    Age-related influences on somatic and physical fitness of elite police agents (Influencias de la edad en la aptitud física y somática de los agentes de policía de élite)

    Background: Elite police officers must be physically fit to perform their job occupational demands but information on the effect of age in their physical fitness and somatic composition is scarce. Our aim is to describe the effect of age on somatic and physical fitness of a Special Police Unit (SPU); and to understand age-related changes. Methods: 117 SPU elements of a total of 218, aged 42.6±4.2 years, were assessed on their somatic (height, weight, circumferences, skinfolds); body composition (BMI, body fat); and physical fitness (maximal, power and endurance strength, aerobic power, and flexibility). T-tests were used for comparing results to other police studies. Regression analysis was used to detect the effect of age for somatic and fitness variable. Results: SPU elements showed a homogeneous and suitable fitness condition. No somatic differences were found along age, but annual age losses were found for physical fitness, namely for strength: left handgrip strength (95%CI -0.70 to -0.12), bench press (95%CI -2.34 to -0.89), squat jump (95%CI -0.70 to -0.12), medicinal ball throw (95%CI -0.62 to -0.25), push-ups (95%CI -1.64 to -0.66), pull-ups (95%CI -0.53 to -0.11), sit-ups (95%CI -1.33 to -0.27), but also on the VO2max (-0.535 to -0.115). Conclusions: Portuguese SPU elements showed a good somatic and physical fitness condition according to the requirements of their profession. Regardless the effect of age they were able to maintain a good somatic fitness and a very good aerobic power along the years.     Effect of nacl replacement by other salts on the quality of bísaro pork sausages (Pgi chouriça de vinhais)

    Concerned about the trend to reduce salt consumption, the meat industry has been increasing the strategies to produce and commercialize products where the reduction or even the replacement of NaCl is an important goal. The aim of this study was to test the effect of partial NaCl replacement by KCl and Sub4Salt® on the quality of pork sausages. Three different formulations (NaCl + KCl, NaCl + Sub4Salt®, and KCl + Sub4Salt®) were considered and compared to the control (2% NaCl). Physicochemical properties, chemical composition, and microbiological and sensory characteristics were evaluated. The replacement of NaCl did not affect pH, water activity (aw) or its chemical composition after eight or 16 days ripening time, while a significant sodium reduction was achieved. The oxidation index expressed in TBARS was also not affected by the NaCl substitution and varied between 0.01 to 0.04 of malonaldehyde (MDA) per kg of sample. Similarly, the NaCl replacement did not change the microbiological quality of the sausages, and the production of healthier meat sausages had also no significant effect on their sensory characteristics.     Physicochemical properties, fatty acid profile and sensory characteristics of sheep and goat meat sausages manufactured with different pork fat levels

    The effect of three pork backfat levels (0% vs. 10% vs. 30%) on chemical composition, fatty acid profile and sensory properties on sheep and goat meat sausages was studied. All physicochemical parameters were affected by the addition of pork backfat in both types of sausages. Sausages manufactured with 30% of pork backfat showed the lowest moisture and protein contents and the highest total fat content. The lower a(w) values in sausages manufactured with higher fat content while in pH happened the reverse situation. The addition of pork backfat modified the total fatty acid profile, prompting a significant drop in the relative percentages of C14:0, C16:0, C17:0, C17:1, C18:0 and TVA (trans-vaccenic acid), together with a marked increase in oleic and linoleic acids. Finally, in goat sausages, the fat content significantly affected sensory parameters: taste, texture and overall acceptability (P<0.05).     Hippo-Independent Activation of YAP by the GNAQ Uveal Melanoma Oncogene through a Trio-Regulated Rho GTPase Signaling Circuitry

    SummaryMutually exclusive activating mutations in the GNAQ and GNA11 oncogenes, encoding heterotrimeric Gαq family members, have been identified in ∼83% and ∼6% of uveal and skin melanomas, respectively. However, the molecular events underlying these GNAQ-driven malignancies are not yet defined, thus limiting the ability to develop cancer-targeted therapies. Here, we focused on the transcriptional coactivator YAP, a critical component of the Hippo signaling pathway that controls organ size. We found that Gαq stimulates YAP through a Trio-Rho/Rac signaling circuitry promoting actin polymerization, independently of phospholipase Cβ and the canonical Hippo pathway. Furthermore, we show that Gαq promotes the YAP-dependent growth of uveal melanoma cells, thereby identifying YAP as a suitable therapeutic target in uveal melanoma, a GNAQ/GNA11-initiated human malignancy

