Vertebrate SLRP family evolution and the subfunctionalization of osteoglycin gene duplicates in teleost fish
Background Osteoglycin (OGN, a.k.a. mimecan) belongs to cluster III of the small leucine-rich proteoglycans (SLRP) of the extracellular matrix (ECM). In vertebrates OGN is a characteristic ECM protein of bone. In the present study we explore the evolution of SLRP III and OGN in teleosts that have a skeleton adapted to an aquatic environment. Results The SLRP gene family has been conserved since the separation of chondrichthyes and osteichthyes. Few gene duplicates of the SLRP III family exist even in the teleosts that experienced a specific whole genome duplication. One exception is ogn for which duplicate copies were identified in fish genomes. The ogn promoter sequence and in vitro mesenchymal stem cell (MSC) cultures suggest the duplicate ogn genes acquired divergent functions. In gilthead sea bream (Sparus aurata) ogn1 was up-regulated during osteoblast and myocyte differentiation in vitro, while ogn2 was severely down-regulated during bone-derived MSCs differentiation into adipocytes in vitro. Conclusions Overall, the phylogenetic analysis indicates that the SLRP III family in vertebrates has been under conservative evolutionary pressure. The retention of the ogn gene duplicates in teleosts was linked with the acquisition of different functions. Novel galanin receptors in teleost fish: identification, expression and regulation by sex steroids
In fish, the onset of puberty, the transition from juvenile to sexually reproductive adult animals, is triggered by the activation of pituitary gonadotrophin secretion and its timing is influenced by
external and internal factors that include the growth/adiposity status of the animal. Kisseptins have been implicated in the activation of puberty but peripheral signals coming from the immature gonad or associated to the metabolic/nutritional status are also thought to be involved. Additionally, there is evidence that the galaninergic system in the brain and testis of pre-pubertal male sea bass is a possible mediator involved in the translation of somatic signals leading to gonadal maturation. Here, the transcripts for four galanin receptors (GALR), named GALR1a, 1b, 2a and 2b, were isolated from
European sea bass, Dicentrarchus labrax. Phylogenetic analysis confirmed the previously reported duplication of GALR1 in teleost fish, and unravelled the duplication of GALR2 in teleost fish and in some tetrapod species. Comparison with human showed that the key amino acids involved in ligand binding are present in the corresponding GALR1 and GALR2 orthologues. Transcripts for all four receptors are expressed in brain and testes of adult fish with GALR1a and GALR1b abundant in testes
15 and hardly detected in ovaries. In order to investigate whether GALR1 dimorphic expression was dependent on steroid context we evaluated the effect of 11-ketotestosterone and 17β-estradiol
treatments on the receptor expression in brain and testes of pre-pubertal males. Interestingly, steroid treatments had no effect on the expression of GALRs in the brain while in the testes, GALR1a and
GALR1b were significantly up regulated by 11KT. Altogether, these results support a role for the galaninergic system, in particular the GALR1 paralog in fish reproductive function
Upconverting nanoparticles as primary thermometers and power sensors
Luminescence thermometry is a spectroscopic technique for remote
temperature detection based on the thermal dependence of the
luminescence of phosphors, presenting numerous applications ranging from
biosciences to engineering. In this work, we use the Er3+ emission of the
NaGdF4/NaGdF4:Yb3+,Er3+/NaGdF4 upconverting nanoparticles upon 980 nm
laser excitation to determine simultaneously the absolute temperature and the
excitation power density. The Er3+ 2H11/2→4
I15/2 and 4
S3/2→4
I15/2 emission bands,
which are commonly used for thermometric purposes, overlap with the 2
H9/2
→4
I13/2 emission band, which can lead to erroneous temperature readout.
