    Analysis of Checking Account Balances and Prices to Determine the Existence and Maintenance of an Optimum Balance That Minimizes Checking Account Costs

    This study was undertaken to determine the relationship between the number of checks written and the account balance maintained. The effects of price changes was also observed. An optimal balance was calculated based upon the price schedule and the number of checks written. A particular account was placed in the appropriate optimum balance based upon the number of checks written. The accounts were sampled in such a way that the exact sane account was not chosen each month, but the same general group was sampled. There was no relationship found between the number of checks written and the balance maintained. When the optimum balance was zero during period one the R2 was 0.007, when the optimum balance was 200 dollars the R was 0.001. The only optimum balance during period two was 200 dollars and the R2 was zero. When the optimum balance was zero during period 2 three the R2 was 0.0092, when the optimum balance was 500 the R was 0.0007. During period one and period three when the optimum balance was zero the balance maintained was 526.55 and 575.40 dollars. During period one and two when the optimum balance was 200 dollars the balance maintained was 479.90 and 467.00 dollars. During period three when the optimum balance was 500 dollars the balance maintained-was 805.63 dollars. The price changes at the beginning of period two and three affected the average number of checks significantly (P.01 ). The average number of checks dropped from 34.06 during period one to 22.4G during period t110 and rose to 31.62 during period three. Throughout this study depositors were not purchasing and maintaining the correct balance. When the correct balance was zero only 5.7 and 3.87 percent purchased and maintained that balance during period one and three. When the optimum balance was 200 dollars during period one and two only 23.65 and 33.44 percent purchased and maintained that balance.     Internet Of Things (IoT) is a continually growing area which aids us to unite diverse objects. The proposed system exhibits the universal notion of utilizing cloud-based intellectual automotive car parking facilities in smart cities as a notable implementation of the IoT. Such services demonstrate to be a noteworthy part of the IoT and thus serving users in no small amount due to its pure commerce positioned qualities. Electromagnetic fields are being used by RFID to detect and track tags ascribed to objects automatically. The RFID technology is used in this system along with suitable IoT protocols to evade human interference, which reduces the cost. Information is bartered using readers and tags. RFID and IoT technologies are mainly used to automate the guide systems and make them strong and more accurate. Open Service Gateways can be effectively used for this module. This system established on the consequence of IoT and the purposes are solving the chaos, bewilderment, and extensive backlogs in parking spaces like malls and business parks that are customary as a consequence of the increased use of automobiles. The proposed work aims to solve these problems and offer car drivers a hassle-free and instantaneous car parking experience. While a number of nodes are positioned depends on topographical restrictions, positioning of prominent anchor sensor nodes in the smart parking is a primary factor against which the efficiency and cost of the parking system hang. A Raspberry Pi would act as a mini-computer in our system. A suitable smallest path methodology would be cast-off to obtain the shortest distance between the user and every car park in the system. Hence, the pausing time of the user is decreased.     Study on validity of a rapid diagnostic test kit versus light microscopy for malaria diagnosis in Ahmedabad city

    ABSTRACT Light microscopy of blood smears for diagnosis of malaria in the field has several limitations, notably delays in diagnosis. This study in Ahmedabad in Gujarat State, India, evaluated the diagnostic performance of a rapid diagnostic test for malaria (SD Bioline Malaria Ag P.f/Pan) versus blood smear examination as the gold standard. All fever cases presenting at 13 urban health centres were subjected to rapid diagnostic testing and thick and thin blood smears. A total of 677 cases with fever were examined; 135 (20.0%) tested positive by rapid diagnostic test and 86 (12.7%) by blood smear. The sensitivity of the rapid diagnostic test for malaria was 98.8%, specificity was 91.5%, positive predictive value 63.0% and negative predictive value 99.8%. For detection of Plasmodium falciparum the sensitivity of rapid diagnostic test was 100% and specificity was 97.3%. The results show the acceptability of the rapid test as an alternative to light microscopy in the field setting

    Energy cost and return for hunting in African wild dogs and Cheetahs

    African wild dogs (Lycaon pictus) are reported to hunt with energetically costly long chase distances. We used high-resolution GPS and inertial technology to record 1,119 high-speed chases of all members of a pack of six adult African wild dogs in northern Botswana. Dogs performed multiple short, high-speed, mostly unsuccessful chases to capture prey, while cheetahs (Acinonyx jubatus) undertook even shorter, higher-speed hunts. We used an energy balance model to show that the energy return from group hunting and feeding substantially outweighs the cost of multiple short chases, which indicates that African wild dogs are more energetically robust than previously believed. Comparison with cheetah illustrates the trade-off between sheer athleticism and high individual kill rate characteristic of cheetahs, and the energetic robustness of frequent opportunistic group hunting and feeding by African wild dogs

    Insufficient OPC migration into demyelinated lesions is a cause of poor remyelination in MS and mouse models

