Avian colibacillosis in Morocco: Threatening multi-layered infection
Les colibacilloses aviaires sont les infections bactériennes les plus fréquentes et parmi les plus importantes en pathologie des volailles. Elles représentent vraisemblablement la première cause de traitement antibiotique dans les élevages et de l’émergence de souches résistantes constitue un problème de santé publique vétérinaire. Cette infection dont la voie d’entrée principale est le tractus respiratoire ou parfois génital affecte essentiellement les élevages de poulets de chair en engendrant des lésions et des pertes économiques lourdes. En effet, elles peuvent entrainer la mortalité, les baisses de performances et les saisies aux abattoirs. En outre, les moyens de lutte contre ces maladies sont très onéreux. La plupart des colibacilloses sont des surinfections suite à des infections virales, bactériennes ou parasitaires. L’importance des colibacilloses est surtout médicale et économique. Les colibacilloses peuvent être classées en colibacilloses primaires dues à des colibacilles spécifiquement pathogènes et des colibacilloses secondaires dues à une infection virale ou bactérienne. La problématique de la colibacillose réside dans son impact à la fois économique et sanitaire. L’émergence de souches de plus en plus résistantes devient préoccupante. L’objectif du présent travail est de dresser une revue bibliographique sur la colibacillose avec un focus sur la pathologie au Maroc.

They are probably the main cause of antibiotic treatment in farms and the emergence of resistant strains is a veterinary public health problem. The main way of entry of this infection is the respiratory and/ or genital tract. It affects essentially broiler chickens, causing serious damage and economic loss. In fact, they can lead to mortality, reduced performance and condemnation at slaughterhouses.In addition, the tools and strategies to control these diseases are very expensive. Most colibacillosis is secondary infection following viral, bacterial or parasitic infections. The importance of colibacillosis is mainly medical and economic. Colibacillosis can be classified into primary colibacillosis due to specific pathogenic colibacilli and colibacillosis secondary due to a viral or bacterial infection. The problem with colibacillosis lies in its impact, both economic and health. The emergence of increasingly resistant strains is of concern. The objective of this work is to draw up a bibliographical review on colibacillosis with a focus on pathology in Morocco.

Activité antibactérienne de l'huile essentielle d'origan et du carvacrol sur des souches d'Escherichia coli d'origine aviaire
Bacterial resistance to antibiotics and the search for new alternatives to antibiotics are of great importance in human and animal health. The main aim of this study was to evaluate the antibacterial pharmacodynamic properties of the essential oil of oregano as well as those of its major component, carvacrol, on strains of avian Escherichia coli resistant to antibiotics of current use in poultry (Colistin and Enrofloxacin). These properties were evaluated using standard agar diffusion and macro-dilution techniques for determination of Minimum Inhibitory Concentration (MIC). The results obtained showed a potent activity of the two tested products in comparison with the two antibiotics used in this study, giving the largest inhibition diameters which ranged respectively from 8 to 40 mm and from 14 to 30 mm for oregano essential oil (HE) and carvacrol. This was confirmed by the determination of the MIC which was 0.0031 % for oregano HE and 0.0625 % for its active ingredient. These results lead us to conclude that the use of oregano oil could be a good alternative to the evolving bacterial resistance. However, it would be interesting to confirm these results on living organisms in order to evaluate in vivo effectiveness of oregano essential oil and its active ingredients.

