    The pitch-heave dynamics of transportation vehicles

    The analysis and design of suspensions for vehicles of finite length using pitch-heave models is presented. Dynamic models for the finite length vehicle include the spatial distribution of the guideway input disturbance over the vehicle length, as well as both pitch and heave degrees-of-freedom. Analytical results relate the vehicle front and rear accelerations to the pitch and heave natural frequencies, which are functions of vehicle suspension geometry and mass distribution. The effects of vehicle asymmetry and suspension contact area are evaluated. Design guidelines are presented for the modification of vehicle and suspension parameters to meet alternative ride quality criteria

    Analysis of normalized radar cross section (sigma-O) signature of Amazon rain forest using SEASAT scatterometer data

    The normalized radar cross section (NRCS) signature of the Amazon rain forest was SEASAT scatterometer data. Statistics of the measured (NRCS) values were determined from multiple orbit passes for three local time periods. Plots of mean normalized radar cross section, dB against incidence angle as a function of beam and polarization show that less than 0.3 dB relative bias exists between all beams over a range of incidence angle from 30 deg to 53 deg. The backscattered measurements analyzed show the Amazon rain forest to be relatively homogeneous, azimuthally isotropic and insensitive to polarization. The return from the rain forest target appears relatively consistent and stable, except for the small diurnal variation (0.75 dB) that occurs at sunrise. Because of the relative stability of the rain forest target and the scatterometer instrument, the response of versus incidence angle was able to detect errors in the estimated yaw altitude angle. Also, small instrument gain biases in some of the processing channels were detected. This led to the development of an improved NRCS algorithm, which uses a more accurate method for estimating the system noise power

    Database Search Strategies for Proteomic Data Sets Generated by Electron Capture Dissociation Mass Spectrometry

    Large data sets of electron capture dissociation (ECD) mass spectra from proteomic experiments are rich in information; however, extracting that information in an optimal manner is not straightforward. Protein database search engines currently available are designed for low resolution CID data, from which Fourier transform ion cyclotron resonance (FT-ICR) ECD data differs significantly. ECD mass spectra contain both z-prime and z-dot fragment ions (and c-prime and c-dot); ECD mass spectra contain abundant peaks derived from neutral losses from charge-reduced precursor ions; FT-ICR ECD spectra are acquired with a larger precursor m/z isolation window than their low-resolution CID counterparts. Here, we consider three distinct stages of postacquisition analysis: (1) processing of ECD mass spectra prior to the database search; (2) the database search step itself and (3) postsearch processing of results. We demonstrate that each of these steps has an effect on the number of peptides identified, with the postsearch processing of results having the largest effect. We compare two commonly used search engines: Mascot and OMSSA. Using an ECD data set of modest size (3341 mass spectra) from a complex sample (mouse whole cell lysate), we demonstrate that search results can be improved from 630 identifications (19% identification success rate) to 1643 identifications (49% identification success rate). We focus in particular on improving identification rates for doubly charged precursors, which are typically low for ECD fragmentation.     SLoMo: automated site localization of modifications from ETD/ECD mass spectra

    Recently, software has become available to automate localization of phosphorylation sites from CID data and to assign associated confidence scores. We present an algorithm, SLoMo (Site Localization of Modifications), which extends this capability to ETD/ECD mass spectra. Furthermore, SLoMo caters for both high and low resolution data and allows for site-localization of any UniMod post-translational modification. SLoMo accepts input data from a variety of formats (e.g., Sequest, OMSSA). We validate SLoMo with high and low resolution ETD, ECD, and CID data

    Development of cyclosporin A mediated immunity in L1210 leukaemia.

    Cyclosporin A (CsA) is an effective modulator of multidrug resistance (MDR) in vitro and in murine tumour systems in vivo. We now report the production of immunity to L1210 leukaemia by the addition of CsA to VP-16 therapy of leukaemic BDF/1 mice. VP-16/cyclosporin A tumour immunity induction arises as a consequence of active therapy independently of immunisation with modified tumour cells. The addition of CsA to VP-16 prolongs survival of BDF/1 host mice bearing L1210 leukaemia beyond that produced by equivalent dose VP-16 alone. A subpopulation of 60-day surviving mice after combined VP-16/CsA are immune to rechallenge with the same leukaemia inoculum to which they were originally exposed. Spleen cells from immune mice adoptively transfer anti-L1210 leukaemia immunity to untreated BDF/1 mice in a dose dependent, statistically significant manner. Adoptive transfer experiments additionally suggest active recruitment of immunologic response in recipient animals: (1) We have been able to perpetuate leukaemia immunity in four sequential cohorts of naive recipient mice. This propogation of adoptive immunity is accomplished by use of spleen cells harvested from each preceeding passively-protected animal cohort; (2) Cyclophosphamide pretreatment of adoptive transfer recipient mice abrogates the ability of their splenocytes to perpetuate passive protection in sequential adoptive transfer experiments

    The interactive effect of leader-member exchange and perceived organizational support on employee adaptive performance

    Examining the role of intra-organizational social exchanges in influencing adaptive performance, the authors hypothesized that leader-member exchange (LMX) quality and organizational support have an interactive effect on employee adaptive performance. We surveyed 175 private sector workers and found that subordinate perceptions of LMX were positively related to supervisor-rated adaptive performance among workers reporting average and high but not low levels of organizational support. Results add to the LMX literature by showing that the LMX-outcome relationship may depend on context, provide support for the divergent validity of POS and LMX, and raise important questions for future social exchange research

    Large scale localization of protein phosphorylation by use of electron capture dissociation mass spectrometry.

