    Integrating Narcissus-derived galanthamine production into traditional upland farming systems

    Alzheimer’s disease (AD) is a disorder associated with progressive degeneration of memory and cognitive function. Galantamine is a licenced treatment for AD but supplies of the plant alkaloid that it is produced from, galanthamine, are limited. This three-year system study tested the potential to combine Narcissus-derived galanthamine production with grassland-based ruminant production. Replicate plots of permanent pasture were prepared with and without bulbs of Narcissus pseudonarcissus sown as lines into the sward. Two different fertiliser regimes were imposed. The above-ground green biomass of N. pseudonarcissus was harvested in early spring and the galanthamine yield determined. In the second harvest year a split-plot design was implemented with lines of N. pseudonarcissus cut annually and biennially. All plots were subsequently grazed by ewes and lambs and animal performance recorded. Incorporation of N. pseudonarcissus into grazed permanent pasture had no detrimental effects on the health or performance of the sheep which subsequently grazed the pasture. There was no consistency to the effects of fertiliser rates on galanthamine yields. There was no difference in overall galanthamine yield if N. pseudonarcissus was cut biennially (1.64 vs. 1.75 kg galanthamine/ha for annual combined vs biennial cuts respectively; s.e.d = 0.117 kg galanthamine/ha; ns).     Organic Centre Wales (OCW) was originally formed to act as an information source for producers enquiring into organic farming; in our second phase, in response to changes in the sector, we have an increased remit on policy issues. It has been a busy time, seeing the production of a second Organic Action Plan, the implementation of the CAP reform and development work on agri-environment reforms. The year also saw the start of organic maintenance payments for farmers. This Annual Report contains accounts of the progress that has been made on many fronts during the last 12 months, particularly in market knowledge due to the research for the recently published report, 'Organic Food: understanding the consumer and increasing sales'. This information, together with continuing product development work, will enable Welsh organic producers to be proactive in building a solid consumer base

    Identification of common genetic variation that modulates alternative splicing

    Alternative splicing of genes is an efficient means of generating variation in protein function. Several disease states have been associated with rare genetic variants that affect splicing patterns. Conversely, splicing efficiency of some genes is known to vary between individuals without apparent ill effects. What is not clear is whether commonly observed phenotypic variation in splicing patterns, and hence potential variation in protein function, is to a significant extent determined by naturally occurring DNA sequence variation and in particular by single nucleotide polymorphisms (SNPs). In this study, we surveyed the splicing patterns of 250 exons in 22 individuals who had been previously genotyped by the International HapMap Project. We identified 70 simple cassette exon alternative splicing events in our experimental system; for six of these, we detected consistent differences in splicing pattern between individuals, with a highly significant association between splice phenotype and neighbouring SNPs. Remarkably, for five out of six of these events, the strongest correlation was found with the SNP closest to the intron-exon boundary, although the distance between these SNPs and the intron-exon boundary ranged from 2 bp to greater than 1,000 bp. Two of these SNPs were further investigated using a minigene splicing system, and in each case the SNPs were found to exert cis-acting effects on exon splicing efficiency in vitro. The functional consequences of these SNPs could not be predicted using bioinformatic algorithms. Our findings suggest that phenotypic variation in splicing patterns is determined by the presence of SNPs within flanking introns or exons. Effects on splicing may represent an important mechanism by which SNPs influence gene function

    Synthesis of a boronic acid anhydride based ligand and its application in beryllium coordination

    The synthesis of a boronic acid anhydride‐based ligand containing one three‐ and one four‐coordinated boron atom is presented. This ligand was successfully employed as a tridentate κ¹N,κ²O‐ligand in the coordination of beryllium chloride and both the ligand and the resulting complex have been structurally characterized. While the boron‐element separations are within the typical range of related homo‐nuclear compounds, the corresponding beryllium‐element distances are rather long, suggesting unexpectedly high electron density at the beryllium center. Incorporation of a beryllium atom in a six‐membered ring causes no more distortion than the corresponding boron atom, suggesting that analogous ligand systems could be used in boron and beryllium coordination chemistry. The generated hetero‐tri‐nuclear complex enables the direct comparison of bond lengths and angles at beryllium and boron atoms in similar coordination environments and can act as a monomolecular model for beryllium borates

