    Downregulation of desmuslin in primary vein incompetence

    ObjectivePrimary vein incompetence is one of the most common diseases of the peripheral veins, but its pathogenesis is unknown. These veins present obvious congenital defects, and examination of gene expression profiles of the incompetent vein specimens may provide important clues. The aim of this study was to screen for genes affecting the primary vein incompetence phenotype and test the differential expression of certain genes.MethodsWe compared gene expression profiles of valvular areas from incompetent and normal great saphenous veins at the saphenofemoral junctions by fluorescent differential display reverse-transcription polymerase chain reaction (FDD RT-PCR). Differentially expressed complimentary DNAs (cDNAs) were confirmed by Northern blotting and semi-quantitative RT-PCR. Similarity of the cDNAs sequences to GenBank sequences was determined. Gene expression status was then determined by Western blot analysis and immunohistochemical techniques.ResultsThere were >30 differentially expressed cDNA bands. Sequence analysis revealed that a cDNA fragment obviously downregulated in incompetent great saphenous vein was a portion of the messenger RNA (mRNA) encoding desmuslin, a newly discovered intermittent filament protein. Northern blotting and semi-quantitative RT-PCR analysis revealed a similar mRNA expression profile of the desmuslin gene in other samples. Western blotting and immunohistochemical techniques localized the desmuslin protein mainly in the cytoplasm of venous smooth muscle cells. The amount of desmuslin was greatly decreased in the smooth muscle cells of incompetent veins.ConclusionsThe expression of many genes is altered in primary vein incompetence. Up- or downregulation of these genes may be involved in the pathogenesis of this disease. Desmuslin expression is downregulated in the abnormal veins. Its effect on the integrity of smooth muscle cells might be related to malformation of the vein wall. Further studies are needed to investigate other differentially expressed cDNAs and the exact role of desmuslin in this disease.Clinical RelevancePrimary vein incompetence is a frequent and refractory disease of the peripheral veins. Exploring its pathogenesis may enhance our comprehension and management of this disease. We used reliable techniques to detect disease-related genes and confirmed downregulation of desmuslin in abnormal veins. Alteration of these genes might be used as disease markers or gene therapy targets

    PDE5 Overexpression in Well-Differentiated Thyroid Carcinomas Is Associated with Lymph Node Metastasis

    Overexpression of PDE5 is observed in certain human cancers, but PDE5 expression in well-differentiated thyroid carcinoma (WDTC) is unknown. We therefore examined PDE5 expression and its relationship with the clinicopathological features of WDTC. Real-time qPCR and Western blotting were performed to analyze the expression of PDE5 mRNA and protein in paired WDTC tumor and adjacent nontumor tissues. Immunohistochemistry was used to analyze the expression of PDE5 in paraffin-embedded tissues obtained from 103 cases of WDTC. Statistical analyses were performed to examine the correlation between PDE5 expression and clinicopathological features. The expression of PDE5 mRNA and protein was upregulated in WDTC lesions compared to their paired noncancerous tissues. The expression of PDE5 was significantly correlated with age (P=0.032), regional lymph node status (P=0.004), and the presence of distant metastasis (P=0.020). High PDE5 expression was more closely associated with lymph node involvement in patients over 45 years (OR = 15.60, P≤0.05). Thus, PDE5 may be a potential biomarker in WDTC, particularly in patients with regional lymph node metastasis, which is associated with disease recurrence, treatment failure, and morbidity. PDE5 expression may also help predict the prognosis and recurrence of WDTC after surgery

    Evolutionary mechanisms of the strong winds associated with an intense cold wave event and their effects on the wind power production

    Cold wave events (CWEs) often cause major economic losses and serious casualties in the cold seasons, making CWEs among the most significant types of disastrous weather. Previous studies have mainly focused on disasters due to abrupt drops in surface temperatures, with less discussion of the strong winds associated with CWEs. Based on an intense CWE that occurred in late December 2020, we investigated the evolutionary mechanisms of the associated strong winds in terms of kinetic energy (KE) budget and evaluated the effects of this CWE on wind power production based on quantitative comparisons with the mean state. The results showed that the CWE occurred under favorable background conditions, which were characterized by a southward-moving transversal trough and a southeastward-moving shortwave trough in the middle troposphere. The surface high ridge that formed around Lake Baikal and the cold front around the southern periphery of the ridge were key factors related to the CWE evolution. The positive work carried out on the horizontal wind by the pressure gradient force that linked a lower tropospheric high-pressure ridge inland and a low-pressure trough offshore and the downward momentum transportation due to the descending motions behind the cold front dominated the enhancement and sustainment of the CWE-associated strong winds. The CWE contributed to wind power production by 1) increasing the wind power density (by an average of ∼1.05-fold) and 2) improving the availability of the wind to generate power, as it reduced the percentage of zero wind power generation by ∼6.4%, while maintaining the high-wind-velocity cut-out percentage

    Location and Capacity Optimization of Waste Recycling Centers:Mathematical Models and Numerical Experiments

