    Identification of an ironā€“hepcidin complex

    Following its identification as a liver-expressed antimicrobial peptide, the hepcidin peptide was later shown to be a key player in iron homoeostasis. It is now proposed to be the 'iron hormone' which, by interacting with the iron transporter ferroportin, prevents further iron import into the circulatory system. This conclusion was reached using the corresponding synthetic peptide, emphasizing the functional importance of the mature 25-mer peptide, but omitting the possible functionality of its maturation. From urine-purified native hepcidin, we recently demonstrated that a proportion of the purified hepcidin had formed iron-hepcidin complexes. This interaction was investigated further by computer modelling and, based on the sequence similarity of hepcidin with metallothionein, a three-dimensional model of hepcidin, containing one atom of iron, was constructed. To characterize these complexes further, the interaction with iron was analysed using different spectroscopic methods. Monoferric hepcidin was identified by MS, as were possibly other complexes containing two and three atoms of iron respectively, although these were present only in minor amounts. UV/visible absorbance and CD studies identified the iron-binding events which were facilitated at a physiological pH. EPR spectroscopy identified the ferric state of the bound metal, and indicated that the iron-hepcidin complex shares some similarities with the rubredoxin iron-sulfur complex, suggesting the presence of Fe(3+) in a tetrahedral sulfur co-ordination. The potential roles of iron binding for hepcidin are discussed, and we propose either a regulatory function in the maturation of pro-hepcidin into active hepcidin or as the necessary link in the interaction between hepcidin and ferroportin

    Urban energy exchanges monitoring from space

    One important challenge facing the urbanization and global environmental change community is to understand the relation between urban form, energy use and carbon emissions. Missing from the current literature are scientific assessments that evaluate the impacts of different urban spatial units on energy fluxes; yet, this type of analysis is needed by urban planners, who recognize that local scale zoning affects energy consumption and local climate. However, satellite-based estimation of urban energy fluxes at neighbourhood scale is still a challenge. Here we show the potential of the current satellite missions to retrieve urban energy budget, supported by meteorological observations and evaluated by direct flux measurements. We found an agreement within 5% between satellite and in-situ derived net all-wave radiation; and identified that wall facet fraction and urban materials type are the most important parameters for estimating heat storage of the urban canopy. The satellite approaches were found to underestimate measured turbulent heat fluxes, with sensible heat flux being most sensitive to surface temperature variation (-64.1, +69.3 W m-2 for Ā±2 K perturbation); and also underestimate anthropogenic heat flux. However, reasonable spatial patterns are obtained for the latter allowing hot-spots to be identified, therefore supporting both urban planning and urban climate modelling

    SummaryObjectivesTo characterize diarrhea and fever as risk factors for anemia among children in developing countries.MethodsWe characterized risk factors for anemia in a sample of 32873 children, aged 6ā€“59 months, from poor families in urban slum areas of Indonesia from 2000 to 2003.ResultsThe prevalence of anemia was 58.7%. In separate multivariate models, after adjusting for age, sex, stunting, maternal age and education, and weekly per capita household expenditure, current diarrhea (OR 1.20, 95% CI 1.07ā€“1.35, p=0.002), current fever (OR 1.44, 95% CI 1.18ā€“1.75, p<0.0001), and a history of diarrhea in the previous seven days (OR 1.12, 95% CI 1.03ā€“1.23, p=0.024) were associated with an increased risk of anemia.ConclusionsDiarrhea and fever are important risk factors for anemia among young children living in urban slum communities in Indonesia

    Field efficacy of a new mosaic long-lasting mosquito net (PermaNetĀ® 3.0) against pyrethroid-resistant malaria vectors: a multi centre study in Western and Central Africa

    <p>Abstract</p> <p>Background</p> <p>Due to the spread of pyrethroid-resistance in malaria vectors in Africa, new strategies and tools are urgently needed to better control malaria transmission. The aim of this study was to evaluate the performances of a new mosaic long-lasting insecticidal net (LLIN), i.e. PermaNet<sup>Ā® </sup>3.0, against wild pyrethroid-resistant <it>Anopheles gambiae s.l</it>. in West and Central Africa.</p> <p>Methods</p> <p>A multi centre experimental hut trial was conducted in Malanville (Benin), VallĆ©e du Kou (Burkina Faso) and Pitoa (Cameroon) to investigate the exophily, blood feeding inhibition and mortality induced by PermaNet<sup>Ā® </sup>3.0 (i.e. a mosaic net containing piperonyl butoxide and deltamethrin on the roof) comparatively to the WHO recommended PermaNet<sup>Ā® </sup>2.0 (unwashed and washed 20-times) and a conventionally deltamethrin-treated net (CTN).</p> <p>Results</p> <p>The personal protection and insecticidal activity of PermaNet 3.0 and PermaNet<sup>Ā® </sup>2.0 were excellent (>80%) in the "pyrethroid-tolerant" area of Malanville. In the pyrethroid-resistance areas of Pitoa (metabolic resistance) and VallĆ©e du Kou (presence of the L1014F <it>kdr </it>mutation), PermaNet<sup>Ā® </sup>3.0 showed equal or better performances than PermaNet<sup>Ā® </sup>2.0. It should be noted however that the deltamethrin content on PermaNet<sup>Ā® </sup>3.0 was up to twice higher than that of PermaNet<sup>Ā® </sup>2.0. Significant reduction of efficacy of both LLIN was noted after 20 washes although PermaNet<sup>Ā® </sup>3.0 still fulfilled the WHO requirement for LLIN.</p> <p>Conclusion</p> <p>The use of combination nets for malaria control offers promising prospects. However, further investigations are needed to demonstrate the benefits of using PermaNet<sup>Ā® </sup>3.0 for the control of pyrethroid resistant mosquito populations in Africa.</p
