    Susceptibility to blackheart disorder in potato tubers is influenced by sugar and phenolic profile

    Blackheart (BH) is a physiological disorder of potato tubers in which internal tissue becomes discoloured during storage. The development of BH has been previously linked with general phenolic accumulation. In this study, five potato stocks cv. Maris Piper with different susceptibility to BH were selected across two consecutive seasons, whereupon targeted analysis of sugar and individual phenolic compounds in two tuber sections (flesh and heart) was conducted after storage at 1.5 °C or after one week at 15 °C. The most susceptible stock to BH had the highest accumulation of reducing sugars, while crypto- and neo-chlorogenic acids (chlorogenic acid isomers) were more abundant in flesh tissue of non-susceptible stocks. It is postulated that these metabolites may represent putative pre-symptomatic predictive biomarkers of stock susceptibility to BH

    Remote monitoring and telemedicine in heart failure: implementation and benefits

    Purpose of review: Remote monitoring (RM) of cardiac implantable electronic devices (CIEDs) is recommended as part of the individualized multidisciplinary follow-up of heart failure (HF) patients. Aim of this article is to critically review recent findings on RM, highlighting potential benefits and barriers to its implementation. Recent findings: Device-based RM is useful in the early detection of CIEDs technical issues and cardiac arrhythmias. Moreover, RM allows the continuous monitoring of several patients' clinical parameters associated with impending HF decompensation, but there is still uncertainty regarding its effectiveness in reducing mortality and hospitalizations. Summary: Implementation of RM strategies, together with a proactive physicians' attitude towards clinical actions in response to RM data reception, will make RM a more valuable tool, potentially leading to better outcomes

    Effect of ethylene on postharvest strawberry fruit tissue biochemistry

    The effect of continuous ethylene supplementation (50 ”L L-1) on cold-stored strawberry fruit physiology and biochemistry, including phytohormone (abscisic acid) metabolism was investigated. In comparison with control fruit which exhibited high sucrose and malic acid contents during storage, ethylene-treated fruits showed increased respiration, sucrose hydrolysis and concomitant reducing sugars accumulation. Ethylene supplementation did not have any effect on phenolic profile. ABA biosynthesis, in both flesh and achenes, was promoted by ethylene. The results herein suggest that controlling ethylene after harvest could suppress senescence and extend shelf-life

    Cancer prevention by targeting angiogenesis

    Healthy individuals can harbour microscopic tumours and dysplastic foci in different organs in an undetectable and asymptomatic state for many years. These lesions do not progress in the absence of angiogenesis or inflammation. Targeting both processes before clinical manifestation can prevent tumour growth and progression. Angioprevention is a chemoprevention approach that interrupts the formation of new blood vessels when tumour cell foci are in an indolent state. Many efficacious chemopreventive drugs function by preventing angiogenesis in the tumour microenvironment. Blocking the vascularization of incipient tumours should maintain a dormancy state such that neoplasia or cancer exist without disease. The current limitations of antiangiogenic cancer therapy may well be related to the use of antiangiogenic agents too late in the disease course. In this Review, we suggest mechanisms and strategies for using antiangiogenesis agents in a safe, preventive clinical angioprevention setting, proposing different levels of clinical angioprevention according to risk, and indicate potential drugs to be employed at these levels. Finally, angioprevention may go well beyond cancer in the prevention of a range of chronic disorders where angiogenesis is crucial, including different forms of inflammatory or autoimmune diseases, ocular disorders, and neurodegeneration

    Assuring potato tuber quality during storage: A future perspective

    Potatoes represent an important staple food crop across the planet. Yet, to maintain tuber quality and extend availability, there is a necessity to store tubers for long periods often using industrial-scale facilities. In this context, preserving potato quality is pivotal for the seed, fresh and processing sectors. The industry has always innovated and invested in improved post-harvest storage. However, the pace of technological change has and will continue to increase. For instance, more stringent legislation and changing consumer attitudes have driven renewed interest in creating alternative or complementary post-harvest treatments to traditional chemically reliant sprout suppression and disease control. Herein, the current knowledge on biochemical factors governing dormancy, the use of chlorpropham (CIPC) as well as existing and chemical alternatives, and the effects of pre- and post-harvest factors to assure potato tuber quality is reviewed. Additionally, the role of genomics as a future approach to potato quality improvement is discussed. Critically, and through a more industry targeted research, a better mechanistic understanding of how the pre-harvest environment influences tuber quality and the factors which govern dormancy transition should lead to a paradigm shift in how sustainable storage can be achieved

    A predation assay using amoebae to screen for virulence factors unearthed the first W. chondrophila inclusion membrane protein.

