    Skeletal muscle extracellular matrix (ECM), surrender of muscle fibers, the amount of which is just <5%, appeals less attention in the field of skeletal muscle physiology. Thus, at one time, the function of skeletal muscle ECM was arbitrarily considered as general structural support that is typical in other tissues. However, an increasing number of recent evidences have indicated that the ECM plays a critical role in muscle fiber force transmission, proliferation, differentiation, migration, and polarization of cells. Alterations of molecules within the ECM are involved in fibrosis, muscle aging, regeneration, and myopathies.     Integrative metabolomic and transcriptomic analysis reveals difference in glucose and lipid metabolism in the longissimus muscle of Luchuan and Duroc pigs

    Luchuan pig, an obese indigenous Chinese porcine breed, has a desirable meat quality and reproductive capacity. Duroc, a traditional western breed, shows a faster growth rate, high feed efficiency and high lean meat rate. Given the unique features these two porcine breeds have, it is of interest to investigate the underlying molecular mechanisms behind their distinctive nature. In this study, the metabolic and transcriptomic profiles of longissimus dorsi muscle from Duroc and Luchuan pigs were compared. A total of 609 metabolites were identified, 77 of which were significantly decreased in Luchuan compared to Duroc, and 71 of which were significantly elevated. Most differentially accumulated metabolites (DAMs) upregulated in Luchuan were glycerophospholipids, fatty acids, oxidized lipids, alcohols, and amines, while metabolites downregulated in Luchuan were mostly amino acids, organic acids and nucleic acids, bile acids and hormones. From our RNA-sequencing (RNA-seq) data we identified a total of 3638 differentially expressed genes (DEGs), 1802 upregulated and 1836 downregulated in Luchuan skeletal muscle compared to Duroc. Combined multivariate and pathway enrichment analyses of metabolome and transcriptome results revealed that many of the DEGs and DAMs are associated with critical energy metabolic pathways, especially those related to glucose and lipid metabolism. We examined the expression of important DEGs in two pathways, AMP-activated protein kinase (AMPK) signaling pathway and fructose and mannose metabolism, using Real-Time Quantitative Reverse Transcription PCR (qRT-PCR). Genes related to glucose uptake, glycolysis, glycogen synthesis, fatty acid synthesis (PFKFB1, PFKFB4, MPI, TPI1, GYS1, SLC2A4, FASN, IRS1, ULK1) are more activated in Luchuan, while genes related to fatty acid oxidation, cholesterol synthesis (CPT1A, HMGCR, FOXO3) are more suppressed. Energy utilization can be a decisive factor to the distinctive metabolic, physiological and nutritional characteristics in skeletal muscle of the two breeds we studied. Our research may facilitate future porcine breeding projects and can be used to reveal the potential molecular basis of differences in complex traits between various breeds

    OLFML3 Expression is Decreased during Prenatal Muscle Development and Regulated by MicroRNA-155 in Pigs

    The Olfactomedin-like 3 (OLFML3) gene has matrix-related function involved in embryonic development. MicroRNA-155 (miR-155), 21- to 23-nucleotides (nt) noncoding RNA, regulated myogenesis by target mRNA. Our LongSAGE analysis suggested that OLFML3 gene was differently expressed during muscle development in pig. In this study, we cloned the porcine OLFML3 gene and detected its tissues distribution in adult Tongcheng pigs and dynamical expression in developmental skeletal muscle (12 prenatal and 10 postnatal stages) from Landrace (lean-type) and Tongcheng (obese-type) pigs. Subsequently, we analyzed the interaction between OLFML3 and miR-155. The OLFML3 was abundantly expressed in liver and pancreas, moderately in lung, small intestine and placenta, and weakly in other tissues and postnatal muscle. There were different dynamical expression patterns between Landrace and Tongcheng pigs during prenatal skeletal muscle development. The OLFML3 was down-regulated (33-50 days post coitus, dpc), subsequently up-regulated (50-70 dpc), and then down-regulated (70-100 dpc) in Landrace pigs, while in Tongcheng pigs, it was down-regulated (33-50 dpc), subsequently up-regulated (50-55 dpc) and then down-regulated (55-100 dpc). There was higher expression in Tongcheng than Landrace in prenatal muscle from 33 to 60 dpc, and opposite situation from 65 to 100 dpc. Dual luciferase assay and real time PCR documented that OLFML3 expression was regulated by miR-155 at mRNA level. Our research indicated that OLFML3 gene may affect prenatal skeletal muscle development and was regulated by miR-155. These finding will help understanding biological function and expression regulation of OLFML3 gene in mammal animals

    Mesona is a special edible and medicinal food in China. The country has an abundant Mesona output and a large number of Mesona consumers. Polysaccharide is one of the important active components in Mesona, which has a variety of biological activities in disease prevention and treatment. Currently, Mesona polysaccharide has been widely used in Chinese Traditional medicine, herbal tea, as well as jelly. However, there are problems such as insufficient development of deep-processing products. In this paper, the domestic and foreign literature in the past recent years are retrieved and collected. This article comprehensively proposes the extraction, isolation and purification technologies of Mesona polysaccharide, as well as their characteristic structures, and rheological and gelling properties. Additionally, their functional activities and the underlying mechanism of action on antioxidant, regulation of gut flora, anti-hyperglycemia, anti-hyperlipidemia, liver protection and immunoregulation are systematically reviewed.     A Colorimetric Sensor for Qualitative Discrimination and Quantitative Detection of Volatile Amines

