    Objetivo General Sintetizar complejos de vanadio (IV) y (V) a fin de obtener compuestos con potencial aplicación farmacológica, estudiar sus efectos biológicos en osteoblastos en cultivo e investigar los mecanismos que puedan mediar sus efectos citotóxicos. Objetivos Específicos 1. Sintetizar y caracterizar fisicoquímicamente derivados de vanadio (IV) y (V) con distintos ligandos de interés biológico y farmacológico. 2. Evaluar los efectos de los complejos de vanadio sobre la proliferación, morfología celular y proteínas del citoesqueleto. 3.     El presente trabajo investigativo es desarrollar la: “Auditoría de gestión de la empresa Jaime Lituma Construcciones & Comercio Cía. Ltda., de la ciudad de Macas periodo 01 de enero al 31 de diciembre del 2014”. La metodología utilizada para el desarrollo se sustentó: en el método Deductivo; y se utilizaron las técnicas de observación y encuesta de datos. El marco propositivo contiene FASE I: Familiarización y revisión de legislación, FASE II: Evaluación del sistema de control interno, FASE III: Desarrollo de hallazgos, FASE IV: Redacción de informe y comunicación de resultados. Uno de los principales problemas que se logró detectar es la ausencia de toma de decisiones oportunas impidiendo así medir y cuantificar los logros alcanzados con la finalidad de mejorar los niveles de responsabilidad administrativa para iniciar acciones correctivas e incrementar oportunidades para el desarrollo óptimo de la compañía dentro del ejercicio de sus funciones. La Auditoría de Gestión, presento como resultados la inexistencia de: indicadores de gestión, reglamento interno, y manual de funciones; no existe políticas para contratar al personal; ineficaz prestación del servicio de monitoreo; en cuanto a los recursos no existe mecanismos de seguridad para su acceso ni políticas de su vida útil. Se determina como conclusión la necesidad de establecer un programa para evaluar el nivel de la Gestión Administrativa que consiste en los cumplimientos de planes, objetivos, metas y políticas. Finalmente se recomienda a la empresa diseñar e implementar a todos los departamentos indicadores de gestión; realizar manuales de funciones y reglamento interno que permitirá mejorar el desempeño en su servicio y responsabilidad a sus clientes.The present research is about Management Audit of Jaime Lituma Construcciones & Comercio Cia. Ltda enterprise in Macas city From January 31 to December 2014. Deductive method was used as methodology and observation and data survey were used as techniques. The proposal frame contains: PHASE I: Familiarization and revision of legislation, PHASE II: Evaluation of the Internal Control System, PHASE III: Development of findings, PHASE IV: Writing of a report and communication of the results. One of the most important problems detected was the lack of making opportune decisions which stops measuring and quantifying the goals in order to improve the levels of administrative responsibility to stard corrective actions and to increase opportunities for the best development of the company. Results gotten from the management audit were: lack of management indicators, internal regulation, function manual, policies to hire the staff, monitoring inefficiency, there are not mechanisms of security to access and there are not policies of useful life. It is concluded that a program must be established to evaluate the level of Administrative Management consisting on the meet of plans, objectives, goals and policies. It is recommended that the enterprise desing and implement management indicators in all the departments, make function manuals and internal regulation which will improve the performance in its service and responsibility to its clients

    Survey for major grapevine viruses in commercial vineyards of Northwestern Argentina

    This study aimed to survey the occurrence of eight grapevine viruses in commercial vine yards located in the Calchaquíes Valleys in the northwest region of Argentina. A total of 103 samples of mature canes of vines showing either none or some viral-like symptoms were randomly collected. The samples were tested by RT-PCR/PCR-based assays for the screening of the following viruses: Grapevine fanleaf virus (GFLV), Grapevine leafroll-associated viruses (GLRaV-1, -2, -3, -4), Grapevine virus A (GVA), Grapevine rupestris stem pitting-associated viruses (GRSPaV), and Grapevine red blotch virus (GRBV). Sixty percent of the analyzed samples showed infection with some of the analyzed viruses, except GRBV. GLRaV-3 and GFLV were the most frequent viruses, present in 34% and 21% of the pos itive samples, respectively. This study represents the first survey report of the presence of grapevine viruses in the region of the Calchaquíes Valleys and contributes to the knowledge to maintain the sanitary status of commercial vineyards in Argentina.EEA SaltaFil: Rivadeneria, Monica.     Novel antibacterial bioactive glass nanocomposite functionalized with tetracycline hydrochloride

