    Electroacupuncture Attenuates Ovalbumin-Induced Allergic Asthma via Modulating CD4+CD25+ Regulatory T Cells

    A mouse pulmonary hypersensitivity experimental model that mimics human asthma was developed, and electroacupuncture (EA) treatment was shown to reduce allergic inflammatory processes. In addition, we also assessed whether the beneficial effects of EA on allergic asthma could be correlated with CD4+CD25+Foxp3+ regulatory T cells (Treg). Cellular profiles and histopathologic analysis demonstrated that peribronchial and perivascular inflammatory cell infiltrates were significantly decreased in the EA-treated groups when compared to the OVA and anti-CD25 Ab-injected (Treg depletion) groups. Furthermore, total BAL cells were reduced in the EA groups when compared to other groups. Interestingly, the population of CD4+CD25+Foxp3+Tregs in pneumonocytes increased in EA-treated group when compared to OVA and Treg depletion groups. These results imply that EA stimulation at ST 36 may affect CD4+CD25+Foxp3+ Treg in an OVA-induced experimental model and may enhance Treg function by suppressing other T cells and limiting the immune response

    Expression of Phospholipase A2 Receptor in Pediatric Hepatitis B Virus-Related Membranous Nephropathy

    Purpose Hepatitis B virus (HBV) infection is among etiologies of secondary membranous nephropathy (MN) in pediatric patients. We evaluated expression of phospholipase A2 receptor (PLA2R), a specific target antigen of primary MN, in pediatric HBV-related MN. Methods We retrospectively reviewed patients with biopsy-proven HBV-related MN from the renal biopsy registry and electronic medical records of Severance Hospital, Seoul, Korea, from 1993 to 2004. Paraffin-embedded human kidney tissues were retrieved and immunohistochemically stained for PLA2R. Results Ten pediatric patients with 13 biopsied specimens were reviewed. The predominant pathological stage was stage II–III, and second was stage II. The intensity of staining for IgG was greatest, with less intense staining for IgM, IgA, C3, C4, and C1q. All the patients had angiotensin-converting enzyme inhibitor combined with glucocorticoid, and four patients converted to cyclosporine treatment from glucocorticoid monotherapy. Urinalysis of all the patients normalized after variable period. PLA2R staining was demonstrated in the outer glomerulus in 3 out of 13 biopsies, 2 of which were obtained from the same patient over a 5-year interval. Conclusions PLA2R was expressed in a small number of cases diagnosed as pediatric HBV-related MN, indicating that some HBV-related MN cases may be primary MN concurrent with HBV infection

    Charge Transport in UV-Oxidized Graphene and Its Dependence on the Extent of Oxidation

    Graphene oxides with different degrees of oxidation are prepared by controlling UV irradiation on graphene, and the charge transport and the evolution of the transport gap are investigated according to the extent of oxidation. With increasing oxygenous defect density [Formula: see text] , a transition from ballistic to diffusive conduction occurs at [Formula: see text] cm [Formula: see text] and the transport gap grows in proportion to [Formula: see text]. Considering the potential fluctuation related to the [Formula: see text] puddle, the bandgap of graphene oxide is deduced to be [Formula: see text] meV. The temperature dependence of conductivity showed metal–insulator transitions at [Formula: see text] cm [Formula: see text] , consistent with Ioffe–Regel criterion. For graphene oxides at [Formula: see text] cm [Formula: see text] , analysis indicated charge transport occurred via 2D variable range hopping conduction between localized [Formula: see text] domain. Our work elucidates the transport mechanism at different extents of oxidation and supports the possibility of adjusting the bandgap with oxygen content

    AlphaTuning: Quantization-Aware Parameter-Efficient Adaptation of Large-Scale Pre-Trained Language Models

    There are growing interests in adapting large-scale language models using parameter-efficient fine-tuning methods. However, accelerating the model itself and achieving better inference efficiency through model compression has not been thoroughly explored yet. Model compression could provide the benefits of reducing memory footprints, enabling low-precision computations, and ultimately achieving cost-effective inference. To combine parameter-efficient adaptation and model compression, we propose AlphaTuning consisting of post-training quantization of the pre-trained language model and fine-tuning only some parts of quantized parameters for a target task. Specifically, AlphaTuning works by employing binary-coding quantization, which factorizes the full-precision parameters into binary parameters and a separate set of scaling factors. During the adaptation phase, the binary values are frozen for all tasks, while the scaling factors are fine-tuned for the downstream task. We demonstrate that AlphaTuning, when applied to GPT-2 and OPT, performs competitively with full fine-tuning on a variety of downstream tasks while achieving >10x compression ratio under 4-bit quantization and >1,000x reduction in the number of trainable parameters.Comment: Findings of EMNLP 202

    Changes in Fruit Physicochemical Characteristics by Fruit Clusters in June-bearing Strawberry Cultivars

