    Diffusion Coefficients of Copper Chloride in Aqueous Solutions at 298.15 K and 310.15 K

    Mutual diffusion coefficients (interdiffusion coefficients) and molar conductivities have been measured for copper(II) chloride in water at 298.15 K and 310.15 K at concentrations between 0.005 mol·dm-3 and 0.05 mol·dm-3. The diffusion coefficients were measured using a conductometric cell. The experimental data are discussed on the basis of the Onsager−Fuoss model. The Nernst diffusion coefficients derived from diffusion (1.297 × 10-9 and 1.690 × 10-9) m2·s-1 and from conductance (1.282 × 10-9 and 1.663 × 10-9) m2·s-1 at two temperatures (298.15 K and 310.15 K, respectively) are in good agreement

    Targeting multiple cellular populations is of high therapeutic relevance for the tackling of solid tumors heterogeneity. Herein, the ability of pegylated and pH-sensitive liposomes, functionalized with the nucleolin-binding F3 peptide and containing doxorubicin (DXR)/C6-ceramide synergistic combination, to target, in vitro, ovarian cancer, including ovarian cancer stem cells (CSC), was assessed. The underlying molecular mechanism of action of the nucleolin-mediated intracellular delivery of C6-ceramide to cancer cells was also explored. The assessment of overexpression of surface nucleolin expression by flow cytometry was critical to dissipate differences identified by Western blot in membrane/cytoplasm of SKOV-3, OVCAR-3 and TOV-112D ovarian cancer cell lines. The former was in line with the significant extent of uptake into (bulk) ovarian cancer cells, relative to non-targeted and non-specific counterparts. This pattern of uptake was recapitulated with putative CSC-enriched ovarian SKOV-3 and OVCAR-3 sub-population (EpCAMhigh/CD44high). Co-encapsulation of DXR:C6-ceramide into F3 peptide-targeted liposomes improved cytotoxic activity relative to liposomes containing DXR alone, in an extent that depended on the intrinsic resistance to DXR and on the incubation time. The enhanced cytotoxicity of the targeted combination was mechanistically supported by the downregulation of PI3K/Akt pathway by C6-ceramide, only among the nucleolin-overexpressing cancer cells presenting a basal p-Akt/total Akt ratio lower than 1

    Reducing the environmental impact of surgery on a global scale: systematic review and co-prioritization with healthcare workers in 132 countries

    Abstract Background Healthcare cannot achieve net-zero carbon without addressing operating theatres. The aim of this study was to prioritize feasible interventions to reduce the environmental impact of operating theatres. Methods This study adopted a four-phase Delphi consensus co-prioritization methodology. In phase 1, a systematic review of published interventions and global consultation of perioperative healthcare professionals were used to longlist interventions. In phase 2, iterative thematic analysis consolidated comparable interventions into a shortlist. In phase 3, the shortlist was co-prioritized based on patient and clinician views on acceptability, feasibility, and safety. In phase 4, ranked lists of interventions were presented by their relevance to high-income countries and low–middle-income countries. Results In phase 1, 43 interventions were identified, which had low uptake in practice according to 3042 professionals globally. In phase 2, a shortlist of 15 intervention domains was generated. In phase 3, interventions were deemed acceptable for more than 90 per cent of patients except for reducing general anaesthesia (84 per cent) and re-sterilization of ‘single-use’ consumables (86 per cent). In phase 4, the top three shortlisted interventions for high-income countries were: introducing recycling; reducing use of anaesthetic gases; and appropriate clinical waste processing. In phase 4, the top three shortlisted interventions for low–middle-income countries were: introducing reusable surgical devices; reducing use of consumables; and reducing the use of general anaesthesia. Conclusion This is a step toward environmentally sustainable operating environments with actionable interventions applicable to both high– and low–middle–income countries

    Binary Mutual Diffusion Coefficients of Aqueous Solutions of Sucrose, Lactose, Glucose, and Fructose in the Temperature Range from (298.15 to 328.15) K

    Binary mutual diffusion coefficients measured by the Taylor dispersion method in two different laboratories (University of Naples, Federico II, Italy, and University of Coimbra, Portugal) are reported for aqueous solutions of lactose, sucrose, glucose, and fructose at various concentrations (0.001 to 0.1) mol·dm-3 and temperatures (298.15 to 328.15) K. The hydrodynamic radius and activation energy for the diffusion of aqueous sugars are calculated from those results.     Cardosins in postembryonic development of cardoon: towards an elucidation of the biological function of plant aspartic proteinases

