    Understanding the Effect of Seal Size in Non-GMO Food Advertisements and Measuring Consumer Attitudes Toward GMOs

    Today, genetically modified organisms have an increased presence in the American food supply. GMOs are a topic of controversy, so it is important to know how consumers feel about them. Attitudes toward GMOs have been studied across the globe, but few studies focus on attitudes toward GMOs in the United States. This study consists of two manuscripts: the first manuscript measured attitudes toward GMOs using a semantic differential scale, and the second manuscript measured the effect a certified non-GMO seal has on the way consumers view food advertisements when the size of the seal is manipulated. This study consisted of 100 faculty and staff members from Oklahoma State University. Attitudes toward GMOs were found to be primarily neutral, potentially revealing participants' lack of knowledge of GMOs. The size of the non-GMO seal was found to be significant for time to first fixation and fixation count, but it was not significant for fixation duration.Agricultural Communication

    Rapid simultaneous identification and quantitation of Staphylococcus aureus and Pseudomonas aeruginosa directly from bronchoalveolar lavage specimens using automated microscopy

    AbstractDiagnosis of ventilator-assisted pneumonia (VAP) requires pathogen quantitation of respiratory samples. Current quantitative culture methods require overnight growth, and pathogen identification requires an additional step. Automated microscopy can perform rapid simultaneous identification and quantitation of live, surface-immobilized bacteria extracted directly from patient specimens using image data collected over 3 h. Automated microscopy was compared to 1 μL loop culture and standard identification methods for Staphylococcus aureus and Pseudomonas spp. in 53 remnant bronchoalveolar lavage specimens. Microscopy identified 9/9 S. aureus and 7/7 P. aeruginosa in all specimens with content above the VAP diagnostic threshold. Concordance for specimens containing targets above the diagnostic threshold was 13/16, with concordance for sub-diagnostic content of 86/90. Results demonstrated that automated microscopy had higher precision than 1 μL loop culture (range ~0.55 log versus ≥1 log), with a dynamic range of ~4 logs (~103 to 106 CFU/mL)

    RING-type E3 ligases: Master manipulators of E2 ubiquitin-conjugating enzymes and ubiquitination

    AbstractRING finger domain and RING finger-like ubiquitin ligases (E3s), such as U-box proteins, constitute the vast majority of known E3s. RING-type E3s function together with ubiquitin-conjugating enzymes (E2s) to mediate ubiquitination and are implicated in numerous cellular processes. In part because of their importance in human physiology and disease, these proteins and their cellular functions represent an intense area of study. Here we review recent advances in RING-type E3 recognition of substrates, their cellular regulation, and their varied architecture. Additionally, recent structural insights into RING-type E3 function, with a focus on important interactions with E2s and ubiquitin, are reviewed.     XROMM analysis of tooth occlusion and temporomandibular joint kinematics during feeding in juvenile miniature pigs

    Like humans, domestic pigs are omnivorous and thus are a common model for human masticatory function. Prior attempts to characterize food–tooth interactions and jaw movements associated with mastication have been limited to aspects of the oral apparatus that are visible externally (with videography) and/or to 2D movements of oral structures (with monoplanar videofluoroscopy). We used XROMM, a 3D technique that combines CT-based morphology with biplanar videofluoroscopy, to quantify mandibular kinematics, tooth occlusion and mandibular condylar displacements within the temporomandibular joint (TMJ) during feeding. We observed that the pig TMJ moved detectably in only three of six possible degrees of freedom during mastication: two rotations, pitch and yaw; and one translation, protraction–retraction. Asymmetrical yaw around a dorsoventral axis produced the observed alternating left–right chewing cycles responsible for food reduction. Furthermore, the relative motions of the upper and lower premolars contained a substantial mesiodistal component in addition to the buccolingual component, resulting in an oblique (rather than a strictly transverse) power stroke. This research demonstrates the capacity of XROMM to explore the kinematic underpinnings of key masticatory movements, such as the occlusal power stroke, by integrating tooth, joint and rigid body jaw movements. XROMM also allowed us to test kinematic hypotheses based on skeletal anatomy with actual kinematics observed during naturalistic feeding behaviors. We observed that the soft tissue structures of the TMJ appear to play a significant role in limiting the range of motion of a joint, and thus analyses based solely on osseous morphology may over-estimate joint mobility

    Identifying and testing adaptive management options to increase river catchment system resilience using a Bayesian Network model

    The cumulative impacts of future climatic and socioeconomic change threaten the ability of freshwater catchments to provide essential ecosystem services. Stakeholders who manage freshwaters require decision-support tools that increase their understanding of catchment system resilience and support the appraisal of adaptive management options to inform decision-making. Our research aims to test the ability of a Bayesian Network model to identify adaptive management scenarios and test their effectiveness across future pathways to 2050. Using the predominantly arable river Eden catchment (320 km2) in eastern Scotland as a case study, we invited stakeholders from multiple sectors to participate in a series of workshops aimed at addressing water quality issues and achieving good ecological status in the catchment both now and in the future. Our participatory methods helped stakeholders overcome multiple layers of complexity and uncertainty associated with future-focused water management. Outputs of a Bayesian Network model simulated both current and future catchment resilience to inform the identification of six management scenarios. The effectiveness of each management scenario was tested using the Bayesian Network model. Two adaptive management scenarios increased catchment resilience and helped achieve good ecological status; a ‘Best Available Technology’ scenario, including aerobic granular sludge treatment, and a management scenario focused on ‘Resource Centre’, including phosphorus recovery from wastewater treatment works and constructed lagoons for crop irrigation. Stakeholders were interested in a 'Nature Based' management scenario including options such as wetland wastewater treatment methods and rural sustainable drainage systems, which improved water quality in the catchment, but had lower certainty in achieving desired outcome. Findings led to a recognition that innovative and collaborative action was required to improve current and future freshwater conditions

