    Electroanalysis of Imidacloprid Insecticide in River Waters Using Functionalized Multi-Walled Carbon Nanotubes Modified Glassy Carbon Electrode

    In this work, a functionalized multi-walled carbon nanotubes modified glassy carbon electrode (GCE/MWCNT-f) was optimized for the direct determination of imidacloprid (IMC) insecticide in river water. The functionalized material was characterized by infrared spectroscopy with Fourier transform (FTIR) and the modified electrode by scanning electron microscopy (SEM) and cyclic voltammetry (CV). Results revealed that the GCE/MWCNT-f effectively increased the response toward IMC reduction by enhancing the reduction peak current and decreasing the peak potential in comparison with the bare electrode. After optimizing the electroanalytical conditions, the GCE/MWCNT-f showed a linear voltammetric response at concentration ranging from 2.40 × 10−7 to 3.50 × 10−6 mol L−1, with detection and quantification limits of 4.15 × 10−7 mol L−1 and 1.38 × 10−6 mol L−1, respectively. The recovery rate of IMC in spiked river water samples varied from 90–95%.     Cross-cultural adaptation of the City Birth Trauma Scale for the Brazilian context

    Background: Posttraumatic stress disorder consists of a set of symptoms that occurs in response to one or more traumatic events and can occur in postpartum, from traumatic situations related to the birth or to the baby’s health in the first days of life. It is important tracking the presence of birth trauma, but there is not available instruments in the Brazilian context for this purpose. Objectives: To present the cross-cultural adaptation of City Birth Trauma Scale (BiTS) into Brazilian portuguese. Methods: Cross-cultural adaptation involved independent translations, synthesis,back-translation, and submission to the original author’s appreciation. After the scale was subjected to face validity, followed by a pilot study with postpartum mothers. Results: All steps were performed for the cross-cultural adaptation. Regarding face validity, items evaluated concerning different types of equivalence, presented satisfactory agreement values (≥4.20). Most of the expert’s suggestions were followed, being the main ones related to adjustments in prepositions, pronouns and verbal subjects. Pilot study showed that the mothers had been able to understand and respond to the instrument without adjustments. Discussion: BiTS’s Brazilian version proved to be cross-culturally adapted, ensuring the possibility of intercultural data comparison from the semantic, idiomatic, cultural, and conceptual perspectives.     Dietary sulfur amino acid restriction upregulates DICER to confer beneficial effects

    Dietary restriction (DR) improves health and prolongs lifespan in part by upregulating type III endoribonuclease DICER in adipose tissue. In this study, we aimed to specifically test which missing dietary component was responsible for DICER upregulation. Methods: We performed a nutrient screen in mouse preadipocytes and validated the results in vivo using different kinds of dietary interventions in wild type or genetically modified mice and worms, also testing the requirement of DICER on the effects of the diets. Results: We found that sulfur amino acid restriction (i.e., methionine or cysteine) is sufficient to increase Dicer mRNA expression in preadipocytes. Consistently, while DR increases DICER expression in adipose tissue of mice, this effect is blunted by supplementation of the diet with methionine, cysteine, or casein, but not with a lipid or carbohydrate source. Accordingly, dietary methionine or protein restriction mirrors the effects of DR. These changes are associated with alterations in serum adiponectin. We also found that DICER controls and is controlled by adiponectin. In mice, DICER plays a role in methionine restriction-induced upregulation of Ucpl in adipose tissue. In C. elegans, DR and a model of methionine restriction also promote DICER expression in the intestine (an analog of the adipose tissue) and prolong lifespan in a DICER-dependent manner.     Perfil dos indivíduos que receberam atendimento pelo SAMU a vítimas de traumatismo crânioencefálico no município de Anápolis-GO

