Nitrogen Fertilizer and Growth Regulator Impacts on Tuber Deformity, Rot, and Yield for Russet Potatoes
Potatoes (Solanum tuberosum) are an important high-value commodity for producers in the Mid-Atlantic Region. Current production recommendations were based on white potatoes, and practices for Russet potatoes have not been researched in this region. The objective of this study was to test impacts of N rate (0, 67, 134, 201, and 268 kg N ha−1), N application timing (100% applied with planter, 2-way split (30% with planter and 70% band applied approximately 30 days after planting at dragoff), and three-way split (30% with planter, 50% band applied prior to drag-off, and 20% band applied at first sight of bloom)), and additions of the growth regulator maleic hydrazide (MH-30). We tested “Goldrush” and “Norkotah” Russet potato varieties on marketability, total yield, tuber deformity, and tuber soft rot incidence for sandy loam soils in the Mid-Atlantic. Overall, year variations were significant with substantial rots (up to 86.5%) occurring in year 3. Maleic hydrazide and N application timing had little consistent effect on any tested parameter. Nitrogen rate and variety factors had the greatest impacts on deformity, tuber rots, and yields for Russet potatoes in the Mid-Atlantic Region with 134 kg N ha−1 producing the highest total yields in 2009 and 2010. If tuber rots can be controlled, both “Goldrush” and “Norkotah” are acceptable varieties under the Mid-Atlantic production practices
Vegetable Soybean and Its Seedling Emergence in the United States
Vegetable soybean or edamame is a specialty soybean (Glycine max (L.) Merr.). Unlike grain-type soybean (mainly for oil and source of protein in animal feeds), edamame pods are harvested at a green and immature stage, and beans are consumed by humans as a vegetable. While originally from China, edamame has recently gained much-increased popularity and expanded market needs in the US. However, domestic edamame production is limited in the US because at least 70% of the edamame consumed is imported. Poor seed germination and seedling emergence are one of the major problems in US edamame production. This review focuses on the introduction of edamame, a high-value niche crop, and its low emergence issue in production. Here, we provide a comprehensive exploration of the factors that influence edamame germination and emergence, including the intrinsic factors related to seeds (seed and seedling characteristics), and extrinsic factors related to the biotic (soil/seed-borne diseases) and abiotic (seedbed physical components as well as their interaction with climate) stresses. This information will help farmers and plant breeders to better understand the causes of the poor edamame emergence and may provide a foundation for improved field management of edamame, to increase production of this valuable specialty crop
Emergent Continuum Spacetime from a Random, Discrete, Partial Order
There are several indications (from different approaches) that Spacetime at
the Plank Scale could be discrete. One approach to Quantum Gravity that takes
this most seriously is the Causal Sets Approach. In this approach spacetime is
fundamentally a discrete, random, partially ordered set (where the partial
order is the causal relation). In this contribution, we examine how timelike
and spacelike distances arise from a causal set (in the case that the causal
set is approximated by Minkowski spacetime), and how one can use this to obtain
geometrical information (such as lengths of curves) for the general case, where

Agricultural Practices Influence Salmonella Contamination and Survival in Pre-harvest Tomato Production
Between 2000 and 2010 the Eastern Shore of Virginia was implicated in four Salmonella outbreaks associated with tomato. Therefore, a multi-year study (2012–2015) was performed to investigate presumptive factors associated with the contamination of Salmonella within tomato fields at Virginia Tech’s Eastern Shore Agricultural Research and Extension Center. Factors including irrigation water sources (pond and well), type of soil amendment: fresh poultry litter (PL), PL ash, and a conventional fertilizer (triple superphosphate – TSP), and production practices: staked with plastic mulch (SP), staked without plastic mulch (SW), and non-staked without plastic mulch (NW), were evaluated by split-plot or complete-block design. All field experiments relied on naturally occurring Salmonella contamination, except one follow up experiment (worst-case scenario) which examined the potential for contamination in tomato fruits when Salmonella was applied through drip irrigation. Samples were collected from pond and well water; PL, PL ash, and TSP; and the rhizosphere, leaves, and fruits of tomato plants. Salmonella was quantified using a most probable number method and contamination ratios were calculated for each treatment. Salmonella serovar was determined by molecular serotyping. Salmonella populations varied significantly by year; however, similar trends were evident each year. Findings showed use of