    To prevent the high frequency of wound infections, anti-bacterial agents can be loaded onto composites. In the present study, the antibiotic tetracycline hydrochloride (TC)was incorporated, for the first time, in collagen type I membranes coated with nano-sized SiO2-CaOP2O5 bioactive glass (n-BG) obtained by a sol-gel chemical route. Collagen membranes coated with n-BG were immersed in simulated body fluid (SBF) containing 0.25, 0.75 or 1.25 mg mL−1 of TC for 48 h at 37∘C following a coprecipitation method. The antibiotic was released in distilledwater at 37∘C for up to 72 h. The antibacterial activity of the composites was evaluated in vitro by the inhibition zone test and plate count method. Two different Staphylococcus aureus strains, S. aureus ATCC29213 and S. aureus ATCC25923, were exposed to the biomaterials. The results showed that the incorporation but not the release of TC was dependent on the initial concentration of TC in SBF. The biomaterials inhibited S. aureus growth, although the efficacy was similar for all the concentrations. The results allow us to conclude that the new composite could have potential in the prevention of wound infections.Fil: Rivadeneira, Josefina.     Evaluation of antibacterial and cytotoxic effects of nano-sized bioactive glass/collagen composites releasing tetracycline hydrochloride

    Aims: To evaluate the antibacterial efficacy of silicate bioactive glass nanoparticles/collagen composites functionalized with tetracycline hydrochloride (TCH). Methods and Results: Different concentrations of tetracycline hydrochloride (TCH) were incorporated on silicate bioactive glass nanoparticles/collagen composites by dipping these biomaterials for 48 h at 37°C in a solution of simulated body fluid (SBF) plus 0 05, 0 20 or 0 35 mg ml1 of the antibiotic. TCH release was assessed in double-distilled water at 37°C up to 72 h. The antibacterial activity of the samples has been evaluated in two ways: inhibition zone test and plate count method. The experiments were performed in vitro up to 48 h on four staphylococci strains (Staphylococcus aureus ATCC29213, ATCC25923, ATCC6538P and Staphylococcus epidermidis ATCC12228). The new composites were also tested for cytotoxicity on MG-63 human osteosarcoma cells. The results showed that the incorporation and release of TCH was dependent on the initial concentration of TCH in SBF. The biomaterials also inhibited the Staph. aureus cell growth even though the efficacy was similar for all concentration. On the other hand, no cytotoxic effects were found on osteoblast-like cells, even at the highest concentration. Conclusions: Considering all results, it can be concluded that the new composite acts as a suitable bioactive carrier of TCH and could have potential in the prevention of biomaterial related infections.     Spectroscopic Characterization of a VO²⁺ Complex of Oxodiacetic Acid and its Bioactivity on Osteoblast-like Cells in Culture

    The oxovanadium(IV) complex of oxodiacetic acid (H₂oda) of stoichiometry [VO(oda)(H₂O)₂], which presents an unprecedented tridentate OOO coordination, was thoroughly characterized by infrared, Raman, electronic, and electron paramagnetic resonance spectroscopies. The biological activity of the complex on the cell proliferation and differentiation was tested on osteoblast-like cells (MC3T3E1 osteoblastic mouse calvaria-derived cells and UMR106 rat osteosarcoma-derived cells) in culture. The complex caused inhibition of cellular proliferation in both osteoblast-like cells in culture, but the cytotoxicity was stronger in the normal (MC3T3E1) than in the tumoral (UMR106) osteoblasts. The effect of the complex in cell differentiation was tested through the specific activity of alkaline phosphatase of the UMR106 cells because they expressed a high activity of this enzyme. What occurs with other vanadium compounds [VO(oda)(H₂O)₂] is an inhibitory agent of osteoblast differentiation.Facultad de Ciencias ExactasCentro de Química Inorgánic

    Neurocognitive Stimulation to Improve the Degree of Independence in The Elderly

    Background: research in the field of neurocognitive therapy to improve visual-motor coordination in the elderly is scarce, so in this study an intervention protocol was implemented through playful exercises that sought to improve coordination between sight and movement execution of this age group, the exercises were worked seeking to integrate the executive functions to them having as; Objective: to improve the visual-motor coordination of elderly residents of the nursing home Sacred Heart of Jesus; Methods:  This research was descriptive based on the quantitative approach with a transversal cut, an initial evaluation was made during the first week through the Barthel index, after this the program was executed for 10 weeks to finally in the last week reapply the Barthel index and thus obtain the results that were obtained with this age group; Results: an improvement was evidenced in the final score compared to the initial where it went from a score of 70. Conclusions: in this work a 12-week neurocognitive training program was established, which showed effectiveness in the improvement of visual-motor coordination and quality of life in general in this age group

    Proteomic and metabolomic approaches unveil relevant biochemical changes in carbohydrate and cell wall metabolisms of two blueberry (Vaccinium corymbosum) varieties with different quality attributes

