    Investigation of the Stock Structure of Atlantic Walrus (Odobenus rosmarus rosmarus) in Canada and Greenland Using Dental Pb Isotopes Derived from Local Geochemical Environments

    The chemical composition of animal tissues such as teeth appears to reflect an individual's exposure to its geochemical environment. In this study, the lead (Pb) isotope composition of dental cementum was used to investigate the stock structure of Atlantic walrus (Odobenus rosmarus rosmarus) in the Canadian Arctic and Greenland. The 12 communities providing walrus samples for this study represent most of the Canadian and Greenlandic villages where walrus still form an important part of the traditional Inuit diet. Significant differences between locations in mean Pb isotope ratios and the limited overlap of the ranges of values indicate that each village harvested walrus herds that exploited substantially different geological/geographical habitats. This geographic segregation based on isotopic signatures suggests that most walrus stocks (i.e., the groups of walrus that interact with hunters at each community) are more localized in their range than previously thought. 208Pb/207Pb and 208Pb/204Pb were the most important stock discriminators, reflecting the influence of local geological Th/U composition (i.e., 208Pb) on Pb isotope composition in walrus teeth. 204Pb-based isotope ratios in walrus were consistently higher (more radiogenic) and more homogeneous than those in regional terrestrial bedrock, a difference probably due to selective leaching of radiogenic Pb from mineral phases into seawater and mixing during weathering and transport. Dental Pb isotope signatures may have widespread application to stock discrimination of other coastal marine mammal species.La composition chimique de tissus animaux tels que les dents semble reflĂ©ter l'exposition d'un individu Ă  son milieu gĂ©ochimique.     Understanding service demand for mental health among Australians aged 16 to 64 years according to their possible need for treatment

    Background: To inform decisions about mental health resource allocation, planners require reliable estimates of people who report service demand (i.e. people who use or want mental health services) according to their level of possible need. Methods: Using data on 6915 adults aged 16-64 years in Australia's 2007 National Survey of Mental Health and Wellbeing, we examined past-year service demand among respondents grouped into four levels of possible need: (a) 12-month mental disorder; (b) lifetime but no 12-month mental disorder; (c) any other indicator of possible need (12-month symptoms or reaction to stressful event, or lifetime hospitalisation); (d) no indicator of possible need. Multivariate logistic regression analyses examined correlates of service demand, separately for respondents in each of levels 1-3. Results: Sixteen per cent of Australian adults reported service demand, of whom one-third did not meet criteria for a 12-month mental disorder (equivalent to 5.7% of the adult population). Treatment patterns tended to follow a gradient defined by level of possible need. For example, service users with a 12-month disorder received, on average, 1.6-3.9 times more consultations than their counterparts in other levels of possible need, and had 1.9-2.2 times higher rates of psychologist consultation. Service users with a lifetime but not 12-month disorder or any other indicator of need consumed a similar average number of services to people with mild 12-month mental disorders, but received relatively fewer services involving the mental health sector. Service demand was associated with increased suicidality and psychological distress in all levels of possible need examined, and with poorer clinical and functional status for those with 12-month or lifetime disorders. Conclusions: Many Australians reporting service demand do not meet criteria for a current mental disorder, but may require services to maintain recovery following a past episode or because they are experiencing symptoms and significant psychological distress

    Invited to dinner, but not to the table: web content accessibility evaluation for persons with disabilities

    Disability is very common and yet not well understood within sub-Saharan African countries. There has been growing attention to the use of research evidence to improve social inclusion of persons living with disabilities. This article reports on a process that can be used to monitor and evaluate evidence databases to encourage improvements in website and content accessibility for people with disabilities. We examined five evidence communities’ online databases by: (1) assessing the accessibility of these website databases; and (2) assessing the resources within these websites. Finally, we aimed to provide feedback from the evaluation to these evidence databases. We carried out a cross-sectional study of the online evidence databases using the Web Content Accessibility Guidelines – a universal standard for web content accessibility assessment. We assessed access to the databases using a purposive sample of 25 resources within them. Resources are meant to improve practice, policy and decision making for all, including people with disabilities. They include systematic reviews, reports and articles. Accessibility is being able to obtain, understand and use resources; addressing barriers that could hinder this is important. Even though these evidence databases are considered as enabling inclusion and diversity within the evidence ecosystem, their contents are not fully accessible to people with disabilities, and they only partially met the recommendations of the Web Content Accessibility Guidelines

