Effects of serum and serum heat-inactivation on human bone derived osteoblast progenitor cells
Generally, heat inactivated foetal calf serum (FCS) containing media are used for the cultivation of animal and human cells. The role of serum source and serum treatment on the behaviour of cells has long been neglected. The present study was performed to investigate the effects of serum heat inactivation and serum source on trabecular bone derived progenitor cells (HBC). Furthermore, it was investigated in how far these reactions differed from those seen in bone marrow derived mesenchymal progenitor cells (HBMC) cultures. We found that HBC cultures performed differently in the presence of FCS and HS with or without heat inactivation. The reactions similar to some degree those observed in HBMC cultures. Felines Sarcoid bei einer 1-jährigen europäischen Hauskatze ausgelöst durch bovines Papillomavirus Typ 14 in der Schweiz
Eine einjährige Bauernhofkatze zeigte einen 5 cm grossen Knoten an der linken Nasenöffnung, der aufgrund seiner Lokalisation nur unvollständig entfernt werden konnte. Hinsichtlich der infiltrativ wachsenden, spindelförmigen Proliferation wurde histologisch ein felines Sarkoid diagnostiziert. Die aus dem Gewebe isolierte DNA konnte durch zwei PCRs als solche von bovinem Papillomavirus 14 (BPV-14) identifiziert werden. Die 194 und 549 Basenpaare (bp) langen, amplifizierten Sequenzen waren 99 bzw. 100% identisch mit einer von einem in den USA isolierten Virus, welches mit felinem Sarkoid in Zusammenhang gebracht wurde. Trotz unvollständiger Exzision, ist bis 10 Monate nach der Operation kein Rezidiv aufgetreten
Overexpression of the p73 gene is a novel finding in high-risk B-cell chronic lymphocytic leukemia
The p73 protein shares structural and functional similarities with the tumour-suppressor p53, but its role in neoplastic transformation is unknown. Alternative splicing leads to the expression of at least nine p73 C-terminal mRNA splice variants (α β γ δ ε ξ η ηl θ). In this survey, we analyse the expression of p73 by real-time quantitative RT-PCR, its known C-terminal variants with an RT-PCR-Southern tech nique and by Western blot in samples of 51 patients with B-CLL, normal B lymphocytes from eight individuals, and five haematopoetic cell lines. p73α protein expression positively correlated with higher risk B-CLL stages (P=0.046). Total p73 mRNA expression was higher (P= 0.01) and p73α protein more frequently detected (P=0.008) in B-CLL compared with normal CD19+—B-lymphocytes. p73 C-terminal mRNA variants were expressed both in B-CLL and in normal B-lymphocytes, but their expression was biased since the γ (P=0.041), the θ (P ≪ 0.001), and the η variant (P=0.033) prevailed in normal B-lymphocytes. In summary, we conclude that the accumulation of p73, the expression pattern of particular p73 variants and its link to progression may play a distinct role in the molecular pathology B-CL
Reduced dose of subcutaneous cladribine induces identical response rates but decreased toxicity in pretreated chronic lymphocytic leukaemia
Purpose To study the efficacy and the safety of cladribine (2-chlorodeoxyadenosine, 2-CDA) administered as 24-hour infusions or as subcutaneous bolus injections at two different doses to patients with relapsing or refractory chronic lymphocytic leukaemia (CLL). Patients and methods In this non randomised 2-cohort study, 20 patients with pretreated CLL received cladribine at a dose of 0.7 mg/kg/cycle as continuous i.v. infusions over seven days (group 1) and 35 patients were treated at a reduced dose of 0.5 mg/kg/cycle given as s.c. bolus injections for five days (group 2). After two cycles of four week duration, response was assessed. In the case of progressive disease, therapy was discontinued, otherwise a maximum of four additional cycles were administered until best response. Results A total of 130 cycles were administered (group 1: 41, group 2: 89). Patient characteristics in both groups were comparable. The median dose intensities were 0.172 mg/kg per week and 0.123 mg/kg per week for groups 1 and 2, respectively (P ≤ 0.0001). The overall response rate for all 55 patients was 38% (95% confidence interval (95% CI): 25%-52%), with 5% CR and 33% PR. Response was similar in both patient groups (35% in group 1, 40% in group 2). No association between cladribine dose intensity and response rate was found, and there was no difference between patients relapsing after or refractory to previous therapies (11 of 24 vs. 10 of 31). Median remission duration was six months in both groups. Toxicity, in particular infections (all WHO grades, 34% in group 1 versus 7% in group 2) and myelosuppression (grade 1-4 neutropenia, 72% versus 41% of cladribine cycles) were statistically significantly more frequent in group 1. Conclusion Cladribine is active in heavily pretreated patients with chronic lymphocytic leukaemias. Dose reduction by 29% led to similar response and remission duration, but to a significant decrease of myelotoxicity and risk of infection. Cladribine administered as s.c. bolus injections at 0.5 mg/kg per cycle is safe and this dose level should not be exceeded in this patient populatio
Real-time source apportionment of organic aerosols in three European cities.
