    Molecular Identification of Genetically Modified Crops for Biosafety and Legitimacy of Transgenes

    Crops undergo artificially DNA modifications for improvements are considered as genetically modified (GM) crops. These modifications could be in indigenous DNA or by introduction of foreign DNA as transgenes. There are 29 different crops and fruit trees in 42 countries, which have been successfully modified for various traits like herbicide tolerance, insect/pest resistance, disease resistance and quality improvement. GM crops are grown worldwide and its area is significantly increasing every year. Many countries have very strict rules and regulations for GM crops and are also a trade barrier in some situations. Hence, identification and testing of crops for GM contents is important for identity and legitimacy of transgene to simplify the international trade. Normally, molecular identification is performed at three different levels, i.e., DNA, RNA and protein, and each level has its own importance in testing about the nature and type of GM crops.     DNA Fingerprinting and Cultivar Identification of Olive (Olea europaea L.) using SSR markers

    Background: Genetic diversity and population structure of the olive varieties cultivated in Pakistan are yet to be explored.Methods: In present study, we studied population structure and genetic diversity and developed DNA fingerprints of 13 olive varieties  using 63 Simple Sequence Repeat  markers.Results: Collectively 618 alleles were amplified among which 582 were polymorphic and 36 were monomorphic. High allelic diversity per locus was found among 63 SSR markers, i.e., one for GAPU-12 to 23 for UDO099-008 and GAPU-47 with an average 9.80 alleles per locus. On the basis of cluster analysis genotypes were grouped into two clusters. Cluster I contained varieties Manzanilla, Sohawa-selection, Koroneki, Bulkasar-selection, Arbequina, Arbosona, Chugtai-selection, whereas cluster II was comprised of varieties Bari-Zatoon, Coratina, Gemlik, Frontaio, Pendolino and Ottobratica. UDO-24 marker alone identified seven olive varieties. Similarly, DCA-07 and EMO-02 identified six olive varieties each.Conclusion: The findings of this manuscript will be helpful for future studies related to DNA fingerprinting and genetic diversity assessment for choice of SSR markers and identification of olive varieties.   Keywords: Cluster analysis; Polymorphic Information Content; Structure analysis; Similarity matrix

    Omega-3 and omega-6 polyunsaturated fatty acids and their potential therapeutic role in protozoan infections

    Protozoa exert a serious global threat of growing concern to human, and animal, and there is a need for the advancement of novel therapeutic strategies to effectively treat or mitigate the impact of associated diseases. Omega polyunsaturated fatty acids (ω-PUFAs), including Omega-3 (ω-3) and omega-6 (ω-6), are constituents derived from various natural sources, have gained significant attention for their therapeutic role in parasitic infections and a variety of essential structural and regulatory functions in animals and humans. Both ω-3 and ω-6 decrease the growth and survival rate of parasites through metabolized anti-inflammatory mediators, such as lipoxins, resolvins, and protectins, and have both in vivo and in vitro protective effects against various protozoan infections. The ω-PUFAs have been shown to modulate the host immune response by a commonly known mechanism such as (inhibition of arachidonic acid (AA) metabolic process, production of anti-inflammatory mediators, modification of intracellular lipids, and activation of the nuclear receptor), and promotion of a shift towards a more effective immune defense against parasitic invaders by regulation the inflammation like prostaglandins, leukotrienes, thromboxane, are involved in controlling the inflammatory reaction. The immune modulation may involve reducing inflammation, enhancing phagocytosis, and suppressing parasitic virulence factors. The unique properties of ω-PUFAs could prevent protozoan infections, representing an important area of study. This review explores the clinical impact of ω-PUFAs against some protozoan infections, elucidating possible mechanisms of action and supportive therapy for preventing various parasitic infections in humans and animals, such as toxoplasmosis, malaria, coccidiosis, and chagas disease. ω-PUFAs show promise as a therapeutic approach for parasitic infections due to their direct anti-parasitic effects and their ability to modulate the host immune response. Additionally, we discuss current treatment options and suggest perspectives for future studies. This could potentially provide an alternative or supplementary treatment option for these complex global health problems

    Discovering the Correlation Between Phishing Susceptibility Causing Data Biases and Big Five Personality Traits Using C-GAN

    Recently, on social media, various kinds of social engineering (SE) have made individuals more susceptible to attacks. A phishing attempt is a widely used SE technique that takes advantage of people’s vulnerabilities to acquire personal or confidential information. These attempts are growing at an astonishing speed, causing harm to both individuals and corporations. According to the latest studies, certain individuals are more vulnerable to such kinds of attacks than others. However, the relationship between psychological characteristics and phishing attacks has not been adequately investigated. This study empirically explores the connection between phishing vulnerability that causes data biases and the Big Five personality traits. Recognizing personality traits that make people more vulnerable to phishing attempts is a key step in developing protection and safeguarding individuals. The individuals who scored high in some traits are more probable to suffer from such assault. To the best of our knowledge, no prior quantitative study has attempted to find many genuine phishing victims and their personality behavior. This problem lacks the availability of publically accessible data. It is also challenging to estimate the probability distribution of rows in tabular data and generate realistic synthetic data to train/test the model on more data. This work employs a conditional generative adversarial network (C-GAN) for both data generation and classification to find the correlation between personality traits and phishing attacks

    A Comparative Evaluation Of Premedication With Flurbiprofen And Prednisolone On Post Endodontic Pain In Teeth With Symptomatic Irreversible Pulpitis: A Randomized Clinical Trial

