The n-acetyl phenylalanine glucosamine derivative attenuates the inflammatory/catabolic environment in a chondrocyte-synoviocyte co-culture system
Osteoarthritis (OA), the most prevalent degenerative joint disease, still lacks a true disease-modifying therapy. The involvement of the NF-κB pathway and its upstream activating kinases in OA pathogenesis has been recognized for many years. The ability of the N-acetyl phenylalanine glucosamine derivative (NAPA) to increase anabolism and reduce catabolism via inhibition of IKKα kinase has been previously observed in vitro and in vivo. The present study aims to confirm the chondroprotective effects of NAPA in an in vitro model of joint OA established with primary cells, respecting both the crosstalk between chondrocytes and synoviocytes and their phenotypes. This model satisfactorily reproduces some features of the previously investigated DMM model, such as the prominent induction of ADAMTS-5 upon inflammatory stimulation. Both gene and protein expression analysis indicated the ability of NAPA to counteract key cartilage catabolic enzymes (ADAMTS-5) and effectors (MCP-1). Molecular analysis showed the ability of NAPA to reduce IKKα nuclear translocation and H3Ser10 phosphorylation, thus inhibiting IKKα transactivation of NF-κB signalling, a pivotal step in the NF-κB-dependent gene expression of some of its targets. In conclusion, our data confirm that NAPA could truly act as a disease-modifying drug in OA
Laser hardening of steel sintered parts
The possibility of applying rapid and localized laser hardening to near-net shape parts, like the ones deriving from powder metallurgy (P/M) is investigated, demonstrating that even low alloyed steels (Fe + 2% Cu + 0,7% C) can be successfully heat treated with minimal or no dimensional variations. Laser hardening conditions have been selected on the basis of the results of the previous research, carried out by means of an Nd-YAG high power system [1]. To avoid some carbon loss, observed on previous activities, the samples have been protected by neutral atmosphere. The microstructural features of the laser hardened steels have been analyzed by optical microscopy, whereas the surface micro-geometry has been characterized by scanning electron microscope. Hardened depth (HD), hardened width (HW) and hardened area (HA) have been measured as well. As expected, the micro-hardness profiles present a sharp drop at low distance from the hardened surface. The typical splitting between hardened zone and heat-Affected zone (HAZ), well known from laser hardened fully dense steels, has been observed also on low-Alloy sintered steels. The use of a protective atmosphere has been helpful to control surface decarburization and to prevent oxidation. The research confirm that Laser transformation Hardening (LTH) is a suitable hardening process of P/M components, through the action of a scanning laser beam. The short heating time and the modest volume fraction structurally modified can contribute to avoid part distortion, in comparison with other hardening methods
The ecology of predatory hoverflies as ecosystem-service providers in agricultural systems
The main contemporary challenge for agriculture is to meet the food demands of the increasing world population while becoming more environmentally sustainable. One way to achieve this is through the promotion of functional biodiversity and the ecosystem (nature’s) services (ES) that it can provide. The Syrphinae subfamily is a widespread group of hoverflies with a high potential for that service. Hence, the present review aims to synthesize the existing literature on this group. This review is divided in three main sections. First, we focus on those aspects of the ecology of hoverflies that are relevant to their role in biological control. These are divided into nutrition, feeding preferences and prey detection of the different developmental stages, overwintering and the effect of landscape on their dispersal and efficacy. Second, we review the ES that predatory hoverflies can provide, particularly pollination and biological control. Finally, we discuss those farming practices that can affect the effectiveness of these Diptera as providers of ES. Overall, this review highlights the potential, as well as limitations and current gaps in knowledge, for enhancing the efficacy of hoverflies as ecosystem-service providers in agricultural systems.info:eu-repo/semantics/acceptedVersio
Evaluation of saddle and driving aptitudes in Monterufoli pony.
