Specific Immunoassays Confirm Association of Mycobacterium avium Subsp. paratuberculosis with Type-1 but Not Type-2 Diabetes Mellitus
Mycobacterium avium subspecies paratuberculosis (MAP) is a versatile pathogen with a broad host range. Its association with type-1 diabetes mellitus (T1DM) has been recently proposed. Rapid identification of infectious agents such as MAP in diabetic patients at the level of clinics might be helpful in deciphering the role of chronic bacterial infection in the development of autoimmune diseases such as T1DM.We describe use of an ELISA method to identify live circulating MAP through the detection of a cell envelope protein, MptD by a specific M13 phage--fMptD. We also used another ELISA format to detect immune response to MptD peptide. Both the methods were tested with blood plasma obtained from T1DM, type-2 diabetes (T2DM) patients and non-diabetic controls. Our results demonstrate MptD and fMptD ELISA assays to be accurate and sensitive to detect MAP bacilli in a large fraction (47.3%) of T1DM patients as compared to non-diabetic controls (12.6%) and those with confirmed T2DM (7.7%). Comparative analysis of ELISA assays performed here with 3 other MAP antigen preparations, namely HbHA, Gsd and whole cell MAP lysates confirmed comparable sensitivity of the MptD peptide and the fMptD based ELISA assays. Moreover, we were successful in demonstrating positive bacterial culture in two of the clinical specimen derived from T1DM patients.The MptD peptide/fMptD based ELISA or similar tests could be suggested as rapid and specific field level diagnostic tests for the identification of MAP in diabetic patients and for finding the explanations towards the occurrence of type-1 or type-2 diabetes in the light of an active infectious trigger
Association of Mycobacterium avium subsp. paratuberculosis with Multiple Sclerosis in Sardinian Patients
Mycobacterium avium subsp. paratuberculosis (MAP) infection is highly spread in the ruminant herds of Sardinia, in the Western Mediterranean. The objective of this study was to investigate prevalence of MAP infection in association with Multiple Sclerosis (MS) using clinical specimen from patients and controls. We analyzed samples for the presence of MAP specific DNA and to demonstrate humoral response to a MAP protein (MAP2694), a predicted homologue of the T-cell receptor gamma-chain/complement component 1 of the host. We found presence of MAP DNA in 42% of the MS patients and an extremely significant humoral immune response revealed by the MS patients against the MAP protein. In our opinion, this is the first report that significantly associates MAP infection with MS. Further studies will be required to confirm if MAP could be one of the triggers of MS, according to the molecular mimicry theory, in susceptible (and genetically at risk) individuals

Pre-Procedural Statin Use Is Associated with Improved Long-Term Survival and Reduced Major Cardiovascular Events in Patients Undergoing Carotid Artery Stenting: A Retrospective Study
Carotid artery stenting (CAS) is a minimal invasive procedure used to resolve carotid occlusion that can be affected by peri-procedural complications. Statin use before CAS has shown to reduce peri-procedural risk and improve survival, though time-dependent cofactors that influence mortality has not been considered. The aim of this study was to evaluate long-term survival of patients who undergo CAS considering new occurred major adverse cardiovascular event (MACE) as time-dependent cofactor. In this study, 171 high cardiovascular risk patients (age 72 \ub1 8 years, 125 males) were enrolled after CAS procedure and were followed for a median of 8.4 years. Death occurred in 44% of patients with a mean time to death of 69 \ub1 39 months and MACE in 34% with a mean time of 35 \ub1 42 months. In patients who used or not statins at baseline, death occurred in 33% and 65%, respectively (p < 0.001). Survival analysis showed that statin use reduced risk of death (hazard ratio HR 0.36, 95% confidence interval CI 0.23\u207b0.58, p < 0.0001). Including MACE as time-dependent variable did not change beneficial effects of statins. Additionally, statin use was associated with a protective effect on MACE (HR 0.48, 95% CI 0.27\u207b0.85, p = 0.012); particularly, the prevalence of stroke was reduced by 59% (p = 0.018). In multivariate analysis, effects of statins were independent of demographic and anthropometric variables, prevalence of cardiovascular risk factors, renal function, antiplatelet use, and MACE occurrence. In conclusion, use of statins before CAS procedure is associated with increased long-term survival and reduced MACE occurrence. This evidence supports the hypothesis that statin use before CAS might be beneficial in high risk patients
The relationship between S. aureus and branched-chain amino acids content in composite cow milk
The early diagnosis of mastitis is an essential factor for the prompt detection of the
animal for further actions. In fact, if not culled, infected cows must be segregated from the milking
herd and milked last, or milked with separate milking units. Besides microbiological analysis, the
somatic cell count (SCC) commonly used as predictor of intramammary infection, frequently lead
to a misclassification of milk samples. To overcome these limitations, more specific biomarkers
are continuously evaluated. The total amino acid content increases significantly in mastitic milk
compared to normal milk. S. aureus requires branched-chain amino acids (BCAAs—isoleucine,
leucine, and valine) for protein synthesis, branched-chain fatty acids synthesis, and environmental
