    Dark energy in hybrid inflation

    The situation that a scalar field provides the source of the accelerated expansion of the universe while rolling down its potential is common in both the simple models of the primordial inflation and the quintessence-based dark energy models. Motivated by this point, we address the possibility of causing the current acceleration via the primordial inflation using a simple model based on hybrid inflation. We trigger the onset of the motion of the quintessence field via the transition field, and find that the fate of the universe depends on the true vacuum energy determined by choosing the parameters.     Sustainability of multi-field inflation and bound on string scale

    We study the effects of the interaction terms between the inflaton fields on the inflationary dynamics in multi-field models. With power law type potential and interactions, the total number of e-folds may get considerably reduced and can lead to unacceptably short period of inflation. Also we point out that this can place a bound on the characteristic scale of the underlying theory such as string theory.     A covariant approach to general field space metric in multi-field inflation

    We present a covariant formalism for general multi-field system which enables us to obtain higher order action of cosmological perturbations easily and systematically. The effects of the field space geometry, described by the Riemann curvature tensor of the field space, are naturally incorporated.     Multiple scalar particle decay and perturbation generation

    We study the evolution of the universe which contains a multiple number of non-relativistic scalar fields decaying into both radiation and pressureless matter. We present a powerful analytic formalism to calculate the matter and radiation curvature perturbations, and find that our analytic estimates agree with full numerical results within an error of less than one percent. Also we discuss the isocurvature perturbation between matter and radiation components, which may be detected by near future cosmological observations, and point out that it crucially depends on the branching ratio of the decay rate of the scalar fields and that it is hard to make any model independent predictions.Comment: (v1) 22 pages, 4 figures, 2 tables; (v2) JHEP3 style, 24 pages, references added and typos corrected, to appear in JCA

    Efficient derivation of dopaminergic neurons from SOX1(-) floor plate cells under defined culture conditions.

    BACKGROUND: Parkinson's disease (PD) is a severe neurodegenerative disease associated with loss of dopaminergic neurons. Derivation of dopaminergic neurons from human embryonic stem cells (hESCs) could provide new therapeutic options for PD therapy. Dopaminergic neurons are derived from SOX(-) floor plate (FP) cells during embryonic development in many species and in human cell culture in vitro. Early treatment with sonic hedgehog (Shh) has been reported to efficiently convert hESCs into FP lineages. METHODS: In this study, we attempted to utilize a Shh-free approach in deriving SOX1(-) FP cells from hESCs in vitro. Neuroectoderm conversion from hESCs was achieved with dual inhibition of the BMP4 (LDN193189) and TGF-ÎČ signaling pathways (SB431542) for 24 h under defined culture conditions. RESULTS: Following a further 5 days of treatment with LDN193189 or LDN193189 + SB431542, SOX1(-) FP cells constituted 70-80 % of the entire cell population. Upon treatment with Shh and FGF8, the SOX1(-) FP cells were efficiently converted to functional Nurr1(+) and TH(+) dopaminergic cells (patterning), which constituted more than 98 % of the entire cell population. However, when the same growth factors were applied to SOX1(+) cells, only less than 4 % of the cells became Nurr1(+), indicating that patterning was effective only if SOX1 expression was down-regulated. After transplanting the Nurr1(+) and TH(+) cells into a hemiparkinsonian rat model, significant improvements were observed in amphetamine induced ipslateral rotations, apomorphine induced contra-lateral rotations and Rota rod motor tests over a duration of 8 weeks. CONCLUSIONS: Our findings thus provide a convenient approach to FP development and functional dopaminergic neuron derivation.published_or_final_versio

    Oxygen radical-mediated oxidation reactions of an alanine peptide motif - density functional theory and transition state theory study

