    Agricultural intensification erodes taxonomic and functional diversity in Mediterranean olive groves by filtering out rare species

    Agri-Environmental Schemes (AES) have been proposed to mitigate the impact of agriculture on both taxonomic and functional biodiversity. However, a better knowledge of the mechanisms involved in the loss of agrobiodiversity is needed to implement efficient AES. An unbalanced effort on research towards arable lands compared to permanent crops, and on fauna relative to plants, is patent, which limits the generalization of AES effectiveness. We evaluated the effects of agricultural management and landscape simplification on taxonomic and functional diversity of the ground herb cover of 40 olive groves. We use a recently developed approach based on Hill numbers (rare, common and dominant species based) to analyse taxonomic and functional dissimilarity between farms with contrasting agricultural practices, and its potential attenuation by landscape complexity. We further explore the filtering effect of agricultural intensification on functional traits, and the relationship between functional and species richness across landscapes. We found that taxonomic and functional dissimilarity of herb assemblages between intensively and low-intensively managed fields was mainly due to rare species. Dissimilarity decreased as landscape complexity increased, evidencing that complex landscapes attenuate the impact of agriculture intensification on herb assemblage composition. Agricultural intensification favoured more functionally homogeneous assemblages and disfavoured the herbs pollinated by insects, while it did not seem to affect wind-pollinated species. Overall, functional richness increased exponentially with species richness across landscapes, but the latter was insufficient to drive any clear enhancement in functional richness in simple landscapes. In contrast, high species richness accelerated the enhancement in functional richness in intermediate and complex landscapes. These results highlight the functional filtering that intensive agriculture has generated for decades in homogeneous olive-dominated landscapes. Synthesis and applications. Herb cover is essential to support the fauna of permanent croplands and their sustainable production. Hence, Agri-Environmental Schemes (AES) in these croplands should promote management practices favouring the diversity and functionality of herb assemblages. Such AES should be particularly prioritized in homogeneous landscapes, where ground herb cover composition and function has long been homogenized to a great extent

    Agricultural extensification enhances functional diversity but not phylogenetic diversity in Mediterranean olive groves: A case study with ant and bird communities

    Agroforests are of well-known importance for biodiversity conservation, especially in the tropics, because they are structurally stable and may resemble natural forests. Previous studies have characterized jointly taxonomic, functional and phylogenetic diversity in these agro-ecosystems to comprehensively examine the mechanisms by which agriculture impacts on biodiversity. However, this approach has been barely applied to other woody crops of economic importance, such as olive grove, which is a remarkable overwintering habitat for frugivorous/insectivorous birds from central and northern Europe, and whose original distribution overlaps with the Mediterranean biodiversity hotspot. We examined the effects of landscape complexity and intensive management practices at a local scale (recurrent plowing and pesticides use) on the functional and phylogenetic diversity of animal communities inhabiting olive groves. Since the response of functional traits or clades may vary across different taxonomic groups, we conducted our study at two levels: ants, which are considered semi-sessile organisms, and birds, which exhibit a high dispersal capacity. In birds, neither management type nor landscape complexity had an effect on phylogenetic diversity (PD) indices. Extensively managed farms harbored bird communities with higher values of functional diversity (FD), but this effect only was evident when considering cultivated (productive) zones within the farm (i.e., infield diversity). Ant assemblages on intensively managed farms exhibited a lower level of phylogenetic clustering than those located in extensive farms, but this effect vanished when excluding non-cultivated zones. Ant functional diversity increased with landscape complexity. Our results indicate that PD and FD exhibit different responses to farming intensification in olive groves. Although intensive management does not erode PD due to the existence of phylogenetic redundancy, the loss of species associated to modern farming leads to a reduction in FD being this indicative of functional complementarity. This study provides evidence that land-use extensification (extensive farming and landscape diversification) promotes more functionally rich assemblages than modern intensive practices in olive groves. Our findings also show the need to set apart the effect of non-cultivated zones (e.g., hedgerows, margins) when evaluating the effectiveness of agri-environment schemes as the joint consideration of non-cultivated and cultivated areas may obscure the benefits of local extensification on infield biodiversity.info:eu-repo/semantics/publishedVersio

    Valoración de la adherencia en el seguimiento del paciente diabético y uso de lenguaje enfermero.

