Effect of Monthly, High-Dose, Long-Term Vitamin D on Lung Function: A Randomized Controlled Trial.
Although observational studies suggest positive vitamin D-lung function associations, randomized trials are inconsistent. We examined effects of vitamin D supplementation on lung function. We recruited 442 adults (50-84 years, 58% male) into a randomized, double-blinded, placebo-controlled trial. Participants received, for 1.1 years (median; range = 0.9-1.5 years), either (1) vitamin D₃ 200,000 IU, followed by monthly 100,000 IU doses (n = 226); or (2) placebo monthly (n = 216). At baseline and follow-up, spirometry yielded forced expiratory volume in 1 s (FEV1; primary outcome). Mean (standard deviation) 25-hydroxyvitamin D increased from 61 (24) nmol/L at baseline to 119 (45) nmol/L at follow-up in the vitamin D group, but was unchanged in the placebo group. There were no significant lung function improvements (vitamin D versus placebo) in the total sample, vitamin D-deficient participants or asthma/chronic obstructive pulmonary disease (COPD) participants. However, among ever-smokers (n = 217), the mean (95% confidence interval) FEV1 increase in the vitamin D versus placebo was 57 (4, 109) mL (p = 0.03). FEV1 increases were larger among vitamin D-deficient ever-smokers (n = 54): 122 (8, 236) mL (p = 0.04). FEV1 improvements were largest among ever-smokers with asthma/COPD (n = 60): 160 (53, 268) mL (p = 0.004). Thus, vitamin D supplementation did not improve lung function among everyone, but benefited ever-smokers, especially those with vitamin D deficiency or asthma/COPD
First-principles study of ternary fcc solution phases from special quasirandom structures
In the present work, ternary Special Quasirandom Structures (SQSs) for a fcc
solid solution phase are generated at different compositions,
and , ,
whose correlation functions are satisfactorily close to those of a random fcc
solution. The generated SQSs are used to calculate the mixing enthalpy of the
fcc phase in the Ca-Sr-Yb system. It is observed that first-principles
calculations of all the binary and ternary SQSs in the Ca-Sr-Yb system exhibit
very small local relaxation. It is concluded that the fcc ternary SQSs can
provide valuable information about the mixing behavior of the fcc ternary solid
solution phase. The SQSs presented in this work can be widely used to study the
behavior of ternary fcc solid solutions.Comment: 20 pages, 7 figure
Sulfate transporters involved in sulfate secretion in the kidney are localized in the renal proximal tubule II of the elephant fish (Callorhinchus milii)
Most vertebrates, including cartilaginous fishes, maintain their plasma SO4 (2-) concentration ([SO4 (2-)]) within a narrow range of 0.2-1 mM. As seawater has a [SO4 (2-)] about 40 times higher than that of the plasma, SO4 (2-) excretion is the major role of kidneys in marine teleost fishes. It has been suggested that cartilaginous fishes also excrete excess SO4 (2-) via the kidney. However, little is known about the underlying mechanisms for SO4 (2-) transport in cartilaginous fish, largely due to the extraordinarily elaborate four-loop configuration of the nephron, which consists of at least 10 morphologically distinguishable segments. In the present study, we determined cDNA sequences from the kidney of holocephalan elephant fish (Callorhinchus milii) that encoded solute carrier family 26 member 1 (Slc26a1) and member 6 (Slc26a6), which are SO4 (2-) transporters that are expressed in mammalian and teleost kidneys. Elephant fish Slc26a1 (cmSlc26a1) and cmSlc26a6 mRNAs were coexpressed in the proximal II (PII) segment of the nephron, which comprises the second loop in the sinus zone. Functional analyses using Xenopus oocytes and the results of immunohistochemistry revealed that cmSlc26a1 is a basolaterally located electroneutral SO4 (2-) transporter, while cmSlc26a6 is an apically located, electrogenic Cl(-)/SO4 (2-) exchanger. In addition, we found that both cmSlc26a1 and cmSlc26a6 were abundantly expressed in the kidney of embryos; SO4 (2-) was concentrated in a bladder-like structure of elephant fish embryos. Our results demonstrated that the PII segment of the nephron contributes to the secretion of excess SO4 (2-) by the kidney of elephant fish. Possible mechanisms for SO4 (2-) secretion in the PII segment are discussed
Potential for energy production from farm wastes using anaerobic digestion in the UK : An economic comparison of different size plants
Anaerobic digestion (AD) plants enable renewable fuel, heat, and electricity production, with their efficiency and capital cost strongly dependent on their installed capacity. In this work, the technical and economic feasibility of different scale AD combined heat and power (CHP) plants was analyzed. Process configurations involving the use of waste produced in different farms as feedstock for a centralized AD plant were assessed too. The results show that the levelized cost of electricity are lower for large-scale plants due to the use of more efficient conversion devices and their lower capital cost per unit of electricity produced. The levelized cost of electricity was estimated to be 4.3 p/kWhe for AD plants processing the waste of 125 dairy cow sized herds compared to 1.9 p/kWhe for AD plants processing waste of 1000 dairy cow sized herds. The techno-economic feasibility of the installation of CO2 capture units in centralized AD-CHP plants was also undertaken. The conducted research demonstrated that negative CO2 emission AD power generation plants could be economically viable with currently paid feed-in tariffs in the UK