    Controls on the location of compressional deformation on the NW European margin

    The distribution of Cenozoic compressional structures along the NW European margin has been compared with maps of the thickness of the crystalline crust derived from a compilation of seismic refraction interpretations and gravity modelling, and with the distribution of high-velocity lower crust and/or partially serpentinized upper mantle detected by seismic experiments. Only a subset of the mapped compressional structures coincide with areas susceptible to lithospheric weakening as a result of crustal hyperextension and partial serpentinization of the upper mantle. Notably, partially serpentinized upper mantle is well documented beneath the central part of the southern Rockall Basin, but compressional features are sparse in that area. Where compressional structures have formed but the upper mantle is not serpentinized, simple rheological modelling suggests an alternative weakening mechanism involving ductile lower crust and lithospheric decoupling. The presence of pre-existing weak zones (associated with the properties of the gouge and overpressure in fault zones) and local stress magnitude and orientation are important contributing factors

    Assembly of the Inner Perivitelline Layer, a Homo log of the Mammalian Zona Pellucida: An Immunohistochemical and Ultrastructural Study

    The avian inner perivitelline layer (IPVL), a homologous structure to the mammalian zona pellucida, is deposited between the granulosa cells and the oocyte cell membrane during folliculogenesis. The glycoprotein meshwork of the IPVL forms a 3-dimensional matrix and possesses important functions in the fertilization process: it contributes to the binding of avian spermatozoa to the oocyte and induces acrosomal exocytosis. In contrast to the zona pellucida of mammals, the IPVL does not prevent the physiological polyspermy found in birds. Previous studies have shown that in the Japanese quail (Cotumix japonica) at least 5 glycoproteins are constituents of the IPVL (ZP1, ZP2, ZP3, ZP4, and ZPD). In this study, we investigated the spatiotennporal assembly pattern of the IPVL during folliculogenesis using immunohistochemical and ultrastructural methods. The obtained results clearly show that these glycoproteins are incorporated into the IPVL at distinct points during follicular development, supporting the hypothesis that ZP2 and ZP4 form a type of prematrix into which ZP1, ZP3, and ZPD are integrated at a later stage of development.     An overview of the Upper Paleozoic-Mesozoic stratigraphy of the NE Atlantic region

    This study describes the distribution and stratigraphic range of the Upper Paleozoic–Mesozoic succession in the NE Atlantic region, and is correlated between conjugate-margins and along the axis of the NE Atlantic rift system. The stratigraphic framework has yielded important new constraints on the timing and nature of sedimentary basin development in the NE Atlantic, with implications for rifting and the breakup of the Pangaean supercontinent. From a regional perspective, the Permian–Triassic succession records a northward transition from an arid interior to a passively-subsiding, mixed carbonate/siliciclastic shelf margin. A Late Permian–earliest Triassic rift pulse has regional expression in the stratigraphic record. A fragmentary paralic to shallow-marine Lower Jurassic succession reflects Early Jurassic thermal subsidence and mild extensional tectonism; this was interrupted by widespread Mid-Jurassic uplift and erosion, and followed by an intense phase of Late Jurassic rifting in some (but not all) parts of the NE Atlantic region. The Cretaceous succession is dominated by thick basinal-marine deposits, which accumulated within and along a broad zone of extension and subsidence between Rockall and NE Greenland. There is no evidence for a substantive and continuous rift system along the proto-NE Atlantic until the Late Cretaceous

It is based on the Finnis-Sinclair second moment tight binding approach, but incorporates two independent bands on each atom. The potential is cast in a form suitable for large-scale molecular dynamics, the computational cost being the evaluation of short ranged pair potentials.     The effect of common genetic variation in 11ÎČ-hydroxysteroid dehydrogenase type 1 on hypothalamic-pituitary-adrenal axis activity and incident depression

    Background: Accumulating evidence suggests that hyperactivity of the hypothalamic-pituitary-adrenal axis (HPA axis) is involved in depression. 11ÎČ-Hydroxysteroid dehydrogenase type 1 (11ÎČ-HSD1) converts inert cortisone to active cortisol and is implicated in HPA axis regulation in animal studies. The aim of our study was to identify polymorphisms in 11ÎČ-HSD1 gene (HSD11B1) with consistent associations with increased HPA axis activity and relate those polymorphisms to depression. Methods: Twelve single-nucleotide polymorphisms (SNPs), including 11 tagging SNPs, were selected using the HapMap database and genotyped in 4228 participants of the population-based Rotterdam Study. The outcome measures were salivary cortisol levels after awakening, 30 min later, at 1700 h, at bedtime, and plasma levels of androstenedione (in women only). SNPs that were significantly associated with cortisol as well as androstenedione levels were also related to incident depression. Results: rs11119328 was associated with higher cortisol saliva samples collected at bedtime as well as higher androstenedione levels (P value after correction for multiple testing: 0.01 and 0.04, respectively). Carriers of this polymorphism had an increased risk of an incident depression (hazard ratio 1.28, 95% confidence interval 1.03-1.59). Two other SNPs, which were in high linkage disequilibrium with rs11119328, were related to higher cortisol levels but not with androstenedione levels. Conclusions: We identified one SNP, which was associated with increased salivary cortisol levels at nadir as well as higher androstenedione levels. Moreover, this SNP was also associated with a higher risk of an incident depression. This suggests that 11ÎČ-HSD1 is implicated in human HPA axis regulation and susceptibility to depression.     The Glasgow BeneïŹt Inventory: a systematic review of the use and value of an otorhinolaryngological generic patient-recorded outcome measure

