Cardiorespiratory Fitness Modulates The Acute Flow-Mediated Dilation Response Following High-Intensity But Not Moderate-Intensity Exercise In Elderly Men.
Impaired endothelial function is observed with ageing and with low cardiorespiratory fitness (VO2peak) whilst improvements in both are suggested to be reliant on higher-intensity exercise in the elderly. This may be due to the flow-mediated dilation (FMD) response to acute exercise of varying intensity. We examined the hypothesis that exercise-intensity alters the FMD response in healthy elderly adults, and would be modulated by VO2peak Forty-seven elderly men were stratified into lower- (VO2peak = 24.3±2.9 ml.kg(-1)min(-1), n=27) and higher-fit groups (VO2peak = 35.4±5.5 ml.kg(-1)min(-1), n=20) after a test of cycling peak power output (PPO). In randomised order, participants undertook 27 min moderate-intensity continuous (MICE; 40% PPO) or high-intensity interval cycling exercise (HIIE; 70% PPO), or no-exercise control. Brachial FMD was assessed at rest, 10 and 60 min after exercise. In control, FMD reduced in both groups (P=0.05). FMD increased after MICE in both groups [increase of 0.86 % (95% CI, 0.17 to 1.56), P=0.01], and normalised after 60 min. In the lower-fit, FMD reduced after HIIE [reduction of 0.85 % (95% CI, 0.12 to 1.58), P=0.02), and remained decreased at 60 min (P=0.05). In the higher-fit FMD was unchanged immediately after HIIE and increased after 60 min [increase of 1.52 % (95% CI, 0.41 to 2.62), P<0.01], which was correlated with VO2peak (r =0.41; P<0.01). Exercise-intensity alters the FMD response in elderly adults, and VO2peak modulates the FMD response following HIIE, but not MICE. The sustained decrease in FMD in the lower-fit may represent a signal for vascular adaptation or endothelial fatigue
Role of β-adrenergic receptors in the oral activity of zinc-α2-glycoprotein (ZAG)
Zinc-a2-glycoprotein (ZAG) is an adipokine with the potential as a therapeutic agent in the treatment of obesity and type 2 diabetes. In this study we show that human ZAG, which is a 41-kDa protein, when administered to ob/ob mice at 50 µg/d-1 orally in the drinking water produced a progressive loss of body weight (5 g after 8 d treatment), together with a 0.5 C increase in rectal temperature and a 40% reduction in urinary excretion of glucose. There was also a 33% reduction in the area under the curve during an oral glucose tolerance test and an increased sensitivity to insulin. These results were similar to those after iv administration of ZAG. However, tryptic digestion was shown to inactivate ZAG. There was no evidence of human ZAG in the serum but a 2-fold elevation of murine ZAG, which was also observed in target tissues such as white adipose tissue. To determine whether the effect was due to interaction of the human ZAG with the ß-adrenergic (ß-AR) in the gastrointestinal tract before digestion, ZAG was coadministered to ob/ob mice together with propanolol (40 mg/kg-1), a nonspecific ß-AR antagonist. The effect of ZAG on body weight, rectal temperature, urinary glucose excretion, improvement in glucose disposal, and increased insulin sensitivity were attenuated by propanolol, as was the increase in murine ZAG in the serum. These results suggest that oral administration of ZAG increases serum levels through interaction with a ß-AR in the upper gastrointestinal tract, and gene expression studies showed this to be in the esophagus
Duffer’s Shoal: A Strategic Dream of the Pacific Command Area of Responsibility
This strategic assessment seeks to go beyond a traditional comparative analysis of the military, technological, political, cultural, and economic factors governing the relationships and capabilities of the Asia Pacific environment. To make sense of the intrinsic complexities unique to this region, we endeavor to broaden our view and rely on a tool often overlooked in government studies: imagination. Moreover, we aim to offer a strategic document that is readable, instructive, and provocative. Pulling from a well-referenced piece of military teaching, this assessment borrows a learning concept first employed in 1904 by Major General Sir Ernest Dunlop Swinton in The Defence of Duffer’s Drift. This fictional story describes the plight of young Lieutenant Backsight Forethought as he commands a 50-man platoon tasked to hold a tactically critical piece of land called Duffer’s Drift. The story unfolds in a series of six dreams, where the blunders of the unwitting lieutenant lead to disaster. Maleidride biosynthesis – construction of dimeric anhydrides – more than just heads or tails
Covering: up to early 2022 Maleidrides are a family of polyketide-based dimeric natural products isolated from fungi. Many maleidrides possess significant bioactivities, making them attractive pharmaceutical or agrochemical lead compounds. Their unusual biosynthetic pathways have fascinated scientists for decades, with recent advances in our bioinformatic and enzymatic understanding providing further insights into their construction. However, many intriguing questions remain, including exactly how the enzymatic dimerisation, which creates the diverse core structure of the maleidrides, is controlled. This review will explore the literature from the initial isolation of maleidride compounds in the 1930s, through the first full structural elucidation in the 1960s, to the most recent in vivo, in vitro, and in silico analyses
Heterologous Production of Fungal Maleidrides Reveals the Cryptic Cyclization Involved in their Biosynthesis
