Nanoscale Structure of Zoned Laurites from the Ojén Ultramafic Massif, Southern Spain
We report the first results of a combined focused ion beam and high-resolution transmission
electron microscopy (FIB/HRTEM) investigation of zoned laurite (RuS2)-erlichmanite (OS2) in
mantle-hosted chromitites. These platinum-group minerals form isolated inclusions (<50 um across)
within larger crystals of unaltered chromite form the Ojén ultramafic massif (southern Spain).
High-magnification electron microscopy (HMEM), high angle-annular dark field (HAADF) and
precession electron diffraction (PED) data revealed that microscale normal zoning in laurite consisting
of Os-poor core and Os-rich rims observed by conventional micro-analytical techniques like field
emission scanning electron microscope and electron microprobe analysis (FE-SEM and EPMA) exist
at the nanoscale approach in single laurite crystals. At the nanoscale, Os poor cores consist of
relatively homogenous pure laurite (RuS2) lacking defects in the crystal lattice, whereas the Os-richer
rim consists of homogenous laurite matrix hosting fringes (10–20 nm thickness) of almost pure
erlichmanite (OsS2). Core-to-rim microscale zoning in laurite reflects a nonequilibrium during
laurite crystal growth, which hampered the intra-crystalline diffusion of Os. The origin of zoning
in laurite is related to the formation of the chromitites in the Earth’s upper mantle but fast cooling
of the chromite-laurite magmatic system associated to fast exhumation of the rocks would prevent
the effective dissolution of Os in the laurite even at high temperatures (~1200 ºC), allowing the
formation/preservation of nanoscale domains of erlichmanite in laurite. Our observation highlights
for the first time the importance of nanoscale studies for a better understanding of the genesis of

Clinical Management of COVID-19 in Cancer Patients with the STAT3 Inhibitor Silibinin
COVID-19 pathophysiology is caused by a cascade of respiratory and multiorgan failures arising, at least in part, from the SARS-CoV-2-driven dysregulation of the master transcriptional factor STAT3. Pharmacological correction of STAT3 over-stimulation, which is at the root of acute respiratory distress syndrome (ARDS) and coagulopathy/thrombosis events, should be considered for treatment of severe COVID-19. In this perspective, we first review the current body of knowledge on the role of STAT3 in the pathogenesis of severe COVID-19. We then exemplify the potential clinical value of treating COVID-19 disease with STAT3 inhibitors by presenting the outcomes of two hospitalized patients with active cancer and COVID-19 receiving oral Legalon(R)-a nutraceutical containing the naturally occurring STAT3 inhibitor silibinin. Both patients, which were recruited to the clinical trial SIL-COVID19 (EudraCT number: 2020-001794-77) had SARS-CoV-2 bilateral interstitial pneumonia and a high COVID-GRAM score, and showed systemic proinflammatory responses in terms of lymphocytopenia and hypoalbuminemia. Both patients were predicted to be at high risk of critical COVID-19 illness in terms of intensive care unit admission, invasive ventilation, or death. In addition to physician's choice of best available therapy or supportive care, patients received 1050 mg/day Legalon(R) for 10 days without side-effects. Silibinin-treated cancer/COVID-19+ patients required only minimal oxygen support (2-4 L/min) during the episode, exhibited a sharp decline of the STAT3-regulated C-reactive protein, and demonstrated complete resolution of the pulmonary lesions. These findings might inspire future research to advance our knowledge and improve silibinin-based clinical interventions aimed to target STAT3-driven COVID-19 pathophysiology
K+ channels expression in hypertension after arterial injury, and effect of selective Kv1.3 blockade with PAP-1 on intimal hyperplasia formation
Producción CientíficaK+ channels are central to vascular pathophysiology. Previous results demonstrated that phenotypic modulation associates with a change in Kv1.3 to Kv1.5 expression, and that Kv1.3 blockade inhibits proliferation of VSMCs cultures.
Purpose: To explore whether the Kv1.3 to Kv1.5 switch could be a marker of the increased risk of intimal hyperplasia in essential hypertension and whether systemic treatment with Kv1.3 blockers can prevent intimal hyperplasia after endoluminal lesion . Methods: Morphometric and immunohistochemical analysis were performed in arterial segments following arterial injury and constant infusion of the Kv1.3 blocker PAP-1 during 28 days. Differential expression of K+ channel genes was studied in VSMC from hypertensive (BPH) and normotensive (BPN) mice, both in control and after endoluminal lesion. Finally, the migration and proliferation rate of BPN and BPH VSMCs was explored in vitro.
Results: Changes in mRNA expression led to an increased Kv1.3/Kv1.5 ratio in BPH VSMC. Consistent with this, arterial injury in BPH mice induced a higher degree of luminal stenosis, (84±4 % vs. 70±5 % in BPN, p<0.01), although no differences in migration and proliferation rate were observed in cultured VSMCs. The in vivo proliferative lesions were significantly decreased upon PAP-1 systemic infusion (18± 6 % vs. 58±20 % with vehicle, p<0.05).
