    Intrinsic tethering activity of endosomal Rab proteins.

    Rab small G proteins control membrane trafficking events required for many processes including secretion, lipid metabolism, antigen presentation and growth factor signaling. Rabs recruit effectors that mediate diverse functions including vesicle tethering and fusion. However, many mechanistic questions about Rab-regulated vesicle tethering are unresolved. Using chemically defined reaction systems, we discovered that Vps21, a Saccharomyces cerevisiae ortholog of mammalian endosomal Rab5, functions in trans with itself and with at least two other endosomal Rabs to directly mediate GTP-dependent tethering. Vps21-mediated tethering was stringently and reversibly regulated by an upstream activator, Vps9, and an inhibitor, Gyp1, which were sufficient to drive dynamic cycles of tethering and detethering. These experiments reveal a previously undescribed mode of tethering by endocytic Rabs. In our working model, the intrinsic tethering capacity Vps21 operates in concert with conventional effectors and SNAREs to drive efficient docking and fusion

    Compton-thick AGN in the NuSTAR era II: A deep NuSTAR and XMM-Newton view of the candidate Compton thick AGN in NGC 1358

    We present the combined NuSTATR and XMM-Newton 0.6-79 keV spectral analysis of a Seyfert 2 galaxy, NGC 1358, which we selected as a candidate Compton thick (CT-) active galactic nucleus (AGN) on the basis of previous Swift/BAT and Chandra studies. According to our analysis, NGC 1358 is confirmed to be a CT-AGN using physical motivated models, at >3 σ\sigma confidence level. Our best-fit shows that the column density along the 'line-of-sight' of the obscuring material surrounding the accreting super-massive black hole is NH\rm _H = [1.96--2.80] ×\times 1024^{24} cm2^{-2}. The high-quality data from NuSTAR gives the best constraints on the spectral shape above \sim10 keV to date on NGC 1358. Moreover, by combining NuSTAR and XMM-Newton data, we find that the obscuring torus has a low covering factor (fcf_c <0.17), and the obscuring material is distributed in clumps, rather than uniformly. We also derive an estimate of NGC 1358's Eddington ratio, finding it to be λEdd\lambda_{\rm Edd} \sim4.70.3+0.34.7_{-0.3}^{+0.3} ×\times 102^{-2}, which is in acceptable agreement with previous measurements. Finally, we find no evidence of short-term variability, over a \sim100 ks time-span, in terms of both 'line-of-sight' column density and flux.Comment: 12 pages, 6 figure

    CO excitation in the Seyfert galaxy NGC7130

    We present a coherent multi-band modelling of the CO Spectral Energy Distribution of the local Seyfert Galaxy NGC7130 to assess the impact of the AGN activity on the molecular gas. We take advantage of all the available data from X-ray to the sub-mm, including ALMA data. The high-resolution (~0.2") ALMA CO(6-5) data constrain the spatial extension of the CO emission down to ~70 pc scale. From the analysis of the archival CHANDRA and NuSTAR data, we infer the presence of a buried, Compton-thick AGN of moderate luminosity, L_2-10keV ~ 1.6x10^{43} ergs-1. We explore photodissociation and X-ray-dominated regions (PDRs and XDRs) models to reproduce the CO emission. We find that PDRs can reproduce the CO lines up to J~6, however, the higher rotational ladder requires the presence of a separate source of excitation. We consider X-ray heating by the AGN as a source of excitation, and find that it can reproduce the observed CO Spectral Energy Distribution. By adopting a composite PDR+XDR model, we derive molecular cloud properties. Our study clearly indicates the capabilities offered by current-generation of instruments to shed light on the properties of nearby galaxies adopting state-of-the art physical modelling.Comment: 5 pages, 3 figures, accepted for publication in MNRAS Letter

