    Influence of microstructure on the enhancement of soft magnetic character and the induced anisotropy of field annealed HITPERM-type alloys

    Hitperm-type rapidly quenched ribbons were submitted to field annealing, both longitudinal field (LF) and transversal field (TF) to the axis of the ribbon. LF annealing yields a reduction of the magnetic anisotropy and results can be explained in the frame of random anisotropy model. A coercivity of 3 A/m is obtained for Fe 39 Co 39 Nb 6 B 15 Cu 1 alloy. The addition of Cu to these Nb-containing Hitperm-type alloys is a key factor to refine the microstructure in order to reach this very low coercivity value. TF annealing produces samples with sheared hysteresis loops suitable for sensor and high frequency applications

    A pályázati támogatás olyan neuroprotekciós eljárások, anyagok kidolgozását szolgálta, mely ischemiás állapotok, traumák után fellépő hiperexcitáció csökkentésével mérsékli az ilyenkor fellépő másodlagos sejtpusztulást. A triptofán egyik metabolitja a kinurénsav (KYNA), mely az NMDA receptorok természetes inhibitoraként viselkedik a NMDA receptorokhoz kapcsolódva. A KYNA azonban nem megy át a vér-agy gáton, viszont előanyaga a kinurenin (KYN) igen. Kidolgoztunk egy módszert, melyben a KYN-t (300 mg/kg, i.p.) probeneciddel (PROB, 200 mg/kg, i.p.) együtt adva, oly mértékig meg tudjuk emelni a KYNA szintet az agyban, hogy az hatékonyan képes gátolni az agyi sérüléseket követő hiperexcitációt. Ezzel a módszerrel hatékony neuroprotekciót értünk el focalis és globalis agyi ischemiában, pentylentetrazollal (PTZ) kiváltott görcstevékenységben, de még migrén modellben is. Kifejlesztettünk két további kinurénsav származékot, a glukozamin-kinurénsavat (KYNA-NH-GLUC) és az SZR-72-nek nevezett szert, melyek szintén hatékony neuroprotektív anyagok, ugyanakkor átjutnak a vér-agy gáton, tehát szisztémásan is adhatók. Kísérleti körülmények között tehát hatékony neuroprotektív eljárást dolgoztunk ki, és hatékony anyagokat állítottunk elő. Továbbá, kimutattuk a KYNA-ról, hogy Janus-arcú, mert gátló hatása mellett, amit ?M-os koncentrációban fejt ki, ennél jóval alacsonyabb koncentrációban (200-250 nM) serkentő hatású. Mindez felveti a KYNA lehetséges neuromodulátor szerepét. | The aim of this project was to develop neuroprotective agents and protocols, which are able to reduce the late neuronal death following ischemic brain attack or brain trauma. The kynurenine pathway converts tryptophan into various compounds, including L-kynurenine (KYN), which in turn can be converted into the excitatory amino acid receptor (namely NMDA receptor) antagonist kynurenic acid (KYNA). The use of KYNA as a neuroprotective agent is rather restricted, however, because KYNA has only a very limited ability to cross the blood-brain barrier. In contrast, KYN crosses this barrier more readily. We have developed a protocol in which KYN (300 mg/kg, i.p.) administration together with probenedid (PROB, 200 mg/kg, i.p.), an organic acid transport blocker, led to significant neuroprotection in both focal and global brain ischemia, in traumatic brain attack, in migraine, and led to the inhibition of pentylenetetrazole (PTZ)-induced seizures. In addition, we have developed two kynurenic acid derivatives; glucosamine-kynurenic acid (KYNA-NH-GLUC) and SZR-72. Both of them cross the blood-brain barrier easily, and are neuroprotective agents. In addition to these experimental results, we have shown that KYNA is a Janus-faced agent, which in ?M concentration behaves as a neuroinhibitor, while in nM concentration, it stimulates the neuronal activity. This result suggests that KYNA might be play a role of a neuromodulator in the central nervoussystem

    Principles of Formation of Flexible Manufacturing Systems

    The article presents points necessary for the production re-prophilation into the flexible one through the generation of so called flexible manufacturing cells. Mentioned are the reasons and effects which are brought by the flexible production. In general, the key to the generation of the flexible production is the group technology based on the principle of the material flow analysis. The variants of the possible solutions based on the material flows confirm that the problem of the flexible manufacturing cells generation is not an easy one and unambiguous either. But considering only this one criterion is not sufficient for the practice because it does not consider for example the number of the pieces. Therefore the present article in the principle points at the application of the multiplied criteria about the part or machine, based on which more specifically stated is the cell profile, as number of machines, type of machines, intercellular transfers and so on

    Cilj je ovoga rada određivanje odgovarajućih parametara za raspršivanje adheziva korištenih u industriji uz primjenu robota. Korišten je robot ABB IRB 1600 u neeksplozivnom okruženju. Adheziv se oslobađao iz tlačne posude i nanosio raspršivačem Krautzberger RA5. U radu se daje detaljan opis dijagrama za automatsko spajanje pištolja za bojenje. Rezultati pojedinačnih testova dani su u tablicama, a ovisnosti pojedinih parametara prikazane su u grafikonima. Ovisno o količini nanesenog mlaza, najbolji uzorci su ocijenjeni u skladu s preporukama proizvođača adheziva.This article aims at setting appropriate parameters for robotic spraying of adhesives used in industry. The robot ABB IRB 1600 was used in a non-explosive environment. The adhesive was dispensed from pressure sweat and applied by a Krautzberger RA5 spray head. The article describes a detailed diagram for automatic connection of a painting gun. The results of the individual tests are shown in the tables and the dependencies of the individual parameters are described in the graphs. Depending on the amount of spray applied, the best samples are evaluated according to the adhesive manufacturer\u27s recommendations