    Quantitative comparison of immunohistochemical staining measured by digital image analysis versus pathologist visual scoring

    <p>Abstract</p> <p>Immunohistochemical (IHC) assays performed on formalin-fixed paraffin-embedded (FFPE) tissue sections traditionally have been semi-quantified by pathologist visual scoring of staining. IHC is useful for validating biomarkers discovered through genomics methods as large clinical repositories of FFPE specimens support the construction of tissue microarrays (TMAs) for high throughput studies. Due to the ubiquitous availability of IHC techniques in clinical laboratories, validated IHC biomarkers may be translated readily into clinical use. However, the method of pathologist semi-quantification is costly, inherently subjective, and produces ordinal rather than continuous variable data. Computer-aided analysis of digitized whole slide images may overcome these limitations. Using TMAs representing 215 ovarian serous carcinoma specimens stained for S100A1, we assessed the degree to which data obtained using computer-aided methods correlated with data obtained by pathologist visual scoring. To evaluate computer-aided image classification, IHC staining within pathologist annotated and software-classified areas of carcinoma were compared for each case. Two metrics for IHC staining were used: the percentage of carcinoma with S100A1 staining (%Pos), and the product of the staining intensity (optical density [OD] of staining) multiplied by the percentage of carcinoma with S100A1 staining (OD*%Pos). A comparison of the IHC staining data obtained from manual annotations and software-derived annotations showed strong agreement, indicating that software efficiently classifies carcinomatous areas within IHC slide images. Comparisons of IHC intensity data derived using pixel analysis software versus pathologist visual scoring demonstrated high Spearman correlations of 0.88 for %Pos (p < 0.0001) and 0.90 for OD*%Pos (p < 0.0001). This study demonstrated that computer-aided methods to classify image areas of interest (e.g., carcinomatous areas of tissue specimens) and quantify IHC staining intensity within those areas can produce highly similar data to visual evaluation by a pathologist.</p> <p>Virtual slides</p> <p>The virtual slide(s) for this article can be found here: <url>http://www.diagnosticpathology.diagnomx.eu/vs/1649068103671302</url></p

    Biomechanical, biochemical, and morphological mechanisms of heat shock-mediated germination in Carica papaya seed.

    Carica papaya (papaya) seed germinate readily fresh from the fruit, but desiccation induces a dormant state. Dormancy can be released by exposure of the hydrated seed to a pulse of elevated temperature, typical of that encountered in its tropical habitat. Carica papaya is one of only a few species known to germinate in response to heat shock (HS) and we know little of the mechanisms that control germination in tropical ecosystems. Here we investigate the mechanisms that mediate HS-induced stimulation of germination in pre-dried and re-imbibed papaya seed. Exogenous gibberellic acid (GA3 ≥250 µM) overcame the requirement for HS to initiate germination. However, HS did not sensitise seeds to GA3, indicative that it may act independently of GA biosynthesis. Seed coat removal also overcame desiccation-imposed dormancy, indicative that resistance to radicle emergence is coat-imposed. Morphological and biomechanical studies identified that neither desiccation nor HS alter the physical structure or the mechanical strength of the seed coat. However, cycloheximide prevented both seed coat weakening and germination, implicating a requirement for de novo protein synthesis in both processes. The germination antagonist abscisic acid prevented radicle emergence but had no effect on papaya seed coat weakening. Desiccation therefore appears to reduce embryo growth potential, which is reversed by HS, without physically altering the mechanical properties of the seed coat. The ability to germinate in response to a HS may confer a competitive advantage to C. papaya, an opportunistic pioneer species, through detection of canopy removal in tropical forests

    Knowledge sharing, problem solving and professional development in a Scottish Ecosystem Services Community of Practice

    The ecosystem services framework has now been embodied in policy and practice, creating the need for governance structures that allow science, policy and practice to come together and facilitate shared learning. We describe five years of progress in developing an Ecosystem Services Community of Practice in Scotland, which brings together over 600 individuals from diverse constituencies to share experiences and learn from each other. We consider the ‘community’ and ‘practice’ aspects to demonstrate the benefits of establishing an Ecosystem Services Community (ESCom). We also demonstrate how the journey involved in the creation and continuing evolution of ESCom has proved valuable to researchers, policy-makers, practitioners and students and as such has contributed to social learning. We reflect on challenges, given the voluntary nature, absence of formal institutional support and emergence of initiatives focusing on overlapping topics. Based on our experience, we provide ten recommendations to help future ecosystem services communities of practice
