    Implementation of a combined association-linkage model for quantitative traits in linear mixed model procedures of statistical packages

    Atransmission disequilibrium test for quantitative traits which combines association and linkage analyses is currently available in several dedicated software packages. We describe how to implement such models in linear mixed model procedures that are available in widely used statistical packages such as SPSS. We also briefly mention a few extensions of the model that become naturally available once the model is implemented in such procedures. Genotyping of many microsatellite markers or single nucleotide polymorphisms (SNPs) over the entire genome is becoming increasingly common in human genetics. In those high-resolution maps the average distance between microsatellite markers may be as small as 5 cM and between SNPs one half cM or less. At those small distances it becomes fairly likely that some markers in the set are in linkage disequilibrium (LD) with a gene affecting the trait (a so-called quantitative trait locus or QTL if the trait or the vulnerability distribution is quantitative). Different alleles or combinations of alleles of the markers or SNPs can then be associated with different trait means. Association studies are conducted to discover such allelic effects. Abecasis et al. (2000) generalized the model proposed by Fulker et al. (1999) for combined linkage and association tests, within and between families.     Dust Temperatures in the Infrared Space Observatory Atlas of Bright Spiral Galaxies

    We examine far-infrared and submillimeter spectral energy distributions for galaxies in the Infrared Space Observatory Atlas of Bright Spiral Galaxies. For the 71 galaxies where we had complete 60-180 micron data, we fit blackbodies with lambda^-1 emissivities and average temperatures of 31 K or lambda^-2 emissivities and average temperatures of 22 K. Except for high temperatures determined in some early-type galaxies, the temperatures show no dependence on any galaxy characteristic. For the 60-850 micron range in eight galaxies, we fit blackbodies with lambda^-1, lambda-2, and lambda^-beta (with beta variable) emissivities to the data. The best results were with the lambda^-beta emissivities, where the temperatures were ~30 K and the emissivity coefficient beta ranged from 0.9 to 1.9. These results produced gas to dust ratios that ranged from 150 to 580, which were consistent with the ratio for the Milky Way and which exhibited relatively little dispersion compared to fits with fixed emissivities.Comment: AJ, 2003, in pres

    La presente investigación tipo instrumental, tuvo como objetivo el poder determinar las evidencias de validez y confiabilidad de la Escala Capital Psicológico en 244 trabajadores de una empresa pública de la provincia de Trujillo. Los resultados evidenciaron la estructura interna, mediante el uso del análisis factorial confirmatorio, donde se obtuvieron datos a través de índices de ajuste comparativo como el TLI=.91, y un CFI=.93, además de un ajuste de parsimonia de X2/gl=2,12, y unos índices de error como RMSEA=.068 y S-RMR=0.59. Por otro lado, la confiabilidad por consistencia interna reportó índices de ω=.86 para el modelo Factor General de segundo orden. Se concluye afirmando que el instrumento presenta una adecuada estructura interna, con índices de ajuste satisfactorios

    A cis-regulatory element promoting increased transcription at low temperature in cultured ectothermic Drosophila cells

    Background Temperature change affects the myriad of concurrent cellular processes in a non-uniform, disruptive manner. While endothermic organisms minimize the challenge of ambient temperature variation by keeping the core body temperature constant, cells of many ectothermic species maintain homeostatic function within a considerable temperature range. The cellular mechanisms enabling temperature acclimation in ectotherms are still poorly understood. At the transcriptional level, the heat shock response has been analyzed extensively. The opposite, the response to sub-optimal temperature, has received lesser attention in particular in animal species. The tissue specificity of transcriptional responses to cool temperature has not been addressed and it is not clear whether a prominent general response occurs. Cis-regulatory elements (CREs), which mediate increased transcription at cool temperature, and responsible transcription factors are largely unknown. Results The ectotherm Drosophila melanogaster with a presumed temperature optimum around 25 °C was used for transcriptomic analyses of effects of temperatures at the lower end of the readily tolerated range (14–29 °C). Comparative analyses with adult flies and cell culture lines indicated a striking degree of cell-type specificity in the transcriptional response to cool. To identify potential cis-regulatory elements (CREs) for transcriptional upregulation at cool temperature, we analyzed temperature effects on DNA accessibility in chromatin of S2R+ cells. Candidate cis-regulatory elements (CREs) were evaluated with a novel reporter assay for accurate assessment of their temperature-dependency. Robust transcriptional upregulation at low temperature could be demonstrated for a fragment from the pastrel gene, which expresses more transcript and protein at reduced temperatures. This CRE is controlled by the JAK/STAT signaling pathway and antagonizing activities of the transcription factors Pointed and Ets97D. Conclusion Beyond a rich data resource for future analyses of transcriptional control within the readily tolerated range of an ectothermic animal, a novel reporter assay permitting quantitative characterization of CRE temperature dependence was developed. Our identification and functional dissection of the pst_E1 enhancer demonstrate the utility of resources and assay. The functional characterization of this CoolUp enhancer provides initial mechanistic insights into transcriptional upregulation induced by a shift to temperatures at the lower end of the readily tolerated range