    Seabed morphology and bed shear stress predict temperate reef habitats in a high energy marine region

    High energy marine regions host ecologically important habitats like temperate reefs, but are less anthropogenically developed and understudied compared to lower energy waters. In the marine environment direct habitat observation is limited to small spatial scales, and high energy waters present additional logistical challenges and constraints. Semi-automated predictive habitat mapping is a cost-effective tool to map benthic habitats across large extents, but performance is context specific. High resolution environmental data used for predictive mapping are often limited to bathymetry, acoustic backscatter and their derivatives. However, hydrodynamic energy at the seabed is a critical habitat structuring factor and likely an important, yet rarely incorporated, predictor of habitat composition and spatial patterning. Here, we used a machine learning classification approach to map temperate reef substrate and biogenic reef habitat in a tidal energy development area, incorporating bathymetric derivatives at multiple scales and simulated tidally induced seabed shear stress. We mapped reef substrate (four classes: sediment (not reef), stony reef (low resemblance), stony reef (medium – high resemblance) and bedrock reef) with overall balanced accuracy of 71.7%. Our model to predict potential biogenic Sabellaria spinulosa reef performed less well with an overall balanced accuracy of 63.4%. Despite low performance metrics for the target class of potential reef in this model, it still provided insight into the importance of different environmental variables for mapping S. spinulosa biogenic reef habitat. Tidally induced mean bed shear stress was one of the most important predictor variables for both reef substrate and biogenic reef models, with ruggedness calculated at multiple scales from 3 m to 140 m also important for the reef substrate model. We identified previously unresolved relationships between temperate reef spatial distribution, hydrodynamic energy and seabed three-dimensional structure in energetic waters. Our findings contribute to a better understanding of the spatial ecology of high energy marine ecosystems and will inform evidence-based decision making for sustainable development, particularly within the growing tidal energy sector

    A calibration protocol for population-specific accelerometer cut-points in children

    PurposeTo test a field-based protocol using intermittent activities representative of children\u27s physical activity behaviours, to generate behaviourally valid, population-specific accelerometer cut-points for sedentary behaviour, moderate, and vigorous physical activity.MethodsTwenty-eight children (46% boys) aged 10&ndash;11 years wore a hip-mounted uniaxial GT1M ActiGraph and engaged in 6 activities representative of children\u27s play. A validated direct observation protocol was used as the criterion measure of physical activity. Receiver Operating Characteristics (ROC) curve analyses were conducted with four semi-structured activities to determine the accelerometer cut-points. To examine classification differences, cut-points were cross-validated with free-play and DVD viewing activities.ResultsCut-points of &le;372, &gt;2160 and &gt;4806 counts&bull;min&minus;1 representing sedentary, moderate and vigorous intensity thresholds, respectively, provided the optimal balance between the related needs for sensitivity (accurately detecting activity) and specificity (limiting misclassification of the activity). Cross-validation data demonstrated that these values yielded the best overall kappa scores (0.97; 0.71; 0.62), and a high classification agreement (98.6%; 89.0%; 87.2%), respectively. Specificity values of 96&ndash;97% showed that the developed cut-points accurately detected physical activity, and sensitivity values (89&ndash;99%) indicated that minutes of activity were seldom incorrectly classified as inactivity.ConclusionThe development of an inexpensive and replicable field-based protocol to generate behaviourally valid and population-specific accelerometer cut-points may improve the classification of physical activity levels in children, which could enhance subsequent intervention and observational studies.<br /

    A cross-curricular physical activity intervention to combat cardiovascular disease risk factors in 11-14 year olds: 'Activity Knowledge Circuit'

