a shelf during the Late Pennsylvanian where relief was very low, and very laterally extensive deposits formed as sea level rose and fell. The alternation of shale-rich and limestone-rich intervals plus the feet-per-mile dip to the WNW create the characteristic topographic expression of cuestas with gently sloping caps that are underlain by limestone. While glacial deposits are mostly restricted to northern and east-central Douglas County, Pleistocene glaciers influenced the erosional and drainage patterns of the area
Alteration of hyaloclastites in the HSDP 2 Phase 1 Drill Core 1. Description and paragenesis

All rights reserved.The core from the Hawaii Scientific Drilling Project 2 Phase 1 provides a unique opportunity for studying the low-temperature alteration processes affecting basalt in suboceanic-island environments. In hyaloclastites, which make up about one half of the lower 2 km of this core (the portion that accumulated below sea level), these processes have resulted in zones of incipient, smectitic, and palagonitic alteration. The alteration of sideromelane in these hyaloclastites has four distinct outcomes: dissolution, replacement by two different textural varieties of smectite (i.e., reddened and green grain-replacive), and conversion to palagonite. All samples show evidence of the incipient stage of alteration, suggesting that every sample passed through that zone. However, most samples that show palagonitic alteration do not also show evidence of smectitic alteration and vice versa, suggesting these two outcomes represent divergent paths of alteration. Incipient alteration (1080 to 1335 m depth) includes fracturing and mechanical reduction of porosity from 40–45% to about 20–30%; growth of one form of pore-lining smectite; dissolution of sideromelane; and formation of sideromelane-grain replacements consisting of Fe-hydroxide-strained smectite, titaniferous nodules, and tubules. DNA-specific stains and morphological features indicate that tubules are the result of microbial activity. Smectitic alteration (1405 to 1573 m) includes growth of a second variety of pore-lining smectite, pore-filling and grain-replacing smectite, and cements of phillipsite and Ca-silicate minerals. Palagonitic alteration (1573 m to the deepest samples) includes replacement of margins of shards with palagonite and growth of pore-filling chabazite. The porosity is reduced by cementation to less than 4% at 1573 m. Porosity does not decrease further down hole, nor does the thickness of palagonite rims on shards increase through the zone of palagonitic alteration. In these samples, palagonite is not an intermediate alteration product in the development of smectite. Rather, in hyaloclastites from the HSDP core, palagonite has formed after all observed smectites. Current downhole temperatures at the boundaries between the three alteration zones are in the range from 12° to 15°C, suggesting that geochemical thresholds or vital effects, not temperature conditions, control different outcomes of alteration

The article draws on bell hooks’ (1992) concept of ‘eating the other’ and the analysis undertakes an interdisciplinary approach that combines visual analysis with Anibal Quijano’s (2007) concept of modernity/coloniality. The discussion explores the trends of global multiculturalism that have been adopted by Sandals in a hybridized cut and mix approach to selling a packaged ideal of the Caribbean. The visual techniques devised to create a culinary holiday package are overlaid onto a manufactured and homogenised or McDonaldized (Ritzer and Liska 1997) Caribbean that provides insight into the way in which global neoliberal multiculturalism is framed by ongoing colonial relations after formal colonial rule has ended in the Caribbean region
Value of percutaneous transluminal coronary angioplasty after unsuccessful intravenous streptokinase therapy in acute myocardial infarction
The effect of sequential high-dose intravenous streptokinase (SK) (1.5 million units) followed by emergency percutaneous transluminal coronary angioplasty (PTCA) on preserving left ventricular function was assessed prospectively in 34 patients with acute myocardial infarction (AMI). Intravenous SK therapy was initiated 2.6 +/- 1.3 hours (mean +/- standard deviation) after the onset of chest pain. Urgent coronary angiography showed persistent total occlusion in 13 patients, significant diameter stenosis (70 to 99%) in 18 patients and a widely patent artery (less than 50% stenosis) in 3 patients. Emergency PTCA was performed in 29 patients 5.0 +/- 2.1 hours after symptom onset. Successful recanalization was achieved in 33 of the 34 patients (97%) treated with sequential therapy. Repeat contrast ventriculograms recorded 7 to 10 days after intervention in 23 patients showed that the left ventricular ejection fraction increased from 53 +/- 12% to 59 +/- 13% (area-length method, p < 0.002). Regional wall motion of the infarcted segments improved from - 2.7 +/- 1.1 to - 1.5 +/- 1.7 SD/chord (centerline method, p < 0.003). In the subgroup of patients with an occluded artery on initial angiography (group A, N = 10), both global left ventricular ejection fraction (49 +/- 12% vs 59 +/- 12%, p < 0.002) and regional wall motion (-3.2 +/- 1.0 vs -1.9 +/- 1.7 SD/chord, p < 0.002) improved significantly. In contrast, no significant improvement was seen in patients with a patent artery on initial angiography (n = 13). Thus, sequential intravenous SK and emergency PTCA is efficacious in achieving coronary reperfusion and in improving both global and regional left ventricular function. When thrombolytic therapy fails, successful recanalization can be achieved by emergency PTCA, resulting in significant myocardial salvage.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/26031/1/0000104.pd
Early reperfusion therapy improves left ventricular function after acute inferior myocardial infarction associated with right coronary artery disease
