Blended not flipped (with apologies to James Bond)
The ability to make learning resources readily available to students via course websites can mean that attendance falls and learning suffers. To address this it is necessary to engage students in different ways which provides an opportunity to change the delivery of courses. But the implementation of blended learning or the flipped classroom with greater engagement and interactivity with peers and learning content is not something that can be simply done. Each approach has its own particular pedagogy and requires particular support mechanisms. Here we present our experience of a coordinated approach to provide students with a comprehensive set of online learning resources and engage them in the classroom to encourage attendance and potentially deeper and more meaningful learning outcomes. The target group was first year environmental science students in their first semester. Three courses were adjusted to various levels to incorporate elements of a flipped classroom approach and/or a blended approach. Course coordinators worked with a pedagogical expert and an experienced science specialist who has considerable experience in digital and innovative approaches to university teaching. The paper presents each person’s goals, approaches and perceptions of the extent to which they were successful. Students’ impressions gleaned from their responses to the standard assessment of teaching will also be presented
Progress, challenges and opportunities for Red Listing
Despite its recognition as an important global resource for conservation, the International Union for Conservation of Nature's (IUCN) Red List of Threatened Species only provides assessments of extinction risk for a small and biased subset of known biodiversity. A more complete Red List can better support species-level conservation by indicating how quickly we need to act on species deemed to be priorities for conservation action.Vascular plants represent one of the Red List knowledge gaps, with only 7% of species currently on the Red List (including in the Data Deficient and Least Concern categories). Using vascular plants as a case study we highlight how recent developments, such as changes to rules, improvements to data management systems, better assessment tools and training, can support Red List assessment activity. We also identify ongoing challenges, such as the need to support regional and national assessment initiatives, the largely voluntary nature of the Red List community, as well as the need to meet core operating costs for the Red List. Finally, we highlight how new opportunities such as automation and batch uploading can fast-track assessments, and how better monitoring of assessment growth can help assess the impact of new developments. Most of our findings are also applicable to other species-rich groups that are under-represented on the Red List.We examine trends in plant Red Listing and conclude that the rate of new assessments has not increased in line with what would be required to reach goals such as the Barometer of Life. This may result partly from a lag between recent changes and their effects, but further progress can be made by realising the opportunities outlined here and by growing the Red List community and strengthening collaboration with IUCN
Designer artificial membrane binding proteins direct stem cells to the myocardium
International audienceWe present a new cell membrane modification methodology where the inherent heart tissue homing properties of the infectious bacteria Streptococcus gordonii are transferred to human stem cells. This is achieved via the rational design of a chimeric protein-polymer surfactant cell membrane binding construct, comprising the cardiac fibronectin (Fn) binding domain of the bacterial adhesin protein CshA fused to a supercharged protein. Significantly, the protein-polymer surfactant hybrid spontaneously inserts into the plasma membrane of stem cells without cytotoxicity, instilling the cells with a high affinity for immobilized fibronectin. Moreover, we show that this cell membrane reengineering approach significantly improves retention and homing of stem cells delivered either intracardially or intravenously to the myocardium in a mouse model
Mythical numbers and the proceeds of organised crime: estimating mafia proceeds in Italy
Organised crime is a field vulnerable to mythical numbers, i.e. exaggerated estimates lacking empirical support, but acquiring acceptance through repetition. The figures on mafia proceeds in Italy are a striking example of this problem. This study proposes an estimation of mafia proceeds in Italy from nine criminal activities (sexual exploitation of women, illicit firearms trafficking, drug trafficking, counterfeiting, the illicit cigarette trade, illicit gambling, illicit waste disposal, loan sharking, and extortion racketeering) by region and type of mafia (Cosa Nostra, Camorra,\u2018Ndrangheta, Apulian
mafias, and other mafias). The results estimate yearly mafia proceeds at approximately \u20ac10.7 bn (0.7% of the Italian GDP), discussing the impact on the regional and national economies and the differences among the types of mafias as to their geographical sources of revenue
Introducing EDEN ISS - A European project on advancing plant cultivation technologies and operations
Plant cultivation in large-scale closed environments is challenging and several key
technologies necessary for space-based plant production are not yet space-qualified
or remain in early stages of development. The EDEN ISS project foresees
development and demonstration of higher plant cultivation technologies, suitable for
future deployment on the International Space Station and from a long-term
perspective, within Moon and Mars habitats. The EDEN ISS consortium will design
and test essential plant cultivation technologies using an International Standard
Payload Rack form factor cultivation system for potential testing on-board the
International Space Station. Furthermore, a Future Exploration Greenhouse will be
designed with respect to future planetary bio-regenerative life support system
deployments. The technologies will be tested in a laboratory environment as well as
at the highly-isolated German Antarctic Neumayer Station III. A small and mobile
container-sized test facility will be built in order to provide realistic mass flow
relationships. In addition to technology development and validation, food safety and
plant handling procedures will be developed. This paper describes the goals and
objectives of EDEN ISS and the different project phases and milestones.