L’objectif principal de cette étude a été d’évaluer les propriétés anti-bactériennes de l’huile essentielle de l’origan, ainsi que celle de son composant majoritaire le carvacrol, sur des souches d’Escherichia coli d’origine aviaire résistantes aux antibiotiques d’utilisation courante en aviculture (Colistin et Enrofloxacin). Ces propriétés ont été évaluées en utilisant des techniques usuelles de diffusion sur gélose et de macro-dilution pour la détermination de la concentration minimale inhibitrice. Les résultats obtenus ont permis de mettre en évidence une puissante activité anti-bactérienne des deux produits testés en comparaison avec les deux antibiotiques utilisés dans cette étude, en donnant les plus grands diamètres d’inhibition qui ont respectivement varié de 8 à 40 mm et de 14 à 30 mm quand pour l’huile essentielle (HE) d’origan et du carvacrol. Ceci a été confirmé par la détermination de la Concentration Minimale Inhibitrice (CMI) qui a été de 0,0031 % pour l’HE d’origan et de 0,0625 % pour son principe actif, le carvacrol. Ces résultats permettent de conclure que l’utilisation de l’huile d’origan ou de son constituant majoritaire le carvacrol, pourrait être une alternative efficace pour pallier aux résistances bactériennes sans cesse en évolution. Cependant, il serait intéressant de concrétiser ces résultats sur le vivant afin d’évaluer l’efficacité in-vivo de cette huile et de ses principes actifs.

The economic and health impact of mycotoxins between now and tomorrow
Par la production des Mycotoxines, les champignons toxinogènes constituent un grand danger pour la santé animal et la santé humain. Ces mycotoxines sont des contaminants naturels des aliments pour animaux. Ils ont un impact significatif sur la santé et la production animal. L’exposition aigue ou chronique de l’animal à ces substances toxiques engendre de très grandes pertes économiques. Plusieurs études récentes ont établi un lien entre l’exposition à divers mycotoxines et des effets néfastes sur la santé animale ainsi que de réelles pertes économiques. La première catégorie est relative au rejet des aliments ou matières première contaminé avec des niveaux élevés en mycotoxines. La deuxième catégorie concerne les effets néfastes sur la santé des animaux avec de faibles performances et un manque à gagner pour l’éleveur. Notre travail consiste en une synthèse bibliographique avec un aperçu sur les principales mycotoxines (Aflatoxines, Trichotecenes, Zeralenone, Fumonisine et l’ochratoxines) et leurs effets sur la santé animale ainsi que sur la règlementation nationale et internationale relative à ces mycotoxines.

These mycotoxins are natural contaminants in animal feed. They have a significant impact on animal health and production. Acute or chronic exposure of animals to these toxic substances results in very great economic loss. Several recent studies have linked exposure to various mycotoxins to adverse effects on animal health as well as real economic losses. The first category relate to the rejection of food or raw materials contaminated with high levels of mycotoxins. The second category concerns the harmful effects on the health of animals with poor performance. Our work consists of a literature review on the main mycotoxins occurring in animal feeds (Aflatoxins, Trichotecenes, Zeralenone, Fumonisin and ochratoxins) and their effects on animal health as well as reviewing the national and international regulation of these myctoxins.

Kinetics of maternal antibodies against low pathogenic avian influenza virus H9N2 and interference with vaccination in broiler chickens
La vaccination systématique des reproducteurs contre l’IAFP H9N2 permet la transmission d’une immunité passive à leur descendance. Cependant, les anticorps maternels anti-H9N2 présents chez les jeunes poussins à l’éclosion, leurs offrent une certaine protection pendant les premiers jours d’âge et peuvent de même provoquer une interférence avec la vaccination. Le but de cette étude est d'évaluer la cinétique des AOMs en présence et/ou en absence de vaccination. Dans la première partie, nous avons suivi la cinétique des anticorps anti-H9N2 chez des poussins non vaccinés élevés en isolement. Dans la deuxième partie, des lots de poulets de chair vaccinés avec 4 différents vaccins, administrés à 3 différents âges, répartis dans plusieurs régions du Maroc ont fait l’objet d’un suivi sérologique, sur l’ensemble des résultats obtenus, un effet âge de vaccination a été prouvé par analyse statistique ce qui met en cause une interférence entre les vaccins anti-IAFP H9N2 et les anticorps d’origine maternelles.

However, the maternal anti-H9N2 antibodies present in day old chicks offer them some protection during the first days of age and may interfere with vaccination. The aim of this study is to assess the kinetics of AOMs in the presence and/or in the absence of vaccination. In the first part, we followed the kinetics of anti-H9N2 antibodies in unvaccinated chicks reared in isolation. In the second part, batches of broilers vaccinated with 4 different vaccines, administered at 3 different ages, distributed in several regions of Morocco were the subject of serological monitoring, on all the results obtained, an effect Age of vaccination has been proven by statistical analysis which calls into question the interference between H9N2 anti-IAFP vaccines and maternal antibodies.