Fungal maleidrides are an important family of bioactive secondary metabolites that consist of 7, 8, or 9-membered carbocycles with one or two fused maleic anhydride moieties. The biosynthesis of byssochlamic acid (a nonadride) and agnestadride A (a heptadride) was investigated through gene disruption and heterologous expression experiments. The results reveal that the precursors for cyclization are formed by an iterative highly reducing fungal polyketide synthase supported by a hydrolase, together with two citrate-processing enzymes. The enigmatic ring formation is catalyzed by two proteins with homology to ketosteroid isomerases, and assisted by two proteins with homology to phosphatidylethanolamine-binding proteins. Ring cycle: The enzymes involved in the cyclization of the maleidride family of bioactive fungal natural products, including agnestadride A and byssochlamic acid, were identified. Effects of Walnut Consumption on Cognitive Performance in Young Adults
Walnuts contain a number of potentially neuroprotective compounds like vitamin E, folate, melatonin, several antioxidative polyphenols and significant amounts of n-3 α-linolenic fatty acid. The present study sought to determine the effect of walnuts on verbal and non-verbal reasoning, memory and mood. A total of sixty-four college students were randomly assigned to two treatment sequences in a crossover fashion: walnuts-placebo or placebo-walnuts. Baseline data were collected for non-verbal reasoning, verbal reasoning, memory and mood states. Data were collected again after 8 weeks of intervention. After 6 weeks of washout, the intervention groups followed the diets in reverse order. Data were collected once more at the end of the 8-week intervention period. No significant increases were detected for mood, non-verbal reasoning or memory on the walnut-supplemented diet. However, inferential verbal reasoning increased significantly by 11.2 %, indicating a medium effect size (P = 0.009; d = 0.567). In young, healthy, normal adults, walnuts do not appear to improve memory, mood or non-verbal reasoning abilities. However, walnuts may have the ability to increase inferential reasoning
A Description of the "Crow's Foot" Tunnel Concept
NASA Langley Research Center has actively pursued the development and the use of pictorial or three-dimensional perspective displays of tunnel-, pathway- or highway-in-the-sky concepts for presenting flight path information to pilots in all aircraft categories (e.g., transports, General Aviation, rotorcraft) since the late 1970s. Prominent among these efforts has been the development of the crow s foot tunnel concept. The crow's foot tunnel concept emerged as the consensus pathway concept from a series of interactive workshops that brought together government and industry display designers, test pilots, and airline pilots to iteratively design, debate, and fly various pathway concepts. Over years of use in many simulation and flight test activities at NASA and elsewhere, modifications have refined and adapted the tunnel concept for different applications and aircraft categories (i.e., conventional transports, High Speed Civil Transport, General Aviation). Aspects of Synthetic Vision Display Systems and the Best Practices of the NASA's SVS Project
NASA s Synthetic Vision Systems (SVS) Project conducted research aimed at eliminating visibility-induced errors and low visibility conditions as causal factors in civil aircraft accidents while enabling the operational benefits of clear day flight operations regardless of actual outside visibility. SVS takes advantage of many enabling technologies to achieve this capability including, for example, the Global Positioning System (GPS), data links, radar, imaging sensors, geospatial databases, advanced display media and three dimensional video graphics processors. Integration of these technologies to achieve the SVS concept provides pilots with high-integrity information that improves situational awareness with respect to terrain, obstacles, traffic, and flight path. This paper attempts to emphasize the system aspects of SVS - true systems, rather than just terrain on a flight display - and to document from an historical viewpoint many of the best practices that evolved during the SVS Project from the perspective of some of the NASA researchers most heavily involved in its execution. The Integrated SVS Concepts are envisagements of what production-grade Synthetic Vision systems might, or perhaps should, be in order to provide the desired functional capabilities that eliminate low visibility as a causal factor to accidents and enable clear-day operational benefits regardless of visibility conditions
A Study of the Diverse T Dwarf Population Revealed by WISE
We report the discovery of 87 new T dwarfs uncovered with the Wide-field
Infrared Survey Explorer (WISE) and three brown dwarfs with extremely red
near-infrared colors that exhibit characteristics of both L and T dwarfs. Two
of the new T dwarfs are likely binaries with L7+/-1 primaries and mid-type T
secondaries. In addition, our follow-up program has confirmed 10 previously
identified T dwarfs and four photometrically-selected L and T dwarf candidates
in the literature. This sample, along with the previous WISE discoveries,
triples the number of known brown dwarfs with spectral types later than T5.
Using the WISE All-Sky Source Catalog we present updated color-color and
color-type diagrams for all the WISE-discovered T and Y dwarfs. Near-infrared
spectra of the new discoveries are presented, along with spectral
classifications. To accommodate later T dwarfs we have modified the integrated
flux method of determining spectral indices to instead use the median flux.
Furthermore, a newly defined J-narrow index differentiates the early-type Y
dwarfs from late-type T dwarfs based on the J-band continuum slope. The K/J
indices for this expanded sample show that 32% of late-type T dwarfs have
suppressed K-band flux and are blue relative to the spectral standards, while
only 11% are redder than the standards. Comparison of the Y/J and K/J index to
models suggests diverse atmospheric conditions and supports the possible
re-emergence of clouds after the L/T transition. We also discuss peculiar brown
dwarfs and candidates that were found not to be substellar, including two Young
Stellar Objects and two Active Galactic Nuclei. The coolest WISE-discovered
brown dwarfs are the closest of their type and will remain the only sample of