    Flip-Flap: A Simple Dual-View Imaging Method for 3D Reconstruction of Thick Plant Samples

    Plant development is a complex process that relies on molecular and cellular events being co-ordinated in space and time. Microscopy is one of the most powerful tools available to investigate this spatiotemporal complexity. One step towards a better understanding of complexity in plants would be the acquisition of 3D images of entire organs. However, 3D imaging of intact plant samples is not always simple and often requires expensive and/or non-trivial approaches. In particular, the inner tissues of thick samples are challenging to image. Here, we present the Flip-Flap method, a simple imaging protocol to produce 3D images of cleared plant samples at the organ scale. This method allows full 3D reconstruction of plant organs suitable for 3D segmentation and further related analysis and can be easily handled by relatively inexperienced microscopists

    Asia-Pacific International Center of Excellence in Malaria Research: Maximizing Impact on Malaria Control Policy and Public Health in Cambodia and Papua New Guinea

    The Asia-Pacific International Center of Excellence in Malaria Research (ICEMR) was funded in 2016 to conduct a coordinated set of field and in-depth biological studies in Cambodia and Papua New Guinea (PNG), in sites that span the range of transmission intensities currently found in the Asia-Pacific regions. The overall objective is to gain an understanding of key parasite, human host, and vector factors involved in maintaining transmission in the face of intensified control and elimination programs, and to develop novel approaches to identify and target residual transmission foci. In this article, we will describe how the ICEMR program was designed to address key knowledge gaps and priority areas for the malaria control programs in each country. In PNG, partners have worked together on two consecutive ICEMR grants (2009-2016 and 2017-2024) and we present a case study of the partnership and engagement approach that has led to stronger coordination of research activities and integration with program, informing country-level strategic planning and prioritization of control activities. In both settings, the ICEMR program has generated insights into transmission foci, risk factors for ongoing transmission, highlighting the hidden burden of vivax malaria, and the need for additional complementary vector control tools. Finally, we will summarize the emerging research questions and priority areas-namely surveillance, vivax malaria, new vector control tools, and community/health systems-oriented approaches-where further tool development and implementation research have been identified as being needed to guide policy

    Insecticide resistance status of Aedes aegypti and Aedes albopictus mosquitoes in Papua New Guinea

    Background: Aedes aegypti and Ae. albopictus are important vectors of infectious diseases, especially those caused by arboviruses such as dengue, chikungunya and Zika. Aedes aegypti is very well adapted to urban environments, whereas Ae. albopictus inhabits more rural settings. Pyrethroid resistance is widespread in these vectors, but limited data exist from the Southwest Pacific Region, especially from Melanesia. While Aedes vector ecology is well documented in Australia, where incursion of Ae. albopictus and pyrethroid resistance have so far been prevented, almost nothing is known about Aedes populations in neighbouring Papua New Guinea (PNG). With pyrethroid resistance documented in parts of Indonesia but not in Australia, it is important to determine the distribution of susceptible and resistant Aedes populations in this region. Methods: The present study was aimed at assessing Aedes populations for insecticide resistance in Madang and Port Moresby, located on the north and south coasts of PNG, respectively. Mosquitoes were collected using ovitraps and reared in an insectary. Standard WHO bioassays using insecticide-treated filter papers were conducted on a total of 253 Ae. aegypti and 768 Ae. albopictus adult mosquitoes. Subsets of samples from both species (55 Ae. aegypti and 48 Ae. albopictus) were screened for knockdown resistance mutations in the voltage-sensitive sodium channel (Vssc) gene, the target site of pyrethroid insecticides. Results: High levels of resistance against pyrethroids were identified in Ae. aegypti from Madang and Port Moresby. Aedes albopictus exhibited susceptibility to pyrethroids, but moderate levels of resistance to DDT. Mutations associated with pyrethroid resistance were detected in all Ae. aegypti samples screened. Some genotypes found in the present study had been observed previously in Indonesia. No Vssc mutations associated with pyrethroid resistance were found in the Ae. albopictus samples. Conclusions: To our knowledge, this is the first report of pyrethroid resistance in Ae. aegypti mosquitoes in PNG. Interestingly, usage of insecticides in PNG is low, apart from long-lasting insecticidal nets distributed for malaria control. Further investigations on how these resistant Ae. aegypti mosquito populations arose in PNG and how they are being sustained are warranted