untreated pond water and raw PL amendment increased the likelihood of Salmonella detection in tomato plots. Salmonella Newport and Typhimurium were the most frequently detected serovars in pond water and PL amendment samples, respectively. Interestingly, while these factors increased the likelihood of Salmonella detection in tomato plots (rhizosphere and leaves), all tomato fruits sampled (n = 4800) from these plots were Salmonella negative. Contamination of tomato fruits was extremely low (< 1%) even when tomato plots were artificially inoculated with an attenuated Salmonella Newport strain (104 CFU/mL). Furthermore, Salmonella was not detected in tomato plots irrigated using well water and amended with PL ash or TSP. Production practices also influenced the likelihood of Salmonella detection in tomato plots. Salmonella detection was higher in tomato leaf samples for NW plots, compared to SP and SW plots. This study provides evidence that attention to agricultural inputs and production practices may help reduce the likelihood of Salmonella contamination in tomato fields
Absence of alpha-synuclein affects dopamine metabolism and synaptic markers in the striatum of aging mice
Despite numerous evidences for neurotoxicity of overexpressed α-synuclein, a protective function was suggested for endogenous α-synuclein and other members of the synuclein family. This protective role is most important for and evident in presynaptic terminals, where synucleins are normally accumulated. However, mice lacking synucleins display no adverse phenotype. In particular, no significant changes in striatal dopamine metabolism and only subtle deficit of dopaminergic neurons in the substantia nigra were found in juvenile or adult mice. To assess whether aging and synuclein deficiency may have additive detrimental effect on the nigrostriatal system, we studied dopaminergic neurons of the substantia nigra and their striatal synapses in 24–26-month-old α-synuclein and γ-synuclein null mutant mice. Significant ∼36% reduction of the striatal dopamine was found in aging α-synuclein, but not γ-synuclein null mutant mice when compared to age-matching wild type mice. This was accompanied by the reduction of TH-positive fibers in the striatum and decrease of striatal levels of TH and DAT. However, no progressive loss of TH-positive neurons was revealed in the substantia nigra of synuclein-deficient aging animals. Our results are consistent with a hypothesis that α-synuclein is important for normal function and integrity of synapses, and suggest that in the aging nervous system dysfunction of this protein could become a predisposition factor for the development of nigrostriatal pathology
Microbial Quality of Agricultural Water Used in Produce Preharvest Production on the Eastern Shore of Virginia
Several produce-borne outbreaks have been associated with the use of contaminated water during preharvest applications. Salmonella has been implicated in a number of these outbreaks. The purpose of this study was to evaluate the microbial quality of agricultural surface water used in preharvest production on the Eastern Shore of Virginia in accordance with the Food Safety Modernization Act's Produce Safety Rule water standards. The study also examined the prevalence, concentration, and diversity of Salmonella in those water sources. Water samples (1 L) from 20 agricultural ponds were collected during the 2015 and 2016 growing seasons ( n = 400). Total aerobic bacteria, total coliforms, and Escherichia coli were enumerated for each sample. Population levels of each microorganism were calculated per 100-mL sample and log transformed, when necessary. Samples (250 mL) were also enriched for Salmonella. Presumptive Salmonella isolates were confirmed by PCR ( invA gene) and were serotyped. In 2016, the concentration of Salmonella in each sample was also estimated by most probable number (MPN). Indicator bacteria and environmental and meteorological factors were analyzed for their association with the detection of a Salmonella-positive water sample by using logistic regression analysis. Seventeen of the 20 ponds met the Food Safety Modernization Act's Produce Safety Rule standards for production agricultural water. Three ponds did not meet the standards because the statistical threshold value exceeded the limit. Salmonella was detected in 19% of water samples in each year (38 of 200 in 2015 and 38 of 200 in 2016). Of the 118 Salmonella isolates serotyped, 14 serotypes were identified with the most prevalent being Salmonella Newport. E. coli concentration, farm, and total aerobic bacteria concentration were significantly associated with the likelihood of detecting a Salmonella-positive sample The average concentration of Salmonella in all samples was 4.44 MPN/100 mL, with the limit of detection being 3.00 MPN/100 mL. The highest concentration of Salmonella was 93.0 MPN/100 mL. These data will assist in a better understanding of the risks that production water poses to produce contamination events

Herpesvirus surveillance and discovery in zoo-housed ruminants.