Les probiotiques: Seraient-ils la nouvelle génération naturelle des cofacteurs promoteurs de croissance chez le poulet de chair ?
Growth promoter antibiotics (GPAs) play an important role in improving growth performances by preventing intestinal pathogenic bacteria proliferation such us Escherichia coli, Salmonella and clostridium. However, the large use of GPAs led to antibio-resistance and residues in both food of poultry origin (offal...) and environment which has negative effects on public health and meat quality. Thus, several clinical trials have been conducted to evaluate the effects of potential alternatives to ban GPAs’ use in poultry breeding. In fact, effects of those alternatives on gut health and growth performances, have been reported to be similar at GPAs especially ensuring intestinal microbiota balance and improving gut health and zootechnical performances. Thus, there is a need for further investigations to determine: strains, doses, synergistic combination, administration’ moment and cost of the probiotics per bird to establish appropriate protocols. The current review paper sheds light on the different aspects that define probiotics, going from their mechanism of action and their effects on zootechnical parameters and the digestive health on broilers to conclude with an overview on the economic indicators that succeed in their use and the revolutionary concept of the Ovo-feed.
Key words: GPA, probiotics, broilers, gut health, growth performancesDepuis leur apparition, les antibiotiques promoteurs de croissance (APC) ont joué un rôle crucial dans l’amélioration des performances zootechniques à travers la prévention de la prolifération des bactéries pathogènes au niveau intestinal du poulet, notamment Escherichia coli, salmonelles et clostridies. Les différentes investigations scientifiques ont prouvé que leur utilisation à grande échelle a engendré une antibio-résistance et des résidus dans l’environnement, la viande et les denrées alimentaires d’origine avicole (abats...) ce qui peut se répercuter négativement sur la santé publique et l’image de qualité de ces produits. Par conséquent, il était primordial de trouver des alternatives potentielles aux antibiotiques promoteurs de croissance afin de bannir leur utilisation en élevage avicole. On compte parmi ces produits précédemment cités, une multitude de substances et de micro-organismes, notamment les probiotiques. En effet, ces substituts ont prouvé leur faculté à égaler les effets escomptés par un APC à savoir assurer un bon équilibre du microbiote intestinal et améliorer la santé digestive et les performances zootechniques. Ainsi, il serait très lucide d’approfondir les investigations afin de déterminer la souche, la dose, le moment d’administration, les associations synergiques avec d’autre produits alternatifs ainsi que le coût de revient par individu des probiotiques afin d’établir un protocole d’usage en fonction des effets souhaités. L’objectif de ce travail est de mettre l’accent sur les différents volets qui définissent les probiotiques et ce en allant de leur mécanisme d’action et leurs effets sur les paramètres zootechniques et la santé digestive du poulet de chair pour conclure avec un aperçu sur les indicateurs économiques qui succèdent à leur usage tout en passant par un léger préambule sur le concept révolutionnaire de l’Ovo-feeding.

Séroprévalence de Mycoplasma gallisepticum et de Mycoplasma synoviae dans les élevages reproducteurs type poulet de chair au Maroc de 1983 au 2005
Les mycoplasmoses aviaires, particulièrement Mycoplasma gallisepticum (MG) et Mycoplasma synoviae (MS) sont des infections insidieuses respiratoires, génitales ou articulaires, entraînant de lourdes pertes économiques dans les différents types d’élevage avicole.L’objectif de cette étude est de tracer l’évolution des infections mycoplasmiques aviaires dues à MG et MS à partir d’une synthèse des enquêtes sérologiques réalisées au Maroc et d’évaluer les retombées de la nouvelle loi sanitaire. Ces enquêtes ont concerné 12, 13 et 15 élevages de poules reproductrices situés dans différentes régions, notamment les principaux axes de productions avicoles réalisées respectivement en 1983, 2002 et 2005. Les sérums recueillis ont été testés par Agglutination Rapide sur Lame (ARL) afin d’identifier les élevages positifs. Les résultats sérologiques ont révélé la présence des anticorps contre MG et MS avec des taux d’infection variables en fonction de l’année d’étude et du germe. En effet, Les prévalences obtenues sont très importantes surtout pour MS qui dépassent les 50% ; un pic a été noté en 2002 (MG:30,76%) et (MS:76,92%), avec une légère régression en 2005 (MG:26,67%) et (MS:66,67 %).Ces taux d’infections non négligeables incitent à l’amélioration des normes d’hygiène et de biosécurité avec un accompagnement vétérinaire rigoureux conformément aux directives de la loi

