    Utilization of glycerin byproduct derived from soybean oil biodiesel as a carbon source for heterologous protein production in Pichia pastoris

    AbstractCrude glycerol, also known as glycerin, is the main byproduct of the biodiesel industry. It has been estimated that up to 40,000tons of glycerin will be produced each year by 2020. This study evaluated the value-added use of crude glycerol derived from soybean biodiesel preparation as a carbon source for heterologous protein production using the yeast Pichia pastoris. Eleven glycerin samples were obtained by methanolysis of soybean oil using different acids or bases as catalysts. Cell growth experiments showed that crude glycerol containing either potassium or sodium hydroxide resulted in 1.5–2 times higher final cell densities when compared to glycerol P.A. Finally, crude glycerol containing sodium hydroxide was successfully utilized for constitutive heterologous α-amylase production in P. pastoris. This study demonstrated that crude glycerol without any purification steps may be directly used as carbon source for protein production in P. pastoris

No rio Sana há migração preferencial do canal em direção à margem esquerda e no rio Dantas o canal se mantém próximo ao eixo central da bacia.     Coriander (Coriandrum sativum L.) is a vegetable widely consumed in Brazil, however, especially in communities across the Brazilian semiarid, it is still cultivated in a rudimentary way, without rationalization of inputs such as seeds and fertilizers. So the objective of this study was to evaluate the impacts of strategies of optimization of inputs and their implications in the production and mineral composition of coriander (cv. Tabocas) grown in low-cost hydroponic system. The treatments consisted of four rates of seeds per cell in seeding (0.5, 1.0, 1.5 and 2.0 g) and three spacing between cells (7.0, 10.0 and 15.0 cm), distributed in a completely randomized design, analyzed in 4×3factorial, with three repetitions, totaling 36 experimental units. The fresh and dry weight of plant, shoot and root, as well as content of Ntot, P, K, Ca, Mg, Cl and Na in the shoot were evaluated, and the results were submitted to analysis of variance. It was concluded that the production of coriander is feasible in the " low cost hydroponic module ". It is recommended the use of 1.0 g of seeds per cell and spacing of 7.0 cm between cells, which means an average production of 5.5 kg m-2 of fresh mass of aerial parts. About the mineral composition, it was found a decreasing trend of Ca and chlorides contents due to the increase in seed weight. © 2016, Universidade Estadual Paulista - UNESP.     Using polyacrylamide with sprinkler irrigation to improve infiltration

    Center-pivot irrigation systems often apply water at rates greater than the soil infiltration rate. Applying high molecular weight, water-soluble, anionic polyacrylamide (PAM) to the soil can improve infiltration and reduce soil erosion The objective of this study was to determine whether single and multiple PAM applications with sprinkler irrigation improved infiltration under field conditions. A two-year study conducted near Kimberly, Idaho, used a solid-set sprinkler system, and a one-year study conducted in Monte dos Alhos near Alvalade do Sado, Portugal, used a center pivot. At Kimberly, applying PAM with four irrigations (total applied PAM was 2.1 kg ha-1 in 2000 and 3.0 kg ha-1 in 2001) significantly reduced total measured runoff, from 5.9 mm (2000) and 9.2 mm (2001) for the control to 2.0 and 2.1 mm. Total measured soil erosion was also reduced from 52 and 34 kg ha 1 for the control to 21 and 5 kg ha 1 for the multiple PAM treatment. Applying similar or greater amounts of PAM with a single irrigation reduced erosion, but not runoff, compared with the control. In the Monte dos Alhos study, runoff was reduced by applying a total of 0.3 kg PAM ha 1 with a single irrigation (43 mm runoff) or three irrigations (65 mm runoff) compared with the control (111 mm runoff). Measured soil erosion was not significantly different among treatments. Applying PAM with multiple irrigations extended its effectiveness as long as the application rate was great enough to adequately stabilize the soil surface during the first irrigation

    Neuroimmunomodulatory and neuroprotective effects of the flavonoid apigenin in in vitro models of neuroinflammation associated with Alzheimer's disease

