    Growers generally use herbicides to efficiently produce high-quality fruit and vegetables for processing or fresh market sales. Due to the smaller acreage of these crops compared to major field crops, fewer herbicides are registered for use in fruit and vegetable crops than for field crops. Each year, new herbicides are evaluated under Arkansas growing conditions with the objective of improving the herbicide technology for the grower, processor, and ultimately the consumer. This report includes studies on the control of many of the more serious weed problems in important crops of this region, including snapbeans, spinach, southern pea, watermelon, cantaloupe, tomato, blackberry, and grape. In addition, the report includes information on the tolerance of selected bedding plants to some effective herbicides

    COVID, Church, & Cuts: A Single Narrative Case Study of Pandemic Impacts on a Bi-vocational Pastor & Barber

    This narrative case study provides an in-depth look into the impacts of the COVID-19 Pandemic from the perspective of an assistant pastor with a small African American church in an urban city. His story indicates that the pandemic had both positive and negative effects on the pastor’s ability to carry out his role of pastoral care and ministry. Some positives were the results of new technological adaptations that helped to improve members’ consistency in attending and participating in weekly services. Some negative effects imposed by COVID-19 was the revelation of imbalanced relationships and a lack of trust in God. The pastor shared how he struggled with trusting God to provide financially for him and his family when the ability to work was taken away. Through a renewed trust in the Lord and re-examination of familial responsibilities, his faith and family connections strengthened. Additionally, this pastor shares his personal and community plans for moving forward with a renewed vision and new wisdom

    Utilization of Free Medication Samples in the United States in a Nationally Representative Sample: 2009-2013

    Background—Manufacturers provide free sample medications as a means to increase use of branded medications. Sample use varies year-to-year as branded product patents expire and new products come to market. Objective—This study sought to describe the use of sample medications during 2009–2013 and assess individual characteristics associated with sample use. Methods—Data from the 2009–2013 U.S. Medical Expenditure Panel Survey (MEPS) were used. MEPS asks participants whether they received each medication they are taking as a sample. The top 10 medications and medication classes used each year by volume were identified as well as the proportion of people who used at least one sample medication. The proportion of new initiators of medications were also classified as the percent who received a sample for the specific medication. Logistic regression was used to assess individual demographics, insurance, and medication characteristics associated with use. Results—Prevalence of sample use ranged from 9.3% in 2009 to 6.2% in 2013. The most widely used sample medications included statins during 2009–2011, which changed to inhaled β-agonists in 2012–2013, as atorvastatin became available as a generic. The overall volume of the top 10 free sample medications decreased by one-third over this study period. In 2013, 12.6% of new insulin analog users and 11.0% of new oral contraceptive users receive these medications through samples. Regression analysis showed that U.S. Medicaid- and Medicare-insured persons were less likely to use samples compared to those with private insurance. Conclusions—Sample medication use has decreased as generic medications are becoming more used in the U.S

    Changes in the Rpb3 Interactome Caused by the Deletion of RPB9 in Saccharomyces cerevisiae

    Indiana University-Purdue University Indianapolis (IUPUI)RNA Polymerase II (Pol II) is the primary actor in the transcription of mRNA from genes. Pol II is a complex composed of twelve protein subunits. This study focused on the changes in the interactome of Rbp3 in S. cerevisiae when the Pol II subunit Rpb9 is removed. Rpb3 is one of the core subunits of Pol II, and any significant changes to the Rpb3 incteractome due to the loss of Rpb9 can be used to infer new information about Rpb9’s role in the Pol II complex. Rpb3 was pulled down using FLAG purification from both wild type and rpb9Δ S. cerevisiae cultures. Rpb3 and the proteins complexed with it were then analyzed using multi-dimensional protein identification technology (MudPIT), a form of liquid chromatography-mass spectrometry (LC-MS). This data was searched using the SEQUEST database search algorithm, and the results were further analyzed for likelihood of interaction using Significance Analysis of INTeractome (SAINT), as well as for post-translational phosphorylation. Deletion of rpb9 did not present any changes in Pol II phosphorylation however it did cause several changes in the interaction network. The rpb9Δ strain showed new interactions with Rtr1, Sen1, Vtc4, Pyc1, Tgl4, Sec61, Tfb2, Hfd1, Erv25, Rib4, Sla1, Ubp15, Bbc1, and Hxk1. The most prominent of these hits are Rtr1, an Rpb1 C-terminal domain phosphatase linked to transcription termination, and Sen1, an RNA/DNA nuclease that terminates transcription. In addition, this mutant showed no interaction with Mtd1, an interaction that is present in the wild type. In all cases, these hits should be considered fuel for future research, rather than conclusive evidence of novel interactions

    Rapid detection of bacteria responsible for foodborne diseases is a growing necessity for public health. Reporter bacteriophages (phages) are robust biorecognition elements uniquely suited for the rapid and sensitive detection of bacterial species. The advantages of phages include their host specificity, ability to distinguish viable and non-viable cells, low cost, and ease of genetic engineering. Upon infection with reporter phages, target bacteria express reporter enzymes encoded within the phage genome. In this study, the T7 coliphage was genetically engineered to express the newly developed luceriferase, NanoLuc (NLuc), as an indicator of bacterial contamination. While several genetic approaches were employed to optimize reporter enzyme expression, the novel achievement of this work was the successful fusion of the NanoLuc reporter to a carbohydrate binding module (CBM) with specificity to crystalline cellulose. This novel chimeric reporter (nluc::cbm) bestows the specific and irreversible immobilization of NanoLuc onto a low-cost, widely available crystalline cellulosic substrate. We have shown the possibility of detecting the immobilized fusion protein in a filter plate which resulted from a single CFU of E. coli. We then demonstrated that microcrystalline cellulose can be used to concentrate the fusion reporter from 100 mL water samples allowing a limit of detection of \u3c10 CFU mL-1 E. coli in 3 hours. Therefore, we conclude that our phage-based detection assay displays significant aptitude as a proof-of-concept drinking water diagnostic assay for the low-cost, rapid and sensitive detection of E. coli. Additional improvements in the capture efficiency of the phage-based fusion reporter should allow a limit of detection of \u3c10 CFU per 100 mL
