    Wound healing in Piaractus mesopotamicus supplemented with chromium carbochelate and Saccharomyces cerevisiae

    A cicatrização e reparação de tecidos são mecanismos essenciais para garantir a sobrevivência e saúde de qualquer indivíduo. O objetivo deste trabalho foi avaliar o impacto da suplementação com carboquelato de cromo (CC) e Saccharomyces cerevisiae (SC) sobre a cicatrização no peixe tropical Piaractus mesopotamicus. Para isto, os peixes foram distribuídos em quatro grupos: controle (sem tratamento), suplementados com 18 mg/kg de carboquelato de cromo, 0,3% de S. cerevisiae e associação de ambos os suplementos. Após 105 dias de alimentação, foram realizadas incisões na pele de espessura completa (2,0 x 1,0 x 0,25 cm) removendo epiderme e derme. Avaliações macroscópicas e microscópicas foram realizadas 1, 3, 7, 14, 21, 28 e 35 dias após a indução das feridas, para monitorar a taxa de cicatrização. As bordas opostas das feridas avançaram gradualmente a cada dia, demonstrando o aumento progressivo do processo de cicatrização ao longo do tempo. O processo inflamatório foi exacerbado e expansivo, com aumento no número de células mucosas e cromatóforos. Apesar deste processo, não foram observadas diferenças significativas na retração das feridas e nos parâmetros microscópicos entre os grupos. Peixes suplementados com CC ou Sc apresentaram rápida reepitelização, maior grau de organização de fibras colágenas e de neovascularização inicial. Concluiu-se que a suplementação com S. cerevisiae ou carboquelato de cromo melhora aspectos específicos do processo cicatricial no pacu.Wound healing and tissue repair are necessary to ensure survival and health of any organism. The aim of this study was to investigate the impact of supplementation with chromium carbochelate (CC) and Saccharomyces cerevisiae (SC) on wound healing in tropical teleost fish Piaractus mesopotamicus. Thus, fish were distributed into four groups: a) control (without supplementation); b) supplemented with 18 mg/kg of chromium carbochelate; c) supplemented with 0.3% of S. cerevisiae and d) supplemented with an association of both supplements. After 105 days of feeding, full-thickness skin incisions (2.0 x 1.0 x 0.25 cm) were performed removing epidermis and dermis. Macroscopic and histologic observations were carried out at 1, 3, 7, 14, 21, 28, and 35 days after wounding to monitor the healing rate. Opposing fronts advanced gradually and faster each day demonstrating a progressive increase in the healing process over time. The inflammatory process was exacerbated and expansive, with an increase in mucous cells and chromatophores. Although no significant differences were observed between groups on wound retraction and microscopic parameters, fish supplemented with CC and SC showed faster re-epithelialization, greater degree of organization of collagen fibers, and higher neovascularization.     Healthy maternal bonding as a resilience factor for depressive disorder

    INTRODUCTION: Depressive disorders (DDs) are very prevalent disorders particularly in women, a high-risk gender group. Determining the risk and protective factors associated with the development of DDs is fundamental to planning preventive and therapeutic strategies. In this study, we evaluated the correlations between healthy maternal attachment and the development of DDs in adulthood. METHODS: We evaluated 52 women at 6 months to 1 year after premature childbirth at Maternidade Vila Nova Cachoeirinha. They were evaluated using the following instruments: Brazilian Criteria of Economic Classification,Parental Bonding Inventory (PBI),Center for Epidemiologic Studies Depression Scale (CES-D), and Edinburgh Postnatal Depression Scale (EPDS). Cut-off scores on the CES-D and EPDS were used to classifythe subjects as currently having a DD or having probable postpartum disorder (PPD) after childbirth. Multiple logistic regression was used to evaluate the risk factors associated with DDs. RESULTS: We found that 49.1% of the sample had a current depressive episode, and 73.6% had probable PPD. Based on logistic regression, current depression (odds ratio = 1.092 [confidence interval: 1.005; 1.186]), and a PPD (odds ratio = 1.108 [confidence interval: 1.011; 1.21]) were negatively correlated with affective maternal relationships. CONCLUSIONS: Women who reported healthy attachment with their mothers did not develop DDs when faced with stressful situations such as premature childbirth

    Integrative genomic analysis of methylphenidate response in attention-deficit/hyperactivity disorder

