    A Novel Root-Knot Nematode Resistance QTL on Chromosome Vu01 in Cowpea.

    The root-knot nematode (RKN) species Meloidogyne incognita and M. javanica cause substantial root system damage and suppress yield of susceptible cowpea cultivars. The narrow-based genetic resistance conferred by the Rk gene, present in some commercial cultivars, is not effective against Rk-virulent populations found in several cowpea production areas. The dynamics of virulence within RKN populations require a broadening of the genetic base of resistance in elite cowpea cultivars. As part of this goal, F1 and F2 populations from the cross CB46-Null (susceptible) x FN-2-9-04 (resistant) were phenotyped for M. javanica induced root-galling (RG) and egg-mass production (EM) in controlled growth chamber and greenhouse infection assays. In addition, F[Formula: see text] families of the same cross were phenotyped for RG on field sites infested with Rk-avirulent M. incognita and M. javanica The response of F1 to RG and EM indicated that resistance to RKN in FN-2-9-04 is partially dominant, as supported by the degree of dominance in the F2 and F[Formula: see text] populations. Two QTL associated with both RG and EM resistance were detected on chromosomes Vu01 and Vu04. The QTL on Vu01 was most effective against aggressive M. javanica, whereas both QTL were effective against avirulent M. incognita Allelism tests with CB46 x FN-2-9-04 progeny indicated that these parents share the same RKN resistance locus on Vu04, but the strong, broad-based resistance in FN-2-9-04 is conferred by the additive effect of the novel resistance QTL on Vu01. This novel resistance in FN-2-9-04 is an important resource for broadening RKN resistance in elite cowpea cultivars

    Division site positioning is critical for both symmetric and asymmetric cell divisions. In many organisms, positive and negative signals cooperate to position the contractile actin ring for cytokinesis. In rod-shaped fission yeast Schizosaccharomyces pombe cells, division at midcell is achieved through positive Mid1/anillin-dependent signaling emanating from the central nucleus and negative signals from the dual-specificity tyrosine phosphorylation-regulated kinase family kinase Pom1 at the cell poles. In this study, we show that Pom1 directly phosphorylates the F-BAR protein Cdc15, a central component of the cytokinetic ring. Pom1-dependent phosphorylation blocks Cdc15 binding to paxillin Pxl1 and C2 domain protein Fic1 and enhances Cdc15 dynamics. This promotes ring sliding from cell poles, which prevents septum assembly at the ends of cells with a displaced nucleus or lacking Mid1. Pom1 also slows down ring constriction. These results indicate that a strong negative signal from the Pom1 kinase at cell poles converts Cdc15 to its closed state, destabilizes the actomyosin ring, and thus promotes medial septation

    On the Origin of the Wide HI Absorption Line Toward Sgr A*

    We have imaged a region of about 5' extent surrounding Sgr A* in the HI 21 cm-line absorption using the Very Large Array. A Gaussian decomposition of the optical depth spectra at positions within about 2' (approx. 5 pc at 8.5 kpc) of Sgr A* detects a wide line underlying the many narrow absorption lines. The wide line has a mean peak optical depth of 0.32 +/- 0.12 centered at a mean velocity of V(lsr) = -4 +/- 15 km/s. The mean full width at half maximum is 119 +/- 42 km/s. Such a wide line is absent in the spectra at positions beyond about 2' from Sgr A*. The position-velocity diagrams in optical depth reveal that the wide line originates in various components of the circumnuclear disk (radius approx. 1.3') surrounding Sgr A*. These components contribute to the optical depth of the wide line in different velocity ranges. The position-velocity diagrams do not reveal any diffuse feature which could be attributed to a large number of HI clouds along the line of sight to Sgr A*. Consequently, the wide line has no implications either to a global population of shocked HI clouds in the Galaxy or to the energetics of the interstellar medium as was earlier thought.Comment: LaTeX, 12 pages and 9 figures, accepted for publication in J.     Comprehensive analysis of type 1 fimbriae regulation in fimB -null strains from the multidrug resistant Escherichia coli ST131 clone

    Summary\ud \ud Uropathogenic Escherichia coli (UPEC) of sequence type 131 (ST131) are a pandemic multidrug resistant clone associated with urinary tract and bloodstream infections. Type 1 fimbriae, a major UPEC virulence factor, are essential for ST131 bladder colonization. The globally dominant sub-lineage of ST131 strains, clade C/H30-R, possess an ISEc55 insertion in the fimB gene that controls phase-variable type 1 fimbriae expression via the invertible fimS promoter. We report that inactivation of fimB in these strains causes altered regulation of type 1 fimbriae expression. Using a novel read-mapping approach based on Illumina sequencing, we demonstrate that ‘off’ to ‘on’ fimS inversion is reduced in these strains and controlled by recombinases encoded by the fimE and fimX genes. Unlike typical UPEC strains, the nucleoid-associated H-NS protein does not strongly repress fimE transcription in clade C ST131 strains. Using a genetic screen to identify novel regulators of fimE and fimX in the clade C ST131 strain EC958, we defined a new role for the guaB gene in the regulation of type 1 fimbriae and in colonisation of the mouse bladder. Our results provide a comprehensive analysis of type 1 fimbriae regulation in ST131, and highlight important differences in its control compared to non-ST131 UPEC