    Background: Root canal treatment is an endodontic procedure that encompasses cleaning & shaping i.e. preparation of root canals followed by obturation of the prepared canals. Pain is a very common sensation that occurs few hours after root canal treatment (RCT). Objective: To evaluate and compare the efficacy of premedication with flurbiprofen and prednisolone on post-endodontic pain in teeth with symptomatic irreversible pulpitis. Materials and Methods: It is a randomized control trial study conducted in the department of Operative Dentistry & Endodontics, Sardar Begum Dental College, & Hospital Peshawar. Patients were randomly allocated into two groups i.e. A and B using coin flip technique. Thirty minutes before starting the endodontic therapy, single pretreatment dose of Flurbiprofen (100mg) was given to patients in Group A and Prednisolone (30mg) to patients in Group B. Patients’ pain intensity experience was measured using the visual analog scale (VAS) after root canal treatment. They were instructed to complete a pain diary at specific intervals (i.e. at 6, 12 and 24 hours after the commencement of treatment) in order to determine efficacy in both groups. Results: As per efficacy in both groups, in Group A, 44 (55%) showed effective results whereas in Group B, only 20 (25%) patients showed effective results. Conclusion: Single pretreatment dose of flurbiprofen has a more sustained effect in reducing post-endodontic pain as compared to prednisolone

    Protocol optimization for efficient in vitro micro-propagation of Stevia rebaudiana Bertoni

    Background: Stevia rebaudiana Bertoni is one of the most amazing herb that is an abundant source of diterpene glycoside a non-caloric sweetener. In recent years alongside its utilization in herbal and folk medicine, commercial exploitation of this herb for weight control and anti-diabetic treatment has started.Methods: In present study healthy nodal segments of Stevia rebaudiana Bertoni were used as explants for optimization of in vitro micro-propagation and regeneration protocol for rapid, fast and efficient multiplication under in-vitro conditions. For shoot proliferation and rooting various concentrations cytokinins NAA (Naphthalene acetic acid), BAP (Benzyl amino purine), IAA (Indole acetic acid) and kinetin (Kn) were employed.Results: Shoots without roots were regenerated from nodal segments via auxiliary shoot induction. The production of multiple shoots from nodal portion was maximum on MS medium complemented with 0.5 mg/L of BAP and 1.5 mg/L of Kn. Maximum rooting was recorded on MS medium supplemented with 0.25 mg/L NAA and 1.0 mg/L of IAA. Successful hardening was achieved under controlled growth room conditions followed by transfer to glass house and open field.Conclusion: Standardization of explant source and culturing media for multiplication of shoot resulted in mass proliferation of homogenous and healthy plantlets of Stevia clones under local conditions. Keywords: Stevia rebaudiana Bertoni; BAP; Kinetin, NAA; IAA; Micro-propagation; Regeneratio

    Revealing the Yield and Quality Responses of Soybean Advanced Lines under Semi-Arid Conditions

    Background: Soybean as human diet is a rich source of protein and oil. It also plays a vital role in livestock and poultry industries. Objective of this work is to exploit the local soybean germplasm for semi-arid conditions.Methods: The experiment was conducted in Randomized Complete Block Design with three replications. Plant × plant and row × row distance was maintained as 4 inch and 1ft respectively. At maturity data for plant height, days to 50% flowering, no. of branches, no. of pods, grains per pod and grain yield per hectare were recorded.  Furthermore, oil percentage, protein percentage, omega-3, omega-6, omega-9, palmitic acid and stearic acids were also measured.Results: All genotypes showed highly significant difference from each other for selected traits. Grain yield per hectare was significant in genotypes such as CN-5, FS-10, E-402 and SH-1274 as compared to Faisal soybean (check). Protein and oil percentage were significantly more in CN-5, HS-17 and FS-10. Branches per plant significantly correlated with the yield but protein and oil percentage negatively correlated with each other. PCA indicated that only four out of 13 PCAs exhibited more than 1 Eigen value and showed 76.53 % variation. All traits for yield and quality were presented in PCA1, PCA2 and PCA3. Biplot indicated that genotype CN-5, SH-1274 and HB-17 falls in the positive portion that perform good.Conclusion: Soybean genotypes CN-5 and FS-10 showed the more yield with high protein and oil percentage as compared to check variety and could be used in semi-arid environments.Keywords: Oilseeds; Soybean; Semi-arid; Yield; Quality

    Extracellular vesicles (EVs) have emerged as key intercellular communication and pathogenesis mediators. Parasitic organisms’ helminths, cause widespread infections with significant health impacts worldwide. Recent research has shed light on the role of EVs in the lifecycle, immune evasion, and disease progression of these parasitic organisms. These tiny membrane-bound organelles including microvesicles and exosomes, facilitate the transfer of proteins, lipids, mRNAs, and microRNAs between cells. EVs have been isolated from various bodily fluids, offering a potential diagnostic and therapeutic avenue for combating infectious agents. According to recent research, EVs from helminths hold great promise in the diagnosis of parasitic infections due to their specificity, early detection capabilities, accessibility, and the potential for staging and monitoring infections, promote intercellular communication, and are a viable therapeutic tool for the treatment of infectious agents. Exploring host-parasite interactions has identified promising new targets for diagnostic, therapy, and vaccine development against helminths. This literature review delves into EVS’s origin, nature, biogenesis, and composition in these parasitic organisms. It also highlights the proteins and miRNAs involved in EV release, providing a comprehensive summary of the latest findings on the significance of EVs in the biology of helminths, promising targets for therapeutic and diagnostic biomarkers
