    Synthesis of dominant plastic microfibre prevalence and pollution control feasibility in Chinese freshwater environments

    Microplastic pollution of freshwaters is known to be a great concern in China and these pollutants can be discharged into the coastal environment through fluvial processes, posing threats to the global marine ecosystem. This paper reviewed the literature measuring microplastic pollution in the Chinese freshwater environment and found that microfibres dominate other plastic morphologies in more than 65% of samples collected in surface water, sediments and effluents of wastewater treatment plants and domestic sewers. Current potential sources of microfibre pollution are identified including fishery activities, laundry sewage, and waste textiles according to previous research. Recommendations are offered using the circular economy management framework, such as textile waste reuse and recycling systems in China, for improving current control measures for microplastics in freshwaters

    Identification of genetic variants associated with Huntington's disease progression: a genome-wide association study

    Background Huntington's disease is caused by a CAG repeat expansion in the huntingtin gene, HTT. Age at onset has been used as a quantitative phenotype in genetic analysis looking for Huntington's disease modifiers, but is hard to define and not always available. Therefore, we aimed to generate a novel measure of disease progression and to identify genetic markers associated with this progression measure. Methods We generated a progression score on the basis of principal component analysis of prospectively acquired longitudinal changes in motor, cognitive, and imaging measures in the 218 indivduals in the TRACK-HD cohort of Huntington's disease gene mutation carriers (data collected 2008–11). We generated a parallel progression score using data from 1773 previously genotyped participants from the European Huntington's Disease Network REGISTRY study of Huntington's disease mutation carriers (data collected 2003–13). We did a genome-wide association analyses in terms of progression for 216 TRACK-HD participants and 1773 REGISTRY participants, then a meta-analysis of these results was undertaken. Findings Longitudinal motor, cognitive, and imaging scores were correlated with each other in TRACK-HD participants, justifying use of a single, cross-domain measure of disease progression in both studies. The TRACK-HD and REGISTRY progression measures were correlated with each other (r=0·674), and with age at onset (TRACK-HD, r=0·315; REGISTRY, r=0·234). The meta-analysis of progression in TRACK-HD and REGISTRY gave a genome-wide significant signal (p=1·12 × 10−10) on chromosome 5 spanning three genes: MSH3, DHFR, and MTRNR2L2. The genes in this locus were associated with progression in TRACK-HD (MSH3 p=2·94 × 10−8 DHFR p=8·37 × 10−7 MTRNR2L2 p=2·15 × 10−9) and to a lesser extent in REGISTRY (MSH3 p=9·36 × 10−4 DHFR p=8·45 × 10−4 MTRNR2L2 p=1·20 × 10−3). The lead single nucleotide polymorphism (SNP) in TRACK-HD (rs557874766) was genome-wide significant in the meta-analysis (p=1·58 × 10−8), and encodes an aminoacid change (Pro67Ala) in MSH3. In TRACK-HD, each copy of the minor allele at this SNP was associated with a 0·4 units per year (95% CI 0·16–0·66) reduction in the rate of change of the Unified Huntington's Disease Rating Scale (UHDRS) Total Motor Score, and a reduction of 0·12 units per year (95% CI 0·06–0·18) in the rate of change of UHDRS Total Functional Capacity score. These associations remained significant after adjusting for age of onset. Interpretation The multidomain progression measure in TRACK-HD was associated with a functional variant that was genome-wide significant in our meta-analysis. The association in only 216 participants implies that the progression measure is a sensitive reflection of disease burden, that the effect size at this locus is large, or both. Knockout of Msh3 reduces somatic expansion in Huntington's disease mouse models, suggesting this mechanism as an area for future therapeutic investigation

    Synthetic fibers as a type of microplastics: pathways for the textile sector act to combat marine litter