Quantitative global and regional ventriculographic analysis was performed acutely and 1 week later in 46 patients undergoing reperfusion procedures within 6 hours of acute inferior myocardial infarction due to right coronary artery disease. While serial improvement in global left ventricular ejection fraction was not demonstrated for the group, infarct zone regional wall motion did improve (-2.7 +/- 0.9 vs -2.3 +/- 1.4 SD/chord, p p p p p < 0.0001). We conclude that significant improvement in global and reglonal left ventricular function in patients with inferior myocardial infarction is possible when reperfusion therapy is begun early or when arterial patency is achieved.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/26625/1/0000166.pd
Fluorescence Lifetime Imaging of Quantum Dot Labeled DNA Microarrays
Quantum dot (QD) labeling combined with fluorescence lifetime imaging microscopy is proposed as a powerful transduction technique for the detection of DNA hybridization events. Fluorescence lifetime analysis of DNA microarray spots of hybridized QD labeled target indicated a characteristic lifetime value of 18.8 ns, compared to 13.3 ns obtained for spots of free QD solution, revealing that QD labels are sensitive to the spot microenvironment. Additionally, time gated detection was shown to improve the microarray image contrast ratio by 1.8, achieving femtomolar target sensitivity. Finally, lifetime multiplexing based on Qdot525 and Alexa430 was demonstrated using a single excitation-detection readout channel
Fast DNA and protein microarray tests for the diagnosis of hepatitis C virus infection on a single platform
Hepatitis C virus (HCV) is a major cause of chronic liver disease and liver cancer, and remains a large health care burden to the world. In this study we developed a DNA microarray test to detect HCV RNA and a protein microarray to detect human anti-HCV antibodies on a single platform. A main focus of this study was to evaluate possibilities to reduce the assay time, as a short time-to-result (TTR) is a prerequisite for a point-of-care test. Significantly reducing hybridisation and washing times did not impair the assay performance. This was confirmed first using artificial targets and subsequently using clinical samples from an HCV seroconversion panel derived from a HCV-infected patient. We were able to reduce the time required for the detection of human anti-HCV antibodies to only 14 min, achieving nanomolar sensitivity. The protein microarray exhibited an analytical sensitivity comparable to that of commercial systems. Similar results were obtained with the DNA microarray using a universal probe which covered all different HCV genotypes. It was possible to reduce the assay time after PCR from 150 min to 16 min without any loss of sensitivity. Taken together, these results constitute a significant step forward in the design of rapid, microarray-based diagnostics for human infectious disease, and show that the protein microarray is currently the most favourable candidate to fill this role
Peptide-tags for enhanced DNA microarray performance
DNA microarrays are powerful tools for gene expression analysis and genotyping studies in research and diagnostic applications. A high sensitivity and short time-to-result are prerequisites for their practical application in the clinic. The hybridization efficiency of DNA microarrays depends on the probe density and the probe orientation and thus their accessibility for target molecules. In order to find an optimal probe immobilization procedure a set of different oligonucleotide modifications was tested on epoxy silane functionalized glass slides. It was found that histidine-tagged oligonucleotides resulted in the highest amount of bound probe and by far the best hybridization efficiencies. The detection limit obtained with histidine-tagged probes was up to two orders of magnitude lower compared to commonly used probe modifications. In order to further investigate the binding mechanism of histidine-tags towards functionalized glass substrates a set of different peptide-tags with and without free terminal amino-groups and with different amino acid compositions was tested. The results indicate an impact of the terminal amino group on the covalent surface binding and of aromatic amino acid residues on the enhanced hybridisation efficiency
Natural history of small index lesions suspicious for prostate cancer on multiparametric MRI: recommendations for interval imaging follow-up
PURPOSEWe aimed to determine the natural history of small index lesions identified on multiparametric-magnetic resonance imaging (MP-MRI) of the prostate by evaluating lesion-specific pathology and growth on serial MP-MRI.MATERIALS AND METHODSWe performed a retrospective review of 153 patients who underwent a minimum of two MP-MRI sessions, on an institutional review board-approved protocol. Index lesion is defined as the lesion(s) with the highest cancer suspicion score based on initial MP-MRI of a patient, irrespective of size. Two study cohorts were identified: (1) patients with no index lesion or index lesion(s) ≤7 mm and (2) a subset with no index lesion or index lesion(s) ≤5 mm. Pathological analysis of the index lesions was performed following magnetic resonance/ultrasound fusion-guided biopsy. Growth rate of the lesions was calculated based on MP-MRI follow-up.RESULTSPatients with small index lesions measuring ≤7 mm (n=42) or a subset with lesions ≤5 mm (n=20) demonstrated either benign findings (86.2% and 87.5%, respectively) or low grade Gleason 6 prostate cancer (13.8% and 12.5%, respectively) on lesion-specific targeted biopsies. These lesions demonstrated no significant change in size (P = 0.93 and P = 0.36) over a mean imaging period of 2.31±1.56 years and 2.40±1.77 years for ≤7 mm and ≤5 mm index lesion thresholds, respectively. These findings held true on subset analyses of patients who had a minimum of two-year interval follow-up with MP-MRI.CONCLUSIONSmall index lesions of the prostate are pathologically benign lesions or occasionally low-grade cancers. Slow growth rate of these small index lesions on serial MP-MRI suggests a surveillance interval of at least two years without significant change