Applying the concept of luminescent primary thermometry to resolve the
overlapping Er3+ transitions, a dual nanosensor synchronously measuring the
temperature and the delivered laser pump power is successfully realized

Oxidative Stress in Neurodegenerative Diseases: Mechanisms and Therapeutic Perspectives
The incidence and prevalence of neurodegenerative diseases (ND) increase with life expectancy. This paper reviews the role of oxidative stress (OS) in ND and pharmacological attempts to fight against reactive oxygen species (ROS)-induced neurodegeneration. Several mechanisms involved in ROS generation in neurodegeneration have been proposed. Recent articles about molecular pathways involved in ROS generation were reviewed. The progress in the development of neuroprotective therapies has been hampered because it is difficult to define targets for treatment and determine what should be considered as neuroprotective. Therefore, the attention was focused on researches about pharmacological targets that could protect neurons against OS. Since it is necessary to look for genes as the ultimate controllers of all biological processes, this paper also tried to identify gerontogenes involved in OS and neurodegeneration. Since neurons depend on glial cells to survive, recent articles about the functioning of these cells in aging and ND were also reviewed. Finally, clinical trials testing potential neuroprotective agents were critically reviewed. Although several potential drugs have been screened in in vitro and in vivo models of ND, these results were not translated in benefit of patients, and disappointing results were obtained in the majority of clinical trials
Developmental expression of DAXI in the European sea bass, Dicentrarchus labrax: lack of evidence for sexual dimorphism during sex differentiation
Background: DAX1 (NR0B1), a member of the nuclear receptors super family, has been shown
to be involved in the genetic sex determination and in gonadal differentiation in several vertebrate
species. In the aquaculture fish European sea bass, Dicentrarchus labrax, and in the generality of
fish species, the mechanisms of sex determination and differentiation have not been elucidated. The
present study aimed at characterizing the European DAX1 gene and its developmental expression
at the mRNA level.
Methods: A full length European sea bass DAX1 cDNA (sbDAX1) was isolated by screening a
testis cDNA library. The structure of the DAX1 gene was determined by PCR and Southern blot.
Multisequence alignments and phylogenetic analysis were used to compare the translated sbDAX1
product to that of other vertebrates. sbDAX1 expression was analysed by Northern blot and
relative RT-PCR in adult tissues. Developmental expression of mRNA levels was analysed in groups
of larvae grown either at 15°C or 20°C (masculinising temperature) during the first 60 days, or two
groups of fish selected for fast (mostly females) and slow growth.
Results: The sbDAX1 is expressed as a single transcript in testis and ovary encoding a predicted
protein of 301 amino acids. A polyglutamine stretch of variable length in different DAX1 proteins
is present in the DNA binding domain. The sbDAX1 gene is composed of two exons, separated by
a single 283 bp intron with conserved splice sites in same region of the ligand binding domain as
other DAX1 genes. sbDAX1 mRNA is not restricted to the brain-pituitary-gonadal axis and is also
detected in the gut, heart, gills, muscle and kidney. sbDAX1 mRNA was detected as early as 4 days
post hatching (dph) and expression was not affected by incubation temperature. Throughout
gonadal sex differentiation (60–300 dph) no dimorphic pattern of expression was observed.
Conclusion: The sbDAX1 gene and putative protein coding region is highly conserved and has a
wide pattern of tissue expression. Although gene expression data suggests sbDAX1 to be
important for the development and differentiation of the gonads, it is apparently not sex specific
Surface crystallization of ionic liquid crystals
The evidence for surface crystallization in ionic liquids is scarce. The existing reports seem to be contradictory
as for its driving forces, since in the two compounds investigated in the literature, the
contribution of coloumbic and van der Waals forces is very different. In this work 1-dodecyl-3-
methylimidazolium tetrafluoroborate was studied and its surface crystallization characterized by surface
tension, ellipsometry and optical microscopy. The results obtained seem to reconcile previous
observations, and it was further shown, using the same techniques, that this phenomenon is prevalent in
other ionic liquids. MD simulation results illustrate the different possibilities of organization, providing
reasonable models to rationalize the experimental observations.This work was developed with the scope of the project CICECO –

Anti-proliferative effects of novel hydroxyamides and triazoles
Chemotherapy is a major cancer treatment option. The synthesis of new compounds with anti-proliferative properties and specificity is a current challenge in drug-discovery today. Our goal was to develop compounds, either hydroxyamides derived from D-glucuronic acid or triazole-cinchone hybrids, and to evaluate their anti-proliferative properties.
Anti-proliferative activity of the newly synthesized compounds was examined against human breast adenocarcinoma (MCF-7) and human colon carcinoma (MDST8) cell-lines. Cell growth and viability was analysed by the Cell-Counting Kit-8 method. The 5-fluoroacil was used as a positive control. The compounds were studied between 10-9-10-5M. Fifteen compounds from the hydroxyamide family and two triazole compounds were investigated.