    Waddlia chondrophila is an intracellular bacterium phylogenetically related to the well-studied human and animal pathogens of the Chlamydiaceae family. In the last decade, W. chondrophila was convincingly demonstrated to be associated with adverse pregnancy outcomes in humans and abortions in animals. All members of the phylum Chlamydiae possess a Type Three Secretion System that they use for delivering virulence proteins into the host cell cytosol to modulate their environment and create optimal conditions to complete their life cycle. To identify W. chondrophila virulence proteins, we used an original screening approach that combines a cosmid library with an assay monitoring resistance to predation by phagocytic amoebae. This technique combined with bioinformatic data allowed the identification of 28 candidate virulence proteins, including Wimp1, the first identified inclusion membrane protein of W. chondrophila

    Effect of selenium enrichment on metabolism of tomato (Solanum lycopersicum) fruit during post‐harvest ripening

    BACKGROUND Selenium (Se) enrichment of plants seems effective in enhancing the health‐related properties of produce, and in delaying plant senescence and fruit ripening. The current study investigated the effects of Se on tomato fruit ripening. Tomato (Solanum lycopersicum L.) plants were grown in hydroponics with different Se‐enriched nutrient solutions. Se, as sodium selenate, was added at rate of 0 mg L‐1 (control), 1 mg L‐1, and 1.5 mg L‐1. RESULTS Selenium was absorbed by roots and translocated to leaves and fruit. Se enrichment did not significantly affect the qualitative parameters of fruit at commercial harvest, instead it delayed ripening by affecting specific ripening‐related processes (respiration, ethylene production, color evolution) during postharvest. In the current experiment 100 g of tomato hydroponically grown with a 1.5 mg Se L‐1 enriched solution provided a total of 23.7 ÎŒg Se. Selenium recommended daily intake is 60 ÎŒg for women and 70 ÎŒg for men, thus the daily consumption of 100 g of enriched tomato would not lead to Se toxicity, but would provide a good Se diet supplementation. CONCLUSIONS The cultivation of tomato plants in a Se‐enriched solution appeared effective in producing tomato fruit with improved performances during storage and postharvest shelf life, and also with greater potential health‐promoting properties

    Double-Tuned Birdcage Radio Frequency Coil for 7 T MRI: Optimization, Construction and Workbench Validation

    The aim of the present study is the optimization, construction, and workbench validation of a double-tuned 1H- 23Na volume radio frequency (RF) coil suitable for human head imaging at 7 T, based on the birdcage geometry. The birdcage-like design which is considered is the four-ring model, in which two standard birdcage-like structures with the same diameters are nested along the longitudinal axis. Simulations based on Maxwell’s equations are performed to evaluate the RF magnetic field homogeneity and the RF coil efficiency varying the coil geometrical parameters. The RF magnetic field homogeneity is evaluated both on the transverse (z = 0) and longitudinal (y = 0) planes without performing the impedance matching procedure, so that the RF coil symmetry is not perturbed by the matching network. The RF coil efficiency is instead dependent on the effective coil input RF power, and it is evaluated after matching the coil, so that the reflected power is minimized, assuming that the stimulation power is totally delivered to the RF coil. Considering the simulation results and the target application, the useful RF coil geometrical parameters are fixed. The four-ring model, which showed the best performances, has been built and tested on a workbench, using a cylindrical phantom filled with a 0.05 M saline solution as load. This provides the first example of a four-ring realization intended 1H- 23Na for human head imaging at 7 T

    Upper limb children action-observation training (UP-CAT): a randomised controlled trial in Hemiplegic Cerebral Palsy

    BACKGROUND: Rehabilitation for children with hemiplegic cerebral palsy (HCP) aimed to improve function of the impaired upper limb (UL) uses a wide range of intervention programs. A new rehabilitative approach, called Action-Observation Therapy, based on the recent discovery of mirror neurons, has been used in adult stroke but not in children. The purpose of the present study is to design a randomised controlled trial (RCT) for evaluating the efficacy of Action-Observation Therapy in improving UL activity in children with HCP.METHODS/DESIGN: The trial is designed according to CONSORT Statement. It is a randomised, evaluator-blinded, match-pair group trial. Children with HCP will be randomised within pairs to either experimental or control group. The experimental group will perform an Action-Observation Therapy, called UP-CAT (Upper Limb-Children Action-Observation Training) in which they will watch video sequences showing goal-directed actions, chosen according to children UL functional level, combined with motor training with their hemiplegic UL. The control group will perform the same tailored actions after watching computer games. A careful revision of psychometric properties of UL outcome measures for children with hemiplegia was performed. Assisting Hand Assessment was chosen as primary measure and, based on its calculation power, a sample size of 12 matched pairs was established. Moreover, Melbourne and ABILHAND-Kids were included as secondary measures. The time line of assessments will be T0 (in the week preceding the onset of the treatment), T1 and T2 (in the week after the end of the treatment and 8 weeks later, respectively). A further assessment will be performed at T3 (24 weeks after T1), to evaluate the retention of effects. In a subgroup of children enrolled in both groups functional Magnetic Resonance Imaging, exploring the mirror system and sensory-motor function, will be performed at T0, T1 and T2.DISCUSSION: The paper aims to describe the methodology of a RCT for evaluating the efficacy of Action-Observation Therapy in improving UL activity in children with hemiplegia. This study will be the first to test this new type of treatment in childhood. The paper presents the theoretical background, study hypotheses, outcome measures and trial methodology