    Three Korean-bred strawberry cultivars 'Maehyang', 'Seolhyang', and 'Keumhyang', and a Japanese cultivar 'Tochiotome' were grown in a greenhouse and their physicochemical characteristics were investigated. Fruit weight of 'Seolhyang' and 'Keumhyang' in the first and second fruit clusters were greater than those of other cultivars and that of 'Tochiotome' was the greatest in the fifth fruit cluster. Fruit firmness generally decreased at later fruit clusters, and was the lowest in 'Seolhyang'. The sugars/organic acids ratios in the first and third fruit clusters of 'Maehyang' were 4.9 and 8.0, respectively, representing the highest values among all cultivars. The ascorbic acid content was the greatest in the second fruit cluster for 'Seolhyang', 'Keumhyang', and 'Tochiotome' cultivars and that of 'Maehyang' was the greatest at the third fruit cluster. The anthocyanin content was higher in later fruit clusters and was the highest in 'Keumhyang' overall.     The Effects of Maekmoondong-Tang on Cockroach Extract-Induced Allergic Asthma

    Maekmoondong-tang (MMDT) has long been used in Asian countries to treat respiratory diseases. However, the precise mechanisms underlying its effects on asthma are unknown. This study was conducted to evaluate the protective effects of MMDT in a cockroach allergen (CKA-)induced animal model of allergic asthma. After being challenged with CKA, the number of macrophages, eosinophils, neutrophils, lymphocytes, and total cells in the bronchoalveolar lavage fluid (BALF) was evaluated. The Th2 specific cytokines IL-4, IL-5, and IL-13 were also analyzed in BALF along with IgE levels in serum. For histological analysis, hematoxylin and eosin (H&E) staining, periodic acid-Schiff (PAS) staining, and immunohistochemical staining were performed. In addition, airway hyperresponsiveness was assessed by noninvasive plethysmography. The cellular profiles and histopathologic analysis demonstrated that peribronchial and perivascular inflammatory cell infiltrates were significantly decreased in the MMDT-treated groups compared with the cockroach extract-injected (CKA) groups. In addition, the IgE, IL-4, IL-5, and IL-13 levels were significantly decreased in the MMDT group. MMDT treatment also significantly attenuated airway hyperresponsiveness. These results demonstrated that MMDT significantly reduced the hallmark signs of asthma: elevated serum IgE, airway eosinophilia, airway remodeling, mucus hypersecretion, and airway hyperresponsiveness. The remarkable antiasthmatic effects of MMDT suggest its therapeutic potential in allergic asthma treatment

    The Effect of Environmental Enrichment on Glutathione-Mediated Xenobiotic Metabolism and Antioxidation in Normal Adult Mice

    Olfactory bulb (OB) plays an important role in protecting against harmful substances via the secretion of antioxidant and detoxifying enzymes. Environmental enrichment (EE) is a common rehabilitation method and known to have beneficial effects in the central nervous system. However, the effects of EE in the OB still remain unclear. At 6 weeks of age, CD-1® (ICR) mice were assigned to standard cages or EE cages. After 2 months, we performed proteomic analysis. Forty-four up-regulated proteins were identified in EE mice compared to the control mice. Gene Ontology analysis and Kyoto Encyclopedia of Genes and Genomes Pathway demonstrated that the upregulated proteins were mainly involved in metabolic pathways against xenobiotics. Among those upregulated proteins, 9 proteins, which participate in phase I or II of the xenobiotic metabolizing process and are known to be responsible for ROS detoxification, were validated by qRT-PCR. To explore the effect of ROS detoxification mediated by EE, glutathione activity was measured by an ELISA assay. The ratio of reduced glutathione to oxidized glutathione was significantly increased in EE mice. Based on a linear regression analysis, GSTM2 and UGT2A1 were found to be the most influential genes in ROS detoxification. For further analysis of neuroprotection, the level of iNOS and the ratio of Bax to Bcl-2 were significantly decreased in EE mice. While TUNEL+ cells were significantly decreased, Ki67+ cells were significantly increased in EE mice, implicating that EE creates an optimal state for xenobiotic metabolism and antioxidant activity. Taken together, our results suggested that EE protects olfactory layers via the upregulation of glutathione-related antioxidant and xenobiotic metabolizing enzymes, eventually lowering ROS-mediated inflammation and apoptosis and increasing neurogenesis. This study may provide an opportunity for a better understanding of the beneficial effects of EE in the OB

    Diffusion Decay Coefficient for Chloride Ions of Concrete Containing Mineral Admixtures

    The diffusion coefficient for chloride ions and the diffusion decay coefficient for chloride ions are essential variables for a service life evaluation of concrete structures. They are influenced by water-binder ratio, exposure condition, curing temperature, cement type, and the type and use of mineral admixture. Mineral admixtures such as ground granulated blast furnace slag, fly ash, and silica fume have been increasingly used to improve resistance against chloride ions penetration in concrete structures built in an offshore environment. However, there is not enough measured data to identify the statistical properties of diffusion decay coefficient for chloride ions in concrete using mineral admixtures. This paper is aimed at evaluating the diffusion decay coefficient for chloride ions of concrete using ordinary Portland cement or blended cement. NT BUILD 492 method, an electrophoresis experiment, was used to measure the diffusion coefficient for chloride ions with ages. It was revealed from the test results that the diffusion decay coefficient for chloride ions was significantly influenced by W/B and the replacement ratio of mineral admixtures