Most of the compounds from the hydroxyamide family revealed mild (~20%) to moderate (50%) anti-proliferative effects in both cell-lines, with the exception of Hydroxyamide B1 which did not affect MDST8 proliferation, and hydroxyamide B3 where proliferation of MDST8 was inhibited by 90%. Triazoles (A and B) evoked a strong (~100%) anti-proliferative effect of MDST8 cell-lines. Proliferation of MCF-7 was selectively and effectively (~98%) inhibited by triazole B while triazole A had a mild effect.
In conclusion, when compared to hydroxyamides, triazoles evoked a stronger anti-proliferative effect and might be promising anti-tumoral drugs
Bio-based solar energy harvesting for onsite mobile optical temperature sensing in Smart Cities
The Internet of Things (IoT) fosters the development of smart city systems for sustainable living and increases comfort for people. One of the current challenges for sustainable buildings is the optimization of energy management. Temperature monitoring in buildings is of prime importance, as heating account for a great part of the total energy consumption. Here, a solar optical temperature sensor is presented with a thermal sensitivity of up to 1.23% °C-1 based on sustainable aqueous solutions of enhanced green fluorescent protein and C-phycocyanin from biological feedstocks. These photonic sensors are presented under the configuration of luminescent solar concentrators widely proposed as a solution to integrate energy-generating devices in buildings, as windows or façades. The developed mobile sensor is inserted in IoT context through the development of a self-powered system able to measure, record, and send data to a user-friendly website.publishe
Photoperiodic Modulation of Circadian Clock and Reproductive Axis Gene Expression in the Pre-Pubertal European Sea Bass Brain
The acquisition of reproductive competence requires the activation of the brain-pituitary-gonad (BPG) axis, which in most vertebrates, including fishes, is initiated by changes in photoperiod. In the European sea bass long-term exposure to continuous light (LL) alters the rhythm of reproductive hormones, delays spermatogenesis and reduces the incidence of precocious males. In contrast, an early shift from long to short photoperiod (AP) accelerates spermatogenesis. However, how photoperiod affects key genes in the brain to trigger the onset of puberty is still largely unknown. Here, we investigated if the integration of the light stimulus by clock proteins is sufficient to activate key genes that trigger the BPG axis in the European sea bass. We found that the clock genes clock, npas2, bmal1 and the BPG genes gnrh, kiss and kissr share conserved transcription factor frameworks in their promoters, suggesting co-regulation. Other gene promoters of the BGP axis were also predicted to be co-regulated by the same frameworks. Co-regulation was confirmed through gene expression analysis of brains from males exposed to LL or AP photoperiod compared to natural conditions: LL fish had suppressed gnrh1, kiss2, galr1b and esr1, while AP fish had stimulated npas2, gnrh1, gnrh2, kiss2, kiss1rb and galr1b compared to NP. Recursos locais para aplicações multifuncionais de captação de energia com base em tetraedrite
CIES2020 - XVII Congresso Ibérico e XIII Congresso Ibero-americano de Energia SolarRESUMO: Este trabalho descreve as principais linhas de investigação que estão em curso no âmbito do projeto LocalEnergy (http://localenergy.lneg.pt). LocalEnergy é um projeto multidisciplinar de Investigação, Desenvolvimento e Inovação (I&D&I), envolvendo atividades no domínio da ciência dos materiais, da geologia e das energias renováveis. Essas atividades estão orientadas para a indústria e assentam numa abordagem disruptiva visando o desenvolvimento de materiais termoelétricos, através da utilização de materiais à base de tetraedrites naturais e sintéticas, e o desenvolvimento de novos absorsores para células solares de filmes finos, usando materiais à base de tetraedrites sintéticas. Considerando que Portugal é um dos países europeus com maior nível de irradiação de energia solar e que a tetraedrite é um recurso mineral local, presente na Zona Sul Portuguesa da Faixa Piritosa Ibérica, o projeto LocalEnergy representa uma oportunidade para o desenvolvimento de sistemas energéticos sustentáveis baseados na maximização e exploração de dois importantes recursos endógenos (energia solar e recursos minerais).ABSTRACT: This work describes the main lines of investigation that are underway within LocalEnergy project (http://localenergy.lneg.pt).