The Monterufoli pony is an endangered Tuscan breed. In the 80's began a project for the conservation of the breed and at present there are roughly 200 individuals. The equine was once utilized for saddle and driving and this study deals with the training for these two aptitudes. The mor- phologic type of the pony seems suited for saddle, in particular for children and beginners, and driving. The ponies showed developed chest, strong legs with short shanks: all these characters were useful for trot and driving. In this trial 3-4 years old never tamed Monterufoli ponies were opportunely choose and subsequently trained for saddle and driving. The ponies were submitted to the "aptitude test" for the two aptitudes and the results were good both for practical and character sides. The marks for sad- dle and driving were 8.16 and 8.06 respectively. Also the 3 ponies showed good results for the Aptitude Index: 7.60, 7.87 and 7.89. The results of the trial showed the excellent ability of the Monterufoli pony for saddle and driving. The good results of the test are important for the diffusion of the breed in the territory and in particular in horse centres and in equestrian tourism sites
Pulsed Current Effect on the Hard Anodizing of an AlSi10Mg Aluminum Alloy Obtained via Additive Manufacturing
The hard anodizing treatments of cast Al-Si alloys are notoriously difficult. Indeed, their microstructural features hinder the growth of a uniform, compact, and defect-free anodic oxide. In this paper, AlSi10Mg samples, produced via Gravity Casting (GC) and Additive Manufacturing, i.e., Laser Powder Bed Fusion (L-PBF), were hard anodized in a sulfuric acid bath, in order to verify how the particular microstructure obtained via L-PBF affects the thickness, hardness, compactness, and defectiveness of the anodic oxide. Moreover, for the first time, Pulsed Direct Current (PDC) procedures were used to perform the hard anodizing treatments on additively manufactured AlSi10Mg alloy. Several combinations of temperature and electrical parameters, i.e., current density, frequency, and Duty Cycle, were tested. The anodized samples were characterized through optical microscopy analysis, Scanning Electron Microscopy (SEM) analysis, and accelerated corrosion tests, i.e., Potentiodynamic Polarization (POL) and Electrochemical Impedance Spectroscopy (EIS) measurements. The PDC procedures allowed improvement of the compromise between evenness, compactness, and defectiveness. Among the attempted PDC procedures, a specific combination of electrical parameters and temperature allowed the best results to be obtained, i.e., the highest hardness and the lowest volumetric expansion values without compromising the oxide quality rating and the corrosion resistance behavior. However, none of the attempted PCD strategies allowed the hardness values obtained on samples produced via GC to be reached
High entropy alloys obtained by field assisted powder metallurgy route: SPS and microwave heating
The aim of this work was to investigate the field assisted powder metallurgy route for producing HEAs at equimolar composition, i.e. FeCoNiCrAl, starting from metal powders. Both mixed, mechanically activated and mechanically alloyed powders have been used. The powders obtained by mechanical alloying were synthesized only by SPS, whereas the remaining ones were sintered by SPS or microwave heating. The investigated field assisted sintering techniques allowed an extremely short alloying time, high energy density on the load and negligible contamination by the surrounding environment. Both the conducted sintering-synthesis technology resulted not definitive to produce chemical homogeneity and to obtain a single stable structure. Thus a subsequently heat treatment was required. The post heat treatment, indeed, led to a single crystalline structure (FCC) and the material was fully recrystallized. After heat treatment samples are isomorphic: they exhibit two different phases with the same FCC cell, but different chemical composition, in detail Fe-Cr richer and Al-Ni richer. SPS-ed samples present a reduced porosity, while microwave processed ones are much more porous and this is reflected in the mechanical properties
Microwave sintering of SiAlON ceramics with TiN addition
α-β SiAlON/TiN composites with nominal composition of α:= β25:75 were fabricated by microwave sintering. The effect of titanium nitride addition on the phases, microstructure, microwave absorption ability and mechanical properties (Vickers hardness and fracture toughness) of the SiAlON-based composites were studied. Finite Difference Time Domain (FDTD) software was used for the numerical simulation in order to assess the most suitable experimental setup. Sintering trials were performed in a single mode microwave furnace operating at 2.45 GHz and a power output of 660W, for a reaction time of 30 min. SiC blocks were used as a susceptor to accelerate the microwave processing by hybrid heating, with reduced heat losses from the surface of the material of the α- βSiAlON/TiN composites. The optimum comprehensive mechanical properties, corresponding to a relative density of 96%, Vickers hardness of 12.98 ± 1.81 GPa and Vickers indentation fracture toughness of 5.52 ± 0.71 MPa.m1/2 were obtained at 850°C when the content of TiN was 5 wt.%
Prognosis based on primary breast carcinoma instead of pathological nodal status.