    The structure of the X-ray absorber in Mrk 915 revealed by Swift

    In this paper we present the results obtained with a monitoring programme (23 days long) performed with Swift-XRT on the local Seyfert galaxy Mrk 915. The light-curve analysis shows a significant count rate variation (about a factor of 2-3) on a time-scale of a few days, while the X-ray colours show a change in the spectral curvature below 2 keV and the presence of two main spectral states. From the spectral analysis we find that the observed variations can be explained by the change of the intrinsic nuclear power (about a factor of 1.5) coupled with a change of the properties of an ionized absorber. The quality of the data prevents us from firmly establishing if the spectral variation is due to a change in the ionization state and/or in the covering factor of the absorbing medium. The latter scenario would imply a clumpy structure of the ionized medium. By combining the information provided by the light curve and the spectral analyses, we can derive some constraints on the location of the absorber under the hypotheses of either homogeneous or clumpy medium. In both cases, we find that the absorber should be located inside the outer edge of an extended torus and, in particular, under the clumpy hypothesis, it should be located near, or just outside, to the broad emission line region.Comment: 8 pages, 6 figures, 1 table.     XMM-Newton and NuSTAR joint observations of Mrk 915: a deep look into the X-ray properties

    We report on the X-ray monitoring programme (covering slightly more than 11 days) carried out jointly by XMM-Newton and NuSTAR on the intermediate Seyfert galaxy Mrk 915. The light curves extracted in different energy ranges show a variation in intensity but not a significant change in spectral shape. The X-ray spectra reveal the presence of a two-phase warm absorber: a fully covering mildly ionized structure [log xi/(erg cm/s)~2.3, NH~1.3x10^21 cm-2] and a partial covering (~90 per cent) lower ionized one [log xi/(erg cm/s)~0.6, NH~2x10^22 cm-2]. A reflection component from distant matter is also present. Finally, a high-column density (NH~1.5x10^23 cm-2) distribution of neutral matter covering a small fraction of the central region is observed, almost constant, in all observations. Main driver of the variations observed between the datasets is a decrease in the intrinsic emission by a factor of ~1.5. Slight variations in the partial covering ionized absorber are detected, while the data are consistent with no variation of the total covering absorber. The most likely interpretation of the present data locates this complex absorber closer to the central source than the narrow line region, possibly in the broad line region, in the innermost part of the torus, or in between. The neutral obscurer may either be part of this same stratified structure or associated with the walls of the torus, grazed by (and partially intercepting) the line of sight.Comment: 14 pages, 10 figures, 4 tables.     Sustainable and Resistant Road Infrastructures: The Role of the Envision Framework as a Guide to a New Design Approach

    In Italy, resilience and sustainability concepts are generally applied to road infrastructures (mainly in terms of pavements) in order to minimize the environmental impacts of their construction and maintenance. In this paper the Envision framework has been adopted to evaluate the sustain-ability of a road project, optimizing its resilience for both short-term and long-term impacts. The rating approach has been applied considering not only pavements analysis but also the complete project process. The authors have upgraded the original Envision framework to road infrastructures, in compliance with the Italian regulatory framework. Four scenarios have been considered. The first allowed us to understand which credits can be evaluated with the available project documentation. The second represents a snapshot of the state of the project. The third evaluates the effect of the improvement in the performance level obtained by analyzing the potential increases. Finally, the last considers also for “pending” credits

    Optimization of a perishable inventory system: A simulation study in a Ho.Re.Ca. company

    The main goal of this paper is to describe the optimization of the inventory management process in a real context of perishable food products. The study involves one of the largest Italian HO.RE.CA. companies, located in the north of Italy and operating as a provider of the catering, commercial and welfare services. A simulation model was set up with the purpose of adapting three traditional reordering policies (i.e. Re-Order Point, Re-Order Cycle, and (s,S)) to a set of products belonging to company's assortment and evaluating the resulting economic outcomes. To this end, each policy was modelled on Microsoft ExcelTM, so as to compute the total cost of inventory management and determine of the minimum cost strategy. A comparison with the current company's performance and that achievable with the optimized policy is also proposed