    Na poluição plástica do meio ambiente, há mais de uma década, lavagens domésticas são reconhecidas como atividade promotora de fragmentos têxteis. Mais estudadas em relação a outras fontes, são responsáveis pela emissão de milhares de toneladas de fibras, anualmente, em corpos d´água e no oceano. Com consequências ecológicas e socioeconômicas, fomentam interesse dos setores público e privado. Ao enfrentamento de suas intercorrências, dependem de uma série de fatores que, em âmbito global, envolvem toda sorte de atores. Neste contexto, surgem estratégias ligadas, por exemplo, a dispositivos para máquinas de lavar, modos de operação do equipamento, infraestrutura de tratamento de esgoto e parâmetros têxteis. Em relação a estes últimos, ocorre maior potencial de atuação, posto que se encontram na base deste tipo de poluição. Neste cenário, o presente estudo se propõe a refletir sobre as bases teóricas e empíricas de um programa de certificação para produtos têxteis, além de outras possibilidades de intervenção, dentro do contexto nacional. De tal modo, a presente tese se desenvolve em três capítulos. O primeiro, elabora uma revisão da literatura relacionada às fibras liberadas em lavagens domésticas, com foco nos parâmetros têxteis e suas implicações ambientais. O segundo, apresenta uma análise crítica sobre certificações têxteis e outras intervenções traçando um paralelo com a literatura visitada no primeiro capítulo. O terceiro e último, verifica a aplicabilidade e aceitação voluntária deste tipo de certificado junto ao setor têxtil brasileiro, além de outras soluções. Para tanto, conduz entrevistas semiestruturadas com atores desta cadeia. Estas entrevistas expõem que apenas parte das empresas do setor têxtil brasileiro estão alinhadas aos principais projetos e discussões sobre os microplásticos, globalmente. Neste contexto, a aplicação de uma certificação é viável para aqueles que já trabalham com algum tipo de programa. De todo modo, faltam dados para que este tipo de iniciativa seja implementada. Em termos de intervenções no processo produtivo, ações como a comercialização de fibras sintéticas biodegradáveis são mais comuns. Por outro lado, faltam análises sobre seus contrapontos e estimativas de emissão para o sul global. Para todas as intervenções relacionadas ao processo produtivo, abordagens interdisciplinares, integradas e sistêmicas são necessárias para atuar no combate ao lixo no marIn environmental plastic pollution, for more than a decade, domestic washings have been recognized, as an activity that promotes textile fragments. More studied than other sources, they are annually responsible for the emission of thousand tons of fibers in water bodies and the ocean. With ecologic and socioeconomic consequences, they receive attention from both, the public and private sectors. In this way, to deal with its outcomes, several factors and actors take place. Here, main strategies rely on washing machine devices, equipment choices and modes of operation, wastewater treatment infrastructure and textile parameters. About the latter, there is a higher potential of action as it is on the base of this type of pollution. In this scenario, the present study intends to reflect, theoretically and empirically about a certification program for textile products and other interventions on a national context. In this way, the current thesis is developed in three chapters. The first one, elaborates a literature review based on textile fibers released from domestic washings, with a focus on textile parameters and its environmental consequences. The second one, presents a critical analysis about textile certifications and other solutions, utilizing literature from the first chapter. The third and last one verifies the applicability and voluntary acceptance of this type of certificate within the Brazilian textile sector. It also explores other interventions. For this purpose, it conduces semi structured interviews, with stakeholders from this chain. These interviews expose that only part of companies from Brazilian textile sector are aligned to the main projects and discussions related to microplastics globally. In this context, the application of a certificate is viable to those who already work with this type of program. Either way, data to execute this initiative are still lacking. In terms of interventions related to manufacturing process, actions like the commercialization of biodegradable synthetic fibers are common. On the other way, counterpoints and estimations of emission for the global south are not enough available. For all the interventions related to production, interdisciplinary, integrated and systemic approaches are necessary to act on the combat of marine litter