    Methylphenidate (MPH) is the most frequently used pharmacological treatment in children with attention-deficit/hyperactivity disorder (ADHD). However, a considerable interindividual variability exists in clinical outcome. Thus, we performed a genome-wide association study of MPH efficacy in 173 ADHD paediatric patients. Although no variant reached genome-wide significance, the set of genes containing single-nucleotide polymorphisms (SNPs) nominally associated with MPH response (P < 0.05) was significantly enriched for candidates previously studied in ADHD or treatment outcome. We prioritised the nominally significant SNPs by functional annotation and expression quantitative trait loci (eQTL) analysis in human brain, and we identified 33 SNPs tagging cis-eQTL in 32 different loci (referred to as eSNPs and eGenes, respectively). Pathway enrichment analyses revealed an over-representation of genes involved in nervous system development and function among the eGenes. Categories related to neurological diseases, psychological disorders and behaviour were also significantly enriched. We subsequently meta-analysed the association with clinical outcome for the 33 eSNPs across the discovery sample and an independent cohort of 189 ADHD adult patients (target sample) and we detected 15 suggestive signals. Following this comprehensive strategy, our results provide a better understanding of the molecular mechanisms implicated in MPH treatment effects and suggest promising candidates that may encourage future studies

    Integrative proteomics and pharmacogenomics analysis of methylphenidate treatment response

    Transcriptomics and candidate gene/protein expression studies have indicated several biological processes modulated by methylphenidate (MPH), widely used in attention-deficit/hyperactivity disorder (ADHD) treatment. However, the lack of a differential proteomic profiling of MPH treatment limits the understanding of the most relevant mechanisms by which MPH exerts its pharmacological effects at the molecular level. Therefore, our aim is to investigate the MPHinduced proteomic alterations using an experimental design integrated with a pharmacogenomic analysis in a translational perspective. Proteomic analysis was performed using the cortices of Wistar-Kyoto rats, which were treated by gavage with MPH (2 mg/kg) or saline for two weeks (n = 6/group). After functional enrichment analysis of the differentially expressed proteins (DEP) in rats, the significant biological pathways were tested for association with MPH response in adults with ADHD (n = 189) using genome-wide data. Following MPH treatment in rats, 98 DEPs were found (P 1.0). The functional enrichment analysis of the DEPs revealed 18 significant biological pathways (gene-sets) modulated by MPH, including some with recognized biological plausibility, such as those related to synaptic transmission. The pharmacogenomic analysis in the clinical sample evaluating these pathways revealed nominal associations for gene-sets related to neurotransmitter release and GABA transmission. Our results, which integrate proteomics and pharmacogenomics, revealed putative molecular effects of MPH on several biological processes, including oxidative stress, cellular respiration, and metabolism, and extended the results involving synaptic transmission pathways to a clinical sample. These findings shed light on the molecular signatures of MPH effects and possible biological sources of treatment response variability

    Análise dos irrigantes endodônticos contra o biofilme de Enterococcus faecalis: uma revisão integrativa

    Um dos principais desafios no sucesso do tratamento endodôntico é a eliminação do microrganismo Enterococcus faecalis (EF), que é o principal responsável pelas infecções persistentes e secundárias nos canais radiculares, em decorrência disso cada vez mais os irrigantes são analisados em combate ao EF. Dessa forma, foi realizada uma revisão integrativa buscando comparar os irrigantes de diversas naturezas, tanto à base de plantas quanto sintéticos, contra o Enteroccocus faecalis. Nessa revisão foi inicialmente selecionado 84 artigos, que fossem dos últimos 5 anos, na plataforma PubMed que após os critérios de inclusão e exclusão foram selecionados 8 artigos para compor a tabela de resultados; ao discutir estes trabalhos foi observado que as nanoparticuladas não apresentaram resultados satisfatórios em relação aos convencionais, também foi observado diversos irrigantes fitoterápicos, que são pouco populares, apresentam resultados iguais e em alguns casos até superiores aos irrigantes convencionais, demonstrando assim que deve-se aumentar as pesquisas e desenvolvimento de irrigantes alternativos que podem apresentar uma grande eficácia clínica e baixa citotoxidade

    SARS-CoV-2 uses CD4 to infect T helper lymphocytes

    The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the agent of a major global outbreak of respiratory tract disease known as Coronavirus Disease 2019 (COVID-19). SARS-CoV-2 infects mainly lungs and may cause several immune-related complications, such as lymphocytopenia and cytokine storm, which are associated with the severity of the disease and predict mortality. The mechanism by which SARS-CoV-2 infection may result in immune system dysfunction is still not fully understood. Here, we show that SARS-CoV-2 infects human CD4+ T helper cells, but not CD8+ T cells, and is present in blood and bronchoalveolar lavage T helper cells of severe COVID-19 patients. We demonstrated that SARS-CoV-2 spike glycoprotein (S) directly binds to the CD4 molecule, which in turn mediates the entry of SARS-CoV-2 in T helper cells. This leads to impaired CD4 T cell function and may cause cell death. SARS-CoV-2-infected T helper cells express higher levels of IL-10, which is associated with viral persistence and disease severity. Thus, CD4-mediated SARS-CoV-2 infection of T helper cells may contribute to a poor immune response in COVID-19 patients.</p