97% of the urban population in the EU in 2019 were exposed to an annual fine particulate matter level higher than the World Health Organization (WHO) guidelines (5 ÎĽg/m3). Organic aerosol (OA) is one of the major air pollutants, and the knowledge of its sources is crucial for designing cost-effective mitigation strategies. Positive matrix factorization (PMF) on aerosol mass spectrometer (AMS) or aerosol chemical speciation monitor (ACSM) data is the most common method for source apportionment (SA) analysis on ambient OA. However, conventional PMF requires extensive human labor, preventing the implementation of SA for routine monitoring applications. This study proposes the source finder real-time (SoFi RT, Datalystica Ltd.) approach for efficient retrieval of OA sources. The results generated by SoFi RT agree remarkably well with the conventional rolling PMF results regarding factor profiles, time series, diurnal patterns, and yearly relative contributions of OA factor on three year-long ACSM data sets collected in Athens, Paris, and Zurich. Although the initialization of SoFi RT requires a priori knowledge of OA sources (i.e., the approximate number of factors and relevant factor profiles) for the sampling site, this technique minimizes user interactions. Eventually, it could provide up-to-date trustable information on timescales useful to policymakers and air quality modelers
Designing visual analytics methods for massive collections of movement data
Exploration and analysis of large data sets cannot be carried out using purely visual means but require the involvement of database technologies, computerized data processing, and computational analysis methods. An appropriate combination of these technologies and methods with visualization may facilitate synergetic work of computer and human whereby the unique capabilities of each “partner” can be utilized. We suggest a systematic approach to defining what methods and techniques, and what ways of linking them, can appropriately support such a work. The main idea is that software tools prepare and visualize the data so that the human analyst can detect various types of patterns by looking at the visual displays. To facilitate the detection of patterns, we must understand what types of patterns may exist in the data (or, more exactly, in the underlying phenomenon). This study focuses on data describing movements of multiple discrete entities that change their positions in space while preserving their integrity and identity. We define the possible types of patterns in such movement data on the basis of an abstract model of the data as a mathematical function that maps entities and times onto spatial positions. Then, we look for data transformations, computations, and visualization techniques that can facilitate the detection of these types of patterns and are suitable for very large data sets – possibly too large for a computer's memory. Under such constraints, visualization is applied to data that have previously been aggregated and generalized by means of database operations and/or computational techniques
Massive habitat-specific genomic response in D. melanogaster populations during experimental evolution in hot and cold environments
Experimental evolution in combination with whole-genome sequencing (evolve and resequence [E&R]) is a promising approach to define the genotype–phenotype map and to understand adaptation in evolving populations. Many previous studies have identified a large number of putative selected sites (i.e., candidate loci), but it remains unclear to what extent these loci are genuine targets of selection or experimental noise. To address this question, we exposed the same founder population to two different selection regimes—a hot environment and a cold environment—and quantified the genomic response in each. We detected large numbers of putative selected loci in both environments, albeit with little overlap between the two sets of candidates, indicating that most resulted from habitat-specific selection. By quantifying changes across multiple independent biological replicates, we demonstrate that most of the candidate SNPs were false positives that were linked to selected sites over distances much larger than the typical linkage disequilibrium range of Drosophila melanogaster. We show that many of these mid- to long-range associations were attributable to large segregating inversions and confirm by computer simulations that such patterns could be readily replicated when strong selection acts on rare haplotypes. In light of our findings, we outline recommendations to improve the performance of future Drosophila E&R studies which include using species with negligible inversion loads, such as D. mauritiana and D. simulans, instead of D. melanogaster