Détection de souches multi-résistantes d’Escherichia coli d’origine aviaire dans la région de Rabat-Salé-Zemmour-Zaer
Antimicrobial resistance surveillance is a standard measure in the sanitary policies. In order to monitor multi-resistance within APEC strains, a study was conducted on 370 broiler chickens farms from the Rabat-Salé-Zemmour-Zaer region in Morocco. The isolation of Escherichia coli was made from lesions of avian colibacillosis. Biochemical identification, serotyping and antimicrobial susceptibility tests as well as phenotypic identifications on lactamases resistance were carried out on the isolates. Serogrouping showed that 66.7 % of the isolates belong to the dominant serotypes O78, O2 and O1. Drug sensitivity tests showed extremely high levels of resistance to oxytetracyclin (100 %), amoxicillin (90.9%), trimethoprim + sulphamethoxazol (82.2 %), enrofloxacin (75.9 %) and florfenicol (61.5 %). Low frequencies of resistances were noted for gentamicin (24.8 %), fosfomycin (16.1 %) and colistin (2%). In parallel, tests for the determination of the phenotype of resistance to beta-lactamases and cephalosporinases on 250 strains revealed that 66.1% of the strains were classified as class A penicillinases; 14.8% were class C cephalosporinases. The ESBL class A was found in 11.1% of the tested stains and only 3.7% were classified as wild-type strains. The multi-resistance is very high since 97.4% and 55.1% of the strains were respectively resistant to three and five antibacterial drugs.

La surveillance de l’antibio-résistance de germes spécifiques comme E. coli, Salmonella spp., Staph. aureus ou Campylobacter requiert une importance particulière dans la politique sanitaire humaine et vétérinaire des pays. Dans cette optique, une étude rétrospective sur l’antibio-résistance de 370 cultures d’Escherichia coli a été réalisée dans les élevages avicoles de la région de Rabat-Salé -Zemmour-Zaer. Les bactéries ont été identifiées par les mini galeries API et sérotypées par les immunsérums aviaires O1, O2 et O78. Les tests de sensibilité aux antibiotiques ont permis de distinguer entre antibiotiques connaissant de forts taux de résistance à savoir: l’oxytétracycline (100 %), l’amoxicilline (90,9 %), trimethoprim + sulphamethoxazole (82,2 %), l’enrofloxacine (75,9 %) et le florfénicol (61,5 %) et des molécules à faibles taux de résistance comme la gentamicine (24,8 %), la fosfomycine à (16,1 %) et la colistine (2,94 %). En parallèle, des tests de détermination du phénotype de résistance aux béta-lactamases et aux céphalosporinases sur 250 souches ont permis de mettre en évidence que 66,1 % des souches testées étaient classées parmi les pénicillinases classe A; 14,8 % étaient parmi les céphalosporinases C, les BLSE classe A comptaient 11,1 % et seulement 3,7 % des souches testées étaient classées phénotype sauvage. La multi-résistance reste très élevée avec une fréquence des souches résistantes à trois molécules d’antibactériens de l’ordre de 97 % et 55 % des souches sont résistantes à cinq antibiotiques. Ces résultats sont utiles aux vétérinaires praticiens, pour éviter les échecs thérapeutiques, et constituent une base de données importante en matière de pharmaco-vigilance et de l’épidémio-surveillance de l’antibio-résistance dans notre pays.