    Background: Cardiovascular disease is the leading cause of mortality worldwide. Risk factors associated with cardiovascular disease have been shown to track from childhood through to adulthood. Previous school-based physical activity interventions have demonstrated modest improvements to cardiovascular disease risk factors by implementing extra-curricular activities or improving current physical education curriculum. Few have attempted to increase physical activity in class-room taught curriculum subjects. This study will outline a school-based cross-curricular physical activity intervention to combat cardiovascular disease risk factors in 11-14 year old children. Method/Design: A South Wales Valley school of low socio-economic status has been selected to take part. Participants from year eight (12-13 years) are to be assigned to an intervention group, with maturation-matched participants from years seven (11-12 years) and nine (13-14 years) assigned to a control group. A cross-curricular physical activity intervention will be implemented to increase activity by two hours a week for 18 weeks. Participants will briskly walk 3200 m twice weekly during curriculum lessons (60 minutes duration). With the exception of physical education, all curriculum subjects will participate, with each subject delivering four intervention lessons. The intervention will be performed outdoors and on school premises. An indoor course of equal distance will be used during adverse weather conditions. Cardiovascular disease risk factors will be measured pre- and post-intervention for intervention and control groups. These will take place during physical education lessons and will include measures of stature, mass, waist, hip, and neck circumferences, together with skinfold measure's taken at four sites. Blood pressure will be measured, and fitness status assessed via the 20 m multi-stage fitness test. Questionnaires will be used to determine activity behaviour (physical activity questionnaire for adolescence), diet (seven day food diary) and maturation status. Fasting blood variables will include total cholesterol, lowdensity lipoprotein cholesterol, high density lipoprotein cholesterol, triglycerides, insulin, glucose, high-sensitivity C-reactive protein, interleukin-6, adiponectin, and fibrinogen. Motivational variables and psychological well-being will be assessed by questionnaire. Discussion: Our study may prove to be a cost effective strategy to increase school time physical activity to combat cardiovascular disease risk factors in children.</p

    Endovascular strategy or open repair for ruptured abdominal aortic aneurysm: one-year outcomes from the IMPROVE randomized trial.

    AIMS: To report the longer term outcomes following either a strategy of endovascular repair first or open repair of ruptured abdominal aortic aneurysm, which are necessary for both patient and clinical decision-making. METHODS AND RESULTS: This pragmatic multicentre (29 UK and 1 Canada) trial randomized 613 patients with a clinical diagnosis of ruptured aneurysm; 316 to an endovascular first strategy (if aortic morphology is suitable, open repair if not) and 297 to open repair. The principal 1-year outcome was mortality; secondary outcomes were re-interventions, hospital discharge, health-related quality-of-life (QoL) (EQ-5D), costs, Quality-Adjusted-Life-Years (QALYs), and cost-effectiveness [incremental net benefit (INB)]. At 1 year, all-cause mortality was 41.1% for the endovascular strategy group and 45.1% for the open repair group, odds ratio 0.85 [95% confidence interval (CI) 0.62, 1.17], P = 0.325, with similar re-intervention rates in each group. The endovascular strategy group and open repair groups had average total hospital stays of 17 and 26 days, respectively, P < 0.001. Patients surviving rupture had higher average EQ-5D utility scores in the endovascular strategy vs. open repair groups, mean differences 0.087 (95% CI 0.017, 0.158), 0.068 (95% CI -0.004, 0.140) at 3 and 12 months, respectively. There were indications that QALYs were higher and costs lower for the endovascular first strategy, combining to give an INB of £3877 (95% CI £253, £7408) or €4356 (95% CI €284, €8323). CONCLUSION: An endovascular first strategy for management of ruptured aneurysms does not offer a survival benefit over 1 year but offers patients faster discharge with better QoL and is cost-effective.     In the year marking the centenary since the foundation of the Azerbaijani Diplomatic Service, Baku’s foreign policy is increasingly characterised by a broader understanding of diplomacy, shaped by the gradual yet steady expansion of both areas and the tools for intervention. Guided by the attempt to develop a ‘niche strategy’ aiming at safeguarding and promoting Azerbaijani national interest, the Humanitarian Diplomacy emerges as a privileged field for Baku to adopt a pro-active and creative foreign policy. Building upon the debate around the interests behind the aid-providing activities of traditional and emerging donors, the article aims at introducing the motivations and the aims behind Azerbaijani aid policy. In particular, it aims at demonstrating that Baku’s Humanitarian Diplomacy aims chiefly at achieving immaterial benefits, having to do with international prestige and with the construction and international projection of a Good International Citizenship