A comparison of lead-based and lead-free bullets for shooting sambar deer (Cervus unicolor) in Australia
Context. In response to the toxic health threats posed by lead (Pb), there is currently a focus on transitioning to lead-free bullets for shooting wild animals. Aim. We aimed to quantify the killing efficiency and animal welfare outcomes of lead-based and lead-free (copper-based) bullets for ground-based shooting of sambar deer (Cervus unicolor) in Victoria, south-eastern Australia. Methods. We used shooter-collected data from recreational diurnal hunting and professional nocturnal culling during 2020–2021. Shooters recorded rifle calibre, cartridge type, bullet mass, bullet type, shooting outcomes (miss, wound or kill), shooting distance, flight distance (the distance between where the animal was shot and where it died) as an assumed positive correlate of time to incapacitation, anatomical zones struck by bullets, and frequency of bullet exit wounds. We used flight distance as our response variable, assuming that it is positively correlated with time to incapacitation. To examine the role of several predictor variables (including bullet type) potentially influencing flight distance, the dataset was reduced to those deer killed with a single thoracic shot. Key results. Our data captured shooting events involving 276 deer, with 124 deer shot at with lead-based bullets and 152 with copper-based bullets. Most (87%) of the deer were killed with a single shot. The frequency of non-fatal wounding was <4% for both bullet types and there was no distinct difference in the probability of a single shot kill for deer shot with either bullet type. For those deer killed with a single thoracic shot (n = 198), there was no evidence that bullet energy or shooting distance influenced flight distance. After accounting for differences in terminal kinetic energy, the mean flight distance of deer shot with lead-free bullets (35 m) was 56% greater than that of deer shot with lead-based bullets (22 m). Conclusions. Lead-based and lead-free bullets produced similar animal welfare outcomes for shooting sambar deer. Implications. The sound of street corner society: UK grime music as ethnography
This article explores the ways in which popular music can be linked to ethnography. While there is a tradition of connecting popular music with sociology, this article posits a further resonance that is not so much theoretical as methodological. The article suggests that forms of contemporary popular music parallel key facets of ethnography, not simply in terms of sociological analysis, but with regard to popular music as an ethnographic resource, as ‘data’, and as the reflexive expression of Paul Willis’ conception of the ‘ethnographic imagination’; and the article argues that contemporary British hip-hop in the form of ‘grime’ is a potent exemplar. This is due to the resolutely cultural, spatial nature of grime music: a factor that marks out grime as a distinctive musical genre and a distinctive ethnographic form, as it is an experientially rooted music about urban locations, made from within those urban locations

Shwachman-Bodian-Diamond syndrome (SBDS) protein is a direct inhibitor of protein phosphatase 2A (PP2A) activity and overexpressed in acute myeloid leukaemia.
Protein phosphatase 2A (PP2A) is a serine/threonine phosphatase inactivated in many cancers including acute myeloid leukaemia (AML). Activation of PP2A is emerging as a therapeutic strategy, however the mechanisms underpinning PP2A inhibition are not well understood. Using myeloid progenitor cells harbouring oncogenic mutant c-KIT and characterised by PP2A inhibition, we have identified the ribosome biogenesis protein SBDS, as a target of the PP2A activating drugs FTY720 and AAL(S). We show SBDS binds to PP2A complexes comprised of the B55α regulatory subunit of PP2A. shRNA mediated knockdown of SBDS increased PP2A activity and induced apoptosis. At diagnosis, AML patients expressed significantly more SBDS mRNA than healthy controls, with relapsed patients expressing significantly more SBDS mRNA than both healthy controls and patients at diagnosis. Together, our data presents a role for SBDS in the dysregulation of PP2A in AML
Processes and experiences of creative cognition in seven Western classical composers
In a qualitative study, we explored the range of reflections and experiences involved in the composition of score-based music by administering a 15-item, open-ended, questionnaire to seven professional composers from Europe and North America. Adopting a grounded theory approach, we organized six different codes emerging from our data into two higher-order categories ( the act of composing and establishing relationships). Our content analysis, inspired by the theoretical resources of 4E cognitive science, points to three overlapping characteristics of creative cognition in music composition: it is largely exploratory, it is grounded in bodily experience, and it emerges from the recursive dialogue of agents and their environment. More generally, such preliminary findings suggest that musical creativity may be advantageously understood as a process of constant adaptation – one in which composers enact their musical styles and identities by exploring novel interactivities hidden in their contingent and historical milieux