    The Glasgow Benefit Inventory (GBI) is a validated, generic patient-recorded outcome measure widely used in otolaryngology to report change in quality of life post-intervention.To date, no systematic review has made (i) a quality assessment of reporting of Glasgow Benefit Inventory outcomes; (ii) a comparison between Glasgow Benefit Inventory outcomes for different interventions and objectives; (iii) an evaluation of subscales in describing the area of benefit; (iv) commented on its value in clinical practice and research.Systematic review.'Glasgow Benefit Inventory' and 'GBI' were used as keywords to search for published, unpublished and ongoing trials in PubMed, EMBASE, CINAHL and Google in addition to an ISI citation search for the original validating Glasgow Benefit Inventory paper between 1996 and January 2015.Papers were assessed for study type and quality graded by a predesigned scale, by two authors independently. Papers with sufficient quality Glasgow Benefit Inventory data were identified for statistical comparisons. Papers with 50% and gave sufficient Glasgow Benefit Inventory total and subscales for meta-analysis. For five of the 11 operation categories (vestibular schwannoma, tonsillectomy, cochlear implant, middle ear implant and stapes surgery) that were most likely to have a single clear clinical objective, score data had low-to-moderate heterogeneity. The value in the Glasgow Benefit Inventory having both positive and negative scores was shown by an overall negative score for the management of vestibular schwannoma. The other six operations gave considerable heterogeneity with rhinoplasty and septoplasty giving the greatest percentages (98% and 99%) most likely because of the considerable variations in patient selection. The data from these operations should not be used for comparative purposes. Five papers also reported the number of patients that had no or negative benefit, a potentially a more clinically useful outcome to report. Glasgow Benefit Inventory subscores for tonsillectomy were significantly different from ear surgery suggesting different areas of benefitThe Glasgow Benefit Inventory has been shown to differentiate the benefit between surgical and medical otolaryngology interventions as well as 'reassurance'. Reporting benefit as percentages with negative, no and positive benefit would enable better comparisons between different interventions with varying objectives and pathology. This could also allow easier evaluation of factors that predict benefit. Meta-analysis data are now available for comparison purposes for vestibular schwannoma, tonsillectomy, cochlear implant, middle ear implant and stapes surgery. Fuller report of the Glasgow Benefit Inventory outcomes for non-surgical otolaryngology interventions is encouraged

    Alpha-B-Crystallin overexpression is sufficient to promote tumorigenesis and metastasis in mice

    Background: αB-Crystallin is a heat shock chaperone protein which binds to misfolded proteins to prevent their aggregation. It is overexpressed in a wide-variety of cancers. Previous studies using human cancer cell lines and human xenograft models have suggested potential tumor promoter (oncogene) roles for αB-Crystallin in a widespectrum of cancers. Methods: To determine the causal relationship between CRYAB overexpression and cancer, we generated a Cryab overexpression knock-in mouse model and monitor them for development of spontaneous and carcinogen (DMBA)- induced tumorigenesis. In order to investigate the mechanism of malignancies observed in this model multiple techniques were used such as immunohistochemical characterizations of tumors, bioinformatics analysis of publically available human tumor datasets, and generation of mouse embryonic fbroblasts (MEFs) for in vitro assays (clonogenic survival and migration assays and proteome analysis by mass-spectrometry). Results: This model revealed that constitutive overexpression of Cryab results in the formation of a variety of lethal spontaneous primary and metastatic tumors in mice. In vivo, the overexpression of Cryab correlated with the upregulation of epithelial-to-mesenchymal (EMT) markers, angiogenesis and some oncogenic proteins including Basigin. In vitro, using E1A/Ras transformed MEFs, we observed that the overexpression of Cryab led to the promotion of cell survival via upregulation of Akt signaling and downregulation of pro-apoptotic pathway mediator JNK, with subsequent attenuation of apoptosis as assessed by cleaved caspase-3 and Annexin V staining.