Application du système de scoring lésionnel pour contribuer à l’étude de la pathogénicité du virus de l’influenza aviaire faiblement pathogène H9N2 chez le poulet de chair infecté expérimentalement
Following the introduction of the low pathogenic avian influenza virus H9N2 in Morocco in 2016, a study of the pathogenicity of the virus was carried out, to fully understand the pathophysiology of the disease and its role in the damage observed in the field. . The study was carried out under controlled conditions so that the true impact of the H9N2 virus alone could be demonstrated by limiting concomitant infections. To do this, 110 day-old broilers vaccinated to one day of age against the low pathogenic avian influenza virus H9N2 were reared and maintained in isolators. At 21 days, the chicks were divided into three batches, two batches (vC, nvC) were inoculated with the Moroccan strain of the H9N2 virus (GenBank accession number: LT 598532) and an uncontested control batch (vNC). The results of clinical examinations revealed only respiratory discomfort and no mortality. Two autopsies were scheduled for 5 and 12 days post-challenge. Lesionally, tracheitis, thickening of the air sacs, and pulmonary congestion were observed during the first autopsy, during the second, only the thickening of the air sacs persisted. The histopathological study carried out on the organs removed during the autopsies, revealed lymphohistiocytic tracheitis, severe bronchitis, mild lymphocytic nephritis and lymphocyte depletion in the spleen at 5 days post challenge, 12 days post-challenge , the lesions regressed and only a few cases of tracheitis were detected. The present study is the first to have investigated the pathogenic effect of the virus alone under controlled conditions, the results indicate that infection with the virus alone does not reproduce the disease as reported in the field and that of other factors (concomitant infections, breeding conditions and environmental stress, etc.) may be at the origin of the seriousness of the damage reported, the team also concluded that the Moroccan strain of the H9N2 AI virus has an essentially respiratory tropism and a weak nephropathogenic effect.

L’étude a été effectuée dans des conditions contrôlées pour pouvoir ressortir le véritable impact du virus H9N2 seul en limitant les infections concomitantes. Pour ce faire, 110 poulets de chair d’un jour vaccinés à un jour d’âge, contre le virus de l’influenza aviaire faiblement pathogène H9N2, ont été élevés et entretenus dans des isolateurs. A 21 jours, les poussins ont été répartis en trois lots, deux lots (vC, nvC) ont été inoculé par la souche marocaine du virus H9N2 (GenBank accession number : LT 598532) et un lot témoin non challengé (vNC). Les résultats des examens cliniques ont révélé une gêne respiratoire seulement et aucune mortalité. Deux autopsies ont été programmées à 5 et à 12 jours post-challenge. Sur le plan lésionnel, une trachéite, un épaississement des sacs aériens, et une congestion pulmonaire ont été observés pendant la première autopsie, lors de la deuxième, seulement l’épaississement des sacs aériens a persisté. L’étude histopathologique réalisée sur les organes prélevés lors des autopsies, a révélé une trachéite lympho-histiocytaire, une bronchite sévère, une néphrite lymphocytaire légère et une déplétion lymphocytaire au niveau de la rate à 5 jours post challenge, à 12 jours post-challenge, les lésions ont régressé et seulement quelques cas de trachéite ont été détectés. La présente étude est la première à avoir étudié l’effet pathogène du virus seul dans des conditions contrôlées, les résultats indiquent que l’infection par le virus seul ne reproduit pas la maladie telle qu’elle est rapportée dans le terrain et que d’autres facteurs (infections concomitantes, conditions d’élevage et stress environnemental…) peuvent être à l’origine de la gravité des dégâts rapportés, l’équipe a aussi conclu que la souche marocaine du virus de l’IA H9N2 a un tropisme essentiellement respiratoire et un effet néphropathogène faible.