In breast cancer patients, prognostic information required to plan post-surgical therapy is obtained mainly through axillary dissection. This study was designed to establish a new prognostic score based solely on parameters of the primary tumour as an alternative to axillary surgery in assessing prognosis. Eight different prognostic factors, including menopausal status, tumour size, grading, lymphatic invasion, desmoplasia, necrosis, c-erbB-2 and laminin receptor expression, were evaluated retrospectively on a large series of primary breast carcinoma patients. From multivariate analysis, four independent parameters were selected and examined, alone and in combination, for their prognostic potential. These parameters were used to generate a prognostic score that was analysed retrospectively in 467 N0-N1a patients to determine its predictive value for survival. The score, which includes variables such as tumour size, grading, laminin receptor and c-erbB-2 overexpression, was established based on the number of negative prognostic factors: score 1 refers to cases in which all four parameters reflect a good prognosis, scores 2 and 3 refer to tumours in which, respectively, one or two of the four parameters reflect a poor prognosis, whereas score 4 refers to tumours with three or four poor prognosis factors. Analysis of the overall survival of the four score groups shows that patients with score 1 tumours (22% of the total) had the best prognosis with a 15 year survival of 82%, patients with score 2 and 3 had an intermediate prognosis, whereas score 4 patients had the poorest prognosis with a 15 year survival of only 38%. Moreover, survival in the N+ score 1 cases was found to be longer than that in the total N- patients. Our data suggest that the primary tumour score provides more reliable prognostic information than pathological nodal status, and that axillary dissection can be avoided in a large number of patients
The contribution of major risk factors and job strain to occupational class differences in coronary heart disease incidence: the MONICA Brianza and PAMELA population-based cohorts
Objectives We investigated the contribution of major coronary heart disease (CHD) risk factors and job strain to occupational class differences in CHD incidence in a pooled-cohort prospective study in northern Italy.
Methods 2964 men aged 25-74 from four northern Italian population-based cohorts were investigated at baseline and followed for first fatal or non-fatal CHD event (171 events). Standardised procedures were used for baseline risk factor measurements, follow-up and validation of CHD events. Four occupational classes were derived from the Erikson-Goldthorpe-Portocarero social class scheme: higher and lower professionals and administrators, non-manual workers, skilled and unskilled manual workers, and the self-employed. HRs were estimated with Cox models.
Results Among CHD-free subjects, with non-manual workers as the reference group, age-adjusted excess risks were found for professionals and administrators (+84%, p=0.02), the self-employed (+72%, p=0.04) and manual workers (+63%, p=0.04). The relationship was consistent across different CHD diagnostic categories. Adjusting for major risk factors only slightly reduced the reported excess risks. In a sub-sample of currently employed subjects, adjusting for major risk factors, sport physical activity and job strain reduced the excess risk for manual workers (relative change = -71.4%) but did not substantially modify the excess risks of professionals and administrators and the self-employed.
Conclusions In our study, we found higher CHD incidence rates for manual workers, professionals and administrators, and the self-employed, compared to non-manual workers. When the entire spectrum of job categories is considered, the job strain model helped explain the CHD excess risk for manual workers but not for other occupational classes
