    Impacts of in vivo and in vitro exposures to tamoxifen: comparative effects on human cells and marine organisms

    Tamoxifen (TAM) is a first generation-SERM administered for hormone receptor-positive (HER+) breast cancer in both pre- and post-menopausal patients and may undergo metabolic activation in organisms that share similar receptors and thus face comparable mechanisms of response. The present study aimed to assess whether environmental trace concentrations of TAM are bioavailable to the filter feeder M. galloprovincialis (100 ng L-1) and to the deposit feeder N. diversicolor (0.5, 10, 25 and 100 ng L-1) after 14 days of exposure. Behavioural impairment (burrowing kinetic), neurotoxicity (AChE activity), endocrine disruption by alkali-labile phosphate (ALP) content, oxidative stress (SOD, CAT, GPXs activities), biotransformation (GST activity), oxidative damage (LPO) and genotoxicity (DNA damage) were assessed. Moreover, this study also pertained to compare TAM cytotoxicity effects to mussels and targeted human (i.e. immortalized retinal pigment epithelium - RPE; and human transformed endothelial cells - HeLa) cell lines, in a range of concentrations from 0.5 ng L-1 to 50 μg L-1. In polychaetes N. diversicolor, TAM exerted remarkable oxidative stress and damage at the lowest concentration (0.5 ng L-1), whereas significant genotoxicity was reported at the highest exposure level (100 ng L-1). In mussels M. galloprovincialis, 100 ng L-1 TAM caused endocrine disruption in males, neurotoxicity, and an induction in GST activity and LPO byproducts in gills, corroborating in genotoxicity over the exposure days. Although cytotoxicity assays conducted with mussel haemocytes following in vivo exposure was not effective, in vitro exposure showed to be a feasible alternative, with comparable sensitivity to human cell line (HeLa).info:eu-repo/semantics/publishedVersio

The present work describes the synthesis and characterization of novel series of triarylamines isoxazoles (TAA) addressed to the organic photovoltaic materials. Diarylisoxazoles were synthesized by sequential [3+2] 1,3-dipolar cycloaddition reaction between arylnitrile oxides and selected arylalkenes followed by MnO2-oxidation. Isoxazoles were coupled to diarylamines by Büchwald-Hartwig reaction to afford desired compounds 6a-k.     A Novel Technique to Produce Polygenic Resistance to Anthracnose in \u3ci\u3eStylosanthes capitata\u3c/i\u3e

    An improved cultivar, based on 17 genotypes of S. capitata and six of S.macrocephala, was developed at the Embrapa Beef Cattle Research Center, Campo Grande, Brazil. The aim was to create durable, race non-specific resistance to anthracnose controlled by polygenic factors. A mass hybridisation technique was employed to produce a high degree of genetic diversity and sizeable quantities of seed of hybrid-derived progenies of Brazilian and Venezuelan genotypes of S. capitata. Outcrossing resulted in a significant improvement in the forage production of progeny of Venezuelan accessions. The multicross was evaluated in multilocational trials, each representing a large tract of country in the Cerrados ecosystem along a north-south transect from lat.60 S to 200 S. The genetic shift that occurred in S. capitata was a key element in the formation of the new cultivar. It is a complex mixture of two species, and a recombination of much desired forage traits of Brazilian x Venezuelan genotypes, high forage and seed yields coupled with anthracnose resistance. The new cultivar with its diverse genetic make-up has a wide application in the acid-soil savannas of tropical America. It was released by Embrapa for the Cerrados in 2000

    Hybrid stylos (Stylosanthes guianensis var. vulgaris x var. pauciflora) with durable, quantitative resistance to anthracnose, mid-season harvest maturity date (early-July), high DM and seed yields have been selected at the Embrapa Beef Cattle Research Center, Campo Grande, Brazil. The hybrids displayed improved forage traits in Brazil, the native habitat and major center of diversity of the species and its pathogen, as well as in vastly different ecosystems. Dry forage yields and anthracnose resistance of superior selections and their composites were equal, in some instances, significantly better, than those of cv. Mineirão in multilocational trials situated in the Cerrados from lat. 6° S to lat. 20° S. Selected hybrids performed well in comparison with the highly successful CIAT184 (cv.Reyan II) on Hainan Island, China. Composites have also shown good promise in seed multiplication plots in Queensland, Australia. A positive attribute of composite hybrids is their great genetic diversity in contrast to pure-line cultivars with a relatively narrow genetic base.     We use high signal-to-noise ratio long-slit spectra in the 3600-4700A range of the twenty brightest northern Seyfert 2 galaxies to study the variation of the stellar population properties as a function of distance from the nucleus. In order to characterize the stellar population and other continuum sources (e.g. featureless continuum FC) we have measured equivalent widths Ws of six absorption features, four continuum colours and their radial variations, and performed spectral population synthesis as a function of distance from the nucleus. About half the sample has CaIIK and G-band W values smaller at the nucleus than at 1 kpc from it, due to a younger population and/or FC. The stellar population synthesis shows that, while at the nucleus, 75% of the galaxies present contribution > 20% of ages younger or equal than 100Myr and/or of a FC, this proportion decreases to 45% at 3 kpc. In particular, 55% of the galaxies have contribution > 10% of the 3 Myr/FC component (a degenerate component in which one cannot separate what is due to a FC or to a 3 Myr stellar population) at the nucleus, but only 25% of them have this contribution at 3 kpc. As reference, the stellar population of 10 non-Seyfert galaxies, spanning the Hubble types of the Seyfert (from S0 to Sc) was also studied. A comparison between the stellar population of the Seyferts and that of the non-Seyferts shows systematic differences: the contribution of ages younger than 1 Gyr is in most cases larger in the Seyfert galaxies than in non-Seyferts, not only at the nucleus but up to 1 kpc from it.Comment: 23 pages, 18 figures, MNRAS in pres

    Does EU Public Support to Firm Investments Boost High Quality Jobs? Evidence from Linked Employer-Employee Microdata and Natural-Experiment Conditions

    In this paper we estimate the impact on job quality of the direct support to firm- investment projects provided through the EU cohesion policy funds. We use linked employer-employee micro-data, from administrative sources, that cover the universe of Portuguese firms over the years 2000s. The analysis exploits the existence of natural experiment conditions in the form of treatment assignment procedures based on exogenous cut-offs in the ranking of the applicant firms. Our preferred estimation strategy is a dynamic exact matching procedure that is combined with a difference-in-difference scheme to mimic a block-randomised pretest-posttest design. The procedure is adopted separately for each of the successive cohorts of applicants and solves the small sample issues that would be encountered with a multiple-ranking regression discontinuity design. Our results indicate that the firm-level support co-funded by EU structural funds in Portugal has contributed to improving job quality compared to the counterfactual scenario of no public support, with each treated firm creating on average 4.9 additional standard-working-time jobs, 2.9 skilled jobs, and 2.0 permanent contract jobs. We also estimate that the average remuneration per hour in treated firms is €2.2 higher than in similar non-treated applicant firms. These results are adequately robust to a wide range of sensitivity analyses and bear some relevant policy implications for the EU.FC