Semi-quantitative scoring system for the evaluation of microscopic lesions in the trachea of turkeys following experimental infection with LPAI H9N2
Dans le cadre de l’étude de la pathogénicité et de l’évaluation de la vaccination du virus de l’Influenza Aviaire faiblement pathogène H9N2 chez la dinde ; une étude histopathologique des lésions engendrées par ce virus suite à l’infection expérimentale de la dinde a été réalisée. Ainsi, 80 dindonneaux ont été mis en essai, et vaccinés contre H9N2 à 8 jours d’âge et le challenge à 26 jours. Un monitoring clinique a été effectué après le challenge, avec la réalisation de deux autopsies à 5 jours et à 10 jours post-challenge. Les dindonneaux ont manifesté des signes respiratoires avec des râles trachéaux, la toux, une sinusite avec des écoulements nasaux et oculaires mousseux, associés à des symptômes généralisés y compris l’entassement, la léthargie et un plumage ébouriffé. Des taux de mortalité de 50 et 20% ont été relevés respectivement chez le groupe de dindonneaux non vaccinés et vaccinés. Sur le plan lésionnel, une sinusite fibrineuse, une trachéite, et une aérosacculite fibrineuse ont été notés chez les deux groupes challengés, à moindre degré chez les vaccinés, avec une pneumonie fibrineuse détectée seulement chez les non vaccinés. L’étude histpathologique a révélé des lésions similaires à la fois chez les groupes vaccinés et non vaccinés à 5 et à 10 jours post-challenge, qui ne différent que par leur gravité. Ainsi un système de scoring de 4 grades (de 0 à 3) a été adopté pour évaluer les lésions trachéales chez les lots d’oiseaux étudiés, la comparaison de ceux-ci s’est basée principalement, sur l’évolution des grades et la précocité de retour à la normale (grade 0). Les lésions ont été plus sévères pendant l’autopsie de 5 JPC, une différence entre les groupes a été relevée, mais aucune différence statistique n’a été démontrée. En conclusion, le système de scoring procure des résultats cohérents ce qui prouve sa fiabilité en tant qu’outil supplémentaire dans les études expérimentales en pathologie aviaire.
Mots-clés: Lésions Histopathologiques, scoring lésionnel, dinde, H9N2, infection expérimentaleThe present study has been designed to contribute both in the evaluation of the pathogenicity of the Moroccan strain of low pathogenic avian influenza virus (LPAI) H9N2 in turkeys and vaccine efficacy against it. For this purpose, clinical monitoring and examination of gross and hitopathological lesions following vaccination at 8 days and a challenge at 26 days of age were carried out. The study was conducted on 80 turkeys, which were divided into 4 groups (A: vaccinated and challenged, B: vaccinated and unchallenged, C: unvaccinated and challenged, and D: unvaccinated and unchallenged). Birds were observed daily for 15 days after the challenge for clinical signs. 10 poults from each group were euthanized and autopsied 5 days post-challenge (DPC) and at 10 DPC. Gross lesions were evaluated during necropsy, and trachea samples were collected, and placed in a 10% neutral buffered formalin solution for histopathological examination. All challenged birds exhibited respiratory signs during the observation period which consisted of rales, coughing, sinusitis and ocular and nasal discharges, with lethargy and ruffled feathers. Mortalities of 20 % and 50 % starting 7 DPC were recorded in group A and B respectively. Necropsy revealed fibrinous sinusitis, tracheitis, and fibrinous aerosacculitis in groups A and B with pneumonia only in group B at 5 DPC. Histopathology of the trachea in all the infected birds revealed a lympho-plasmocytic tracheitis with mononuclear cell infiltrates, hyperplasia of epithelial cells, metaplasia, and loss of cilia and mucous glands. Lesions exhibited a varying range of severity depending on days post challenge and groups. Thus a scoring system of 4 grades was created to evaluate and compare groups. Score 0 corresponds to normal tracheal structure, histologic scores of 1 and 2 are characterized mainly by congested blood vessels, depletion of specialized cells, and mononuclear infiltrates, while score 3 corresponds to a histologic pattern of notable thickening of mucosal lining by hyperplasia of epithelial cells, severely congested blood vessels, and metaplasia of surface epithelial cells. Lesions were significantly more severe at 5 DPC, and a visual but non-statistical difference between groups at the same necropsy was noted. The scoring system showed consistent results and helped broaden the perspectives of the present study, and proved to be a reliable tool in the setting of experimental studies in avian pathology.