    Peroxisome Proliferator Activated Receptor Gamma Controls Mature Brown Adipocyte Inducibility through Glycerol Kinase

    Peroxisome proliferator-activated receptors (PPARs) have been suggested as the master regulators of adipose tissue formation. However, their role in regulating brown fat functionality has not been resolved. To address this question, we generated mice with inducible brown fat-specific deletions of PPARα, β/δ, and γ, respectively. We found that both PPARα and β/δδ are dispensable for brown fat function. In contrast, we could show that ablation of PPARγ in vitro and in vivo led to a reduced thermogenic capacity accompanied by a loss of inducibility by β-adrenergic signaling, as well as a shift from oxidative fatty acid metabolism to glucose utilization. We identified glycerol kinase (Gyk) as a partial mediator of PPARγ function and could show that Gyk expression correlates with brown fat thermogenic capacity in human brown fat biopsies. Thus, Gyk might constitute the link between PPARγ-mediated regulation of brown fat function and activation by β-adrenergic signaling.</p

    Impact of safety-related dose reductions or discontinuations on sustained virologic response in HCV-infected patients: Results from the GUARD-C Cohort

    BACKGROUND: Despite the introduction of direct-acting antiviral agents for chronic hepatitis C virus (HCV) infection, peginterferon alfa/ribavirin remains relevant in many resource-constrained settings. The non-randomized GUARD-C cohort investigated baseline predictors of safety-related dose reductions or discontinuations (sr-RD) and their impact on sustained virologic response (SVR) in patients receiving peginterferon alfa/ribavirin in routine practice. METHODS: A total of 3181 HCV-mono-infected treatment-naive patients were assigned to 24 or 48 weeks of peginterferon alfa/ribavirin by their physician. Patients were categorized by time-to-first sr-RD (Week 4/12). Detailed analyses of the impact of sr-RD on SVR24 (HCV RNA <50 IU/mL) were conducted in 951 Caucasian, noncirrhotic genotype (G)1 patients assigned to peginterferon alfa-2a/ribavirin for 48 weeks. The probability of SVR24 was identified by a baseline scoring system (range: 0-9 points) on which scores of 5 to 9 and <5 represent high and low probability of SVR24, respectively. RESULTS: SVR24 rates were 46.1% (754/1634), 77.1% (279/362), 68.0% (514/756), and 51.3% (203/396), respectively, in G1, 2, 3, and 4 patients. Overall, 16.9% and 21.8% patients experienced 651 sr-RD for peginterferon alfa and ribavirin, respectively. Among Caucasian noncirrhotic G1 patients: female sex, lower body mass index, pre-existing cardiovascular/pulmonary disease, and low hematological indices were prognostic factors of sr-RD; SVR24 was lower in patients with 651 vs. no sr-RD by Week 4 (37.9% vs. 54.4%; P = 0.0046) and Week 12 (41.7% vs. 55.3%; P = 0.0016); sr-RD by Week 4/12 significantly reduced SVR24 in patients with scores <5 but not 655. CONCLUSIONS: In conclusion, sr-RD to peginterferon alfa-2a/ribavirin significantly impacts on SVR24 rates in treatment-naive G1 noncirrhotic Caucasian patients. Baseline characteristics can help select patients with a high probability of SVR24 and a low probability of sr-RD with peginterferon alfa-2a/ribavirin

    BATLAS: Deconvoluting Brown Adipose Tissue

    Recruitment and activation of thermogenic adipocytes have received increasing attention as a strategy to improve systemic metabolic control. The analysis of brown and brite adipocytes is complicated by the complexity of adipose tissue biopsies. Here, we provide an in-depth analysis of pure brown, brite, and white adipocyte transcriptomes. By combining mouse and human transcriptome data, we identify a gene signature that can classify brown and white adipocytes in mice and men. Using a machine-learning-based cell deconvolution approach, we develop an algorithm proficient in calculating the brown adipocyte content in complex human and mouse biopsies. Applying this algorithm, we can show in a human weight loss study that brown adipose tissue (BAT) content is associated with energy expenditure and the propensity to lose weight. This online available tool can be used for in-depth characterization of complex adipose tissue samples and may support the development of therapeutic strategies to increase energy expenditure in humans

    Azimuthal anisotropy of charged jet production in root s(NN)=2.76 TeV Pb-Pb collisions

    We present measurements of the azimuthal dependence of charged jet production in central and semi-central root s(NN) = 2.76 TeV Pb-Pb collisions with respect to the second harmonic event plane, quantified as nu(ch)(2) (jet). Jet finding is performed employing the anti-k(T) algorithm with a resolution parameter R = 0.2 using charged tracks from the ALICE tracking system. The contribution of the azimuthal anisotropy of the underlying event is taken into account event-by-event. The remaining (statistical) region-to-region fluctuations are removed on an ensemble basis by unfolding the jet spectra for different event plane orientations independently. Significant non-zero nu(ch)(2) (jet) is observed in semi-central collisions (30-50% centrality) for 20 <p(T)(ch) (jet) <90 GeV/c. The azimuthal dependence of the charged jet production is similar to the dependence observed for jets comprising both charged and neutral fragments, and compatible with measurements of the nu(2) of single charged particles at high p(T). Good agreement between the data and predictions from JEWEL, an event generator simulating parton shower evolution in the presence of a dense QCD medium, is found in semi-central collisions.
