Mice Lacking Thyroid Hormone Receptor β Show Enhanced Apoptosis and Delayed Liver Commitment for Proliferation after Partial Hepatectomy

This is an open-access article distributed under the terms of the Creative Commons Attribution License.[Background]: The role of thyroid hormones and their receptors (TR) during liver regeneration after partial hepatectomy (PH) was studied using genetic and pharmacologic approaches. Roles in liver regeneration have been suggested for T3, but there is no clear evidence distinguishing the contribution of increased amounts of T3 from the modulation by unoccupied TRs. [Methodology/Principal Findings]: Mice lacking TR alpha 1/TR beta or TR beta alone fully regenerated liver mass after PH, but showed delayed commitment to the initial round of hepatocyte proliferation and transient but intense apoptosis at 48h post-PH, affecting similar to 30% of the remaining hepatocytes. Pharmacologically induced hypothyroidism yielded similar results. Loss of TR activity was associated with enhanced nitrosative stress in the liver remnant, due to an increase in the activity of the nitric oxide synthase (NOS) 2 and 3, caused by a transient decrease in the concentration of asymmetric dimethylarginine (ADMA), a potent NOS inhibitor. This decrease in the ADMA levels was due to the presence of a higher activity of dimethylarginineaminohydrolase-1 (DDAH-1) in the regenerating liver of animals lacking TR alpha 1/TR beta or TR beta. DDAH-1 expression and activity was paralleled by the activity of FXR, a transcription factor involved in liver regeneration and up-regulated in the absence of TR. [Conclusions/Significance]: We report that TRs are not required for liver regeneration; however, hypothyroid mice and TR beta-or TR alpha 1/TR beta-deficient mice exhibit a delay in the restoration of liver mass, suggesting a specific role for TRb in liver regeneration. Altered regenerative responses are related with a delay in the expression of cyclins D1 and E, and the occurrence of liver apoptosis in the absence of activated TRb that can be prevented by administration of NOS inhibitors. Taken together, these results indicate that TRb contributes significantly to the rapid initial round of hepatocyte proliferation following PH, and improves the survival of the regenerating liver at later times. This work was supported by grants BFU2008-02161, BFU2007-62402, SAF2007-60511, and SAF2007-60551 from MICINN; S-BIO-0283/2006 from Comunidad de Madrid and FIS-RECAVA RD06/0014/0025 to L.B.; and PI05.0050, PI080070, and the Fundacion Mutua Madrileña to S.H. This work investigates the timing, paleoclimatic framework and inter-hemispheric teleconnections inferred from the glaciers last maximum extension and the deglaciation onset in the Arid Tropical Andes. A study area was selected to the northeastward of the Nevado Coropuna, the volcano currently covered by the largest tropical glacier on Earth. The current glacier extent, the moraines deposited in the past and paleoglaciers at their maximum extension have been mapped. The present and past Equilibrium Line Altitudes (ELA and paleoELA) have been reconstructed and the chlorine-36 ages have been calculated, for preliminary absolute dating of glacial and volcanic processes. The paleoELA depression, the thermometers installed in the study area and the accumulation data previously published allowed development of paleotemperature and paleoprecipitation models. The Coropuna glaciers were in maximum extension (or glacial standstill) ~20–12 ka ago (and maybe earlier). This last maximum extension was contemporary to the Heinrich 2–1 and Younger Dryas events and the Tauca and Coipasa paleolake transgressions on Bolivian Altiplano. The maximum paleoELA depression (991 m) shows a colder (-6.4 °C) and moister climate with precipitation ×1.2–×2.8 higher than the present. The deglaciation onset in the Arid Tropical Andes was 15–11 ka ago, earlier in the most southern, arid, and low mountains and later in the northernmost, less arid, and higher mountains