Conclusions: Hypertension leads to a higher degree of luminal stenosis in our arterial injury model, that correlates with a decreased expression of Kv1.5 channels. Kv1.3 blockers decreased in vitro VSMCs proliferation, migration, and in vivo intimal hyperplasia formation, pointing to Kv1.3 channels as promising therapeutical targets against restenosis.La versión original del artículo contiene un error. Cost-effectiveness of a programme to address sedentary behaviour in older adults: results from the SITLESS RCT
Background:
This study details the within-trial economic evaluation and long-term economic model of SITLESS, a multi-country, three-armed randomized controlled trial comparing a combined intervention of exercise referral schemes (ERS) enhanced by self-management strategies (SMS) against ERS alone and usual care (UC).
Methods:
A cost-utility analysis, conducted from the base-case perspective of the National Health Service and personal and social services, estimated the incremental cost per incremental quality-adjusted life year (QALY) and years in full capability (YFC). A secondary analysis combined the costs with a broad set of outcomes within a cost-consequence framework, from a societal perspective. A Markov-type decision-analytic model was developed to project short-term changes in physical activity to long-term outcomes and costs, over a 5- and 15-year time horizon.
Results:
The results of the within-trial analysis show that SMS+ERS is highly likely to be cost-effective compared to ERS alone (ICER €4270/QALY), but not compared to UC. Participants allocated to the SMS+ERS group also showed an improvement in YFC compared to ERS alone and UC. The long-term analysis revealed that SMS+ERS is likely to be a cost-effective option compared to ERS and UC over a 5-year, but not with a 15-year horizon, being then dominated by ERS alone.
Conclusion:
This research provides new evidence that SMS is a cost-effective add-on to ERS strategies. This economic evaluation informs the case for further, cost-effective, refinement of lifestyle change programmes targeted to older adults, with the aim of ultimately reducing the impact of non-communicable diseases in this population
Respiratory syncytial virus: diagnosis, prevention and management.
Respiratory syncytial virus (RSV) is responsible for a large burden of disease globally and can present as a variety of clinical syndromes in children of all ages. Bronchiolitis in infants under 1 year of age is the most common clinical presentation hospitalizing 24.2 per 1000 infants each year in the United Kingdom. RSV has been shown to account for 22% of all episodes of acute lower respiratory tract infection in children globally. RSV hospitalization, that is, RSV severe disease, has also been associated with subsequent chronic respiratory morbidity. Routine viral testing in all children is not currently recommended by the United Kingdom National Institute for Health and Care Excellence (NICE) or the American Academy of Pediatrics (AAP) guidance and management is largely supportive. There is some evidence for the use of ribavirin in severely immunocompromised children. Emphasis is placed on prevention of RSV infection through infection control measures both in hospital and in the community, and the use of the RSV-specific monoclonal antibody, palivizumab, for certain high-risk groups of infants. New RSV antivirals and vaccines are currently in development. Ongoing work is needed to improve the prevention of RSV infection, not only because of the acute morbidity and mortality, but also to reduce the associated chronic respiratory morbidity after severe infection
Cost-effectiveness of exercise referral schemes enhanced by self-management strategies to battle sedentary behaviour in older adults: Protocol for an economic evaluation alongside the SITLESS three-armed pragmatic randomised controlled trial
Introduction: Promoting physical activity (PA) and reducing sedentary behaviour (SB) may exert beneficial effects on the older adult population, improving behavioural, functional, health and psychosocial outcomes in addition to reducing health, social care and personal costs. This paper describes the planned economic evaluation of SITLESS, a multicountry three-armed pragmatic randomised controlled trial (RCT) which aims to assess the short-term and long-term effectiveness and cost-effectiveness of a complex intervention on SB and PA in community-dwelling older adults, based on exercise referral schemes enhanced by a group intervention providing self-management strategies to encourage lifestyle change.
Methods and analysis: A within-trial economic evaluation and long-term model from both a National Health Service/personal social services perspective and a broader societal perspective will be undertaken alongside the SITLESS multinational RCT. Healthcare costs (hospitalisations, accident and emergency visits, appointment with health professionals) and social care costs (eg, community care) will be included in the economic evaluation. For the cost-utility analysis, quality-adjusted life-years will be measured using the EQ-5D-5L and capability well-being measured using the ICEpop CAPability measure for Older people (ICECAP-O) questionnaire. Other effectiveness outcomes (health related, behavioural, functional) will be incorporated into a cost-effectiveness analysis and cost-consequence analysis.
The multinational nature of this RCT implies a hierarchical structure of the data and unobserved heterogeneity between clusters that needs to be adequately modelled with appropriate statistical and econometric techniques. In addition, a long-term population health economic model will be developed and will synthesise and extrapolate within-trial data with additional data extracted from the literature linking PA and SB outcomes with longer term health states.
Methods guidance for population health economic evaluation will be adopted including the use of a long-time horizon, 1.5% discount rate for costs and benefits, cost consequence analysis framework and a multisector perspective.