    Use of biodegradable polyester-based microvessels for micropropagation of mature Eucalyptus microcorys

    Background: Micropropagation, an in vitro vegetative propagation technique using small propagules is one of the main applications of plant tissue culture. It can be used to clone specific plants with desired traits and reduce the cost of plant propagation. In this study, we developed a protocol for micropropagation of Eucalyptus microcorys F.Muell using a selected mature tree, in which we tested various combinations of different culture media and evaluated the use of biodegradable polyester-based microvessels during the adventitious rooting and acclimatisation phases. Methods: Epicormic shoots were used as an explant source. After the in vitro explant establishment and multiplication, we tested 8 combinations of BAP, NAA and IBA in the elongation phase. Three types of microvessels were tested in the adventitious rooting phase and acclimatisation of the microcuttings. Results: Epicormic shoots had an establishment percentage of 40.6% and a total of 820 explants were generated by the 11th subculture, with an average of 12 buds per explant. Best shoot elongation results were achieved with BAP (0.05 mg L-1) + NAA (1 mg L-1) and BAP (0.05 mg L-1) + NAA (1 mg L-1) + IBA (1 mg L-1) combinations, whereas microvessel types M2 and M3 provided higher rooting and acclimatisation. According to the results of ISSR markers, at the end of 535 days of in vitro cultivation, cloning was successful between acclimatised micro-plantlets and the parent plant. Conclusions: The micropropagation protocol using microvessels was efficient in producing E. microcorys clonal microplantlets and is recommended for further studies with this species, and for testing in the micropropagation of other species.Peer reviewe

    Effects of nitric oxide-related compounds in the acute ketamine animal model of schizophrenia

    Abstract\ud \ud Background\ud Better treatments for schizophrenia are urgently needed. The therapeutic use of the nitric oxide (NO)-donor sodium nitroprusside (SNP) in patients with schizophrenia has shown promising results. The role of NO in schizophrenia is still unclear, and NO modulation is unexplored in ketamine (KET) animal models to date. In the present study, we compared the behavioral effects of pre- and post-treatment with SNP, glyceryl trinitrate (GTN), and methylene blue (MB) in the acute KET animal model of schizophrenia.\ud The present study was designed to test whether acute SNP, GTN, and MB treatment taken after (therapeutic effect) or before (preventive effect) a single KET injection would influence the behavior of rats in the sucrose preference test, object recognition task and open field.\ud \ud \ud Results\ud The results showed that KET induced cognitive deficits and hyperlocomotion. Long- term memory improvement was seen with the therapeutic GTN and SNP treatment, but not with the preventive one. MB pretreatment resulted in long-term memory recovery. GTN pre-, but not post-treatment, tended to increase vertical and horizontal activity in the KET model. Therapeutic and preventive SNP treatment consistently decreased KET-induced hyperlocomotion.\ud \ud \ud Conclusion\ud NO donors – especially SNP – are promising new pharmacological candidates in the treatment of schizophrenia. In addition, we showed that the potential impact of NO-related compounds on KET-induced behavioral changes may depend on the temporal window of drug administration.FAPESPCNPqCAPE

    Epidemiology of Invasive Fungal Infections in Latin America

    The pathogenic role of invasive fungal infections (IFIs) has increased during the past two decades in Latin America and worldwide, and the number of patients at risk has risen dramatically. Working habits and leisure activities have also been a focus of attention by public health officials, as endemic mycoses have provoked a number of outbreaks. An extensive search of medical literature from Latin America suggests that the incidence of IFIs from both endemic and opportunistic fungi has increased. The increase in endemic mycoses is probably related to population changes (migration, tourism, and increased population growth), whereas the increase in opportunistic mycoses may be associated with the greater number of people at risk. In both cases, the early and appropriate use of diagnostic procedures has improved diagnosis and outcome
