    Flow induced ultrasound scattering: experimental studies

    Sound scattering by a finite width beam on a single rigid body rotation vortex flow is detected by a linear array of transducers (both smaller than a flow cell), and analyzed using a revised scattering theory. Both the phase and amplitude of the scattered signal are obtained on 64 elements of the detector array and used for the analysis of velocity and vorticity fields. Due to averaging on many pulses the signal-to-noise ratio of the phases difference in the scattered sound signal can be amplified drastically, and the resolution of the method in the detection of circulation, vortex radius, vorticity, and vortex location becomes comparable with that obtained earlier by time-reversal mirror (TRM) method (P. Roux, J. de Rosny, M. Tanter, and M. Fink, {\sl Phys. Rev. Lett.} {\bf 79}, 3170 (1997)). The revised scattering theory includes two crucial steps, which allow overcoming limitations of the existing theories. First, the Huygens construction of a far field scattering signal is carried out from a signal obtained at any intermediate plane. Second, a beam function that describes a finite width beam is introduced, which allows using a theory developed for an infinite width beam for the relation between a scattering amplitude and the vorticity structure function. Structure functions of the velocity and vorticity fields deduced from the sound scattering signal are compared with those obtained from simultaneous particle image velocimetry (PIV) measurements.     Novel synovial fluid recovery method allows for quantification of a marker of arthritis in mice

    SummaryObjectiveWe evaluated three methodologies – a calcium sodium alginate compound (CSAC), polyacrylate beads (PABs), and Whatman paper recovery (WPR) – for the ability to recover synovial fluid (SF) from mouse knees in a manner that facilitated biochemical marker analysis.MethodsPilot testing of each of these recovery vehicles was conducted using small volumes of waste human SF. CSAC emerged as the method of choice, and was used to recover and quantify SF from the knees of C57BL/6 mice (n=12), six of which were given left knee articular fractures. SF concentrations of cartilage oligomeric matrix protein (COMP) were measured by enzyme-linked immunosorbent assay.ResultsThe mean concentration ratio [(COMPleft knee)/(COMPright knee)] was higher in the mice subjected to articular fracture when compared to the non-fracture mice (P=0.026). The mean total COMP ratio (taking into account the quantitative recovery of SF) best discriminated between fracture and non-fracture knees (P=0.004).ConclusionsOur results provide the first direct evidence of accelerated joint tissue turnover in a mouse model responding to acute joint injury. These data strongly suggest that mouse SF recovery is feasible and that biomarker analysis of collected SF samples can augment traditional histological analyses in mouse models of arthritis

    Antiviral efficacy of lobucavir (BMS-180194), a cyclobutyl-guanosine nucleoside analogue, in the woodchuck (Marmota monax) model of chronic hepatitis B virus (HBV) infection

    Abstract Lobucavir (BMS-180194), a cyclobutyl-guanosine nucleoside analogue, effectively reduced WHV-viremia in chronically infected carrier woodchucks (Marmota monax) by daily per os treatment. WHV-viremia in the animals was measured by the serum content of hybridizable WHV-genomic DNA. Lobucavir, given at daily doses of 10 and 20 mg/kg body weight, reduced WHV-viremia by a 10-to 200-fold range during therapy. Lobucavir, given at 5 mg/kg, suppressed WHV-viremia by a 10-to 30-fold range, whereas a 0.5 mg/kg dose had no significant effect. WHV-viremia was also measured by hepadnaviral endogenous polymerase activity (EPA) in sera of animals treated for 6 weeks at 5 and 0.5 mg/kg. Changes in EPA in sera of lobucavir treated animals were comparable to changes in WHV DNA levels. Viremia in treated carriers recrudesced to pretreatment levels by 2 weeks of therapy cessation. These results indicated that the minimally effective antiviral daily per os dose of lobucavir in WHV-carrier woodchucks was : 5 mg/kg

    Estudio de Seeing en la Zona del Cerro Champaquí (I)

    Resumen: Se reportan los resultados del estudio más reciente de seeing que hemos realizado en la zona del Cerro Champaquí (2800 m) en el cordón de las Sierras Grandes en la Provincia de Córdoba. Se describe además, el dispositivo DIMM de alta frecuencia construido para este trabajo. Este instrumento se empleó en otros muestreos de seeing realizados en las provincias de San Juan y San Luis como parte de un proyecto de búsqueda de sitios con muy baja turbulencia atmosférica y adecuados para observaciones con óptica activa y adaptable en el rango infrarrojo cercano, iniciado en 2006. Dos de estos sitios han sido monitoreados un total de 46 noches distribuidas en seis meses del periodo 2011-2012. Los resultados indican que se darían condiciones de seeing estable, inferior al segundo de arco por prolongados períodos de tiempo, en al menos un sitio del cordón montañoso del SO de Córdoba y NE de San Luis.Abstract: Abstract. We report the results of the most recent seeing feasibility study performed in the region of Cerro Champaquí (2800 m) at the mountain range of Sierras Grandes at the Province of Córdoba. We also describe the high frequency DIMM device built for this and other seeing samplings obtained at the provinces of San Juan, San Luis and Córdoba. This work is part of a long term project started in 2006, in search of sites of very low turbulence and suited for near infrared telescopes with active and adaptive optics. Two of the sites have been monitored during a total of 46 nights distributed in six months of the period 2011-2012.     Multi-Particle Collision Dynamics -- a Particle-Based Mesoscale Simulation Approach to the Hydrodynamics of Complex Fluids