    Background: Oxygen-base (O-base) oxidation in protein backbone is important in the protein backbone fragmentation due to the attack from reactive oxygen species (ROS). In this study, an alanine peptide was used model system to investigate this O-base oxidation by employing density functional theory (DFT) calculations combining with continuum solvent model. Detailed reaction steps were analyzed along with their reaction rate constants. Results: Most of the O-base oxidation reactions for this alanine peptide are exothermic except for the bond-breakage of the C-alpha-N bond to form hydroperoxy alanine radical. Among the reactions investigated in this study, the activated energy of OH alpha-H abstraction is the lowest one, while the generation of alkylperoxy peptide radical must overcome the highest energy barrier. The aqueous situation facilitates the oxidation reactions to generate hydroxyl alanine peptide derivatives except for the fragmentations of alkoxyl alanine peptide radical. The C-alpha-C-beta bond of the alkoxyl alanine peptide radical is more labile than the peptide bond. Conclusion: the rate-determining step of oxidation in protein backbone is the generation of hydroperoxy peptide radical via the reaction of alkylperoxy peptide radical with HO2. The stabilities of alkylperoxy peptide radical and complex of alkylperoxy peptide radical with HO2 are crucial in this O-base oxidation reaction

    Hip fracture risk assessment: Artificial neural network outperforms conditional logistic regression in an age- and sex-matched case control study

In this age- and sex-matched case control study, we examined the risk factors of hip fractures and assessed the fracture risk by conditional logistic regression (CLR) and ensemble artificial neural network (ANN). The performances of these two classifiers were compared. Methods - The study population consisted of 217 pairs (149 women and 68 men) of fractures and controls with an age older than 60 years. All the participants were interviewed with the same standardized questionnaire including questions on 66 risk factors in 12 categories. Univariate CLR analysis was initially conducted to examine the unadjusted odds ratio of all potential risk factors. The significant risk factors were then tested by multivariate analyses. For fracture risk assessment, the participants were randomly divided into modeling and testing datasets for 10-fold cross validation analyses. The predicting models built by CLR and ANN in modeling datasets were applied to testing datasets for generalization study. The performances, including discrimination and calibration, were compared with non-parametric Wilcoxon tests. Results - In univariate CLR analyses, 16 variables achieved significant level, and six of them remained significant in multivariate analyses, including low T score, low BMI, low MMSE score, milk intake, walking difficulty, and significant fall at home. For discrimination, ANN outperformed CLR in both 16- and 6-variable analyses in modeling and testing datasets (p?<?0.005). For calibration, ANN outperformed CLR only in 16-variable analyses in modeling and testing datasets (p?=?0.013 and 0.047, respectively). Conclusions - The risk factors of hip fracture are more personal than environmental. With adequate model construction, ANN may outperform CLR in both discrimination and calibration.     Non-Gaussianity from false vacuum inflation: Old curvaton scenario

    We calculate the three-point correlation function of the comoving curvature perturbation generated during an inflationary epoch driven by false vacuum energy. We get a novel false vacuum shape bispectrum, which peaks in the equilateral limit. Using this result, we propose a scenario which we call "old curvaton". The shape of the resulting bispectrum lies between the local and the false vacuum shapes.     Effective theories of single field inflation when heavy fields matter

    We compute the low energy effective field theory (EFT) expansion for single-field inflationary models that descend from a parent theory containing multiple other scalar fields. By assuming that all other degrees of freedom in the parent theory are sufficiently massive relative to the inflaton, it is possible to derive an EFT valid to arbitrary order in perturbations, provided certain generalized adiabaticity conditions are respected. These conditions permit a consistent low energy EFT description even when the inflaton deviates off its adiabatic minimum along its slowly rolling trajectory. By generalizing the formalism that identifies the adiabatic mode with the Goldstone boson of this spontaneously broken time translational symmetry prior to the integration of the heavy fields, we show that this invariance of the parent theory dictates the entire non-perturbative structure of the descendent EFT. The couplings of this theory can be written entirely in terms of the reduced speed of sound of adiabatic perturbations. The resulting operator expansion is distinguishable from that of other scenarios, such as standard single inflation or DBI inflation. In particular, we re-derive how certain operators can become transiently strongly coupled along the inflaton trajectory, consistent with slow-roll and the validity of the EFT expansion, imprinting features in the primordial power spectrum, and we deduce the relevant cubic operators that imply distinct signatures in the primordial bispectrum which may soon be constrained by observations.Comment: (v1) 25 pages, 1 figure; (v2) references added and typos corrected, to appear in Journal of High Energy Physic