    Failure of remyelination of multiple sclerosis (MS) lesions contributes to neurodegeneration that correlates with chronic disability in patients. Currently, there are no available treatments to reduce neurodegeneration, but one therapeutic approach to fill this unmet need is to promote remyelination. As many demyelinated MS lesions contain plentiful oligodendrocyte precursor cells (OPCs), but no mature myelinating oligodendrocytes, research has previously concentrated on promoting OPC maturation. However, some MS lesions contain few OPCs, and therefore, remyelination failure may also be secondary to OPC recruitment failure. Here, in a series of MS samples, we determined how many lesions contained few OPCs, and correlated this to pathological subtype and expression of the chemotactic molecules Semaphorin (Sema) 3A and 3F. 37 % of MS lesions contained low numbers of OPCs, and these were mostly chronic active lesions, in which cells expressed Sema3A (chemorepellent). To test the hypothesis that differential Sema3 expression in demyelinated lesions alters OPC recruitment and the efficiency of subsequent remyelination, we used a focal myelinotoxic mouse model of demyelination. Adding recombinant (r)Sema3A (chemorepellent) to demyelinated lesions reduced OPC recruitment and remyelination, whereas the addition of rSema3F (chemoattractant), or use of transgenic mice with reduced Sema3A expression increased OPC recruitment and remyelination. We conclude that some MS lesions fail to remyelinate secondary to reduced OPC recruitment, and that chemotactic molecules are involved in the mechanism, providing a new group of drug targets to improve remyelination, with a specific target in the Sema3A receptor neuropilin-1.     <p>Abstract</p> <p>Background</p> <p>Protein conformation and protein/protein interaction can be elucidated by solution-phase Hydrogen/Deuterium exchange (sHDX) coupled to high-resolution mass analysis of the digested protein or protein complex. In sHDX experiments mutant proteins are compared to wild-type proteins or a ligand is added to the protein and compared to the wild-type protein (or mutant). The number of deuteriums incorporated into the polypeptides generated from the protease digest of the protein is related to the solvent accessibility of amide protons within the original protein construct.</p> <p>Results</p> <p>In this work, sHDX data was collected on a 14.5 T FT-ICR MS. An algorithm was developed based on combinatorial optimization that predicts deuterium exchange with high spatial resolution based on the sHDX data of overlapping proteolytic fragments. Often the algorithm assigns deuterium exchange with single residue resolution.</p> <p>Conclusions</p> <p>With our new method it is possible to automatically determine deuterium exchange with higher spatial resolution than the level of digested fragments.</p

    A single nuclear transcriptomic characterisation of mechanisms responsible for impaired angiogenesis and blood-brain barrier function in Alzheimer's disease

    Brain perfusion and blood-brain barrier (BBB) integrity are reduced early in Alzheimer's disease (AD). We performed single nucleus RNA sequencing of vascular cells isolated from AD and non-diseased control brains to characterise pathological transcriptional signatures responsible for this. We show that endothelial cells (EC) are enriched for expression of genes associated with susceptibility to AD. Increased β-amyloid is associated with BBB impairment and a dysfunctional angiogenic response related to a failure of increased pro-angiogenic HIF1A to increased VEGFA signalling to EC. This is associated with vascular inflammatory activation, EC senescence and apoptosis. Our genomic dissection of vascular cell risk gene enrichment provides evidence for a role of EC pathology in AD and suggests that reducing vascular inflammatory activation and restoring effective angiogenesis could reduce vascular dysfunction contributing to the genesis or progression of early AD.</p

    Impact of Immunization Technology and Assay Application on Antibody Performance – A Systematic Comparative Evaluation

    Antibodies are quintessential affinity reagents for the investigation and determination of a protein's expression patterns, localization, quantitation, modifications, purification, and functional understanding. Antibodies are typically used in techniques such as Western blot, immunohistochemistry (IHC), and enzyme-linked immunosorbent assays (ELISA), among others. The methods employed to generate antibodies can have a profound impact on their success in any of these applications. We raised antibodies against 10 serum proteins using 3 immunization methods: peptide antigens (3 per protein), DNA prime/protein fragment-boost (“DNA immunization”; 3 per protein), and full length protein. Antibodies thus generated were systematically evaluated using several different assay technologies (ELISA, IHC, and Western blot). Antibodies raised against peptides worked predominantly in applications where the target protein was denatured (57% success in Western blot, 66% success in immunohistochemistry), although 37% of the antibodies thus generated did not work in any of these applications. In contrast, antibodies produced by DNA immunization performed well against both denatured and native targets with a high level of success: 93% success in Western blots, 100% success in immunohistochemistry, and 79% success in ELISA. Importantly, success in one assay method was not predictive of success in another. Immunization with full length protein consistently yielded the best results; however, this method is not typically available for new targets, due to the difficulty of generating full length protein. We conclude that DNA immunization strategies which are not encumbered by the limitations of efficacy (peptides) or requirements for full length proteins can be quite successful, particularly when multiple constructs for each protein are used