Gammaherpesvirus infections are ubiquitous in captive and free-ranging ruminants and are associated with a variety of clinical diseases ranging from subclinical or mild inflammatory syndromes to fatal diseases such as malignant catarrhal fever. Gammaherpesvirus infections have been fully characterized in only a few ruminant species, and the overall diversity, host range, and biologic effects of most are not known. This study investigated the presence and host distribution of gammaherpesviruses in ruminant species at two facilities, the San Diego Zoo and San Diego Zoo Safari Park. We tested antemortem (blood, nasal or oropharyngeal swabs) or postmortem (internal organs) samples from 715 healthy or diseased ruminants representing 96 species and subspecies, using a consensus-based herpesvirus PCR for a segment of the DNA polymerase (DPOL) gene. Among the 715 animals tested, 161 (22.5%) were PCR and sequencing positive for herpesvirus, while only 11 (6.83%) of the PCR positive animals showed clinical signs of malignant catarrhal fever. Forty-four DPOL genotypes were identified of which only 10 have been reported in GenBank. The data describe viral diversity within species and individuals, identify host ranges of potential new viruses, and address the proclivity and consequences of interspecies transmission during management practices in zoological parks. The discovery of new viruses with wide host ranges and presence of co-infection within individual animals also suggest that the evolutionary processes influencing Gammaherpesvirus diversity are more complex than previously recognized

Soil can be a route for contamination of fresh fruits and vegetables. While growers routinely manage soil nutrient levels, little research exists on the synergistic or antagonistic effects of soil nutrients on foodborne pathogens. Data on foodborne pathogen prevalence in unamended soils is also relatively limited in literature. This study evaluated foodborne pathogen prevalence (Salmonella, Listeria monocytogenes) and concentration of indicator bacteria (total coliforms, generic Escherichia coli) in agricultural soils, and characterized associations between soil properties (e.g., macro- and micro-nutrient levels) and each microbial target. Three Virginia produce farms, representing different regions and soil types, were sampled four times over 1 year (October 2021–November 2022). For each individual farm visit, composite soil samples were collected from 20 sample sites (25 m2) per farm per visit for microbial and nutrient analysis (n = 240). Samples (25 g) were processed for Listeria spp. and Salmonella using a modified FDA BAM method; samples (5 g) were enumerated for generic E. coli and total coliforms (TC) using Petrifilm. Presumptive Listeria spp. and Salmonella isolates were confirmed by PCR using the sigB and invA genes, respectively. Soil nutrients from each sample were tested and evaluated for their association with each microbial target by Bayesian Mixed Models. Salmonella prevalence was 4.2% (10/240), with 90% (9/10) recovered on Farm C. Listeria spp. and L. monocytogenes prevalence were 10% (24/240) and 2.5% (6/240), respectively. When samples were positive for generic E. coli (107/240), the average concentration was 1.53 ± 0.77 log10 CFU/g. Soil pH was positively associated with L. monocytogenes [Odds Ratio (OR) = 5.5] and generic E. coli (OR = 4.9) prevalence. There was no association between Salmonella prevalence and any evaluated factor; however, Salmonella was 11.6 times more likely to be detected on Farm C, compared to other farms. Results show pathogen prevalence was relatively low in unamended soils, and that factors influencing prevalence and concentration varied by microbial target and farm.</p
