    Meeting the Needs of Multilingual Students: Using Teacher-Reported Challenges and Successes for Teacher Preparation

    Preparing teachers to meet the needs of multilingual students is the goal of TESOL and Bilingual education programs in higher education. What these programs use to determine what these needs are can vary by location, faculty, and population of learners. This qualitative study surveyed in-service teachers applying for their TESOL or Bilingual endorsements in a college in the southwest United States. Research questions asked about the challenges and successes teachers face in meeting the needs of multilingual students and used this data to determine themes. The data showed that teachers encounter challenges meeting the needs of multilingual students in the areas of language differentiation, and Special Education. Conversely, teachers experienced success in building relationships and utilizing structural support. With this research, the aim is to use what teachers reported to make recommendations to teacher education programs and adapt our course content to what they need

    Underestimating Our Influence over Others’ Unethical Behavior and Decisions

    We examined the psychology of “instigators,” i.e., people who surround an unethical act and influence the wrongdoer (the “actor”) without directly committing the act themselves. In four studies, we found that instigators of unethical acts underestimated their influence over actors. In Studies 1 and 2, university students enlisted other students to commit a “white lie” (Study 1) or commit a small act of vandalism (Study 2) after making predictions about how easy it would be to get their fellow students to do so. In Studies 3 and 4, online samples of participants responded to hypothetical vignettes, e.g., about buying children alcohol, and taking office supplies home for personal use. In all 4 studies, instigators failed to recognize the social pressure they levied on actors through simple unethical suggestions, i.e., the discomfort actors would experience by making a decision that was inconsistent with the instigator’s suggestion

    Glioblastoma multiforme (GBM) is the most frequent and lethal primary brain tumor in adults. Despite intense biomedical research, the median survival after diagnosis is 15 months. One factor contributing to this poor prognosis is the immune protection afforded by the tumor microenvironment. Tumors have a diverse repertoire of immune-evasive techniques. One method of evasion not well explored is the release of tumor-derived exosomes. Exosomes are tiny membrane-bound vesicles of endocytic origin that contain viable mRNA and functional proteins that can affect the physiology of recipient cells. Exosome release has been reported for numerous cancer types, including GBM. Exosomes from colon cancer have been shown to carry Fas ligand (FasL) and to induce apoptosis of activated T cells. The aim of this study was to elucidate whether the same immune-evasive technique is used in GBM. GBM exosomes were isolated from the serum-free culture medium of U87 MG and U138 MG cells by using differential ultracentrifugation and were then resuspended in phosphate-buffered saline. The protein concentration of the resulting exosome pellet was determined, and subsequent exosome treatments were based on protein concentration. A3T T cells were plated at a concentration of 10,000 cells per well in 96-well plates and were treated with quantified exosome fractions or with recombinant FasL, and T cell proliferation was determined. Our data demonstrated that tumor-derived exosomes significantly inhibited the proliferation of T cells and that the cellular inhibition resulting from the exosomes was comparable to that seen with the recombinant FasL. These results suggest that targeting FasL in GBM could greatly decrease the amount of immune suppression that occurs at the tumor site

    Multidrug-resistant Candida glabrata strains obtained by induction of anidulafungin resistance in planktonic and biofilm cells

    Candida glabrata has emerged as a common cause of serious life-threatening fungal infections, largely owing to their low susceptibility to azole antifungals. Recent guidance indicates the use of echinocandins as the first-choice drug for the treatment of systemic infections of C. glabrata; however, C. glabrata resistance to echinocandins is reportedly increasing. Herein, we present the induction of anidulafungin resistance in planktonic and sessile cells of C. glabrata and the development of fluconazole crossresistance. MICs of 21 clinical C. glabrata strains were determined by a broth microdilution method using anidulafungin and fluconazole. Biofilm formation on a tracheal catheter was determined using 1- × 1-cm2 polyvinyl polychloride catheter fragments. Induction of anidulafungin resistance in planktonic and sessile cells and evaluation of its stability were performed by exposing the strains to successively higher concentrations of the antifungal. The induction resulted in strains strongly resistant to anidulafungin (MICs: 1−2 ÎŒg/mL) and fluconazole (≄64 ÎŒg/mL). Most of the sessile cells of C. glabrata presented slightly reduced susceptibility compared with the planktonic cells. Clinically, this cross-resistance could lead to therapeutic failure while using fluconazole in patients previously exposed to subinhibitory concentrations of anidulafungin for extended periods

    The TWA 3 Young Triple System: Orbits, Disks, Evolution

    We have characterized the spectroscopic orbit of the TWA 3A binary and provide preliminary families of probable solutions for the TWA 3A visual orbit as well as for the wide TWA 3A--B orbit. TWA 3 is a hierarchical triple located at 34 pc in the ∌\sim10 Myr old TW Hya association. The wide component separation is 1."55; the close pair was first identified as a possible binary almost 20 years ago. We initially identified the 35-day period orbital solution using high-resolution infrared spectroscopy which angularly resolved the A and B components. We then refined the preliminary orbit by combining the infrared data with a re-analysis of our high-resolution optical spectroscopy. The orbital period from the combined spectroscopic solution is ∌\sim35 days, the eccentricity is ∌\sim0.63, and the mass ratio is ∌\sim0.84; although this high mass ratio would suggest that optical spectroscopy alone should be sufficient to identify the orbital solution, the presence of the tertiary B component likely introduced confusion in the blended optical spectra. Using millimeter imaging from the literature, we also estimate the inclinations of the stellar orbital planes with respect to the TWA 3A circumbinary disk inclination and find that all three planes are likely misaligned by at least ∌\sim30 degrees. The TWA 3A spectroscopic binary components have spectral types of M4.0 and M4.5; TWA 3B is an M3.     Women's reproductive rights in the inter-American system of human rights: conclusions from the Field, June - September 2014