    Improving the low treatment adherence in chronic patients increases patient ‘security and effectiveness of the health system. Nurses play an important role in this objective and they have their own language to describe this situation.Goals: To know the number of diabetic patients that came to the nurse consultancy throughout the year 2011; whether nurses valued their pharmacologic adherence and if they gave advise about medication, nutrition and physical exercise. Another goal was to know if all this information was expressed using nurse language.Methods: It is a cross-sectional descriptive study. Population of study: Diabetic patients that came to the nurse consultancy throughout the year 2011. Data were acquired from OMI-AP and review of medical records. Variables were expressed as absolute frequencies and percentages.Results: From a total of 719 diabetic patients, 465 (64, 6%) attended the nurse consultancy. 369 (80, 7%) received advice about nutrition and 307 (67%) about physical exercise. 390 (85, 3%) were under pharmacological treatment, in 336 (72, 2%) pharmacologic al adherence was registered, the advice: “adequate use of medication” was present in 209 (45%). NIC 5616 “Teaching on prescribed drugs” appeared in 27 patients and 16 had care plans in that period.Conclusions: Evaluation of pharmacologic adherence is an activity included in the follow up made by nurses in diabetic patients. Advice about nutrition is usually given. Advice about physical exercise and adequate use of medication must be reinforced. The use of nurse language is not incorporated in our nurse practicing. This would be facilitated by improving informatics tools and incorporating this item in management strategy.Mejorar la baja adherencia terapéutica de los pacientes crónicos, aumenta la seguridad del paciente y la efectividad del sistema sanitario; las enfermeras tienen en ello una función importante y disponen de un lenguaje propio para denominar esta situación.OBJETIVOS:Conocer el número de pacientes diabéticos que durante el año 2011, acudieron a la consulta de su enfermera; si éstas les valoraron la adherencia farmacológica y dieron consejo sobre medicación, alimentación y ejercicio físico. Saber si se expresa mediante lenguaje enfermero.MÉTODO: Estudio descriptivo transversal. Población a estudio: diabéticos que acuden a consulta de enfermería durante el año 2011. Datos extraídos de OMI-AP y revisión de historias clínicas. Variables expresadas como frecuencias absolutas y porcentajes. RESULTADOSPacientes diabéticos 719, acudieron a consulta de enfermería 465 (64,6%). Tienen recogido consejo sobre alimentación 369 (80,7%) y sobre ejercicio 307 (67%). Tienen tratamiento farmacológico 390 (85,3%), recogida la adherencia farmacológica 336 (72,2%), consejo “uso adecuado de medicación” 209 (45%). La NIC 5616 “Enseñanza medicamentos prescritos”, figura en 27 pacientes y 16 tienen planes de cuidados en ese periodo.CONCLUSIONESLa valoración de la adherencia farmacológica es una actividad integrada por las enfermeras en el seguimiento de los pacientes diabéticos. El consejo sobre alimentación se realiza de forma habitual. Se debe incrementar el consejo sobre ejercicio físico y uso adecuado de medicamentos. La utilización de lenguaje enfermero, no está incorporada en nuestra práctica enfermera. Lo facilitaría mejoras en la herramienta informática y que formara parte de los objetivos de las gerencias.

    Finding immunological differences to help diagnosis and early treatment of Kawasaki Disease and MIS-C (Multisystem Inflammatory Syndrome in Children)