    With rapid urbanization growth, considerable amounts of construction waste are generated on an annual basis, posing significant economic and environmental challenges worldwide. Re-cycling construction waste is a sustainable way for waste disposal, leading to the necessity of meticulous planning of recycling centers. A well-designed plan for constructing recycling centers can effectively improve the recycling rate of construction waste while minimizing investment. This paper formulates a two-stage stochastic model for planning recycling centers with the objective of maximizing the recycling rate under different scenarios. This study comprehensively considers various uncertain factors, such as the amount of construction waste generated and the demand for recycled materials. A case study of Guangzhou is used for validation, which demonstrates the effectiveness of the developed model in planning recycling center construction. The comparison between the proposed model and a conventional mean value model shows the importance of accounting for uncertainties. Specifically, the derived results indicate that 7% more construction waste is recycled with the same investment in constructing recycling centers. Additionally, via a sensitivity analysis, valuable managerial insights on investing resources in recycling center construction are provided to decision makers. Ultimately, the research findings are expected to enhance the recycling rate of construction waste, thereby contributing to sustainable industry development

    Micro RNA expression profile and functional analysis reveal that mi R ‐382 is a critical novel gene of alcohol addiction

    Alcohol addiction is a major social and health concern. Here, we determined the expression profile of microRNAs (miRNAs) in the nucleus accumbens (NAc) of rats treated with alcohol. The results suggest that multiple miRNAs were aberrantly expressed in rat NAc after alcohol injection. Among them, miR‐382 was down‐regulated in alcohol‐treated rats. In both cultured neuronal cells in vitro and in the NAc in vivo , we identified that the dopamine receptor D1 ( Drd1 ) is a direct target gene of miR‐382. Via this target gene, miR‐382 strongly modulated the expression of DeltaFosB. Moreover, overexpression of miR‐382 significantly attenuated alcohol‐induced up‐regulation of DRD1 and DeltaFosB, decreased voluntary intake of and preference for alcohol and inhibited the DRD1‐induced action potential responses. The results indicated that miRNAs are involved in and may represent novel therapeutic targets for alcoholism. The underlying molecular causes of alcohol addiction remain unclear. Many miRNAs are found modulated in the nucleus accumbens of rats chronically treated with alcohol.     Deep Drilling in the Time Domain with DECam: Survey Characterization

    This paper presents a new optical imaging survey of four deep drilling fields (DDFs), two Galactic and two extragalactic, with the Dark Energy Camera (DECam) on the 4 meter Blanco telescope at the Cerro Tololo Inter-American Observatory (CTIO). During the first year of observations in 2021, >>4000 images covering 21 square degrees (7 DECam pointings), with \sim40 epochs (nights) per field and 5 to 6 images per night per filter in gg, rr, ii, and/or zz, have become publicly available (the proprietary period for this program is waived). We describe the real-time difference-image pipeline and how alerts are distributed to brokers via the same distribution system as the Zwicky Transient Facility (ZTF). In this paper, we focus on the two extragalactic deep fields (COSMOS and ELAIS-S1), characterizing the detected sources and demonstrating that the survey design is effective for probing the discovery space of faint and fast variable and transient sources. We describe and make publicly available 4413 calibrated light curves based on difference-image detection photometry of transients and variables in the extragalactic fields. We also present preliminary scientific analysis regarding Solar System small bodies, stellar flares and variables, Galactic anomaly detection, fast-rising transients and variables, supernovae, and active galactic nuclei.Comment: 22 pages, 17 figures, 2 tables.     Genome-Wide Association Study in East Asians Identifies Novel Susceptibility Loci for Breast Cancer

    Genetic factors play an important role in the etiology of both sporadic and familial breast cancer. We aimed to discover novel genetic susceptibility loci for breast cancer. We conducted a four-stage genome-wide association study (GWAS) in 19,091 cases and 20,606 controls of East-Asian descent including Chinese, Korean, and Japanese women. After analyzing 690,947 SNPs in 2,918 cases and 2,324 controls, we evaluated 5,365 SNPs for replication in 3,972 cases and 3,852 controls. Ninety-four SNPs were further evaluated in 5,203 cases and 5,138 controls, and finally the top 22 SNPs were investigated in up to 17,423 additional subjects (7,489 cases and 9,934 controls). SNP rs9485372, near the TGF-β activated kinase (TAB2) gene in chromosome 6q25.1, showed a consistent association with breast cancer risk across all four stages, with a P-value of 3.8×10−12 in the combined analysis of all samples. Adjusted odds ratios (95% confidence intervals) were 0.89 (0.85–0.94) and 0.80 (0.75–0.86) for the A/G and A/A genotypes, respectively, compared with the genotype G/G. SNP rs9383951 (P = 1.9×10−6 from the combined analysis of all samples), located in intron 5 of the ESR1 gene, and SNP rs7107217 (P = 4.6×10−7), located at 11q24.3, also showed a consistent association in each of the four stages. This study provides strong evidence for a novel breast cancer susceptibility locus represented by rs9485372, near the TAB2 gene (6q25.1), and identifies two possible susceptibility loci located in the ESR1 gene and 11q24.3, respectively