adaptation by responding to their availability via transcriptional regulators. The increase of BCAAs
in composite milk has been postulated to be linked to mammary infection by S. aureus. The aim
of this work is to demonstrate, by a direct ion-pairing reversed-phase method, based on the use of
the evaporative light-scattering detector (IP-RP-HPLC-ELSD), applied to 65 composite cow milk
samples, a correlation between the concentration of isoleucine and leucine, and S. aureus load. The
correlation coe cient, r, was found to be 0.102 for SCC (p = 0.096), 0.622 for isoleucine (p < 0.0001),
0.586 for leucine (p < 0.0001), 0.013 for valine (p = 0.381), and 0.07 for tyrosine (p = 0.034), standing for
a positive correlation between S. aureus and isoleucine and leucine concentration. The link between
the content of BCAAs, isoleucine and leucine, and udder infection by S. aureus demonstrated with
our study has an important clinical value for the rapid diagnosis of S. aureus mastitis in cows.http://www.mdpi.com/journal/animalsam2020Paraclinical Science
Isotope correlations as a probe for freeze-out characterization: central 124Sn+64Ni, 112Sn+58Ni collisions
124Sn+64Ni and 112Sn+58Ni reactions at 35 AMeV incident energy were studied
with the forward part of CHIMERA multi-detector. The most central collisions
were selected by means of a multidimensional analysis. The characteristics of
the source formed in the central collisions, as size, temperature and volume,
were inspected. The measured isotopes of light fragments (3 <= Z <=8) were used
to examine isotope yield ratios that provide information on the free neutron to

The human diabetes proteome project (HDPP): The 2014 update
Diabetes is an increasing worldwide problem leading to major associated health issues and increased health care costs. In 2012, 9.3% of the American population was affected by diabetes, according to the American Diabetes Association, with 1.7 million of new cases since during the year (www.diabetes.org). Proteome initiatives can provide a deeper understanding of the biology of this disease and help develop more effective treatments. The collaborative effort of the Human Diabetes Proteome Project (HDPP) brings together a wide variety of complementary resources to increase the existing knowledge about both type 1 and type 2 diabetes and their related complications. The goals are to identify proteins and protein isoforms associated with the pathology and to characterize underlying disease-related pathways and mechanisms. Moreover, a considerable effort is being made on data integration and network biology. Sharing these data with the scientific community will be an important part of the consortium. Here we report on: the content of the HDPP session held at the 12th HUPO meeting in Yokohama; recent achievements of the consortium; discussions of several HDPP workshops; as well as future HDPP directions as discussed at the 13th HUPO congress in Madrid, with a special attention given to the lists of prioritized, diabetes-related proteins and the proteomic means to study them.</p
The Ariel mission: a mission of the European Space Agency for the characterization of exoplanets
Ariel, part of the European Space Agency's (ESA) Cosmic Vision science program, is an innovative medium-class mission designed for atmospheric remote sensing of exoplanets. It is the first mission solely dedicated to investigating the atmospheres of more than 500 transiting exoplanets, ranging from gas giants to super-Earths, using a combination of transit photometry and spectroscopy. The mission's primary goal is to analyze these exoplanets' chemical composition and thermal structures, paving the way for large-scale, comparative planetology. Ariel is scheduled for launch in 2029 aboard Ariane 6.2. It will operate from an orbit around the Sun-Earth system's second Lagrange point. The mission has a nominal lifetime of four years, with the potential for a two-year extension. The spacecraft comprises two main modules: the Service Module (SVM) and the Payload Module (PLM). The SVM manages platform elements, including attitude control, power, data handling, and communication systems. The PLM incorporates an all-aluminium cryogenic telescope with two scientific instruments, the Ariel IR Spectrometer (AIRS) and the Fine Guidance System (FGS). The Operational Ground Segment consists of ground stations and the Mission Operation Centre (MOC) located at ESOC, responsible for the operations of the spacecraft and instruments. The Science Ground Segment (SGS) consists of the Science Operation Centre (SOC), located at ESAC, along with the Instrument Operations and Science Data Centre (IOSDC) provided by the Ariel Mission Consortium (AMC). The SGS will perform the science mission planning as well as processing of the data to generate the mission data products and provision of the Ariel mission archive for the user community. While ESA holds overall responsibility for Ariel, the Ariel Mission Consortium is responsible for the procurement of the payload units, as well as managing the IOSDC. This collaborative effort aims to unlock the mysteries of exoplanetary atmospheres and deepen our understanding of these distant worlds
Primary aldosteronism: A Japanese perspective
Primary aldosteronism (PA) is the most common cause of secondary hypertension, accounting for 10% of all hypertension. Far from being benign, hypertension due to PA is associated with high cardiovascular morbidity and mortality. However, PA is still underdiagnosed in general practice. Recent reports strongly recommend that identifying patients with PA is cost-beneficial based on improved cardiovascular outcomes afforded by specific surgical and medical treatment.