    The Inter-American System of Human Rights has proven to be a forum for the advancement of women’s reproductive rights in the Inter-American region. However, the Inter-American System faces significant challenges in promoting structural transformative change that enables women’s enjoyment of their reproductive health rights. This report examines three reproductive rights cases from the Inter- American Commission on Human Rights and the Inter-American Court of Human Rights: María Mamerita Mestanza Chávez v. Peru; Paulina Ramirez Jacinto v. Mexico; and Artavia Murillo et al. v. Costa Rica. In the summer of 2014, interviews were conducted with representatives in each of the case study countries, with the objective of the research being two-fold: (1) to understand how each of the cases developed, and the subsequent challenges and advancements; and (2) to learn from these cases in order to suggest recommendations for how actors can make better use of the Inter-American System as one of several avenues for protecting, promoting and fulfilling women’s reproductive rights. The report first discusses challenges in implementing women’s reproductive health rights, and then explores how the Inter-American System can strengthen its work on women’s reproductive health rights

    Defective Regulation Of Pomc Precedes Hypothalamic Inflammation In Diet-induced Obesity

This anabolic state results from the defective activity of hypothalamic neurons involved in the sensing and response to adiposity. However, it is currently unknown what the earliest obesity-linked hypothalamic defect is and how it orchestrates the energy imbalance present in obesity. Using an outbred model of diet-induced obesity we show that defective regulation of hypothalamic POMC is the earliest marker distinguishing obesity-prone from obesity-resistant mice. The early inhibition of hypothalamic POMC was sufficient to transform obesity-resistant in obesity-prone mice. In addition, the post-prandial change in the blood level of beta-endorphin, a POMC-derived peptide, correlates with body mass gain in rodents and humans.     The Impact of Vulvar Lichen Sclerosus on Sexual Dysfunction

    Background: Lichen sclerosus (LS) is a chronic inflammatory condition that is known to arise on the vulva. Many women with LS report vulvar pain, often affecting a patient's quality of life. In this study, the sexual function of LS patients, with and without pain, was compared to control populations. Materials and Methods: A case-control study to examine the relationship between LS and sexual dysfunction was conducted. A total of 335 women presenting to the gynecology clinic were included in the study: 197 women with biopsy confirmed LS were compared to two control groups (95 asymptomatic women were ?healthy? controls and 43 women had vulvovaginal candidiasis) on self-reported current health complaints, medical and surgical history and current symptoms such as pain and itching, type and frequency of sexual activity, and satisfaction with sexual activity. Results: Women with LS reported less frequent sexual activity than healthy controls (p=0.007) and Candida controls (p=0.04). Currently sexually active women with LS were significantly less likely to report vaginal intercourse (71.6%) than healthy controls (89.0%, p=0.003) or Candida controls (100%, p=0.0003), even though similar proportions of all three groups reported that vaginal intercourse was important. Satisfaction towards the quality of current sexual activity was significantly lower among women with LS compared with both the healthy and Candida control groups. 23.7% of women with LS reported that sexual activity was rarely or never satisfactory as compared with 0% of healthy controls (p<0.0001) and 6.5% of Candida controls (p=0.03).     Swift XRT Observations of the Afterglow of XRF 050416A

    Swift discovered XRF 050416A with the BAT and began observing it with its narrow field instruments only 64.5 s after the burst onset. Its very soft spectrum classifies this event as an X-ray flash. The afterglow X-ray emission was monitored up to 74 days after the burst. The X-ray light curve initially decays very fast, subsequently flattens and eventually steepens again, similar to many X-ray afterglows. The first and second phases end about 172 and 1450 s after the burst onset, respectively. We find evidence of spectral evolution from a softer emission with photon index Gamma ~ 3.0 during the initial steep decay, to a harder emission with Gamma ~ 2.0 during the following evolutionary phases. The spectra show intrinsic absorption in the host galaxy. The consistency of the initial photon index with the high energy BAT photon index suggests that the initial phase of the X-ray light curve may be the low-energy tail of the prompt emission. The lack of jet break signatures in the X-ray afterglow light curve is not consistent with empirical relations between the source rest-frame peak energy and the collimation-corrected energy of the burst. The standard uniform jet model can give a possible description of the XRF 050416A X-ray afterglow for an opening angle larger than a few tens of degrees, although numerical simulations show that the late time decay is slightly flatter than expected from on-axis viewing of a uniform jet.     Increased levels of RNA oxidation enhance the reversion frequency in aging pro-apoptotic yeast mutants

    Despite recent advances in understanding the complexity of RNA processes, regulation of the metabolism of oxidized cellular RNAs and the mechanisms through which oxidized ribonucleotides affect mRNA translation, and consequently cell viability, are not well characterized. We show here that the level of oxidized RNAs is markedly increased in a yeast decapping Kllsm4Δ1 mutant, which accumulates mRNAs, ages much faster that the wild type strain and undergoes regulated-cell-death. We also found that in Kllsm4Δ1 cells the mutation rate increases during chronological life span indicating that the capacity to han- dle oxidized RNAs in yeast declines with aging. Lowering intracellular ROS levels by antioxidants recovers the wild- type phenotype of mutant cells, including reduced amount of oxidized RNAs and lower mutation rate. Since mRNA oxidation was reported to occur in different neurodegen- erative diseases, decapping-deficient cells may represent a useful tool for deciphering molecular mechanisms of cell response to such conditions, providing new insights into RNA modification-based pathogenesis