    We have developed a novel colorimetric sensor based on a digital camera and white LED illumination. Colorimetric sensor arrays (CSAs) were made from a set of six chemically responsive dyes impregnated on an inert substrate plate by solution casting. Six common amine aqueous solutions, including dimethylamine, triethylamine, diisopropylamine, aniline, cyclohexylamine, and pyridine vaporized at 25 °C and six health-related trimethylamine (TMA) concentrations including 170 ppm, 51 ppm, 8 ppm, 2 ppm, 125 ppb and 50 ppb were analyzed by the sensor to test its ability for the qualitative discrimination and quantitative detection of volatile amines. We extracted the feature vectors of the CSA's response to the analytes from a fusional color space, which was obtained by conducting a joint search algorithm of sequential forward selection and sequential backward selection (SFS&SBS) based on the linear discriminant criteria (LDC) in a mixed color space composed of six common color spaces. The principle component analysis (PCA) followed by the hierarchical cluser analysis (HCA) were utilized to discriminate 12 analytes. Results showed that the colorimetric sensor grouped the six amine vapors and five TMA concentrations correctly, while TMA concentrations of 125 ppb and 50 ppb were indiscriminable from each other. The limitation of detection (LOD) of the sensor for TMA was found to be lower than 50 ppb. The CSAs were reusable for TMA concentrations below 8 ppm

    Genotypic and Environmental Effects on the Volatile Chemotype of Valeriana jatamansi Jones

    Valeriana jatamansi Jones is an aromatic medicinal herb and important alternative to V. officinalis, which is utilized for medicinal purposes in China and India and also as spices in India. Bioactive ingredients of V. jatamansi vary in different regions. However, no information is currently available on influence of genotype and environmental factors in the volatile compounds, especially when germplasms and planting locations need to be selected. Based on the results of SNP and volatile constituents from GC-MS analysis, this study found various genotypes and chemotypes of V. jatamansi for wild plants from seven regions in China and common-garden samples; correlations between genotype and chemotype were revealed for the plants. Two distinct populations (PX, FY) were distinguishable from five others (GJ, YL, SY, DD, DY) according to their genotypes and volatile profiles, the consistency of which was observed showing that genotype could significantly influence chemotype. Wild populations and common-garden samples were also separated in their volatile profiles, demonstrating that environmental factors strongly affected their chemotypes. Compounds contributing to the discrimination were identified as discriminatory compounds. This investigation has explored and provided essential information concerning the correlation between genotype and chemotype as well as environmental factors and chemotype of V. jatamansi in some regions of China. Feasible plantation and conservation strategies of V. jatamansi could be further explored based on these results

    Characterization analysis and polymorphism detection of the porcine Myd88 gene

    The myeloid differentiation primary response protein 88 (Myd88) is an essential adaptor protein, which mediates in all Toll-like receptor (TLR) members signal transduction, except for TLR3. In this study, the 4464 bp genomic sequence of porcine Myd88 was first isolated, whereupon tissue distribution, chromosome mapping and single nucleotide polymorphism (SNP) were analyzed. Our results revealed that porcine Myd88 gene, which was located at chromosome 13 linked with marker S0288 (distance = 40 cR; LOD = 8.66), was widely expressed in all the examined tissues. There were 16 potential SNPs in the isolated genome fragment. SNP 797T/C in the first intron was studied, with no significant association being found between the genotype and immune traits in pigs (p > 0.05). The porcine Myd88 protein contained both the death domain (DD) and the Toll/IL-1 receptor domain (TIR). Leu residues, essential for its structure, were the most abundant encountered in the DD. The TIR contained two conserved motifs which may play important roles in the Myd88 function

    A compendium of genetic regulatory effects across pig tissues

    The Farm Animal Genotype-Tissue Expression (FarmGTEx) project has been established to develop a public resource of genetic regulatory variants in livestock, which is essential for linking genetic polymorphisms to variation in phenotypes, helping fundamental biological discovery and exploitation in animal breeding and human biomedicine. Here we show results from the pilot phase of PigGTEx by processing 5,457 RNA-sequencing and 1,602 whole-genome sequencing samples passing quality control from pigs. We build a pig genotype imputation panel and associate millions of genetic variants with five types of transcriptomic phenotypes in 34 tissues. We evaluate tissue specificity of regulatory effects and elucidate molecular mechanisms of their action using multi-omics data. Leveraging this resource, we decipher regulatory mechanisms underlying 207 pig complex phenotypes and demonstrate the similarity of pigs to humans in gene expression and the genetic regulation behind complex phenotypes, supporting the importance of pigs as a human biomedical model.</p