    Há tempos a ubiquidade dos materiais plásticos no meio ambiente é assunto de discussão, com destaque para as partículas menores, ditas microplásticos (< 5 milímetros). Fibras provenientes de materiais têxteis são um subgrupo dos microplásticos e têm origem em diversas fontes, incluindo lavagens domesticas, uma vez que filtros de lavadoras e sistemas de tratamento de esgoto não são desenhados especificamente para retê-las. Quando no meio ambiente, estes materiais podem alcançar concentrações até milhares de unidades por metro cúbico, ficando disponíveis a uma gama de espécies. Neste cenário, o presente estudo teve como objetivo avaliar parâmetros de lavagem, e características têxteis que pudessem influenciar no desprendimento de fibras em efluentes de lavadoras domesticas. Foram realizados experimentos com dez sucessivas lavagens individuais, com e sem detergente, para quatro tipos de artigo: algodão (como padrão de comparação), acrílico, poliéster e poliamida. Os efluentes foram então filtrados (< 1 milímetro, 500 mícrons, 63 mícrons, 8 mícrons) e pesados. Resultados demonstram que todos os artigos têxteis liberaram fibras na lavagem. Dez sucessivas lavagens representaram queda na massa desprendida, bem como o uso de detergentes em comparação a lavagens sem detergente. Diferenças entre artigos sugeriram variação conforme características têxteis, onde algodão liberou mais fibras, seguido de acrílico, poliamida e poliéster. Em relação ao tamanho das fibras, a maioria ficou retida no filtro da lavadora (< 1 milímetro) e na peneira de 63 mícrons, mostrando o potencial de diminuição de porosidade do filtro. A visualização de fibras em papel-filtro de 8 mícrons sugere a existência de fibras micro e nano. Convertendo massa para numero de unidades, a lavagem individual de um artigo têxtil mostrou desprender entre milhares e centenas de milhares de fibras. Para uma extrapolação mundial, cerca de 40,4 mil toneladas de algodão e 21,5 mil toneladas de fibras sintéticas seriam liberadas em efluentes de esgoto. No Brasil estes valores corresponderiam, respectivamente, a 1,6 mil e 860 toneladas ano. Caso fossem tratadas em estações de tratamento de esgoto em condições ideais, seriam liberadas, em um ano, cerca de 737 toneladas de fibras sintéticas em escala mundial e 29 toneladas em escala nacional. Uma vez em cursos d\'água, estas fibras atingiriam, em ultima instância, o ambiente marinho, indicando a necessidade por soluções que combatam este tipo de poluição, sem antes deixar de explorar as lacunas do conhecimento, relacionadas, no âmbito têxtil, especialmente às diferenças metodológicas entre os estudos.Since decades, the ubiquity of plastic materials in the environment has been a matter of discussion. Smaller pieces, named microplastic (< 5 millimeters) gained more attention recently and are now the focus of several studies. Textile fibers are a subgroup of microplastics and can be originated from several sources, including domestic washings, once filters and sewage treatment plants are not specifically designed to retain them. In the environment, these materials can reach concentrations up to millions of units per cubic meter, being available to many species. The objective of the present study was to evaluate washing parameters and also textile characteristics, which could influence in fibers emission from domestic washing machines. Experiments were done in ten successive times, with and without detergent, for four types of articles: cotton (as a pattern for comparison), acrylic, polyester and polyamide. Resulting effluents were then filtered in different porosities (< 1 millimeter, 500 microns, 63 microns, 8 microns), weighted, related to mass of textile articles and simulated regarding number of fibers. Results demonstrated that all textile articles emitted fibers during domestic washings. Ten successive washings represented a decrease in the emitted mass, as well as the use of detergent in relation to washings without the product. Differences between articles suggested variation of results according to textile characteristics, where the ranking of emission was: cotton, acrylic, polyamide and polyester. When considering the size of fibers, the majority was retained in the filter of the washing machine (porosity < 1 millimiter) and in 63 microns sieve, when compared to 500 microns sieve, showing the importance of the filter of the washing machine and the potential to decrease its porosity. Fibers retained in the filter- paper of 8 microns, couldnt be weighted because of its low mass, but their visualization suggests the existence of fibers in micro and nano scales. Converting mass to number of fibers, one individual washing was responsible for something between thousands and hundred of thousands of units. When extrapolating values to a global perspective, it reaches something around 40.4 thousand tonnes of cotton per year and 21.5 thousand tonnes of synthetic fibers per year. In a Brazilian perspective, these values correspond, respectively, to 1.6 thousand tonnes year and 860 tonnes year. If this effluents were all treated in sewage treatment plants, in ideal conditions, still they would release something like 737 tonnes of synthetic fibers per year in a global scale and 29 tonnes of synthetic fibers per year in a national scale. Once in water bodies, those fibers would reach the marine environment, indicating the necessity of solutions that could impair this kind of pollution, not before solving knowledge gaps, specially related, in the textile area, to methodology differences between studie

    Seizure disorder is a chronic condition in the brain that affects approximately 50 million people worldwide. Oxidative stress plays a crucial role in the pathophysiology of this disorder and can cause neuronal injury. Approximately one in three treated patients suffers from seizures regardless of pharmacological intervention, which results in oxidative damage. The present study aims to investigate the possible protective effect of antioxidant-rich Vitis labrusca extract on pentylenetetrazol-induced oxidative damage in Wistar rats. Possible behavioral alterations, genotoxic and mutagenic effects of the extract were also evaluated. The results showed that V. labrusca extract provides a significant protective effect against oxidative damage to lipids and proteins induced by pentylenetetrazol in the cerebral cortex, cerebellum, hippocampus and liver of rats. Also, the extract did not alter locomotor behavior. Moreover, it was non-genotoxic and non-mutagenic. Our results suggest the possibility of using V. labrusca extract as a therapeutic agent to minimize neuronal damage associated with seizures