Background: Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS) is characterized by persistent physical and mental fatigue. The post-COVID-19 condition patients refer physical fatigue and cognitive impairment sequelae. Given the similarity between both conditions, could it be the same pathology with a different precipitating factor? Objective: To describe the cognitive impairment, neuropsychiatric symptoms, and general symptomatology in both groups, to find out if it is the same pathology. As well as verify if the affectation of smell is related to cognitive deterioration in patients with post-COVID-19 condition. Methods: The sample included 42 ME/CFS and 73 post-COVID-19 condition patients. Fatigue, sleep quality, anxiety and depressive symptoms, the frequency and severity of different symptoms, olfactory function and a wide range of cognitive domains were evaluated. Results: Both syndromes are characterized by excessive physical fatigue, sleep problems and myalgia. Sustained attention and processing speed were impaired in 83.3% and 52.4% of ME/CFS patients while in post-COVID-19 condition were impaired in 56.2% and 41.4% of patients, respectively. Statistically significant differences were found in sustained attention and visuospatial ability, being the ME/CFS group who presented the worst performance. Physical problems and mood issues were the main variables correlating with cognitive performance in post-COVID-19 patients, while in ME/CFS it was anxiety symptoms and physical fatigue. Conclusions: The symptomatology and cognitive patterns were similar in both groups, with greater impairment in ME/CFS. This disease is characterized by greater physical and neuropsychiatric problems compared to post-COVID-19 condition. Likewise, we also propose the relevance of prolonged hyposmia as a possible marker of cognitive deterioration in patients with post-COVID-19.This study has been funded by Instituto de Salud Carlos III (ISCIII) through the project PI20/01076 and co-funded by the European Union, EITB maratoia (BIOS21/COV/006) and grants for health research projects from the Basque Government (2021111006). In a world that is increasingly unequal and always in crisis, the Nursing profession has been recognized as key to contributing to the achievement of the Sustainable Development Goals (SDGs). From the perspective of the Social Determinants of Health (SDH), the profession faces challenges to make proposals that respond to the improvement of care for the most vulnerable groups of the population. The following reflection attempts to answer the question: What are the sociopolitical challenges of nursing in the current context of crisis? This is based on the systematization of work carried out by a cohort of nurses who are studying the Master of Nursing at the Universidad de la Frontera. The resulting manifesto proposes and analyzes nine thematic axes that contribute to a positive and hopeful vision of the future of the profession.

Abstract

Background

Genetic admixture is a common caveat for genetic association analysis. Therefore, it is important to characterize the genetic structure of the population under study to control for this kind of potential bias.

Results

In this study we have sampled over 800 unrelated individuals from the population of Spain, and have genotyped them with a genome-wide coverage. We have carried out linkage disequilibrium, haplotype, population structure and copy-number variation (CNV) analyses, and have compared these estimates of the Spanish population with existing data from similar efforts.

Conclusions

In general, the Spanish population is similar to the Western and Northern Europeans, but has a more diverse haplotypic structure. Moreover, the Spanish population is also largely homogeneous within itself, although patterns of micro-structure may be able to predict locations of origin from distant regions. Finally, we also present the first characterization of a CNV map of the Spanish population. These results and original data are made available to the scientific community.</p

Challenge 1: Past global changes: a context to the Anthropocene

    Get PDF
Facing current climate and environmental crises needs long-term series of Earth Dynamics and anthropogenic pressures on the Planet. Numerous geological, chemical and biological natural archives capture large-scale, multi-temporal, abrupt, and often irreversible shifts in environmental and climate systems, providing an opportunity to better understand and therefore predict potential future impacts of the present anthropogenic warming and Humankind impact on the Planet. By providing robust, reliable, quantitative, detailed, high-resolution and long paleoclimate and paleoenvironmental data series, paleoclimatology and paleoenvironmental research place present climate variability and ecological crises in a long-term perspective to understand climate forcing mechanisms and environmental processes and responses. The success of science-based solutions to the global risks in the 21st century will strongly rely on our capacity to transfer this knowledge to politicians, managers, and society.Peer reviewe