    Objetivo: traçar o perfil dos indivíduos que receberam atendimento pelo Serviço de Atendimento Móvel de Urgência (SAMU) vítimas de traumatismo crânio encefálico (TCE), no município de Anápolis-GO.  Métodos: Este estudo foi do tipo descritivo, documental e retrospectivo. Este estudo foi pautado na abordagem quantitativa de 63 fichas de atendimento de usuários do serviço com traumatismo crânioencefálico no período de janeiro a dezembro de 2014. Os dados colhidos das fichas dos usuários foram tabulados em planilhas do programa Microsoft Excel. A partir do banco de dados foi realizada a estatística descritiva com frequência absoluta e relativa. Resultados: Dos 63 atendimentos realizados pelo SAMU com diagnóstico de TCE 80,9% eram do sexo masculino e 19,1% do sexo feminino. A faixa etária prevaleceu no intervalo entre 21 a 40 anos (31,7%) e quanto à classificação de risco de TCE foi observado que 41,3% dos indivíduos estavam com risco moderado causado por perfuração por arma de fogo (20,6%) seguido de queda ao mesmo nível; e em relação à evolução clínica dos usuários 19% dos casos foram a óbito por TCE. Conclusão: Concluiu-se que no estudo o sexo masculino com faixa etária entre 21 a 40 anos com risco moderado de TCE causado por perfuração por arma de fogo e queda ao mesmo nível compõe grupo de risco. Os perfis de atendimentos realizados pelo SAMU as vítimas de TCE são muito importantes para que haja uma melhoria no atendimento pré e intra-hospitalar e em campanhas educativas mais eficientes para a população

    H1N1pdm Influenza Infection in Hospitalized Cancer Patients: Clinical Evolution and Viral Analysis

    BACKGROUND: The novel influenza A pandemic virus (H1N1pdm) caused considerable morbidity and mortality worldwide in 2009. The aim of the present study was to evaluate the clinical course, duration of viral shedding, H1N1pdm evolution and emergence of antiviral resistance in hospitalized cancer patients with severe H1N1pdm infections during the winter of 2009 in Brazil. METHODS: We performed a prospective single-center cohort study in a cancer center in Rio de Janeiro, Brazil. Hospitalized patients with cancer and a confirmed diagnosis of influenza A H1N1pdm were evaluated. The main outcome measures in this study were in-hospital mortality, duration of viral shedding, viral persistence and both functional and molecular analyses of H1N1pdm susceptibility to oseltamivir. RESULTS: A total of 44 hospitalized patients with suspected influenza-like illness were screened. A total of 24 had diagnosed H1N1pdm infections. The overall hospital mortality in our cohort was 21%. Thirteen (54%) patients required intensive care. The median age of the studied cohort was 14.5 years (3-69 years). Eighteen (75%) patients had received chemotherapy in the previous month, and 14 were neutropenic at the onset of influenza. A total of 10 patients were evaluated for their duration of viral shedding, and 5 (50%) displayed prolonged viral shedding (median 23, range=11-63 days); however, this was not associated with the emergence of a resistant H1N1pdm virus. Viral evolution was observed in sequentially collected samples. CONCLUSIONS: Prolonged influenza A H1N1pdm shedding was observed in cancer patients. However, oseltamivir resistance was not detected. Taken together, our data suggest that severely ill cancer patients may constitute a pandemic virus reservoir with major implications for viral propagation

    A Model of DENV-3 Infection That Recapitulates Severe Disease and Highlights the Importance of IFN-γ in Host Resistance to Infection

    There are few animal models of dengue infection, especially in immunocompetent mice. Here, we describe alterations found in adult immunocompetent mice inoculated with an adapted Dengue virus (DENV-3) strain. Infection of mice with the adapted DENV-3 caused inoculum-dependent lethality that was preceded by several hematological and biochemical changes and increased virus dissemination, features consistent with severe disease manifestation in humans. IFN-γ expression increased after DENV-3 infection of WT mice and this was preceded by increase in expression of IL-12 and IL-18. In DENV-3-inoculated IFN-γ−/− mice, there was enhanced lethality, which was preceded by severe disease manifestation and virus replication. Lack of IFN-γ production was associated with diminished NO-synthase 2 (NOS2) expression and higher susceptibility of NOS2−/− mice to DENV-3 infection. Therefore, mechanisms of protection to DENV-3 infection rely on IFN-γ-NOS2-NO-dependent control of viral replication and of disease severity, a pathway showed to be relevant for resistance to DENV infection in other experimental and clinical settings. Thus, the model of DENV-3 infection in immunocompetent mice described here represents a significant advance in animal models of severe dengue disease and may provide an important tool to the elucidation of immunopathogenesis of disease and of protective mechanisms associated with infection