    Neurodegenerative disorders (ND) are characterized by the progressive and irreversible loss of neurons. Alzheimer’s Disease (AD) is the most incident age-related ND, in which the presence of a chronic inflammatory compound seems to be related to its pathogenesis. Different stimuli in the central nervous system (CNS) can induce activation, proliferation, and changes in phenotype and glial function, which can be modulated by anti-inflammatory agents. Apigenin (4,5,7–trihydroxyflavone) is a flavonoid found in abundance in many fruits and vegetables, that has shown important effects upon controlling the inflammatory response. This study evaluated the neuroprotective and neuroimmunomodulatory potential of apigenin using in vitro models of neuroinflammation associated with AD. Co-cultures of neurons and glial cells were obtained from the cortex of newborn and embryonic Wistar rats. After 26 days in vitro, cultures were exposed to lipopolysaccharide (LPS; 1 μg/ml), or IL-1β (10 ng/ml) for 24 h, or to Aβ oligomers (500 nM) for 4 h, and then treated with apigenin (1 μM) for further 24 h. It was observed that the treatment with apigenin preserved neurons and astrocytes integrity, determined by Rosenfeld’s staining and immunocytochemistry for β-tubulin III and GFAP, respectively. Moreover, it was observed by Fluoro-Jade-B and caspase-3 immunostaining that apigenin was not neurotoxic and has a neuroprotective effect against inflammatory damage. Additionally, apigenin reduced microglial activation, characterized by inhibition of proliferation (BrdU+ cells) and modulation of microglia morphology (Iba-1 + cells), and decreased the expression of the M1 inflammatory marker CD68. Moreover, as determined by RT-qPCR, inflammatory stimuli induced by IL-1β increased the mRNA expression of IL-6, IL-1β, and CCL5, and decreased the mRNA expression of IL-10. Contrary, after treatment with apigenin in inflammatory stimuli (IL-1β or LPS) there was a modulation of the mRNA expression of inflammatory cytokines, and reduced expression of OX42, IL-6 and gp130. Moreover, apigenin alone and after an inflammatory stimulus with IL-1β also induced the increase in the expression of brain-derived neurotrophic factor (BDNF), an effect that may be associated with anti-inflammatory and neuroprotective effects. Together these data demonstrate that apigenin presents neuroprotective and anti-inflammatory effects in vitro and might represent an important neuroimmunomodulatory agent for the treatment of neurodegenerative conditions

    A new measurement of J/psi suppression in Pb-Pb collisions at 158 GeV per nucleon

    We present a new measurement of J/psi production in Pb-Pb collisions at 158 GeV/nucleon, from the data sample collected in year 2000 by the NA50 Collaboration, under improved experimental conditions with respect to previous years. With the target system placed in vacuum, the setup was better adapted to study, in particular, the most peripheral nuclear collisions with unprecedented accuracy. The analysis of this data sample shows that the (J/psi)/Drell-Yan cross-sections ratio measured in the most peripheral Pb-Pb interactions is in good agreement with the nuclear absorption pattern extrapolated from the studies of proton-nucleus collisions. Furthermore, this new measurement confirms our previous observation that the (J/psi)/Drell-Yan cross-sections ratio departs from the normal nuclear absorption pattern for semi-central Pb-Pb collisions and that this ratio persistently decreases up to the most central collisions.Comment: 19 pages, 10 figures.     Bottomonium and Drell-Yan production in p-A collisions at 450 GeV

    The NA50 Collaboration has measured heavy-quarkonium production in p-A collisions at 450 GeV incident energy (sqrt(s) = 29.1 GeV). We report here results on the production of the Upsilon states and of high-mass Drell-Yan muon pairs (m > 6 GeV). The cross-section at midrapidity and the A-dependence of the measured yields are determined and compared with the results of other fixed-target experiments and with the available theoretical estimates. Finally, we also address some issues concerning the transverse momentum distributions of the measured dimuons.Comment: 18 pages, 9 figures, to be published in Phys. Lett.

    Implant of Port-o-Cath for antineoplastic chemotherapy in a canine: case report

    O Port-o-Cath é um cateter venoso central totalmente implantável, o qual permite a infusão de quimioterápicos, hemoderivados, nutrição parenteral e coleta de sangue. Este trabalho relata o caso de um canino com um tumor no membro pélvico que foi tratado cirurgicamente e recebeu o cateter para a quimioterapia antineoplásica. A paciente possuía um sarcoma sinovial, tumor incomum em cães, que acomete normalmente as articulações e exige terapia multimodal. O tratamento foi feito em duas etapas; na primeira, o membro foi amputado e, na segunda, foi realizado o implante do Port-o-Cath e exérese de nódulo metastático. A cadela respondeu satisfatoriamente à cirurgia e realizou as sessões de quimioterapia com perfeito funcionamento do cateter, indo a óbito por outras causas 90 dias após o procedimento. O implante de Port-o-Cath para a quimioterapia é um grande aliado à terapia multimodal preconizada para o câncer, podendo o cateter ser considerado em pacientes que necessitem de medicação quimioterápica por longo período.The Port-a-Cath is a totally implantable central venous catheter that allows the infusion of chemotherapeutic agents, blood products, parenteral nutrition, and blood sample collection. This work reports the case of a dog with a tumor in the pelvic limb that was treated surgically and received the catheter for cancer chemotherapy. The patient was diagnosed with synovial sarcoma, an uncommon tumor in dogs that usually affects the joints and requires multimodal therapy. The treatment was done in two steps, in the first the limb was amputated and in the second the Port-a-Cath implantation and the excision of the metastatic nodule were performed. The dog responded well to surgery and chemotherapy sessions held with proper functioning of the catheter, and died from other causes 90 days after the procedure. The implantation of Port-a-Cath for chemotherapy is a great ally of multimodal therapy recommended for cancer, so this catheter may be considered for patients requiring chemotherapy medication for a long period