    In this review, we describe and analyze a mesoscale simulation method for fluid flow, which was introduced by Malevanets and Kapral in 1999, and is now called multi-particle collision dynamics (MPC) or stochastic rotation dynamics (SRD). The method consists of alternating streaming and collision steps in an ensemble of point particles. The multi-particle collisions are performed by grouping particles in collision cells, and mass, momentum, and energy are locally conserved. This simulation technique captures both full hydrodynamic interactions and thermal fluctuations. The first part of the review begins with a description of several widely used MPC algorithms and then discusses important features of the original SRD algorithm and frequently used variations. Two complementary approaches for deriving the hydrodynamic equations and evaluating the transport coefficients are reviewed. It is then shown how MPC algorithms can be generalized to model non-ideal fluids, and binary mixtures with a consolute point. The importance of angular-momentum conservation for systems like phase-separated liquids with different viscosities is discussed. The second part of the review describes a number of recent applications of MPC algorithms to study colloid and polymer dynamics, the behavior of vesicles and cells in hydrodynamic flows, and the dynamics of viscoelastic fluids

    Vitamin D3 Regulates Follicular Development and Intrafollicular Vitamin D Biosynthesis and Signaling in the Primate Ovary

    There is an increasing recognition that vitamin D plays important roles in female reproduction. Recent studies demonstrated that 1α,25-dihydroxyvitamin D3 (VD3), the biologically active form of vitamin D, improved ovarian follicle survival and growth in vitro. Therefore, we investigated the direct effects of VD3 at the specific preantral and antral stages of follicular development, and tested the hypothesis that vitamin D receptor (VDR) and enzymes critical for vitamin D biosynthesis are expressed in the primate ovary. Fourteen adult rhesus macaques provided ovarian tissue. Secondary and antral follicles were isolated for PCR analysis on VDR, vitamin D3 25-hydroxylase, and 25-hydroxyvitamin D3-1α-hydroxylase. VDR protein localization was determined by immunohistochemistry on ovarian sections. Isolated secondary follicles were cultured under conditions of control and VD3 supplementation during the preantral or antral stage. Follicle survival, growth, steroid and anti-Müllerian hormone (AMH) production, as well as oocyte maturation were evaluated. In vivo- and in vitro-developed follicles were also assessed for genes that are critical for vitamin D biosynthesis and signaling, gonadotropin signaling, steroid and paracrine factor production, and oocyte quality. The mRNA encoding VDR, 25-hydroxylase, and 1α-hydroxylase was detectable in in vivo- and in vitro-developed preantral and antral follicles. The 25-hydroxylase was elevated in cultured follicles relative to in vivo-developed follicles, which further increased following VD3 exposure. VD3 treatment increased 1α-hydroxylase in in vitro-developed antral follicles. The absence of VD3 during culture decreased VDR expression in in vitro-developed antral follicles, which was restored to levels comparable to those of in vivo-developed antral follicles by VD3 supplementation. Positive immunostaining for VDR was detected in the nucleus and cytoplasm of granulosa cells and oocytes. While only survival was improved in preantral follicles treated with VD3, VD3 supplementation promoted both survival and growth of antral follicles with increased estradiol and AMH production, as well as oocyte maturation. Thus, Vitamin D biosynthesis and signaling systems are expressed in primate ovarian follicles. Our findings support a role for VD3 in regulating follicular development in a stage-dependent manner, as well as the intrafollicular vitamin D biosynthesis and signaling, directly in the ovary

    Robots in education and care of children with developmental disabilities : a study on acceptance by experienced and future professionals

    Research in the area of robotics has made available numerous possibilities for further innovation in the education of children, especially in the rehabilitation of those with learning difficulties and/or intellectual disabilities. Despite the scientific evidence, there is still a strong scepticism against the use of robots in the fields of education and care of people. Here we present a study on the acceptance of robots by experienced practitioners (specialized in the treatment of intellectual disabilities) and university students in psychology and education sciences (as future professionals). The aim is to examine the factors, through the Unified Theory of Acceptance and Use of Technology (UTAUT) model, that may influence the decision to use a robot as an instrument in the practice. The overall results confirm the applicability of the model in the context of education and care of children, and suggest a positive attitude towards the use of the robot. The comparison highlights some scepticism among the practitioners, who perceive the robot as an expensive and limited tool, while students show a positive perception and a significantly higher willingness to use the robot. From this experience, we formulate the hypothesis that robots may be accepted if more integrated with standard rehabilitation protocols in a way that benefits can outweigh the costs