    We used on-line electron capture dissociation (ECD) for the large scale identification and localization of sites of phosphorylation. Each FT-ICR ECD event was paired with a linear ion trap collision-induced dissociation (CID) event, allowing a direct comparison of the relative merits of ECD and CID for phosphopeptide identification and site localization. Linear ion trap CID was shown to be most efficient for phosphopeptide identification, whereas FT-ICR ECD was superior for localization of sites of phosphorylation. The combination of confident CID and ECD identification and confident CID and ECD localization is particularly valuable in cases where a phosphopeptide is identified just once within a phosphoproteomics experiment

    Design and fabrication of a long-life Stirling cycle cooler for space application. Phase 3: Prototype model

    A second-generation, Stirling-cycle cryocooler (cryogenic refrigerator) for space applications, with a cooling capacity of 5 watts at 65 K, was recently completed. The refrigerator, called the Prototype Model, was designed with a goal of 5 year life with no degradation in cooling performance. The free displacer and free piston of the refrigerator are driven directly by moving-magnet linear motors with the moving elements supported by active magnetic bearings. The use of clearance seals and the absence of outgassing material in the working volume of the refrigerator enable long-life operation with no deterioration in performance. Fiber-optic sensors detect the radial position of the shafts and provide a control signal for the magnetic bearings. The frequency, phase, stroke, and offset of the compressor and expander are controlled by signals from precision linear position sensors (LVDTs). The vibration generated by the compressor and expander is cancelled by an active counter balance which also uses a moving-magnet linear motor and magnetic bearings. The driving signal for the counter balance is derived from the compressor and expander position sensors which have wide bandwidth for suppression of harmonic vibrations. The efficiency of the three active members, which operate in a resonant mode, is enhanced by a magnetic spring in the expander and by gas springs in the compressor and counterbalance. The cooling was achieved with a total motor input power of 139 watts. The magnetic-bearing stiffness was significantly increased from the first-generation cooler to accommodate shuttle launch vibrations

    The SAMI Galaxy Survey: Satellite galaxies undergo little structural change during their quenching phase

    At fixed stellar mass, satellite galaxies show higher passive fractions than centrals, suggesting that environment is directly quenching their star formation. Here, we investigate whether satellite quenching is accompanied by changes in stellar spin (quantified by the ratio of the rotational to dispersion velocity V/σ\sigma) for a sample of massive (M>M_{*}>1010^{10} M_{\odot}) satellite galaxies extracted from the SAMI Galaxy Survey. These systems are carefully matched to a control sample of main sequence, high V/σV/\sigma central galaxies. As expected, at fixed stellar mass and ellipticity, satellites have lower star formation rate (SFR) and spin than the control centrals. However, most of the difference is in SFR, whereas the spin decreases significantly only for satellites that have already reached the red sequence. We perform a similar analysis for galaxies in the EAGLE hydro-dynamical simulation and recover differences in both SFR and spin similar to those observed in SAMI. However, when EAGLE satellites are matched to their `true' central progenitors, the change in spin is further reduced and galaxies mainly show a decrease in SFR during their satellite phase. The difference in spin observed between satellites and centrals at zz\sim0 is primarily due to the fact that satellites do not grow their angular momentum as fast as centrals after accreting into bigger halos, not to a reduction of V/σV/\sigma due to environmental effects. Our findings highlight the effect of progenitor bias in our understanding of galaxy transformation and they suggest that satellites undergo little structural change before and during their quenching phase.Comment: 11 pages, 7 figures.     Effects of the tat and nef gene products of human immunodeficiency virus type 1 (HIV-1) on transcription controlled by the HIV-1 long terminal repeat and on cell growth in macrophages

    The RAW264 murine macrophage cell line was used as a model to examine the role of the tat and nef gene products in the transcription regulation of the human immunodeficiency virus type 1 (HIV-1) long terminal repeat (LTR) in macrophages. Contrary to claims that the activity of the HIV-1 LTR responds poorly in rodent cells to trans activation by the viral tat gene product, cotransfection of RAW264 cells with a tat expression plasmid in transient transfection assays caused a > 20-fold increase in reporter gene expression that was inhibited by mutations in the TAR region. RAW264 cells stably transfected with the tat plasmid displayed similarly elevated HIV-1 LTR-driven reporter gene activity. By contrast to previous reports indicating a negative role for nef in HIV transcription, cotransfection of RAW264 cells with a nef expression plasmid trans activated the HIV-1 LTR driving either a chloramphenicol acetyltransferase or a luciferase reporter gene. The action of nef was specific to the LTR, as expression of nef had no effect on the activity of the simian virus 40, c-fms, urokinase plasminogen activator, or type 5 acid phosphatase promoter. trans-activating activity was also manifested by a frameshift mutant expressing only the first 35 amino acids of the protein. The effects of nef were multiplicative with those of tat gene product and occurred even in the presence of bacterial lipopolysaccharide, which itself activated LTR-directed transcription. Examination of the effects of selected mutations in the LTR revealed that neither the kappa B sites in the direct repeat enhancer nor the TAR region was required as a cis-acting element in nef action. The action of nef was not species restricted; it was able to trans activate in the human monocyte-like cell line Mono Mac 6. The presence of a nef expression cassette in a neomycin phosphotransferase gene expression plasmid greatly reduced the number of G418-resistant colonies generated in stable transfection of RAW264 cells, and many of the colonies that were formed exhibited very slow growth. The frameshift mutant was also active in reducing colony generation. Given the absence of any effect of the frameshift mutation on nef function, its actions on macrophage growth and HIV transcription are discussed in terms of the role of the N-terminal 30 amino acids and of stable secondary structures in the mRNA