    Resumen del trabajo presentado en el 43 Congreso De La Sociedad Española de Inmunología, celebrado en León (España) del 22 al 24 de septiembre de 2022.The recent COVID-19 pandemic was first thought to spare children from health deprivation caused by infection with SARS-CoV-2. However, soon a new syndrome resembling Kawasaki Disease (KD) was reported: Multisystem Inflammatory Syndrome in Children (MIS-C). The aim of this study is to provide new biomarkers for both diseases in order to facilitate diagnosis and reduce the time-lapse until treatment is provided – which will reduce the risk of developing severe cardiovascular complications. An extensive immune system characterization by flow cytometry and serum protein characterization by a multiplex technology (Olink) was performed from fresh blood samples of patients with acute MIS-C (n=19) and KD (n=10). For protein characterization we also analysed recovery samples for these groups (n=19 and n=8, respectively). Besides the already described lymphopenia in MIS-C, we found additional significant immune differences in both groups. Although lymphocyte numbers (cells/ml) were lower in MIS-C, percentages of activated T-CD4+ and T-CD8+ cells were higher compared to KD. Moreover, when comparing activated T cells in MIS-C and KD individually, regulatory T cells (Treg) showed the highest levels. These data suggest a stronger response of T cells in MIS-C, and higher Treg activity in both groups, which could reflect the response to the excessive inflammation. Ratios previously described in other inflammatory conditions were evaluated: MIS-C showed higher neutrophil/lymphocyte and Th17/Treg ratios than KD, suggesting higher inflammatory conditions in this group. In addition, monocyte and dendritic cells (DCs) numbers were decreased in MIS-C relative to KD. Parallel to these inflammatory cellular profiles, we identified increased levels of inflammatory cytokines in plasma of patients during the acute phase of the disease compared to recovery samples. Moreover, IL-6, which is one of the main cytokines involved in cytokine storm in adult COVID-19, was higher in MIS-C suggesting, again, stronger inflammatory conditions in this pathology compared to KD

    Treatment of early borderline lesions in low immunological risk kidney transplant patients : a Spanish multicenter, randomized, controlled parallel-group study protocol: the TRAINING study

    Subclinical inflammation, including borderline lesions (BL), is very common (30-40%) after kidney transplantation (KT), even in low immunological risk patients, and can lead to interstitial fibrosis/tubular atrophy (IFTA) and worsening of renal function with graft loss. Few controlled studies have analyzed the therapeutic benefit of treating these BL on renal function and graft histology. Furthermore, these studies have only used bolus steroids, which may be insufficient to slow the progression of these lesions. Klotho, a transmembrane protein produced mainly in the kidney with antifibrotic properties, plays a crucial role in the senescence-inflammation binomial of kidney tissue. Systemic and local inflammation decrease renal tissue expression and soluble levels of α-klotho. It is therefore important to determine whether treatment of BL prevents a decrease in α-klotho levels, progression of IFTA, and loss of kidney function. The TRAINING study will randomize 80 patients with low immunological risk who will receive their first KT. The aim of the study is to determine whether the treatment of early BL (3rd month post-KT) with polyclonal rabbit antithymocyte globulin (Grafalon®) (6 mg/kg/day) prevents or decreases the progression of IFTA and the worsening of graft function compared to conventional therapy after two years post-KT, as well as to analyze whether treatment of BL with Grafalon® can modify the expression and levels of klotho, as well as the pro-inflammatory cytokines that regulate its expression.     Oligonucleotide array-CGH identifies genomic subgroups and prognostic markers for tumor stage mycosis fungoides

    Mycosis fungoide (MF) patients who develop tumors or extracutaneous involvement usually have a poor prognosis with no curative therapy available so far. In the present European Organization for Research and Treatment of Cancer (EORTC) multicenter study, the genomic profile of 41 skin biopsies from tumor stage MF (MFt) was analyzed using a high-resolution oligo-array comparative genomic hybridization platform. Seventy-six percent of cases showed genomic aberrations. The most common imbalances were gains of 7q33.3q35 followed by 17q21.1, 8q24.21, 9q34qter, and 10p14 and losses of 9p21.3 followed by 9q31.2, 17p13.1, 13q14.11, 6q21.3, 10p11.22, 16q23.2, and 16q24.3. Three specific chromosomal regions, 9p21.3, 8q24.21, and 10q26qter, were defined as prognostic markers showing a significant correlation with overall survival (OS) (P=0.042, 0.017, and 0.022, respectively). Moreover, we have established two MFt genomic subgroups distinguishing a stable group (0-5 DNA aberrations) and an unstable group (>5 DNA aberrations), showing that the genomic unstable group had a shorter OS (P=0.05). We therefore conclude that specific chromosomal abnormalities, such as gains of 8q24.21 (MYC) and losses of 9p21.3 (CDKN2A, CDKN2B, and MTAP) and 10q26qter (MGMT and EBF3) may have an important role in prognosis.     P53 wild-type colorectal cancer cells that express a fetal gene signature are associated with metastasis and poor prognosis