    HI observations of luminous infrared galaxy mergers

    A total of 19 luminous infrared galaxy mergers, with L_IR~2 10^11 L_sun for H_0=75 km/s/Mpc, have been observed in the HI line at Nancay and four of them were observed at Arecibo as well. Of these 19, ten had not been observed before. Six were clearly detected, one of which for the first time. The objective was to statistically sample the \HI gas mass in luminous infrared mergers along a starburst merger sequence where the molecular CO gas content is already known.Comment: 11 pages, accepted for Astron.     The prevalence of compulsive buying: a meta-analysis

    Aims: To estimate the pooled prevalence of compulsive buying behaviour (CBB) indifferent populations and to determine the effect of age, gender, location and screening instrument on the reported heterogeneity inestimates of CBB and whether publication bias could be identified. Methods: Three databases were searched(Medline, PsychInfo, Web of Science) using the terms 'compulsive buying' 'pathological buying' and 'compulsive shopping' to estimate the pooled prevalence of CBB in different populations. Forty studies reporting 49 prevalence estimates from 16 countries were located (n=32000). To conduct the meta-analysis, data from non-clinical studies regarding mean age and gender proportion, geographical study location and screening instrument used to assess CBB were extracted by multiple independent observers and evaluated using a random-effects model. Four a priori subgroups were analysed using pooled estimation (Cohen's Q) and covariate testing (moderator and meta-regression analysis). Results: The CBB pooled prevalence of adult representative studies was 4.9% (3.4–6.9%, eight estimates, 10 102 participants), although estimates were higher among university students: 8.3% (5.9–11.5%,19 estimates, 14 947 participants) in adult non-representative samples: 12.3% (7.6–19.1%, 11 estimates, 3929 participants) and in shopping-specific samples: 16.2% (8.8–27.8%, 11 estimates, 4686 participants). Being young and female were associated with increased tendency, but not location (United States versus non-United States). Meta-regression revealed large heterogeneity within subgroups,due mainly to diverse measures and time-frames(current versus life-time) used to assess CBB. Conclusions: A pooled estimate of compulsive buying behaviour in the populations studied is approximately 5%, but there is large variation between samples accounted for largely by use of different time-frames and measures

    A cloudy model for the Circumnuclear Disk in the Galactic Centre

    We present a first attempt to construct an analytic model for a clumped gas and dust disk and apply it to the Galactic Centre. The clumps are described as isothermal spheres partially ionized by the external UV radiation field. The disk structure formed by the clouds is described as a quasi standard continuous accretion disk using adequately averaged parameters of the discrete cloud model. The viscosity in the Circumnuclear Disk is due to partially inelastic cloud-cloud collisions. We find two different solutions for the set of equations corresponding to two stable cloud regimes: (i) the observed molecular clouds and (ii) much lighter and smaller clouds which correspond to the stripped cores of the observed clouds. It is shown that the resulting physical characteristics of the heavy clouds and the disk are in very good agreement with all comparable observations at multiple wavelengths. A mass accretion rate of approx. 10^-4 M_solar/yr for the isolated Circumnuclear Disk is inferred.     Trematodes of the Great Barrier Reef, Australia: emerging patterns of diversity and richness in coral reef fishes

    The Great Barrier Reef holds the richest array of marine life found anywhere in Australia, including a diverse and fascinating parasite fauna. Members of one group, the trematodes, occur as sexually mature adult worms in almost all Great Barrier Reef bony fish species. Although the first reports of these parasites were made 100 years ago, the fauna has been studied systematically for only the last 25 years. When the fauna was last reviewed in 1994 there were 94 species known from the Great Barrier Reef and it was predicted that there might be 2,270 in total. There are now 326 species reported for the region, suggesting that we are in a much improved position to make an accurate prediction of true trematode richness. Here we review the current state of knowledge of the fauna and the ways in which our understanding of this fascinating group is changing. Our best estimate of the true richness is now a range, 1,100–1,800 species. However there remains considerable scope for even these figures to be incorrect given that fewer than one-third of the fish species of the region have been examined for trematodes. Our goal is a comprehensive characterisation of this fauna, and we outline what work needs to be done to achieve this and discuss whether this goal is practically achievable or philosophically justifiable

    Susceptibility to tuberculosis is associated with variants in the ASAP1 gene encoding a regulator of dendritic cell migration

    Human genetic factors predispose to tuberculosis (TB). We studied 7.6 million genetic variants in 5,530 people with pulmonary TB and in 5,607 healthy controls. In the combined analysis of these subjects and the follow-up cohort (15,087 TB patients and controls altogether), we found an association between TB and variants located in introns of the ASAP1 gene on chromosome 8q24 (P = 2.6 × 10−11 for rs4733781; P = 1.0 × 10−10 for rs10956514). Dendritic cells (DCs) showed high ASAP1 expression that was reduced after Mycobacterium tuberculosis infection, and rs10956514 was associated with the level of reduction of ASAP1 expression. The ASAP1 protein is involved in actin and membrane remodeling and has been associated with podosomes. The ASAP1-depleted DCs showed impaired matrix degradation and migration. Therefore, genetically determined excessive reduction of ASAP1 expression in M. tuberculosis–infected DCs may lead to their impaired migration, suggesting a potential mechanism of predisposition to TB
