    Slab control on the mega-sized North Pacific ultra-low velocity zone.

    Ultra-low velocity zones (ULVZs) are localized small-scale patches with extreme physical properties at the core-mantle boundary that often gather at the margins of Large Low Velocity Provinces (LLVPs). Recent studies have discovered several mega-sized ULVZs with a lateral dimension of ~900 km. However, the detailed structures and physical properties of these ULVZs and their relationship to LLVP edges are not well constrained and their formation mechanisms are poorly understood. Here, we break the degeneracy between the size and velocity perturbation of a ULVZ using two orthogonal seismic ray paths, and thereby discover a mega-sized ULVZ at the northern edge of the Pacific LLVP. The ULVZ is almost double the size of a previously imaged ULVZ in this region, but with half of the shear velocity reduction. This mega-sized ULVZ has accumulated due to stable mantle flow converging at the LLVP edge driven by slab-debris in the lower mantle. Such flow also develops the subvertical north-tilting edge of the Pacific LLVP

    A Chinese SCA36 pedigree analysis of NOP56 expansion region based on long-read sequencing

    Introduction: Spinocerebellar ataxias 36 (SCA36) is the neurodegenerative disease caused by the GGCCTG Hexanucleotide repeat expansions in NOP56, which is too long to sequence using short-read sequencing. Single molecule real time (SMRT) sequencing can sequence across disease-causing repeat expansion. We report the first long-read sequencing data across the expansion region in SCA36.Methods: We collected and described the clinical manifestations and imaging features of Han Chinese pedigree with three generations of SCA36. Also, we focused on structural variation analysis for intron 1 of the NOP56 gene by SMRT sequencing in the assembled genome.Results: The main clinical features of this pedigree are late-onset ataxia symptoms, with a presymptomatic presence of affective and sleep disorders. In addition, the results of SMRT sequencing showed the specific repeat expansion region and demonstrated that the region was not composed of single GGCCTG hexanucleotides and there were random interruptions.Discussion: We extended the phenotypic spectrum of SCA36. We applied SMRT sequencing to reveal the correlation between genotype and phenotype of SCA36. Our findings indicated that long-read sequencing is well suited to characterize known repeat expansion

    Increased transgene expression mediated by recombinant adeno-associated virus in human neuroglia cells under microgravity conditions

    The space environment has the special characteristics of radiation, noise particularity and weightlessness, all of which have adverse effects on astronauts’ muscles, bones, neurons and immune system. Some reports have shown that chemotherapy and radiotherapy can increase the activity of the recombinant adeno-associated virus (AAV) which is widely used in gene therapy. In this paper, recombinant AAV2 (rAAV2) was first packaged with the enhanced green fluorescence protein (eGFP) gene and used to infect neuroglia cells including the U87 and U251 cell lines, under microgravity conditions; it was then detected by fluorescence microscopy and flow cytometry. The results show that microgravity affects the adhesion ability of cells, promotes transgene expression induced by rAAV2 and causes changes of viral infection receptors at different time points. These findings broaden the current understanding of the microgravity effects on rAAV, with significant implications in gene therapy and the mechanisms of increased virus pathogenicity under space microgravity.

    Noninvasive suspicious liquid detection using wireless signals

    Conventional liquid detection instruments are very expensive and not conducive to large-scale deployment. In this work, we propose a method for detecting and identifying suspicious liquids based on the dielectric constant by utilizing the radio signals at a 5G frequency band. There are three major experiments: first, we use wireless channel information (WCI) to distinguish between suspicious and nonsuspicious liquids; then we identify the type of suspicious liquids; and finally, we distinguish the different concentrations of alcohol. The K-Nearest Neighbor (KNN) algorithm is used to classify the amplitude information extracted from the WCI matrix to detect and identify liquids, which is suitable for multimodal problems and easy to implement without training. The experimental result analysis showed that our method could detect more than 98% of the suspicious liquids, identify more than 97% of the suspicious liquid types, and distinguish up to 94% of the different concentrations of alcohol

    Sensitizing Leukemia Stem Cells to NF-ÎşB Inhibitor Treatment in Vivo by Inactivation of Both TNF and IL-1 Signaling

    We previously reported that autocrine TNF-α (TNF) is responsible for JNK pathway activation in a subset of acute myeloid leukemia (AML) patient samples, providing a survival/proliferation signaling parallel to NF-κB in AML stem cells (LSCs). In this study, we report that most TNF-expressing AML cells (LCs) also express another pro-inflammatory cytokine, IL1β, which acts in a parallel manner. TNF was produced primarily by LSCs and leukemic progenitors (LPs), whereas IL1β was mainly produced by partially differentiated leukemic blasts (LBs). IL1β also stimulates an NF-κB-independent pro-survival and proliferation signal through activation of the JNK pathway. We determined that co-inhibition of signaling stimulated by both TNF and IL1β synergizes with NF-κB inhibition in eliminating LSCs both ex vivo and in vivo.     Warm It Up with Love: The Effect of Physical Coldness on Liking of Romance Movies

    Are romance movies more desirable when people are cold? Building on research on (bodily) feeling-as-information and embodied cognition, we hypothesize that physical coldness activates a need for psychological warmth, which in turn leads to an increased liking for romance movies. Four laboratory experiments and an analysis of online movie rental data provide support for our hypothesis. Specifically, studies 1A and 1B show that physical coldness increases the liking of and willingness to pay for romance movies. Study 2 shows that the effect of physical coldness on liking of romance movies only occurs for people who associate romance movies with psychological warmth. Study 3 shows that people correct for the influence of physical coldness on their liking of romance movies when physical coldness is made salient. In study 4, using data on online movie rentals and historical temperature, we found a negative relationship between weather temperature and preference for romance movies. The movie industry is characterized by strong seasonality in viewer demand (Einav 2007). In the United States, the majority of total box office revenues are generated around six major holidays (Valentine’s Day, Memorial Day, Independence Day, Labor Day, Thanksgiving, and Christmas; Foutz and Kadiyali 2006). This highlights the importance of studios taking seasonality into account in timing movie releases, which is one of the most important decisions in the movie industry considering that the opening weekend typically accounts for 40 % and the first four weekends 80% of a movie’s box office (Foutz and Kadiyali 2006).     Optimization of AICD water control completion technology for horizontal well in bottom water gas reservoir

    At present, adaptive inflow control (AICD) water-controlled completion technology is mainly applied to reservoir water control. In view of the gas reservoir AICD water control completion technology research is still blank, this paper carried out a suitable for gas reservoir water control water control mechanism of the new type runner AICD and flow characteristics analysis, X oilfield bottom water reservoir is established new port type AICD water control completion. The structure parameter combination of the new AICD was analyzed and optimized by orthogonal test, and the water control effect of the new AICD water control completion was analyzed. The results show that compared with conventional perforation completions, the new AICD controlled water completions cause little bottom hole pressure loss and have little effect on condensate production. New type runner AICD completion can prolong the bottom water reservoir water control in low yielding water gas recovery period of about 365 days, predict 20 years of accumulated water rate reduced about 27%, water saturation near wellbore area is generally lower, has good effect of water control.