    Summary. Following on from previous work, the temporal and spatial accumulation of the aspartic proteinases (EC 3.4.23) cardosin A and cardosin B during postembryonic seed development of cardoon (Cynara cardunculus) was studied. mRNA and protein analyses of both cardosins suggested that the proteins accumulate during seed maturation, and that cardosin A is later synthesised de novo at the time of radicle emergence. Immunocytochemistry revealed that the precursor form of cardosin A accumulates in protein bodies and cell walls. This localisation in seeds is different from that previously described for cardoon flowers, suggesting a tissue-dependent targeting of the protein. It is known that procardosins are active and may have a role in proteolysis and processing of storage proteins. However, the presence of procardosin A in seeds could be related to the proposed role of the plant-specific insert in membrane lipid conversion during water uptake and solute leakage in actively growing tissues. This is in accordance with the recently proposed bifunctional role of aspartic proteinase precursor molecules that possess a membrane-destabilising domain in addition to a protease domain. Mature cardosin B, but not its mRNA, was detected in the first hours after seed imbibition and disappeared at the time of radicle emergence. This extracellular aspartic protease has already been implicated in cell wall loosening and remodelling, and its role in seed germination could be related to loosening tissue constraints for radicle protusion. The described pattern of cardosin A and B expression suggests a finely tuned developmental regulation and prompts an analysis of their possible roles in the physiology of postembryonic development

    Mannosylglycerate is essential for osmotic adjustment in Thermus thermophilus strains HB27 and RQ-1

    Abstract We disrupted the mpgS encoding mannosyl-3-phosphoglycerate synthase (MpgS) of Thermus thermophilus strains HB27 and RQ-1, by homologous recombination, to assess the role of the compatible solute mannosylglycerate (MG) in osmoadaptation of the mutants, to examine their ability to grow in NaCl-containing medium and to identify the intracellular organic solutes. Strain HB27 accumulated only MG when grown in defined medium containing 2% NaCl; mutant HB27M9 did not grow in the same medium containing more than 1% NaCl. When trehalose or MG was added, the mutant was able to grow up to 2% of NaCl and accumulated trehalose or MG, respectively, plus amino acids. T. thermophilus RQ-1 grew in medium containing up to 5% NaCl, accumulated trehalose and lower amounts of MG. Mutant RQ-1M1 lost the ability to grow in medium containing more than 3% NaCl and accumulated trehalose and moderate levels of amino acids. Exogenous MG did not improve the ability of the organism to grow above 3% NaCl, but caused a decrease in the levels of amino acids. Our results show that MG serves as a compatible solute primarily during osmoadaptation at low levels of NaCl while trehalose is primarily involved in osmoadaptation during growth at higher NaCl levels

Their employees, particularly those exposed to second-hand smoke, are at increased risk for developing chronic respiratory diseases such as chronic obstructive pulmonary disease (COPD), asthma and lung cancer. Objectives In this work, we aimed at evaluating relationships between occupational ETS exposure, biological damage (DNA or proteome alterations) and putative respiratory dysfunctions. Materials and Methods A group of restaurants located in Lisbon has been studied, in which fine particles (smaller than 2.5”m; PM2.5), indicative of environmental tobacco smoke (ETS) contamination, were measured. After informed consent, workers were evaluated for acute exposure to ETS based on cotinine levels in urine and clinically evaluated for their respiratory health by spirometry measurements and chestpiece auscultation. Effects of ETS exposure on genotoxic lesions were evaluated by measuring DNA/chromosomes breaks in peripheral blood lymphocytes and buccal mucosa cells. Effects of ETS exposure on plasma proteins is being studied using a 2D-DIGE-MALDI-TOF/TOF approach. To achieve that goal, global proteome characterization is being carried based on the same individual plasma samples collected for genotoxic studies and were pooled according to previous criteria. Results Results have confirmed higher respirable particle levels in smoking-designated areas of those entertainment places, indicating an ETS contamination. Leukocytes from ETS-exposed-workers presented lower levels of genotoxic-induced damage in comparison with non-exposed workers, suggesting an ETS-induced stress adaption response in exposed-workers. By proteomics, we are now to investigating those workers for putative alterations on their plasma proteome to provide additional insights on the adaptative response mechanisms that might be activated by ETS exposition. Conclusion Altogether, this study provides information on indoor air quality of Lisbon smoking entertainment places, in particular ETS contamination, and may provide biomarker candidates for occupational ETS-exposure which might precede respiratory diseases on their employees.Work supported by Fundação Calouste Gulbenkian, Administração Central do Sistema de SaĂșde, FCT/PolyAnnual Funding Program and FEDER/SaudeXXI Program, Portugal.
