    Development of a multiplex DNA-based traceability tool for crop plant materials

    The authenticity of food is of increasing importance for producers, retailers and consumers. All groups benefit from the correct labelling of the contents of food products. Producers and retailers want to guarantee the origin of their products and check for adulteration with cheaper or inferior ingredients. Consumers are also more demanding about the origin of their food for various socioeconomic reasons. In contrast to this increasing demand, correct labelling has become much more complex because of global transportation networks of raw materials and processed food products. Within the European integrated research project ‘Tracing the origin of food’ (TRACE), a DNA-based multiplex detection tool was developed—the padlock probe ligation and microarray detection (PPLMD) tool. In this paper, this method is extended to a 15-plex traceability tool with a focus on products of commercial importance such as the emmer wheat Farro della Garfagnana (FdG) and Basmati rice. The specificity of 14 plant-related padlock probes was determined and initially validated in mixtures comprising seven or nine plant species/varieties. One nucleotide difference in target sequence was sufficient for the distinction between the presence or absence of a specific target. At least 5% FdG or Basmati rice was detected in mixtures with cheaper bread wheat or non-fragrant rice, respectively. The results suggested that even lower levels of (un-)intentional adulteration could be detected. PPLMD has been shown to be a useful tool for the detection of fraudulent/intentional admixtures in premium foods and is ready for the monitoring of correct labelling of premium foods worldwide

    Morphological evaluation of cardoon (Cynara cardunculus L.): assessing biodiversity for applications based on tradition, innovation and sustainability

    Selected cardoon cultivars established in the “Serra da Estrela” region were analysed to evaluate morphological characteristics related principally to the production of cardoon flowers, which are a compulsory ingredient for particular Mediterranean PDO cheese regions. The biodiversity of twelve cardoon cultivars installed in an experimental field were evaluated over three growing seasons using thirty-four morphological descriptors. Statistically significant differences were found between cultivars for twenty-four morphological characteristics which indicate a wide genetic diversity. The relationship among the cultivars and characteristics was analysed using principal component analysis. A three dimensional template was found to be very significant and explained 71% of the total variation. The first component is dominated positively by plant height, diameter of stalk, inflorescence characteristics and flower production, while the second component is positively dominated by leaf characteristics. Cultivars A26, D32 and D33, present a plant architecture simultaneously well adapted for flower production, ease of harvesting and plant biomass. These characterizations and understandings can be useful for a plant breeding programme to develop cultivars for innovative potential applications besides flowers, and also for application by other cheese producers in Mediterranean regions.info:eu-repo/semantics/publishedVersio

    Impact of Human Management on the Genetic Variation of Wild Pepper, Capsicum annuum var. glabriusculum

    Management of wild peppers in Mexico has occurred for a long time without clear phenotypic signs of domestication. However, pre-domestication management could have implications for the population's genetic richness. To test this hypothesis we analysed 27 wild (W), let standing (LS) and cultivated (C) populations, plus 7 samples from local markets (LM), with nine polymorphic microsatellite markers. Two hundred and fifty two alleles were identified, averaging 28 per locus. Allele number was higher in W, and 15 and 40% less in LS and C populations, respectively. Genetic variation had a significant population structure. In W populations, structure was associated with ecological and geographic areas according to isolation by distance. When LM and C populations where included in the analysis, differentiation was no longer apparent. Most LM were related to distant populations from Sierra Madre Oriental, which represents their probable origin. Historical demography shows a recent decline in all W populations. Thus, pre-domestication human management is associated with a significant reduction of genetic diversity and with a loss of differentiation suggesting movement among regions by man. Measures to conserve wild and managed populations should be implemented to maintain the source and the architecture of genetic variation in this important crop relative