Image informatics strategies for deciphering neuronal network connectivity
Brain function relies on an intricate network of highly dynamic neuronal connections that rewires dramatically under the impulse of various external cues and pathological conditions. Among the neuronal structures that show morphologi- cal plasticity are neurites, synapses, dendritic spines and even nuclei. This structural remodelling is directly connected with functional changes such as intercellular com- munication and the associated calcium-bursting behaviour. In vitro cultured neu- ronal networks are valuable models for studying these morpho-functional changes. Owing to the automation and standardisation of both image acquisition and image analysis, it has become possible to extract statistically relevant readout from such networks. Here, we focus on the current state-of-the-art in image informatics that enables quantitative microscopic interrogation of neuronal networks. We describe the major correlates of neuronal connectivity and present workflows for analysing them. Finally, we provide an outlook on the challenges that remain to be addressed, and discuss how imaging algorithms can be extended beyond in vitro imaging studies
Identificación de flujos entre acuíferos a nivel regional en base a datos hidroquímicos (Alt Empordà, NE España)
A escala regional, los flujos de agua subterránea entre formaciones hidrogeológicas condicionan su ba lance hídrico y, con él, su régimen de explotación. En este estudio se caracteriza la dinámica hidrogeológica a nivel regional de la depresión del Alt Empordà (Girona, NE España) en base a datos hidroquímicos, con el objetivo de identificar flujos de agua subterránea entre las formaciones que constituyen la zona de recarga (relieves circundantes) y la depresión. En este sistema, el ión sulfato resulta un trazador adecuado para identificar los procesos hidroquímicos y de mezcla que acontecen en este zona. Los datos presentados sugieren que la presencia de sulfato, y por consiguiente la recarga, se debe a aportaciones de flujos profundos, en algunos casos desde las unidades geológicas inferiores, ajenos a la recarga local por precipitación desde la superficie.At a regional scale, groundwater flow between aquifer systems controls their water budget and, therefore, its exploitation regime. This study characterizes the hydrogeology at a regional level of the Alt Empordà Basin (Girona, NE Spain). Our goal consists in recognizing recharge flowpaths, based on hydrochemical data, between the aquifer systems located in the ranges (recharge) areas and these within the basin. Sulfate stands as an appropriate tracer to identify hydrochemical as well as mixing processes and, therefore, recharge relationships. Field data suggest that sulfate occurrence is related to deep groundwater fluxes unrelated to the local rainfall recharge.Universidad Nacional de La Plat
Inorganic hydrophobic coatings: Surfaces mimicking the nature
Added value products are being developed in ceramic industry. Different optical effects as bright metallic shine or new functionalities as hydrophobicity or bactericide characteristics are the new properties searched on the tiles. In this study, we prepare glassy coatings for tiles based on copper pigment by a conventional industrial process. The obtained coatings present different aesthetical aspects, including bright metallic aspect which confers a high decorative value to the tile. Furthermore, these metallic coatings present hydrophobic properties with contact angles with water as high as 115 degrees and also bactericide characteristics. Superficial microstructure and nanoparticles were found in the bactericide-hydrophobic samples, resembling the surface of hydrophobic leaf surfaces. This structure was formed by the crystallization of CuO nanoparticles as Tenorite due to the copper saturation of the glassy matrix at the surface of the coatings
Polymorphism of the FABP2 gene: a population frequency analysis and an association study with cardiovascular risk markers in Argentina
<p>Abstract</p> <p>Background</p> <p>The FABP2 gene encodes for the intestinal FABP (IFABP) protein, which is expressed only in intestinal enterocytes. A polymorphism at codon 54 in exon 2 of the FABP2 gene exchanges an Alanine (Ala), in the small helical region of the protein, for Threonine (Thr). Given the potential physiological role of the Ala54Thr FABP2 polymorphism, we assess in this study the local population frequency and analyze possible associations with five selected markers, i.e. glycemia, total cholesterol, body mass index (BMI), hypertension, and high Cardiovascular Risk Index (CVR index).</p> <p>Methods</p> <p>We studied 86 men and 116 women. DNA was extracted from a blood drop for genotype analysis. Allele frequencies were calculated by direct counting. Hardy Weinberg Equilibrium was evaluated using a Chi-square goodness of fit test.</p> <p>For the polymorphism association analysis, five markers were selected, i.e. blood pressure, Framingham Risk Index, total cholesterol, BMI, and glycemia.</p> <p>For each marker, the Odds Ratio (OR) was calculated by an online statistic tool.</p> <p>Results</p> <p>Our results reveal a similar population polymorphism frequency as in previous European studies, with <b>q = 0.277 </b>(95% confidence limits 0.234–0.323). No significant association was found with any of the tested markers in the context of our Argentine nutritional and cultural habits. We did, however, observe a tendency for increased Cholesterol and high BMI in Thr54 carriers.</p> <p>Conclusion</p> <p>This is the first study to look at the population frequency of the Thr54 allele in Argentina. The obtained result does not differ from previously reported frequencies in European populations. Moreover, we found no association between the Thr54 allele and any of the five selected markers. The observed tendency to increased total cholesterol and elevated BMI in Thr54 carriers, even though not significant for p < 0.1 could be worth of further investigation to establish whether the Thr54 variant should be taken into consideration in cardiovascular prevention strategies.</p
