    Project-based learning module on creativity and entrepreneurship products subject: Validity and empirical effect

    The learning process for Creativity and Entrepreneurship Products subjects in Vocational High Schools still uses printed books which are verbal in nature so this will make it difficult for students to learn the learning material. Apart from that, the learning method used the lecture method which is certainly less effective in supporting students' understanding of the material so that learning is more teacher-centred. Meanwhile, the demand for the current ‘Merdeka’ curriculum is that learning must be student-centered. This research aimed at developing learning tools of PjBL (Project Based Learning) based teaching modules on the Product Creativity and Entrepreneurship subjects. This research was a type of research and development using the ADDIE development model. There were 12 experts who validate this module from 3 aspects, namely material, media and model, and language aspects and there were 30 students for limited trials. The results of this research showed that this module was valid and has a positive effect in supporting the learning process for the Product Creativity and Entrepreneurship subjects. This research had implications for the effectiveness of Creativity and Entrepreneurship Products learning and being one method that can be used by teachers in this learning

    Constructivist Computer-Based Instruction (CBI) Approach: A CBI Flipped Learning Integrated Problem Based and Case Method (PBL-cflip) in Clinical Refraction Course

    Based on the preliminary study conducted, it was revealed that critical thinking skills, creativity, and student learning outcomes were low in clinical refraction courses. Whereas in 21st-century learning, students must master the skills of critical thinking, communication, collaboration, and creativity (4Cs) as recommended. Because of this phenomenon, it is critical to use the appropriate learning approach; one solution offered is CBI Flipped Learning Integrated Problem-Based and Case Method (PBL-cflip). The purpose of this study is to describe the effectiveness of the CBI Flipped Learning Integrated Problem Based and Case Method (PBL-cflip) in improving student learning outcomes in clinical refraction courses. This research is quantitative, using a quasi-experimental method. Participants in this study were 61 vocational education students who took the clinical refraction course. The results showed an increase in students' critical thinking, collaboration, communication, and creativity skills, which helped them achieve the expected learning outcomes. It can be concluded that this PBL-cflip is very useful for students to understand and solve cases in clinical refraction courses and can create an interesting learning atmosphere according to student interests, making students more enthusiastic about participating in learning.     Enhancing Sesquiterpenoid Production from Methane via Synergy of the Methylerythritol Phosphate Pathway and a Short-Cut Route to 1-Deoxy-D-xylulose 5-Phosphate in Methanotrophic Bacteria

    Sesquiterpenoids are one of the most diverse classes of isoprenoids which exhibit numerous potentials in industrial biotechnology. The methanotrophs-based methane bioconversion is a promising approach for sustainable production of chemicals and fuels from methane. With intrinsic high carbon flux though the ribulose monophosphate cycle in Methylotuvimicrobium alcaliphilum 20Z, we demonstrated here that employing a short-cut route from ribulose 5-phosphate to 1-deoxy-d-xylulose 5-phosphate (DXP) could enable a more efficient isoprenoid production via the methylerythritol 4-phosphate (MEP) pathway, using α-humulene as a model compound. An additional 2.8-fold increase in α-humulene production yield was achieved by the fusion of the nDXP enzyme and DXP reductase. Additionally, we utilized these engineering strategies for the production of another sesquiterpenoid, α-bisabolene. The synergy of the nDXP and MEP pathways improved the α-bisabolene titer up to 12.24 ± 0.43 mg/gDCW, twofold greater than that of the initial strain. This study expanded the suite of sesquiterpenoids that can be produced from methane and demonstrated the synergistic uses of the nDXP and MEP pathways for improving sesquiterpenoid production in methanotrophic bacteria

    Riboswitches and toehold switches are considered to have potential for implementation in various fields, i.e., biosensing, metabolic engineering, and molecular diagnostics. The specific binding, programmability, and manipulability of these RNA-based molecules enable their intensive deployments in molecular detection as biosensors for regulating gene expressions, tracking metabolites, or detecting RNA sequences of pathogenic microorganisms. In this review, we will focus on the development of riboswitches and toehold switches in biosensing and molecular diagnostics. This review introduces the operating principles and the notable design features of riboswitches as well as toehold switches. Moreover, we will describe the advances and future directions of riboswitches and toehold switches in biosensing and molecular diagnostics