Furthermore, the project consortium will be introduced and the role of each partner
within the project is explained
Minimal information for studies of extracellular vesicles 2018 (MISEV2018):a position statement of the International Society for Extracellular Vesicles and update of the MISEV2014 guidelines
The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles (“MISEV”) guidelines for the field in 2014. We now update these “MISEV2014” guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. Sex difference and intra-operative tidal volume: Insights from the LAS VEGAS study
BACKGROUND: One key element of lung-protective ventilation is the use of a low tidal volume (VT). A sex difference in use of low tidal volume ventilation (LTVV) has been described in critically ill ICU patients.OBJECTIVES: The aim of this study was to determine whether a sex difference in use of LTVV also exists in operating room patients, and if present what factors drive this difference.DESIGN, PATIENTS AND SETTING: This is a posthoc analysis of LAS VEGAS, a 1-week worldwide observational study in adults requiring intra-operative ventilation during general anaesthesia for surgery in 146 hospitals in 29 countries.MAIN OUTCOME MEASURES: Women and men were compared with respect to use of LTVV, defined as VT of 8 ml kg-1 or less predicted bodyweight (PBW). A VT was deemed 'default' if the set VT was a round number. A mediation analysis assessed which factors may explain the sex difference in use of LTVV during intra-operative ventilation.RESULTS: This analysis includes 9864 patients, of whom 5425 (55%) were women. A default VT was often set, both in women and men; mode VT was 500 ml. Median [IQR] VT was higher in women than in men (8.6 [7.7 to 9.6] vs. 7.6 [6.8 to 8.4] ml kg-1 PBW, P < 0.001). Compared with men, women were twice as likely not to receive LTVV [68.8 vs. 36.0%; relative risk ratio 2.1 (95% CI 1.9 to 2.1), P < 0.001]. In the mediation analysis, patients' height and actual body weight (ABW) explained 81 and 18% of the sex difference in use of LTVV, respectively; it was not explained by the use of a default VT.CONCLUSION: In this worldwide cohort of patients receiving intra-operative ventilation during general anaesthesia for surgery, women received a higher VT than men during intra-operative ventilation. The risk for a female not to receive LTVV during surgery was double that of males. Height and ABW were the two mediators of the sex difference in use of LTVV.TRIAL REGISTRATION: The study was registered at Clinicaltrials.gov, NCT01601223
Minimal information for studies of extracellular vesicles (MISEV2023): From basic to advanced approaches
Extracellular vesicles (EVs), through their complex cargo, can reflect the state of their cell of origin and change the functions and phenotypes of other cells. These features indicate strong biomarker and therapeutic potential and have generated broad interest, as evidenced by the steady year-on-year increase in the numbers of scientific publications about EVs. Important advances have been made in EV metrology and in understanding and applying EV biology. However, hurdles remain to realising the potential of EVs in domains ranging from basic biology to clinical applications due to challenges in EV nomenclature, separation from non-vesicular extracellular particles, characterisation and functional studies. To address the challenges and opportunities in this rapidly evolving field, the International Society for Extracellular Vesicles (ISEV) updates its 'Minimal Information for Studies of Extracellular Vesicles', which was first published in 2014 and then in 2018 as MISEV2014 and MISEV2018, respectively. The goal of the current document, MISEV2023, is to provide researchers with an updated snapshot of available approaches and their advantages and limitations for production, separation and characterisation of EVs from multiple sources, including cell culture, body fluids and solid tissues. In addition to presenting the latest state of the art in basic principles of EV research, this document also covers advanced techniques and approaches that are currently expanding the boundaries of the field. MISEV2023 also includes new sections on EV release and uptake and a brief discussion of in vivo approaches to study EVs. Compiling feedback from ISEV expert task forces and more than 1000 researchers, this document conveys the current state of EV research to facilitate robust scientific discoveries and move the field forward even more rapidly
