FORMULATION DEVELOPMENT AND STATISTICAL OPTIMIZATION OF IVABRADINE HYDROCHLORIDE FLOATING PULSATILE PELLETS BY FLUIDIZED BED COATING TECHNIQUE
ABSTRACTObjective: The objective of the current work was to develop Ivabradine hydrochloride (HCl) floating pulsatile pellets containing drug loaded calciumalginate pellets coated with pH-dependent polymer Eudragit S100 oil dispersion.Methods: Fluidized bed coating technique was used to develop pellets. A 2factorial design was employed to study the effect of independent variables(inlet air temperature, spray rate), on dependent variables (% entrapment efficiency, % friability, and average particle size). Optimization was doneby fitting experimental data to the software program (Minitab). Obtained pellets were subjected to different evaluation parameters which are criticalin the development of the dosage form. An in vitro lag phase study was carried out for all batches in simulated gastric fluid (0.1N HCl) for 5 hrs andin vitro drug release study was carried out for optimized batch (B4) of two different sizes (10/12#, 12/16#) in simulated intestinal fluid (pH 7.4phosphate buffer).2 Results: The optimized batch (B4) showed satisfactory % entrapment efficiency of 92.66±1.52; % friability of 0.57±0.03; and average particle sizeof 1424±16 (µm). All batches maintained lag phase for 5 hrs in 0.1N HCl. An optimized batch of two different sizes exhibited a burst release within30 minutes in simulated intestinal fluid with no significant difference in release rate constant (*p>0.05) and followed first order kinetics.Conclusion: Thus, ivabradine HCl floating pulsatile pellets was successfully developed for treating angina pectoris which is an underlying cause ofheart attack by fluidized bed coating technique employing factorial design.Keywords: Ivabradine hydrochloride, Sodium alginate, Eudragit S100, Pellets, Fluidized bed coating, Optimization, Central composite desig

ANTIOXIDANT STUDIES OF ONE AYURVEDIC MEDICINE ASWAGANDHARISHTAM
Objective: The present study deals with the antioxidant study of one Ayurvedic medicine Aswagandhrishtam by three different methods, namely reducing power, 2,2-diphenyl-1-picrylhydrazyl (DPPH), and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assays.Methods: Aswagandharishtam, which is a liquid medicine, was taken as such at various concentrations for all the three assays.Results: The results show that Aswagandharistam has good antioxidant potential when compared with ascorbic acid as standard. The IC50 values of reducing power assay were 250.142, that of DPPH were 103.607, and of ABTS assay were 197.79 as compared with that of ascorbic acid being 19.59.Conclusion: All the three assays indicated that Aswagandharishtam showed very good antioxidant results
Dynamic hip screw technique in the management of trochanteric fracture
Background: Study was conducted to find the results of dynamic hip screw in the management of trochanteric fracture by analyzing the factors which influence post-operative mobility.Methods: Study was conducted in the department of orthopedics, GSL Medical College. Individuals >18 years, both genders who were diagnosed having trochanteric type I and II Boyd and Griffin stable fractures were included in the study. All surgeries were performed under spinal anesthesia, internal fixation with dynamic hip screw and 135o angled blade plate. Injectable third generation cephalosporins were used 24 hours preoperatively, intra- operatively and 5 days post-operatively, and oral antibiotics till suture removal. Patients allowed to sit on bed on 2nd and 3rd day and static quadriceps exercises were started from 2nd day onwards, hip and knee flexion exercises from 6 or 7th day and weight bearing walking form 10th day.Results: The average age was of the participants was 61.53 years, ranged between 41 to 80 years; 65% were female participants and 35% were male patients. In the study, 20 (50%) patients had right side fracture and left sided affection of trochanteric fracture to the remaining 50%. Most of the patients (67.5%) in this study were classified as type II Boyd and Griffin criteria, and 32.5% were type I. The clinical and functional outcome was calculated using the Kyle's criteria; 25% (10) showed excellent response, followed by good (50%), fair (15%) and poor (10%) results.Conclusions: Dynamic hip screw is the operative treatment of choice for stable trochanteric fractures. However, studies on large sample for long time are recommended
Efficiency Improvement of Rotary Compressor by Improving the Discharge path through Simulation
To help raise consumer awareness on saving energy and create more energy efficient appliances, the government is upgrading the requirements of star rating of air conditioners. The enhanced star rating in air conditioners will significantly support the energy conservation and protect the environment by reducing greenhouse gas emissions in the fight against climate change. The increasing demand for star rated air-conditioners is compelling the compressor designers to develop & optimize more energy-efficient components. This paper deals with discharge port optimization in a rotary compressor, which is heart of an air-conditioner. The existing discharge port is studied & analyzed for the performance in the aspect of its capacity, EER, mass flow rate, over pressure power, area of PV diagram, motor power, valve functioning & stability. The port is redesigned to obtain improved performance, by analyzing the compressor performance parameters. Analysis is performed to study the stresses on the discharge valve and valve lift. Computational fluid dynamics is used to observe the flow behavior & pressure phenomenon in the rotary compressor, since the compressor shell accumulates the discharge gas pressure. The results are validated with the test and improved EER is observed
A case series of sarcomatoid transitional cell carcinoma of renal pelvis and collecting duct carcinoma of kidney
Sarcomatoid transitional cell carcinoma (TCC) of the renal pelvis and collecting duct carcinoma (CDC) of the kidney are rare types of malignant renal tumors with aggressive behavior. Here, we report a case series of these tumors, in males of age 65 years and 72 years, respectively. In both the cases, the left kidney was affected and they underwent the left-sided radical nephroureterectomy. Histologically, sarcomatoid TCC showed two components with invasive TCC and areas with spindle cells and osteoclastic type of giant cells. The immunohistochemistry showed diffuse positivity for CK7 and negativity for CK20 and p63 in both the components. CDC showed tubule-papillary patterns with marked desmoplasia. The patients were disease free after a follow-up of 12 months and 2½ years, respectively, without any adjuvant therapy