    Quality maintenance in rapidly decaying fruit such as blueberries (Vaccinium corymbosum) is of essential importance to guarantee the economic success of the crop. Fruit quality is a multifaceted subject that encompasses flavor, aroma, visual and physical issues as main factors. In this paper we report an ample characterization of different biochemical and physical aspects in two varieties (O'Neal and Emerald) of blueberries that differ in firmness, aspect, flavor and harvesting times, at two different phenological stages (fruit set vs. ripe), with the intention of unveiling how the metabolic signature of each contributes to their contrasting quality. To this effect a metabolomic, ionomic and proteomic approach was selected. The results presented here show marked differences in several variables at the two stages and between varieties. Emerald is an early variety with a large, good taste and firm fruit, while O'Neal is soft, medium sized and very sweet. Proteomic data comparison between both cultivars showed that, at fruit set, processes related with the response to inorganic compounds and small molecule metabolisms are relevant in both varieties. However, solute accumulation (mainly amino acids and organic acids), enzymes related with C: N balance, water transport and cell wall recycling are enhanced in Emerald. In ripe fruit, Emerald showed an enrichment of proteins associated with TCA, nitrogen, small molecules and cell wall in muro recycling processes, while mannitol and fatty acid metabolism were enhanced in the soft variety. The measured variation in metabolite levels gave strong support to the precedent results. This study suggests that at fruit set, a composite scenario of active metabolic recycling of the cell wall, improved C: N balance and solute accumulation give place to a more efficient carbon and water resource management. During the ripe stage, an increased and efficient in muro and metabolic recycling of the cell wall, added to enhanced inositol and secondary metabolism may be responsible for a best turgor conservation in Emerald. These findings may yield clues for improvements in fertilization practices, as well as to assist the guided development of new varieties based on biochemical quality.Fil: Montecchiarini, Marina Lucía.     45S5 Bioglass® concentrations modulate the release of vancomycin hydrochloride from gelatin-starch films: evaluation of antibacterial and cytotoxic effects

    The aim of this work was to evaluate the release profile of vancomycinhydrochloride (VC), as well as the degradation, in vitro antistaphylococcal effect and cytotoxicity in MG-63 osteoblast-like cells of gelatin–starch (GS) films added with different concentrations of microparticles of the bioactive glass 45S5 (m-BG). The biomaterials were obtained through the gel-casting method. Four different composites were prepared at four different weight percentages of m-BG: 0, 5, 10, and 15 %. Glutaraldehyde 0.25 wt% (GA) was used as the cross-linker. The composites were characterized by scanning electron microscopy and the in vitro degradation of the filmswas studied bymeasuring thewater uptake and weight loss. The drug release kinetics was quantified spectrophotometrically. The inhibition zone test and the plate count method were used to evaluate the antibacterial activity of the samples. Three staphylococcus strains were evaluated: Staphylococcus aureus ATCC6538, S. aureus ATCC29213, and Staphylococcus epidermidis ATCC12228. Cytotoxicity effects were evaluatedthrough the MTT assay.The addition of m-BG to GS films showedno effects on the amount of water uptake, but led to an increase in the weight loss over time, even with m-BG content. The release rate of VC was also affected by the increasing concentration of m-BG in the composite films.However, the antibacterial effects of the composites were not improved by this modulation. All composites strongly inhibited staphylococcal cells with similar strength. On the other hand, liquid extracts from the composites resultedin cytotoxic effects onMG-63 osteoblastlike cells due to the presence of GA, but not to the concentration of VC or m-BG.Facultad de Ciencias Exacta

    Cyto- and genotoxicity of a vanadyl(IV) complex with oxodiacetate in human colon adenocarcinoma (Caco-2) cells: potential use in cancer therapy

    The complex of vanadyl(IV) cation with oxodiacetate, VO(oda) caused an inhibitory effect on the proliferation of the human colon adenocarcinoma cell line Caco-2 in the range of 25–100 μM (P < 0.001). This inhibition was partially reversed by scavengers of free radicals. The difference in cell proliferation in the presence and the absence of scavengers was statistically significant in the range of 50–100 μM (P < 0.05). VO(oda) altered lysosomal and mitochondria metabolisms (neutral red and MTT bioassays) in a dose–response manner from 10 μM (P < 0.001). Morphological studies showed important transformations that correlated with the disassembly of actin filaments and a decrease in the number of cells in a dose response manner. Moreover, VO(oda) caused statistically significant genotoxic effects on Caco-2 cells in the low range of concentration (5–25 μM) (Comet assay). Increment in the oxidative stress and a decrease in the GSH level are the main cytotoxic mechanisms of VO(oda). These effects were partially reversed by scavengers of free radicals in the range of 50–100 μM (P < 0.05). Besides, VO(oda) interacted with plasmidic DNA causing single and double strand cleavage, probably through the action of free radical species.
