    Magnetic resonance and bioluminescence imaging of macrophage homing to experimental abdominal aortic aneurysms

    Macrophage infiltration is a prominent feature of abdominal aortic aneurysm (AAA) progression. We used a combined imaging approach with bioluminescence (BLI) and magnetic resonance imaging (MRI) to study macrophage homing and accumulation in experimental AAA disease. Murine AAAs were created via intra-aortic infusion of porcine pancreatic elastase. Mice were imaged over 14 days after injection of prepared peritoneal macrophages. For BLI, macrophages were from transgenic mice expressing luciferase. For MRI, macrophages were labeled with iron oxide particles. Macrophage accumulation during aneurysm progression was observed by in situ BLI and by in vivo 7T MRI. Mice were sacrificed after imaging for histologic analysis. In situ BLI ( n = 32) demonstrated high signal in the AAA by days 7 and 14, which correlated significantly with macrophage number and aortic diameter. In vivo 7T MRI ( n = 13) at day 14 demonstrated T 2 * signal loss in the AAA and not in sham mice. Immunohistochemistry and Prussian blue staining confirmed the presence of injected macrophages in the AAA. BLI and MRI provide complementary approaches to track macrophage homing and accumulation in experimental AAAs. Similar dual imaging strategies may aid the study of AAA biology and the evaluation of novel therapies

    Hematopoietic cell transplant is a curative therapy for many pediatric patients with high risk acute lymphoblastic leukemia. Its therapeutic mechanism is primarily based on the generation of an alloreactive graft-versus-leukemia effect that can eliminate residual leukemia cells thus preventing relapse. However its efficacy is diminished by the concurrent emergence of harmful graft-versus-host disease disease which affects healthly tissue leading to significant morbidity and mortality.     A Total Lightning Perspective of the 20 May 2013 Moore, Oklahoma Supercell

    In the early afternoon of 20 May 2013, a storm initiated to the westsouthwest of Newcastle, Oklahoma. This storm would rapidly intensify into the parent supercell of the tornado that struck the city of Moore, Oklahoma. This article describes what contributions total lightning observations from the Oklahoma Lightning Mapping Array could provide to operational forecasters had these observations been available in realtime. This effort includes a focus on the GOESR pseudogeostationary lightning mapper demonstration product as well as the NASA SPoRT / Meteorological Development Laboratory's total lightning tracking tool. These observations and tools identified several contributions. Two distinct lightning jumps at 1908 and 1928 UTC provided a lead time of 19 minutes ahead of severe hail and 26 minutes ahead of the Moore, Oklahoma tornado's touchdown. These observations provide strong situational awareness to forecasters, as the lightning jumps are related to the rapid strengthening of the storm's updraft and mesocyclone and serve as a precursor to the stretching of the storm vortex ahead severe weather

    Biocrust Amendments to Topsoils Facilitate Biocrust Restoration in a Post-mining Arid Environment

    Soil cryptogamic biocrusts provide many ecological functions in arid zone ecosystems, though their natural reestablishment in disturbed areas is slow. Accelerating reestablishment of biocrusts may facilitate the establishment of vascular plant communities within the timeframes of restoration targets (typically 5–15 years). One technique is to inoculate the soil surface using slurries of biocrust material harvested from another site. However, this is destructive to donor sites, and hence the potential to dilute slurries will govern the feasibility of this practice at large spatial scales. We conducted a replicated experiment on a disturbed mine site to test the individual and combined effects of two strategies for accelerating soil cryptogamic biocrust reestablishment: (1) slurry inoculation using biocrust material harvested from native vegetation; and (2) the use of psyllium husk powder as a source of mucilage to bind the soil surface, and to potentially provide a more cohesive substrate for biocrust development. The experiment comprised 90 experimental plots across six treatments, including different dilutions of the biocrust slurries and treatments with and without psyllium. Over 20 months, the reestablishing crust was dominated by cyanobacteria (including Tolypothrix distorta and Oculatella atacamensis), and these established more rapidly in the inoculated treatments than in the control treatments. The inoculated treatments also maintained this cover of cyanobacteria better through prolonged adverse conditions. The dilute biocrust slurry, at 1:100 of the biocrust in the remnant vegetation, performed as well as the 1:10 slurry, suggesting that strong dilution of biocrust slurry may improve the feasibility of using this technique at larger spatial scales. Psyllium husk powder did not improve biocrust development but helped to maintain a soil physical crust through hot, dry, and windy conditions, and so the potential longer-term advantages of psyllium need to be tested

    The genomes of two key bumblebee species with primitive eusocial organization

    Background: The shift from solitary to social behavior is one of the major evolutionary transitions. Primitively eusocial bumblebees are uniquely placed to illuminate the evolution of highly eusocial insect societies. Bumblebees are also invaluable natural and agricultural pollinators, and there is widespread concern over recent population declines in some species. High-quality genomic data will inform key aspects of bumblebee biology, including susceptibility to implicated population viability threats. Results: We report the high quality draft genome sequences of Bombus terrestris and Bombus impatiens, two ecologically dominant bumblebees and widely utilized study species. Comparing these new genomes to those of the highly eusocial honeybee Apis mellifera and other Hymenoptera, we identify deeply conserved similarities, as well as novelties key to the biology of these organisms. Some honeybee genome features thought to underpin advanced eusociality are also present in bumblebees, indicating an earlier evolution in the bee lineage. Xenobiotic detoxification and immune genes are similarly depauperate in bumblebees and honeybees, and multiple categories of genes linked to social organization, including development and behavior, show high conservation. Key differences identified include a bias in bumblebee chemoreception towards gustation from olfaction, and striking differences in microRNAs, potentially responsible for gene regulation underlying social and other traits. Conclusions: These two bumblebee genomes provide a foundation for post-genomic research on these key pollinators and insect societies. Overall, gene repertoires suggest that the route to advanced eusociality in bees was mediated by many small changes in many genes and processes, and not by notable expansion or depauperation
