Serologic and Molecular Biologic Methods for SARS-associated Coronavirus Infection, Taiwan
Severe acute respiratory syndrome (SARS) has raised a global alert since March 2003. After its causative agent, SARS-associated coronavirus (SARS-CoV), was confirmed, laboratory methods, including virus isolation, reverse transcriptaseâpolymerase chain reaction (RT-PCR), and serologic methods, have been quickly developed. In this study, we evaluated four serologic tests ( neutralization test, enzyme-linked immunosorbent assay [ELISA], immunofluorescent assay [IFA], and immunochromatographic test [ICT]) for detecting antibodies to SARS-CoV in sera of 537 probable SARS case-patients with correlation to the RT-PCR . With the neutralization test as a reference method, the sensitivity, specificity, positive predictive value, and negative predictive value were 98.2%, 98.7%, 98.7%, and 98.4% for ELISA; 99.1%, 87.8%, 88.1% and 99.1% for IFA; 33.6%, 98.2%, 95.7%, and 56.1% for ICT, respectively. We also compared the recombinant-based western blot with the whole virusâbased IFA and ELISA; the data showed a high correlation between these methods, with an overall agreement of >90%. Our results provide a systematic analysis of serologic and molecular methods for evaluating SARS-CoV infection
Asymmetric porous membranes formed by coagulation-induced phase separation in poly(ether sulfone)/poly(vinyl pyrrolidone)/genistein blends
Development of asymmetric channel morphology driven by coagulation-induced phase separation of genistein (G) modified poly(ether sulfone)/poly(vinyl pyrrolidone) (PES/PVP) blends has been examined in relation to their miscibility phase diagram. PES/G pairs turned out to be miscible in the amorphous state, whereas solid-liquid phase separation occurred at high genistein compositions. The solid liquid phase diagram involving the liquidus and solidus lines were computed self-consistently in the framework of the combined free energy of Flory-Huggins for liquid liquid phase separation and phase field free energy for crystal solidification. The ternary phase diagram of PES/PVP/G blends was subsequently established that consisted of various coexistence regions. The actual amounts of genistein incorporated in the PES/PVP membranes were determined as a function of weight percent of genistein in feed. On the basis of UV-vis spectroscopy, the extent of genistein leaching during incubation in human blood was evaluated in conjunction with the PVP leaching from the blend membrane. Optimal Post-Operative Nalbuphine Dose Regimen: A Randomized Controlled Trial in Patients with Laparoscopic Cholecystectomy
Background and Objectives: Optimal opioid analgesia is an excellent analgesia that does not present unexpected adverse effects. Nalbuphine, acting on the opioid receptor as a partial mu antagonist and kappa agonist, is considered a suitable option for patients undergoing laparoscopic surgery. Therefore, we aim to investigate the appropriate dosage of nalbuphine for post-operative pain management in patients with laparoscopic cholecystectomy. Materials and Methods: Patients were randomly categorized into low, medium, and high nalbuphine groups. In each group, a patient control device for post-operative pain control was programed with a low (0.05 mg/kg), medium (0.10 mg/kg), or high (0.20 mg/kg) nalbuphine dose as a loading dose and each bolus dose with a lockout interval of 7 min and without background infusion. Primary and secondary outcomes included the post-operative pain scale and nalbuphine consumption, and episodes of post-operative opioid-related adverse events and satisfactory scores. Results: The low-dosage group presented a higher initial self-reported pain score in comparison to the other two groups for the two hours post-op (p = 0.039) but presented lower nalbuphine consumption than the other two groups for four hours post-op (p = 0.047). There was no significant difference in the analysis of the satisfactory score and adverse events. Conclusions: An appropriate administration of nalbuphine could be 0.1 to 0.2 mg/kg at the initial four hours; this formula could be modified to a lower dosage (0.05 mg/kg) in the post-operative management of laparoscopic cholecystectomy
Rapid and Highly Sensitive Coxsackievirus A Indirect Immunofluorescence Assay Typing Kit for Enterovirus Serotypingâż
We describe the development and evaluation of an indirect immunofluorescence assay (IFA) kit for rapid and sensitive detection of coxsackievirus A2, -4, -5, -6, and -10. This IFA kit was determined to have 95.9 to 100% sensitivity and 95.8 to 97.2% specificity. It also proved to be beneficial in reducing the number of enteroviruses that are untypeable in the clinical virology laboratory
The Ratio of Hmox1/Nrf2 mRNA Level in the Tumor Tissue Is a Predictor of Distant Metastasis in Colorectal Cancer
Heme oxygenase 1 (Hmox1) plays an important role in the growth and spread of tumor, and its expression is regulated positively by Nrf2 [nuclear factor (erythroid-derived 2)-like 2; NFE2L2] and negatively by kelch-like ECH-associated protein 1 (Keap1) and by BTB and CNC homology 1 (Bach1). Both Hmox1 and Nrf2 contribute to distant metastasis of cancer. The mRNA levels of Hmox1, Nrf2, Keap1, and Bach1 in the tumor and normal tissues of 84 subjects with colorectal cancer (CRC) were determined by real-time polymerase chain reaction. The tumor had lower Hmox1 but higher Bach1 mRNA levels than the normal tissue. The correlations of Hmox1 with components of the Nrf2 pathway were not significant in the tumor tissue of CRC subjects with distant metastasis. The ratio of Hmox1/Nrf2 mRNA level (by percentage) in the tumor tissue was lower in the subjects with distant metastasis (97.4% (84.4â111.1%)) than in those without (101.0% (92.7â136.5%)) and was a predictor for distant metastasis in CRC (odds ratio: 0.83; 95% confidence interval: 0.68â0.97) along with serum carcinoembryonic antigen (1.0027, 1.006â1.064). The mRNA level of Hmox1 in the tumor tissue of CRC is not correlated with that of the Nrf2 pathway molecules, and its ratio to the Nrf2 level may be useful for suggesting distant metastasis in CRC
Inhibitions of melanogenesis via Phyllanthus emblica fruit extract powder in B16F10 cells
Nowadays, it is well-known that medicines and cosmetics have potential to handle different skin diseases improving the cutis semblances. The pigment accumulation not only leads to appear dark spots on the derma surface, but also causes some disorders including freckles, melanin precipitation, chloasma, and melanosis. Phyllanthus emblica is an edible nutraceutical and functional food in Asia area with medicinal and nutritive importances, and there is no literature mentioning that P. emblica fruit extract powder has a whitening effect and pigment accumulation inhibition. We chose suitable concentrations at 0.001âŻââŻ1âŻmg/mL of the fruit extract powder to treat with B16F10 cells, presenting no significant cytotoxicity. To investigate the melanin synthesis suppression, we confirmed it by quantitative reverse transcription polymerase chain reaction (qRT-PCR) and western blot in microphthalmia-associated transcription factor (MITF), tyrosinase, tyrosinase-related protein-1 (TRP-1) and dopachrome tautomerase-2 (TRP-2). Notably, as long as the extract concentration was up to 1âŻmg/mL, it has the effect of inhibiting melanin and mushroom tyrosinase. Overall, the extract displayed biofunctions of melanogenesis suppression, so there was a possibility for its application as a food additive and skin-whitening agent