    Current therapy against colorectal cancer (CRC) is based on DNA-damaging agents that remain ineffective in a proportion of patients. Whether and how non-curative DNA damage-based treatment affects tumor cell behavior and patient outcome is primarily unstudied. Using CRC patient-derived organoids (PDO)s, we show that sublethal doses of chemotherapy (CT) does not select previously resistant tumor populations but induces a quiescent state specifically to TP53 wildtype (WT) cancer cells, which is linked to the acquisition of a YAP1-dependent fetal phenotype. Cells displaying this phenotype exhibit high tumor-initiating and metastatic activity. Nuclear YAP1 and fetal traits are present in a proportion of tumors at diagnosis and predict poor prognosis in patients carrying TP53 WT CRC tumors. We provide data indicating the higher efficacy of CT together with YAP1 inhibitors for eradication of therapy resistant TP53 WT cancer cells. Together these results identify fetal conversion as a useful biomarker for patient prognosis and therapy prescription. The failure of chemotherapy in colorectal cancer is currently unclear.     Molecular Characterization of the Region 7q22.1 in Splenic Marginal Zone Lymphomas

    Splenic marginal zone lymphomas (SMZL) are an uncommon type of B-cell non-Hodgkin's lymphoma (NHL-B) in which no specific chromosomal translocations have been described. In contrast, the most frequent cytogenetic abnormality is the loss of the long arm of chromosome 7 (7q). Previous reports have located this loss in the 7q32 region. In order to better characterize the genomic imbalances in SMZL, molecular studies were carried out in 73 patients with SMZL. To gain insight into the mapping at 7q a tiling array was also used. The results confirmed the loss of 7q as the most frequent change. In addition, several abnormalities, including 4q22.1, 1q21.3–q22, 6q25.3, 20q13.33, 3q28, 2q23.3–q24.1 and 17p13, were also present. A loss of 7q22.1 at 99925039–101348479 bp was observed in half of the cases. The region of 7q22.1 has not previously been characterised in SMZL.     Assessment of a New ROS1 Immunohistochemistry Clone (SP384) for the Identification of ROS1 Rearrangements in Patients with Non–Small Cell Lung Carcinoma: the ROSING Study

    Introduction: The ROS1 gene rearrangement has become an important biomarker in NSCLC. The College of American Pathologists/International Association for the Study of Lung Cancer/Association for Molecular Pathology testing guidelines support the use of ROS1 immunohistochemistry (IHC) as a screening test, followed by confirmation with fluorescence in situ hybridization (FISH) or a molecular test in all positive results. We have evaluated a novel anti-ROS1 IHC antibody (SP384) in a large multicenter series to obtain real-world data. Methods: A total of 43 ROS1 FISH-positive and 193 ROS1 FISH-negative NSCLC samples were studied. All specimens were screened by using two antibodies (clone D4D6 from Cell Signaling Technology and clone SP384 from Ventana Medical Systems), and the different interpretation criteria were compared with break-apart FISH (Vysis). FISH-positive samples were also analyzed with next-generation sequencing (Oncomine Dx Target Test Panel, Thermo Fisher Scientific). Results: An H-score of 150 or higher or the presence of at least 70% of tumor cells with an intensity of staining of 2+ or higher by the SP384 clone was the optimal cutoff value (both with 93% sensitivity and 100% specificity). The D4D6 clone showed similar results, with an H-score of at least 100 (91% sensitivity and 100% specificity). ROS1 expression in normal lung was more frequent with use of the SP384 clone (p < 0.0001). The ezrin gene (EZR)-ROS1 variant was associated with membranous staining and an isolated green signal FISH pattern (p = 0.001 and p = 0.017, respectively). Conclusions: The new SP384 ROS1 IHC clone showed excellent sensitivity without compromising specificity, so it is another excellent analytical option for the proposed testing algorithm
